    'Tannat' (Vitis vinifera L.) as a model of responses to climate variability

    Climate variability influence on the vine is widely studied for its impact on grape final composition and quality. During 1994-2016, thermal and water regimes and their influence on grapevine yield, sanitary status and berry composition were analyzed for 'Tannat' grown in commercial vineyards in the south of Uruguay (Lat 34° 37' S; 56° 17' W). Statistical analysis showed that the principal component analysis (PCA) separated years in three groups: Group 1: rainfall over the growing season higher than the average, limited sanitary status, acidity and yield higher than average, lower sugar content, late harvest. Group 2: greater thermal conditions and water component lower than average, better sanitary status, sugar contents and acidity lower than average, early harvest. Group 3: thermal conditions lower than average, rainfall higher during budbreak-fruitset period and lower than average in the month before harvest, berry size and sugar contents greater than average. Correlations between climate, yield and berry quality variables were established and stages of greater sensitivity to these climate elements were determined. In the studied years, climate variability within the region was high and 'Tannat' showed to be strongly influenced by such variability

    Association of Circulating Tumor DNA Testing Before Tissue Diagnosis With Time to Treatment Among Patients With Suspected Advanced Lung Cancer: The ACCELERATE Nonrandomized Clinical Trial.

    IMPORTANCE Liquid biopsy has emerged as a complement to tumor tissue profiling for advanced non-small cell lung cancer (NSCLC). The optimal way to integrate liquid biopsy into the diagnostic algorithm for patients with newly diagnosed advanced NSCLC remains unclear. OBJECTIVE To evaluate the use of circulating tumor DNA (ctDNA) genotyping before tissue diagnosis among patients with suspected advanced NSCLC and its association with time to treatment. DESIGN, SETTING, AND PARTICIPANTS This single-group nonrandomized clinical trial was conducted among 150 patients at the Princess Margaret Cancer Centre-University Health Network (Toronto, Ontario, Canada) between July 1, 2021, and November 30, 2022. Patients referred for investigation and diagnosis of lung cancer were eligible if they had radiologic evidence of advanced lung cancer prior to a tissue diagnosis. INTERVENTIONS Patients underwent plasma ctDNA testing with a next-generation sequencing (NGS) assay before lung cancer diagnosis. Diagnostic biopsy and tissue NGS were performed per standard of care. MAIN OUTCOME AND MEASURES The primary end point was time from referral to treatment initiation among patients with advanced nonsquamous NSCLC using ctDNA testing before diagnosis (ACCELERATE [Accelerating Lung Cancer Diagnosis Through Liquid Biopsy] cohort). This cohort was compared with a reference cohort using standard tissue genotyping after tissue diagnosis. RESULTS Of the 150 patients (median age at diagnosis, 68 years [range, 33-91 years]; 80 men [53%]) enrolled, 90 (60%) had advanced nonsquamous NSCLC. The median time to treatment was 39 days (IQR, 27-52 days) for the ACCELERATE cohort vs 62 days (IQR, 44-82 days) for the reference cohort (P < .001). Among the ACCELERATE cohort, the median turnaround time from sample collection to genotyping results was 7 days (IQR, 6-9 days) for plasma and 23 days (IQR, 18-28 days) for tissue NGS (P < .001). Of the 90 patients with advanced nonsquamous NSCLC, 21 (23%) started targeted therapy before tissue NGS results were available, and 11 (12%) had actionable alterations identified only through plasma testing. CONCLUSIONS AND RELEVANCE This nonrandomized clinical trial found that the use of plasma ctDNA genotyping before tissue diagnosis among patients with suspected advanced NSCLC was associated with accelerated time to treatment compared with a reference cohort undergoing standard tissue testing.     Biology of moderately halophilic aerobic bacteria

    The moderately halophilic heterotrophic aerobic bacteria form a diverse group of microorganisms. The property of halophilism is widespread within the bacterial domain. Bacterial halophiles are abundant in environments such as salt lakes, saline soils, and salted food products. Most species keep their intracellular ionic concentrations at low levels while synthesizing or accumulating organic solutes to provide osmotic equilibrium of the cytoplasm with the surrounding medium. Complex mechanisms of adjustment of the intracellular environments and the properties of the cytoplasmic membrane enable rapid adaptation to changes in the salt concentration of the environment. Approaches to the study of genetic processes have recently been developed for several moderate halophiles, opening the way toward an understanding of haloadaptation at the molecular level. The new information obtained is also expected to contribute to the development of novel biotechnological uses for these organisms

    Objectives: Diagnostic assessment programs (DAPs) can reduce wait times for cancer diagnosis but optimal DAP design is unknown. This study explored how organizational characteristics influenced multidisciplinary teamwork and diagnostic service delivery in lung cancer DAPs. Design: A mixed methods approach integrated data from descriptive qualitative interviews and medical record abstraction at four lung cancer DAPs. Findings were analyzed with the Integrated Team Effectiveness Model. Setting: Four DAPs at two teaching and two community hospitals in Canada Participants: Twenty-two staff were interviewed about organizational characteristics, target service benchmarks, and teamwork processes, determinants and outcomes; 314 medical records were reviewed for actual service benchmarks. Results: Formal, informal and asynchronous team processes enabled service delivery and yielded many perceived benefits at the patient, staff and service levels. However, several DAP characteristics challenged teamwork and service delivery: referral volume/workload, time since launch, days per week of operation, rural-remote population, number and type of full-time/part-time human resources, staff co-location, information systems. As a result, all sites failed to meet target benchmarks (from referral to consultation median 4.0 visits, median wait time 35.0 days). Recommendations included improved information systems, more staff in all specialties, staff co-location and expanded roles for patient navigators. Findings were captured in a conceptual framework of lung cancer DAP teamwork determinants and outcomes. Conclusions: This study identified several DAP characteristics that could be improved to facilitate teamwork and enhance service delivery, thereby contributing to knowledge of organizational determinants of teamwork and associated outcomes. Findings can be used to update existing DAP guidelines, and by managers to plan or evaluate lung cancer DAPs. Ongoing research is needed to identify ideal roles for navigators, and staffing models tailored to case volumes

    Pathogens, parasites, and parasitoids associated with bumble bees (Bombus spp.) from Uruguay

    As elsewhere in the world, bumble bees play a vital role as pollinators in Uruguay, but knowledge on their health status is still limited. Between September 2012 and May 2013, 403 adult individuals of the two speciesof Bombus known for the country (Bombus atratus , Bombus bellicosus ) were collected in six localities.We found that 177 (119 B. atratus, 58 B. bellicosus ) were harboring one or two types of pathogens, parasites, or parasitoids.Identification of these natural enemies carried out by morphological or molecular procedures revealed the presence of two species of Microsporidia [Nosema ceranae (prevalence: 18.2 % in B. atratus; 44.9 % in B. bellicosus ),Tubulinosema pampeana (prevalence: 13 % in B. atratus )], two species of Nematoda [Sphaerularia bombi (prevalence: 40.4 % in B. atratus; 40% in B. bellicosus ) and an unidentified Mermithidae (prevalence: 0.8 % inB. bellicosus )], and one species of Diptera parasitoid (prevalence: 3.2 % in B. atratus ; 4.2 % in B. bellicosus ).
