    Human factors assessments of the STS-57 SpaceHab-1 mission

    SpaceHab-1 (STS-57) was the first of six scheduled Commercial Middeck Augmentation Module (CMAM) missions seeking to offer entrepreneurial companies an opportunity to use the resource of microgravity. The SpaceHab module, which occupies about one-fourth of the payload bay, is approximately 2-3/4 meters (9 feet) long and 4 meters (13.5 feet) in diameter. It provides a shirt-sleeve working environment and contains the storage space equivalent of 50 middeck lockers, considerably over and above the number of experiments that can be carried in the orbiter middeck alone. A modified Spacelab tunnel links the SpaceHab module to the middeck. While in orbit, the orbiter payload bay doors remain open, exposing the padded exterior of the lab and tunnel to space until preparation for reentry at the end of the flight. The crew for SpaceHab-1 was comprised of four males and two females, each of whom participated in some part of the human factors assessment (HFA) evaluation. The HFA was one of over twenty experiments manifested on this maiden flight of the SpaceHab module. HFA consisted of HFA-EPROC, HFA-LIGHT, HFA-SOUND, HFA-QUEST, and HFA-TRANS. The goal of HFA-EPROC was to assess the advantages and disadvantages of paper versus computer presentation for procedural tasks. The next two evaluations investigated the module's lighting and acoustic environment. HFA-TRANS sought to evaluate the design of the SpaceHab tunnel and to characterize translation through it. HFA-QUEST represented a consolidation of the in-flight questions generated by the HFA principal investigators involved in the acoustic, lighting, and translation studies

    Rapid sympathetic cooling to Fermi degeneracy on a chip

    Neutral fermions present new opportunities for testing many-body condensed matter systems, realizing precision atom interferometry, producing ultra-cold molecules, and investigating fundamental forces. However, since their first observation, quantum degenerate Fermi gases (DFGs) have continued to be challenging to produce, and have been realized in only a handful of laboratories. In this Letter, we report the production of a DFG using a simple apparatus based on a microfabricated magnetic trap. Similar approaches applied to Bose-Einstein Condensation (BEC) of 87Rb have accelerated evaporative cooling and eliminated the need for multiple vacuum chambers. We demonstrate sympathetic cooling for the first time in a microtrap, and cool 40K to Fermi degeneracy in just six seconds -- faster than has been possible in conventional magnetic traps. To understand our sympathetic cooling trajectory, we measure the temperature dependence of the 40K-87Rb cross-section and observe its Ramsauer-Townsend reduction.Comment: 5 pages, 4 figures (v3: new collision data, improved atom number calibration, revised text, improved figures.

    Inhalation of the Rho-kinase inhibitor Y-27632 reverses allergen-induced airway hyperresponsiveness after the early and late asthmatic reaction

    BACKGROUND: In guinea pigs, we have previously demonstrated that the contribution of Rho-kinase to airway responsiveness in vivo and ex vivo is enhanced after active sensitization with ovalbumin (OA). Using conscious, unrestrained OA-sensitized guina pigs, we now investigated the role of Rho-kinase in the development of airway hyperresponsiveness (AHR) after the allergen-induced early (EAR) and late asthmatic reaction (LAR) in vivo. METHODS: Histamine and PGF(2α )PC(100)-values (provocation concentrations causing 100% increase in pleural pressure) were assessed before OA-challenge (basal airway responsiveness) and after the OA-induced EAR (5 h after challenge) and LAR (23 h after challenge). Thirty minutes later, saline or the specific Rho-kinase inhibitor Y-27632 (5 mM, nebulizer concentration) were nebulized, after which PC(100)-values were reassessed. RESULTS: In contrast to saline, Y-27632 inhalation significantly decreased the basal responsiveness toward histamine and PGF(2α )before OA-challenge, as indicated by increased PC(100 )-values. Both after the allergen-induced EAR and LAR, AHR to histamine and PGF(2α )was present, which was reversed by Y-27632 inhalation. Moreover, there was an increased effectiveness of Y-27632 to reduce airway responsiveness to histamine and PGF(2α )after the EAR and LAR as compared to pre-challenge conditions. Saline inhalations did not affect histamine or PGF(2α )PC(100)-values at all. Interestingly, under all conditions Y-27632 was significantly more effective in reducing airway responsiveness to PGF(2α )as compared to histamine. Also, there was a clear tendency (P = 0.08) to a more pronounced degree of AHR after the EAR for PGF(2α )than for histamine. CONCLUSION: The results indicate that inhalation of the Rho-kinase inhibitor Y-27632 causes a considerable bronchoprotection to both histamine and PGF(2α). Moreover, the results are indicative of a differential involvement of Rho-kinase in the agonist-induced airway obstruction in vivo. Increased Rho-kinase activity contributes to the allergen-induced AHR to histamine and PGF(2α )after both the EAR and the LAR, which is effectively reversed by inhalation of Y-27632. Therefore, Rho-kinase can be considered as a potential pharmacotherapeutical target in allergic asthma

    Folate catabolites in spot urine as non-invasive biomarkers of folate status during habitual intake and folic acid supplementation.

    Folate status, as reflected by red blood cell (RCF) and plasma folates (PF), is related to health and disease risk. Folate degradation products para-aminobenzoylglutamate (pABG) and para-acetamidobenzoylglutamate (apABG) in 24 hour urine have recently been shown to correlate with blood folate. Since blood sampling and collection of 24 hour urine are cumbersome, we investigated whether the determination of urinary folate catabolites in fasted spot urine is a suitable non-invasive biomarker for folate status in subjects before and during folic acid supplementation. Immediate effects of oral folic acid bolus intake on urinary folate catabolites were assessed in a short-term pre-study. In the main study we included 53 healthy men. Of these, 29 were selected for a 12 week folic acid supplementation (400 µg). Blood, 24 hour and spot urine were collected at baseline and after 6 and 12 weeks and PF, RCF, urinary apABG and pABG were determined. Intake of a 400 µg folic acid bolus resulted in immediate increase of urinary catabolites. In the main study pABG and apABG concentrations in spot urine correlated well with their excretion in 24 hour urine. In healthy men consuming habitual diet, pABG showed closer correlation with PF (rs = 0.676) and RCF (rs = 0.649) than apABG (rs = 0.264, ns and 0.543). Supplementation led to significantly increased folate in plasma and red cells as well as elevated urinary folate catabolites, while only pABG correlated significantly with PF (rs = 0.574) after 12 weeks. Quantification of folate catabolites in fasted spot urine seems suitable as a non-invasive alternative to blood or 24 hour urine analysis for evaluation of folate status in populations consuming habitual diet.     A large West Antarctic Ice Sheet explains early Neogene sea-level amplitude

    Early to Middle Miocene sea-level oscillations of approximately 40-60 m estimated from far-field records1-3 are interpreted to reflect the loss of virtually all East Antarctic ice during peak warmth2. This contrasts with ice-sheet model experiments suggesting most terrestrial ice in East Antarctica was retained even during the warmest intervals of the Middle Miocene4,5. Data and model outputs can be reconciled if a large West Antarctic Ice Sheet (WAIS) existed and expanded across most of the outer continental shelf during the Early Miocene, accounting for maximum ice-sheet volumes. Here we provide the earliest geological evidence proving large WAIS expansions occurred during the Early Miocene (~17.72-17.40 Ma). Geochemical and petrographic data show glacimarine sediments recovered at International Ocean Discovery Program (IODP) Site U1521 in the central Ross Sea derive from West Antarctica, requiring the presence of a WAIS covering most of the Ross Sea continental shelf. Seismic, lithological and palynological data reveal the intermittent proximity of grounded ice to Site U1521. The erosion rate calculated from this sediment package greatly exceeds the long-term mean, implying rapid erosion of West Antarctica. This interval therefore captures a key step in the genesis of a marine-based WAIS and a tipping point in Antarctic ice-sheet evolution

    The value of standards for health datasets in artificial intelligence-based applications

    Artificial intelligence as a medical device is increasingly being applied to healthcare for diagnosis, risk stratification and resource allocation. However, a growing body of evidence has highlighted the risk of algorithmic bias, which may perpetuate existing health inequity. This problem arises in part because of systemic inequalities in dataset curation, unequal opportunity to participate in research and inequalities of access. This study aims to explore existing standards, frameworks and best practices for ensuring adequate data diversity in health datasets. Exploring the body of existing literature and expert views is an important step towards the development of consensus-based guidelines. The study comprises two parts: a systematic review of existing standards, frameworks and best practices for healthcare datasets; and a survey and thematic analysis of stakeholder views of bias, health equity and best practices for artificial intelligence as a medical device. We found that the need for dataset diversity was well described in literature, and experts generally favored the development of a robust set of guidelines, but there were mixed views about how these could be implemented practically. The outputs of this study will be used to inform the development of standards for transparency of data diversity in health datasets (the STANDING Together initiative)

    International audienceGraphs increasingly stand out as an essential data structurein the field of data sciences. To study graphs, or sub-graphs, that char-acterize a set of observations, it is necessary to describe them formally,in order to characterize equivalence relations that make sense in thescope of the considered application domain. Hence we seek to define acanonical graph notation, so that two isomorphic (sub) graphs have thesame canonical form. Such notation could subsequently be used to indexand retrieve graphs or to embed them efficiently in some metric space.Sequential optimized algorithms solving this problem exist, but do notdeal with labeled edges, a situation that occurs in important applicationdomains such as chemistry. We present in this article a new algorithmbased on graph rewriting that provides a general and complete solution tothe graph canonization problem. Although not reported here, the formalproof of the validity of our algorithm has been established. This claim isclearly supported empirically by our experimentation on synthetic com-binatorics as well as natural graphs. Furthermore, our algorithm supportsdistributed implementations, leading to efficient computing perspectives

    Incidence and characteristics of distal radius fractures in a southern Swedish region

    <p>Abstract</p> <p>Background</p> <p>The incidence of distal radius fracture has increased substantially during the last 50 years according to several studies that estimated the overall incidence in various general populations. The incidence of fracture classified according to severity has not been well documented. The aim of this population-based study was to estimate the overall and type-specific incidence rates of distal radius fracture in a representative population in southern Sweden.</p> <p>Methods</p> <p>During 2001, all persons older than 18 years with acute distal radius fracture in the southern Swedish region of Northeastern Scania were prospectively recorded. A radiologist classified the fractures according to the AO system and measured volar tilt and ulnar variance. A fracture with volar tilt outside a range of -5° to 20° and/or ulnar variance of 2 mm or greater was defined as displaced.</p> <p>Results</p> <p>335 persons with acute distal radius fracture were recorded during the 1-year period. The overall incidence rate was 26 (95% confidence interval 23–29) per 10,000 person-years. Among women the incidence rate increased rapidly from the age of 50 and reached a peak of 119 per 10,000 person-years in women 80 years and older. The incidence rate among women 50 to 79 years old (56 per 10,000 person-years) was lower than that reported in previous studies of similar populations. Among men the incidence rate was low until the age of 80 years and older when it increased to 28 per 10,000 person-years. Fractures classified as AO type A comprised about 80% of the fractures in women and 64% in men. Almost two-thirds of all fractures were displaced and among men and women 80 years and older more than 80% of the fractures were displaced.</p> <p>Conclusion</p> <p>The incidence rate of distal radius fracture in women 50 to 79 years old was lower than previously reported, which may indicate declining incidence in this group. In both sexes, the incidence was highest in the age group of 80 years and older. With a growing number of elderly in the general population, the impact of distal radius fracture in the future may be considerable.</p
