    Photoallergic reaction to cyamemazine.

    A 50-year-old man presented with a scaly erythema of the face, upper chest, forearms, and dorsum of the hands. He has been treated with cyamemazine for 6 months. Photopatch tests were performed and the patient was diagnosed with photoallergic reaction to cyamemazine. The drug was discontinued and a course of oral steroids was prescribed. The patient was advised to avoid light exposure. There has been no evidence of recurrence during a six-month follow-up period. Photoallergic reactions are much less frequent than phototoxic disorders. It is well known that several drugs including neuroleptics of the phenothiazine family may produce a skin eruption on light-exposed areas by dose-dependent (phototoxic) or photoallergic mechanisms. It is believed that photopatch testing, which is the clinical investigation of choice for suspected photoallergic reactions, is significantly underused in Europe and probably world-wide

    Caracterização de Erinnyis ello Granulovirus isolado de populações de mandarová da mandioca em Cruzeiro do Sul, Acre.

    Baculovirus são vírus em forma de bastão, envelopados, com DNA dupla fita circular, sendo que a maioria das espécies infecta insetos da ordem Lepidóptera. Devido a seu potencial como agentes de controle de pragas na agricultura e florestas, eles tem sido empregados com sucesso como bioinseticidas. O mandarová da mandioca (Erinnyis ello) é uma importante praga da mandioca e da seringueira com alta capacidade de migração. Neste trabalho é apresentada a caracterização de um baculovírus com ocorrência em populações de Erinnyis ello em área privada (agricultura familiar) no município de Cruzeiro do Sul, Acre (Latitude -7,55086; Longitude -72,72570). Inicialmente partículas virais foram purificadas de larvas com sintomas de infecção através de ultracentrifugação em gradiente de sacarose. Após tratamento para microscopia eletrônica seguido de contraste com acetato de uranila 2%, as partículas foram fotografadas utilizando-se microscópio eletrônico JEOL 1011. O vírus de forma ovalada, apresentando apenas um vírion por envelope e com menos de 0,5 uM, foi identificado como sendo do gênero Betabaculovirus (extinto gênero Granulovirus) da família Baculoviridae. Em seguida o DNA viral foi extraído através de ciclos de fenol e clorofórmio, digerido com diferentes enzimas de restrição e submetido a eletroforese em gel de agarose. A analise dos perfis de restrição obtidos com as enzimas Bam HI e com Hind III revelou um total de sete e de três fragmentos, respectivamente. Na clivagem com Pst I, foram identificados oito fragmentos molares e três submolares. Na digestão com Eco RI foi observado um número superior a vinte fragmentos incluindo várias bandas submolares. Embora tenha apresentado algumas diferenças, o perfil de restrição do DNA do isolado EeGV coletado no Acre foi similar ao descrito para o isolado EeGV proveniente de Itajai, SC, cujo genoma completo possui 102,759 bp.     Sperm quality assessment in Ficopomatus enigmaticus (Fauvel, 1923): Effects of selected organic and inorganic chemicals across salinity levels

    Contamination by organic and inorganic compounds remains one of the most complex problems in both brackish and marine environments, causing potential implications for the reproductive success and survival of several broadcast spawners. Ficopomatus enigmaticus is a tubeworm polychaete that has previously been used as a model organism for ecotoxicological analysis, due to its sensitivity and ecological relevance. In the present study, the effects of five trace elements (zinc, copper, cadmium, arsenic and lead), one surfactant (sodium dodecyl sulfate, SDS) and one polycyclic aromatic hydrocarbon (benzo(a)pyrene, B(a)P) on the sperm quality of F. enigmaticus were investigated. Sperm suspensions were exposed in vitro to different concentrations of each selected contaminant under four salinity conditions (10, 20, 30, 35). Possible adverse effects on sperm function were assessed by measuring oxidative stress, membrane integrity, viability and DNA damage. Sperm quality impairments induced by organic contaminants were more evident than those induced by inorganic compounds. SDS exerted the largest effect on sperm. In addition, F. enigmaticus sperm showed high tolerance to salinity variation, supporting the wide use of this species as a promising model organism for ecotoxicological assays. Easy and rapid methods on polychaete spermatozoids were shown to be effective as integrated sperm quality parameters or as an alternative analysis for early assessment of marine and brackish water pollution

    Development of methodologies for virus detection in soybean and wheat seeds.

    Seeds that contain large amounts of oil, starch, fibers and phenols are the most difficult tissues for RNA extraction. Currently, there are some reports of virus detection in seeds using commercial kits for RNA extraction. However, individual seeds were used, which may not be always suitable for analyses that deal with large amounts of seeds. Sangha [1] described a simple, quick and efficient protocol for RNA extraction and downstream applications in a group of seeds of jatropha (Jatropha curcas), mustard (Brassica sp.) and rice (Oryza sativa). We tested this protocol for soybean (Glycine max), maize (Zea mays), wheat (Triticum aestivum) and triticale (×Triticosecale) seeds and further reverse transcription PCR (RT-PCR)/quantitative real-time PCR (qPCR) in order to have a faster and more practical method for virus detection from seeds than the traditional scheme of seed planting and subsequent Elisa/RT-PCR from leaves. The essential points in the method are: • Some modifications in the protocol [1] were done in order to increase performance: Wheat and triticale seeds are incubated with water prior to maceration. An amount of 1.2 g of dry soybean seeds is used to maceration. • RT-PCR is used for detection of Wheat streak mosaic virus from wheat seeds and RT-qPCR for detection of Soybean mosaic virus from soybean seeds. • The method may be tested for other viruses, however, pre-validation will be needed

    We present the results of a research on Medical Residence in Pediatric Neurology, classifying present information on the teaching and training in the several centers of formation in Brazil. It was possible to contact 17 Institutions with organized services, being 6 accredited by CNRM (National Council of Medical residence), 10 non accredited, and one under diligence. The program content is developed in 3 or 4 years, including the pre-qualification, being the annual schedule load variable, from 1900 to 2880 hours / year.58777-8

    Foraminíferos Recentes do Canal de São Sebastião SP, e suas Relações com os Padrões de Sedimentação

    O presente estudo é parte integrante da primeira fase do projeto de monitoramento do sistema de tratamento de efluentes do Canal de São Sebastião. Este estudo objetivou caracterizar as comunidades de foraminíferos e os sedimentos de fundo do canal, procurando estabelecer a distribuição das associações microfaunísticas e suas relações com o meio, antes da construção desses efluentes no canal. As associações microfaunísticas e os dados sedimentológicos revelaram uma diferenciação entre o ambiente deposicional oeste e leste do canal. O lado oeste caracterizou-se pela presença marcante de minerais micáceos, sedimentos pelíticos com altos teores de carbono e nitrogênio, diversidade baixa de foraminíferos, aparecimento de espécies de tamanho reduzido e dominância de espécies típicas de ambiente rico em matéria orgânica (Buliminella elegantíssima e Fursenkoina pontoni). Tais resultados permitiram inferir domínio de baixa energia nesta região. Já no lado leste do canal, observou-se uma diminuição dos minerais micáceos, abundância de areia e biodetritos marinhos, diversidade relativamente alta e aparecimento da epifauna. Tais dados demonstram que o hidrodinamismo aumentou sensivelmente, sugerindo ambiente de maior energia e oxidação. Além disso, através das associações foi possível detectar uma influência oceânica maior no lado meridional do canal. Quanto ao lado setentrional e central, estes caracterizaram-se por possuir associações típicas de água costeira. Foi observada também a presença de Cassidulina crassa, associada possivelmente à ACAS (Água Central do Atlântico Sul)

    Discovery of spirooxadiazoline oxindoles with dual-stage antimalarial activity

The first-line therapeutic options are based on combinations of fast-acting artemisinin derivatives and longer-acting synthetic drugs. However, the emergence of resistance to these first-line treatments represents a serious risk, and the discovery of new effective drugs is urgently required. For this reason, new antimalarial chemotypes with new mechanisms of action, and ideally with activity against multiple parasite stages, are needed. We report a new scaffold with dual-stage (blood and liver) antiplasmodial activity. Twenty-six spirooxadiazoline oxindoles were synthesized and screened against the erythrocytic stage of the human malaria parasite P. falciparum. The most active compounds were also tested against the liver-stage of the murine parasite P. berghei. Seven compounds emerged as dual-stage antimalarials, with IC50 values in the low micromolar range. Due to structural similarity with cipargamin, which is thought to inhibit blood-stage P. falciparum growth via inhibition of the Na + efflux pump PfATP4, we tested one of the most active compounds for anti-PfATP4 activity.
