    A systematic approach to the interrogation and sharing of standardised biofilm signatures

    Publicado em "6th International Conference on Practical Applications of Computational Biology & Bioinformatics", ISBN 978-3-642-28838-8The study of microorganism consortia, also known as biofilms, is associated to a number of applications in biotechnology, ecotechnology and clinical domains. A public repository on existing biofilm studies would aid in the design of new studies as well as promote collaborative and incremental work. However, bioinformatics approaches are hampered by the limited access to existing data. Scientific publications summarise the studies whilst results are kept in researchers’ private ad hoc files. Since the collection and ability to compare existing data is imperative to move forward in biofilm analysis, the present work has addressed the development of a systematic computer-amenable approach to biofilm data organisation and standardisation. A set of in-house studies involving pathogens and employing different state-of-the-art devices and methods of analysis was used to validate the approach.     Este trabalho caracteriza dois Cambissolos de uma vertente no Espinhaço Meridional/MG a partir de análises texturais e de seções delgadas. Diferenças significativas foram observadas, como cor, espessura do horizonte Bi e presença de mosqueados na média vertente. Diante das diferenças, este perfil foi considerado um Cambissolo em vias de transformação para Latossolo, fato evidenciado pelos indícios de microagregação

    Biostimulants in organic vegetable nurseries: Study case in lettuce

    In order to create better conditions to achieve food safety and sovereignty, agroecology, as a science, looks for solutions for several steps of the technical itineraries of the crops. Crop nutrition and protection are two main crop itinerary components that have been in the center of farmers challenges and consumers concerns, and biopreparations, which have been prepared using natural substances, have been used in agroecological systems, most times based on farmers empirical knowledge. Six biopreparations—purslane vinegar, prickly pear vinegar, orange fermented fruit juice, garlic extract, nettle infusion, and horsetail decoction—were used in this study, for physicochemical analyses and field tests in two different locations (Viseu and Castelo Branco, Portugal) in nurseries of Lactuca sativa L. (lettuce), and aimed to validate its results and uses. The nettle infusion presented the best stimulating results for the length of aerial part and the garlic extract presented the best results for the length of root, though there weren’t significant differences and effects when compared with the control.     Interaction of Genetic Variations in NFE2L2 and SELENOS Modulates the Risk of Hashimoto's Thyroiditis.

    Background: Several single-nucleotide polymorphisms (SNPs) are known to increase the risk of Hashimoto's thyroiditis (HT); such SNPs reside in thyroid-specific genes or in genes related to autoimmunity, inflammation, and/or cellular defense to stress. The transcription factor Nrf2, encoded by NFE2L2, is a master regulator of the cellular antioxidant response. This study aimed to evaluate the impact of genetic variation in NFE2L2 on the risk of developing HT. Methods: In a case-control candidate gene association study, functional SNPs in the NFE2L2 promoter (rs35652124, rs6706649, and rs6721961) were examined either as independent risk factors or in combination with a previously characterized HT risk allele (rs28665122) in the gene SELENOS, encoding selenoprotein S (SelS). A total of 997 individuals from the north of Portugal (Porto) were enrolled, comprising 481 HT patients and 516 unrelated healthy controls. SELENOS and NFE2L2 SNPs were genotyped using TaqMan <sup>®</sup> assays and Sanger sequencing, respectively. Odds ratios (ORs) were calculated using logistic regression, with adjustment for sex and age. Expression of SelS was analyzed by immunohistochemistry in thyroid tissue from HT patients and control subjects. Molecular interactions between the Nrf2 and SelS pathways were investigated in thyroid tissues from mice and in rat PCCL3 thyroid follicular cells. Results: When all three NFE2L2 SNPs were considered together, the presence of one or more minor alleles was associated with a near-significant increased risk (OR = 1.43, p = 0.072). Among subjects harboring only major NFE2L2 alleles, there was no increased HT risk associated with heterozygosity or homozygosity for the SELENOS minor allele. Conversely, in subjects heterozygous or homozygous for the SELENOS risk allele, the presence of an NFE2L2 minor allele significantly increased HT risk by 2.8-fold (p = 0.003). Immunohistochemistry showed reduced expression of SelS in thyroid follicular cells of HT patients. In Nrf2 knockout mice, there was reduced expression of SelS in thyroid follicular cells; conversely, in PCCL3 cells, reducing SelS expression caused reduced activity of Nrf2 signaling. Conclusions: The NFE2L2 promoter genotype interacts with the SELENOS promoter genotype to modulate the risk of HT in a Portuguese population. This interaction may be due to a bidirectional positive feedback between the Nrf2 and SelS pathways

    Biochemical and bioactive phytonutrients changes in tissues of two cultivars of fresh-cut cassava in stick form under refrigerated storage

    The aim of this study was to evaluate the effects of fresh-cut in stick form of two cassava cultivars on their biochemical and antioxidant characteristics, at two harvest times. The fresh cut cassava sticks were packaged in polypropylene, maintained at 5±1ºC, with 90±5% relative humidity for 12 days. The concentration of carotenoids, total soluble phenolic compounds and the activity of phenylalanine ammonia lyase and antioxidant capacity were significantly higher for the sticks of cultivar 'Amarela' cassava than for the sticks of cultivar 'Cacau'. The concentrations of carotenoids, total soluble phenolic compounds, and the activity of phenylalanine ammonia lyase and antioxidant capacity were all significantly greater when harvested at 14-month of age. There was a significant increase in the activity of phenylalanine ammonia lyase during the 12 days of storage at 5ºC. However, there was a decrease in total carotenoids, soluble phenolic compounds, and in the antioxidant capacity of the two cultivars

    Pharmacological activity of the hydroalcoholic extract from Hovenia dulcis thunberg fruit and the flavonoid dihydromyricetin during hypercholesterolemia induced in rats

    Cerebrovascular accidents and coronary artery disease are the leading causes of cardiovascular mortalities in Brazil and high levels of LDL cholesterol are one of the main risk factors. In this context, several plant extracts and natural substances have shown promise as cholesterol-lowering. The objective of this study was to evaluate the potential of the hydroalcoholic extract of the fruit of H. dulcisand of dihydromyricetin in cholesterol reduction in hypercholesterolemic rats. Forty-two Wistar male rats were distributed into seven groups of six animals that received diets supplemented with 1% cholesterol and 0.3% cholic acid, with the exception of the control group, which received conventional diets. Animals were treated with oral suspensions containing: atorvastatin 1.0 mg/kg; H. dulcisextract at 50.0 and 100.0 mg/kg and dihydromyricetin at 25.0 and 50.0 mg/kg vehicle (control group). The following biochemical markers were evaluated; total cholesterol, HDL-C, LDL-C, triglycerides, AST, ALT, and alkaline phosphatase. The hypercholesterolemic diet was effective in inducing hypercholesterolemia, increasing total cholesterol by 112.7% relative to the control group. The treatments with two doses of the extract proved to be promising hypocholesterolemic agents, as they were able to substantially reduce total cholesterol and LDL-C, without significantly altering triglycerides, hepatic transaminases, and alkaline phosphatase, thereby encouraging the studies with the plant H. dulcis. The groups treated with the flavonoid dihydromyricetin, although they showed a significant reduction in total cholesterol and LDL-C, and found increases in triglycerides and hepatic transaminases, which is unwanted in the context of hypercholesterolaemia

    Luteal dynamics in goats: morphological and endocrine features

    The aim of this study was to establish the morphologic and endocrine characteristics of luteal dynamics in goats. It was used Toggenburg female goats that showed natural estrus in a 48-hour interval. After estrus, ultrasonographic evaluations of the ovaries were daily performed during 21 days using a portable device (5MHz probe). Blood sample was collected for plasma progresterone (P4) determination. Corpora lutea were detected for the first time on day 5 and progressively increased in size until D9 (1.26 ± 0.08 cm²), with no variation on subsequent days. In females with one ovulation, the first visualization of the corpora lutea was earlier than in those with multiple ovulation (4.54 ± 0.18 vs 5.74 ± 0.25 days). At the moment of the first visualization, luteal area was smaller in animals with single ovulation. Plasma P4 concentration progressively increased until day 9 and it did not show significant increase until luteolysis, characterized by a sharp decrease in P4 concentration, reaching values below 1 ng/mL in 24 hours. The luteal area slowly and gradually decreased in size. It was observed a significant positive correlation between P4 concentration and area during luteogenesis and luteolysis (r = 0.63 and r = 0.50, respectively). When corpus luteum reached its maximum size (D9), female with more than one corpora lutea, with a greater luteal tissue area, did not show P4 concentration higher than those with one ovulation (5.92 ± 0.59 vs 7.04 ± 0.79 ng/mL). These results show that luteal dynamics in Toggenbur goats follow a similar pattern to those observed in other goat breeds and luteal tissue growth was positively correlated with corpora lutea functionality.Objetivou-se neste estudo estabelecer as características morfológicas e endócrinas da dinâmica luteal em cabras. Foram utilizadas fêmeas da raça Toggenburg que manifestaram estro natural em um intervalo de 48 horas. Após o estro, foram realizadas avaliações ultrassonográficas diárias dos ovários durante 21 dias, utilizando-se um aparelho portátil (5 MHz). Amostras de sangue foram coletadas para dosagem de progesterona (P4) no plasma. Os corpos lúteos foram detectados pela primeira vez no D5 e aumentaram progressivamente de tamanho até o D9 (1,26 ± 0,08 cm²), não havendo variação nos dias subsequentes. Nas fêmeas com uma ovulação, a primeira visualização do corpo lúteo foi mais precoce que naquelas com ovulação múltipla (4,54 ± 0,18 vs 5,74 ± 0,25 dias). No momento da primeira visualização, a área luteal foi menor nos animais com uma ovulação. A concentração plasmática de P4 aumentou progressivamente até o D9 e não apresentou aumento significativo até o momento da luteólise, caracterizada por uma acentuada queda da concentração de P4, atingindo valores inferiores a 1 ng/mL em um intervalo de 24 horas. A área luteal diminuiu de forma lenta e gradual. Foi observada uma correlação positiva significativa entre a área e a concentração de P4 durante a lutegêonese e a luteólise (r = 0,63 e r = 0,50; respectivamente). No dia em que o corpo lúteo atinge sua área máxima (D9), as fêmeas com mais de um corpo lúteo, com maior área de tecido luteal, não apresentaram concentração de P4 superior à daquelas com uma ovulação (5,92 ± 0,59 vs 7,04 ± 0,79 ng/mL). Esses resultados indicam que a dinâmica luteal em caprinos da raça Toggenburg segue padrões semelhantes aos observados em outras raças e em outras espécies e que o crescimento de tecido luteal refletiu positivamente na funcionalidade do corpo lúteo
