    A neuroevolutionary approach to feature selection using multiobjective evolutionary algorithms

    Feature selection plays a central role in predictive analysis where datasets have hundreds or thousands of variables available. It can also reduce the overall training time and the computational costs of the classifiers used. However, feature selection methods can be computationally intensive or dependent of human expertise to analyze data. This study proposes a neuroevolutionary approach which uses multiobjective evolutionary algorithms to optimize neural network parameters in order to find the best network able to identify the most important variables of analyzed data. Classification is done through a Support Vector Machine (SVM) classifier where specific parameters are also optimized.     In vitro inhibition of linoleic acid peroxidation by primary S-nitrosothiols

    Nitric oxide ( NO) is an effective chain-breaking antioxidant in the inhibition of lipid peroxidation and circulates in vivo mainly as primary S-nitrosothiols (RSNOs). In this work, the in vitro peroxidation of linoleic acid-SDS comicelles (LA-SDS) catalyzed by soybean lipoxygenase (SLO) and FeII ions was monitored in the presence and absence of three primary RSNOs: S-nitrosocysteine, S-nitroso-N-acetylcysteyne and S-nitrosoglutathione. Kinetic measurements based on the formation of conjugated double bonds and fluorescent oxidized LA-lysine adducts, showed that RSNOs are more potent antioxidants than their corresponding free thiols (RSHs) in equimolar conditions. These results are consistent with the blocking of LA-SDS peroxidation by RSNOs through the inactivation of peroxyl/alkoxyl (LOO /LO ) radicals, leading to nitrogen-containing products of oxidized LA, which release free NO. These results indicate that endogenous RSNOs may play a major role in the blocking of lipid peroxidation in vivo, through the primary inactivation of alkoxyl/peroxyl radicals and also of preformed lipid hydroperoxides.O óxido nítrico ( NO) é um antioxidante efetivo na inibição da peroxidação lipídica e circula in vivo principalmente como S-nitrosotióis primários (RSNOs). Neste trabalho, a peroxidação in vitro de comicelas do ácido linileico-SDS (LA-SDS) catalisada por lipoxigenase de soja (SLO) e íons FeII foi monitorada na presença e na ausência de três RSNOs: S-nitrosocisteína, S-nitroso-N-acetilcisteína e S-nitrosoglutationa. Medidas cinéticas baseadas na formação de duplas ligações conjugadas e adutos fluorescentes de lisina-La oxidado mostraram que os RSNOs são antioxidantes mais potentes que seus tióis livres (RSHs) correspondentes em condições equimolares. Esses resultados são consistentes com o bloqueio da peroxidação de LA-SDS por RSNOs através da inativação dos radicais peroxila/alcoxila (LOO /LO ), levando a produtos nitrogenados do LA oxidado, os quais liberam NO.     Adicionando a entrevista motivacional e o mapeamento cognitivo à terapia cognitivo-comportamental em grupo: resultados de um ensaio clínico randomizado

    OBJECTIVE: Recent factor-analytic studies of obsessive-compulsive disorder identified consistent symptom dimensions. This study was designed in order to observe which obsessive compulsive symptom dimensions could be changed by adding two individual sessions of motivational interviewing and thought mapping of cognitive-behavioral group therapy using a randomized clinical trial. METHOD: Forty outpatients with a primary diagnosis of obsessive-compulsive disorder were randomly assigned to receive cognitive-behavioral group therapy (control group) or motivational interviewing+thought mapping plus cognitive-behavioral group therapy. To evaluate changes in symptomdimensions, the Dimensional Yale-Brown Obsessive-Compulsive Scale was administered at baseline and after treatment. RESULTS: At post-treatment, there were statistically significant differences between cognitive-behavioral group therapy and motivational interviewing+thought mapping+cognitivebehavioral group therapy groups in the mean total Dimensional Yale-Brown Obsessive-Compulsive Scale score, and in the contamination and aggression dimension score. Hoarding showed a statistical trend towards improvement. CONCLUSION: These findings suggest that adding motivational interviewing+thought mapping to cognitive-behavioral group therapy can facilitate changes and bring about a decrease in the scores in different obsessive-compulsive disorder symptom dimensions, as measured by the Dimensional Yale-Brown Obsessive-Compulsive Scale. Nonetheless, additional trials are needed to confirm these results.OBJETIVO: Recentes estudos utilizando análise fatorial no transtorno obsessivocompulsivo identificaram dimensões consistentes dos sintomas. Este estudo foi delineado para observar quais dimensões dos sintomas obsessivo-compulsivos podem ser modificadas adicionando duas sessões individuais de entrevista motivacional e mapeamento cognitivo à terapia cognitivo-comportamental em grupo usando um ensaio clínico randomizado. MÉTODO: Quarenta pacientes ambulatoriais com diagnóstico primário de transtorno obsessivo-compulsivo foram alocados aleatoriamente para receber terapia cognitivo-comportamental em grupo (grupo controle) ou entrevista motivacional+mapeamento cognitivo+terapia cognitivo-comportamental em grupo. Para avaliar mudanças nas dimensões dos sintomas, foi administrada a Escala Dimensional para Sintomas Obsessivo-Compulsivos de Yale-Brown antes do início e após o tratamento. RESULTADOS: Ao final do tratamento houve diferença estatisticamente significativa entre a terapia cognitivo-comportamental em grupo e entrevista motivacional+mapeamento cognitivo+terapia cognitivocomportamental em grupo na média do escore total da Escala Dimensional para Sintomas Obsessivo-Compulsivos de Yale-Brown e no escore da dimensão de contaminação e agressão. Colecionismo apresentou melhora com tendência estatística. CONCLUSÃO: Esses achados sugerem que acrescentar entrevista motivacional+mapeamento cognitivo à terapia cognitivo-comportamental em grupo pode facilitar mudanças na redução dos escores nas diferentes dimensões dos sintomas, como indicado pela Escala Dimensional para Sintomas Obsessivo-Compulsivos de Yale-Brown.     Microsatellite markers for urochloa Humidicola (poaceae) and their transferability to other urochloa species

    Urochloa humidicola is a warm-season grass commonly used as forage in the tropics and is recognized for its tolerance to seasonal flooding. This grass is an important forage species for the Cerrado and Amazon regions of Brazil. U. humidicola is a polyploid species with variable ploidy (6X-9X) and facultative apomixis with high phenotypic plasticity. However, this apomixis and ploidy, as well as the limited knowledge of the genetic basis of the germplasm collection, have constrained genetic breeding activities, yet microsatellite markers may enable a better understanding of the species' genetic composition. This study aimed to develop and characterize new polymorphic microsatellite molecular markers in U. humidicola and to evaluate their transferability to other Urochloa species. Findings: A set of microsatellite markers for U. humidicola was identified from two new enriched genomic DNA libraries: the first library was constructed from a single sexual genotype and the second from a pool of eight apomictic genotypes selected on the basis of previous results. Of the 114 loci developed, 72 primer pairs presented a good amplification product, and 64 were polymorphic among the 34 genotypes tested. The number of bands per simple sequence repeat (SSR) locus ranged from 1 to 29, with a mean of 9.6 bands per locus. The mean polymorphism information content (PIC) of all loci was 0.77, and the mean discrimination power (DP) was 0.87. STRUCTURE analysis revealed differences among U. humidicola accessions, hybrids, and other Urochloa accessions. The transferability of these microsatellites was evaluated in four species of the genus, U. brizantha, U. decumbens, U. ruziziensis, and U. dictyoneura, and the percentage of transferability ranged from 58.33% to 69.44% depending on the species. Conclusions: This work reports new polymorphic microsatellite markers for U. humidicola that can be used for breeding programs of this and other Urochloa species, including genetic linkage mapping, quantitative trait loci identification, and marker-assisted selection8

    Microsatellite markers for studies with the carnivorous plant Philcoxia minensis (Plantaginaceae)

    Premise of the study: Microsatellite markers were developed for the critically endangered carnivorous species Philcoxia minensis (Plantaginaceae) for further population genetic studies aiming at its conservation. Methods and Results: We identifi ed 29 clones containing 40 microsatellites from a genomic enriched library. A total of 27 primer pairs were developed and evaluated in 30 individuals of a natural P. minensis population. Seventeen markers successfully presented amplifi cation products within the expected size range, of which 12 were polymorphic. The expected and observed heterozygosities ranged from 0.03 to 0.65 and from 0.00 to 0.77, respectively. Positive transferability with the related species P. bahiensis was observed for the same 17 markers.     Verification of the influence of loading and mortar coating thickness on resistance to high temperatures due to fire on load-bearing masonries with clay tiles

    Masonry has been widely used as a construction method. However, there is a lack of information on its fire behavior due to the multitude of variables that could influence this method. Thispaperaimedtoidentifytheinfluenceofloadingandmortarcoatingthicknessonthefirebehavior of masonry. Hence, six masonries made of clay tiles laid with mortar were evaluated. The mortar coatinghadathicknessof25mmonthefacenotexposedtohightemperatures,whilethefire-exposed face had thicknesses of 0, 15, and 25 mm. For each mortar coating thickness, two specimens were tested, with and without loading of 10 tf/m. The real-scale specimens were subjected to the standard ISO 834 fire curve for four hours, during which the properties of stability, airtightness, and thermal insulation were assessed. Results showed that loaded specimens yielded smaller deformations than unloadedones. Samplesthatlackedmortarcoatingonthefire-exposedfaceunderwentfireresistance decreaseof27.5%,whiletheoneswith15mmdecreasedby58.1%,andtheoneswith25mmdecreased by 41.0%. As mortar coating thickness increased, the plane deformations decreased from 40 mm to 29 mm and the thermal insulation properties of the walls improved significantly. For specimens with mortarcoatingthicknessof25mm,theloadapplicationresultedinareductionof23.8%ofthethermal insulation, while the unloaded specimen showed a decrease of 43.3%, as well as a modification of its fire-resistance rating

    Quantitative Proteomic Analysis Reveals Changes in the Benchmark Corynebacterium pseudotuberculosis Biovar Equi Exoproteome after Passage in a Murine Host

    Corynebacterium pseudotuberculosis biovar equi is the etiologic agent of ulcerative lymphangitis. To investigate proteins that could be related to the virulence of this pathogen, we combined an experimental passage process using a murine model and high-throughput proteomics with a mass spectrometry, data-independent acquisition (LC-MSE) approach to identify and quantify the proteins released into the supernatants of strain 258_equi. To our knowledge, this approach allowed characterization of the exoproteome of a C. pseudotuberculosis equi strain for the first time. Interestingly, the recovery of this strain from infected mouse spleens induced a change in its virulence potential, and it became more virulent in a second infection challenge. Proteomic screening performed from culture supernatant of the control and recovered conditions revealed 104 proteins that were differentially expressed between the two conditions. In this context, proteomic analysis of the recovered condition detected the induction of proteins involved in bacterial pathogenesis, mainly related to iron uptake. In addition, KEGG enrichment analysis showed that ABC transporters, bacterial secretion systems and protein export pathways were significantly altered in the recovered condition. These findings show that secretion and secreted proteins are key elements in the virulence and adaptation of C. pseudotuberculosis. Collectively, bacterial pathogenesis-related proteins were identified that contribute to the processes of adherence, intracellular growth and evasion of the immune system.
