    Surface sterilant effect on the regeneration efficiency from cotyledon explants of groundnut (Arachis hypogea L.) varieties adapted to eastern and Southern Africa

    Five groundnut (Arachis hypogea L.) varieties - ICGV-12991, ICGV-99568, ICGV-90704, CG-2 and Chalimbana - that are adapted to Eastern and Southern Africa were compared to variety JL 24 for their regeneration response in tissue culture. Sodium hypochlorite and mercuric chloride were compared for efficiency as sterilizing agents and subsequent effect on regeneration. All five varieties formed shoot buds that elongated well on shoot elongation medium. ICGV-90704 and Chalimbana performed betterthan the other three varieties for shoot organogenesis although all varieties produced healthy rooted plants in vitro that were successfully transferred to the greenhouse where they exhibited normal growth, flowering and seed set. Both sterilizing agents were suitable, but mercuric chloride was lessharmful than sodium hypochlorite. This study established a basis for genetic engineering activities on African groundnuts in the future

    Remote Ischemic Preconditioning (RIPC) is a non-invasive cardioprotective intervention that involves brief cycles of limb ischemia and reperfusion. This is typically delivered by inflating and deflating a blood pressure cuff on one or more limb(s) for several cycles, each inflation-deflation being 3-5Ā min in duration. RIPC has shown potential for protecting the heart and other organs from injury due to lethal ischemia and reperfusion injury, in a variety of clinical settings. The mechanisms underlying RIPC are under intense investigation but are just beginning to be deciphered. Emerging evidence suggests that RIPC has the potential to improve exercise performance, via both local and remote mechanisms. This review discusses the clinical studies that have investigated the role of RIPC in cardioprotection as well as those studying its applicability in improving athletic performance, while examining the potential mechanisms involved

    A comparative study of efficacy and safety of flupirtine versus piroxicamin patients with low back pain

    Background: Low back pain is a common musculoskeletal symptom caused by a variety of disorders that affect the lumbar spine. The most frustrating aspect in the treatment of low back pain is that there is ā€œno magic bulletsā€. The objective of the study was to compare the efficacy and safety of flupirtine versus piroxicam in patients with back pain.Methods: This was prospective, open labeled, randomized, comparative clinical study conducted by the Departments Orthopedics and Pharmacology, BMC&H, Chitradurga. Study was conducted on 60 patients of either sex, aged above 18 years with low back pain. Assessments were done for Finger-to-Floor Distance (FFD), lumbar pain, Lasegueā€™s sign, tenderness of vertebral muscles, pain & sensory disturbance in lower limbs and response to therapy for efficacy. Parametric data was analyzed by t-test and proportions were compared using Chi-square test.Results: 74 patients were randomized to 2 groups of 37 each. Group I patients received flupirtine maleate 100 mg twice daily and Group II patients received piroxicam 20 mg twice daily for 14 days. 30 patients in each group completed the study and were analysed. On intergroup comparison, there was no statistical difference (p>0.05) in the efficacy parameters of finger-to-floor distance (FFD), lumbar pain, Lasegueā€™s sign, tenderness of vertebral muscles, sensory disturbance in lower limbs, VAS scores & global assessment of response to therapy. 13.3% in flupirtine group and 16.6% in piroxicam group reported adverse events. Ā Conclusions: Both flupiritine and piroxicam were equally effective but flupirtine was better tolerated than piroxicam.

    Influence of transport and ocean ice extent on biogenic aerosol sulfur in the Arctic atmosphere

    The recent decline in sea ice cover in the Arctic Ocean could affect the regional radiative forcing via changes in sea ice-atmosphere exchange of dimethyl sulfide (DMS) and biogenic aerosols formed from its atmospheric oxidation, such as methanesulfonic acid (MSA). This study examines relationships between changes in total sea ice extent north of 70 degrees N and atmospheric MSA measurement at Alert, Nunavut, during 1980-2009; at Barrow, Alaska, during 1997-2008; and at Ny-Alesund, Svalbard, for 1991-2004. During the 1980-1989 and 1990-1997 periods, summer (July-August) and June MSA concentrations at Alert decreased. In general, MSA concentrations increased at all locations since 2000 with respect to 1990 values, specifically during June and summer at Alert and in summer at Barrow and Ny-Alesund. Our results show variability in MSA at all sites is related to changes in the source strengths of DMS, possibly linked to changes in sea ice extent as well as to changes in atmospheric transport patterns. Since 2000, a late spring increase in atmospheric MSA at the three sites coincides with the northward migration of the marginal ice edge zone where high DMS emissions from ocean to atmosphere have previously been reported. Significant negative correlations are found between sea ice extent and MSA concentrations at the three sites during the spring and June. These results suggest that a decrease in seasonal ice cover influencing other mechanisms of DMS production could lead to higher atmospheric MSA concentrations

    Effects of epidural lidocaine analgesia on labor and delivery: A randomized, prospective, controlled trial

    BACKGROUND: Whether epidural analgesia for labor prolongs the active-first and second labor stages and increases the risk of vacuum-assisted delivery is a controversial topic. Our study was conducted to answer the question: does lumbar epidural analgesia with lidocaine affect the progress of labor in our obstetric population? METHOD: 395 healthy, nulliparous women, at term, presented in spontaneous labor with a singleton vertex presentation. These patients were randomized to receive analgesia either, epidural with bolus doses of 1% lidocaine or intravenous, with meperidine 25 to 50 mg when their cervix was dilated to 4 centimeters. The duration of the active-first and second stages of labor and the neonatal apgar scores were recorded, in each patient. The total number of vacuum-assisted and cesarean deliveries were also measured. RESULTS: 197 women were randomized to the epidural group. 198 women were randomized to the single-dose intravenous meperidine group. There was no statistical difference in rates of vacuum-assisted delivery rate. Cesarean deliveries, as a consequence of fetal bradycardia or dystocia, did not differ significantly between the groups. Differences in the duration of the active-first and the second stages of labor were not statistically significant. The number of newborns with 1-min and 5-min Apgar scores less than 7, did not differ significantly between both analgesia groups. CONCLUSION: Epidural analgesia with 1% lidocaine does not prolong the active-first and second stages of labor and does not increase vacuum-assisted or cesarean delivery rate

    Production of Ī±1,3-galactosyltransferase-deficient pigs

    The enzyme Ī±1,3-galactosyltransferase (Ī±1,3GT or GGTA1) synthesizes Ī±1,3galactose (Ī±1,3Gal) epitopes (GalĪ±1,3GalĪ²1,4GlcNAc-R), which are the major xenoantigens causing hyperacute rejection in pig-to-human xenotransplantation. Complete removal of Ī±1,3Gal from pig organs is the critical step toward the success of xenotransplantation. We reported earlier the targeted disruption of one allele of the Ī±1,3GT gene in cloned pigs. A selection procedure based on a bacteria[toxin was used to select for cells in which the second allele of the gene was knocked out. Sequencing analysis demonstrated that knockout of the second allele of the Ī±1,3GT gene was caused by a T-to-G single point mutation at the second base of exon 9, which resulted in inactivation of the Ī±1,3GT protein. Four healthy Ī±1,3GT double-knockout female piglets were produced by three consecutive rounds of cloning. The piglets carrying a point mutation in the Ī±1,3GT gene hold significant value, as they would allow production of Ī±1,3Gal-deficient pigs free of antibiotic-resistance genes and thus have the potential to make a safer product for human use

    Towards detection of early response in neoadjuvant chemotherapy of breast cancer using Bayesian intravoxel incoherent motion

Introduction: The early identification of good responders to neoadjuvant chemotherapy (NACT) holds a significant potential in the optimal treatment of breast cancer. A recent Bayesian approach has been postulated to improve the accuracy of the intravoxel incoherent motion (IVIM) model for clinical translation. This study examined the prediction and early sensitivity of Bayesian IVIM to NACT response. Materials and methods: Seventeen female patients with breast cancer were scanned at baseline and 16 patients were scanned after Cycle 1. Tissue diffusion and perfusion from Bayesian IVIM were calculated at baseline with percentage change at Cycle 1 computed with reference to baseline. Cellular proliferative activity marker Ki-67 was obtained semi-quantitatively with percentage change at excision computed with reference to core biopsy. Results: The perfusion fraction showed a significant difference (p = 0.042) in percentage change between responder groups at Cycle 1, with a decrease in good responders [āˆ’7.98% (āˆ’19.47ā€“1.73), n = 7] and an increase in poor responders [10.04% (5.09ā€“28.93), n = 9]. There was a significant correlation between percentage change in perfusion fraction and percentage change in Ki-67 (p = 0.042). Tissue diffusion and pseudodiffusion showed no significant difference in percentage change between groups at Cycle 1, nor was there a significant correlation against percentage change in Ki-67. Perfusion fraction, tissue diffusion, and pseudodiffusion showed no significant difference between groups at baseline, nor was there a significant correlation against Ki-67 from core biopsy. Conclusion: The alteration in tumour perfusion fraction from the Bayesian IVIM model, in association with cellular proliferation, showed early sensitivity to good responders in NACT.     A photovoltaic-fed z-source inverter motor drive with fault-tolerant capability for rural irrigation

In recent years, photovoltaic (PV) systems have emerged as economical solutions for irrigation systems in rural areas. However, they are characterized by low voltage output and less reliable configurations. To address this issue in this paper, a promising inverter configuration called Impedance (Z)-source inverter (ZSI) is designed and implemented to obtain high voltage output with single-stage power conversion, particularly suitable for irrigation application. An improved and efficient modulation scheme and design specifications of the network parameters are derived. Additionally, a suitable fault-tolerant strategy is developed and implemented to improve reliability and efficiency. It incorporates an additional redundant leg with an improved control strategy to facilitate the fault-tolerant operation. The proposed fault-tolerant circuit is designed to handle switch failures of the inverter modules due to the open-circuit and short-circuit faults. The relevant simulation and experimental results under normal, faulty and post-fault operation are presented. The post-fault operation characteristics are identical to the normal operation. The motor performance characteristics such as load current, torque, harmonic spectrum, and efficiency are thoroughly analysed to prove the suitability of the proposed system for irrigation applications.
