    Activity of secondary bacterial metabolites in the control of citrus canker.

    This study investigated the protective effects of secondary bacterial metabolites, produced by Pseudomonas sp. (bacterium strain LN), on citrus canker disease caused by Xanthomonas axonopodis pv. citri (Xac 306). The LN bacteria strain was cultured in liquid medium and the supernatant free-cells was treated with methanol (AMF) and ethyl acetate (AEF), respectively, and then the extract was concentrated, filtrated, lyophilized and fractionated by vacuum liquid chromatography (VLC). After VLC, eight fractions were obtained. All fractions? activity against Xac 306 by agar well diffusion assay and minimum inhibitory concentration but in different concentrations were tested. Cytotoxicity effects were observed in all fractions in 50 ?g?mL?1 concentration. The comet assay demonstrated that the fractions EAF, VLC2 and VLC3 presented no genotoxic effects at tested concentrations. In plants only VLC3 showed significant results (p < 0.05), reducing the incidence of citrus canker lesions

    Summary: Coffee is one of the world?s most traded agricultural products, and its production could be threatened by global warming. The aim of this work was to evaluate the effects of heat on photosynthetic activity and thylakoid membrane lipid dynamics, on genotypes of the two major coffee producing species. Potted plants from C. arabica L. cv. IPR108 and C. canephora Pierre Ex A. Froehner cv. Conilon Clone 153 were grown for 1 year under controlled conditions of temperature (25/20ÂșC, day/night), irradiance (650-800 ?mol m-2 s-1), RH (75%), photoperiod (12 h), and 380 ?L CO2 L-1. Thereafter, temperature was gradually raised to 42/34ÂșC (0.5ÂșC/ day), with a 7 days stabilization step at 31, 37 and 42ÂșC. Studies focused modifications of thylakoid lipid composition and photosynthetic performance. In CL153 photosynthetic capacity (Amax) was not affected until 42ÂșC (40% reduction). In IPR108 it was reduced 35 and 57% at 37ÂșC at 42ÂșC, respectively. Thylakoid electron transport rate for photosystems (PS) I and II increased (ca. 10-25%) up to 37ÂșC in both genotypes. At 42ÂșC only IPR108 presented depressed activities on PSII (15%) and PSI (18%). Under 37 and 42ÂșC, CL153 plants presented digalactosyldiacylglycerol (DGDG) (ca. 42%) and monogalactosyldiacylglycerol (MGDG) (28-34%) increases, while IPR108 showed higher MGDG at all temperatures. In CL153 less unsaturated DGDG and phosphatidylglycerol (PG) along with stable or increased DGDG/MGDG ratio, may have contributed to sustain thylakoid electron flow at 37ÂșC and even 42ÂșC. IPR108 displayed a strong PG rise at all temperatures, in accordance with enhanced PSs activity

    On the distinctive call of a threatened phenotype of Allobates femoralis (Anura: Aromobatidae) and its recognition by allopatric conspecific males

    ABSTRACT The brilliant-thighed frog [Allobates femoralis (Boulenger, 1884)]; is distributed across the Amazon basin and aggregates several allopatric evolutionary lineages, some of which present variation in their advertisement calls. In 2009, an unregistered call phenotype was discovered in the region of Altamira and VitĂłria do Xingu, State of ParĂĄ, Brazil, where males emit advertisement calls formed by six notes, differing from the typical four-note calls described for other A. femoralis populations. In this study, we describe in detail these untypical calls. Additionally, we test whether the aggressive responses of males of a 4-note reference population (Reserva Ducke - RFAD, in Manaus, State of Amazonas) is differential towards the 6-note calls of males recorded in Altamira (ParĂĄ State), and towards 4-note calls recorded in one location at the TapajĂłs-Xingu interfluve (Belterra, ParĂĄ State), and in RFAD. Playback experiments were conducted between 2011-2012, and used standardized stimuli produced from natural call recordings. A total of 30 independent experiments were conducted, 10 for each stimuli class. We measured the phonotaxis of focal males in relation to the loudspeaker, considering the time to orientation and the time to approach the loudspeaker. We found that not all A. femoralis males at RFAD promptly recognize calls from males recorded in Altamira. However, when considering only males who approached the loudspeaker, differences in aggressive reactions were not seen between stimuli classes. Our findings show that the ability to recognize calls from Altamira as belonging to co-specific males is not universal among males at RFAD. The new A. femoralis phenotype occurs in areas potentially impacted by the Belo Monte hydroelectric complex and complementary studies indicate that no gene flow exists between this group and A. femoralis from adjacent regions. Hence, developments in Altamira may put this incipient speciation process at risk

    Development of the preterm gut microbiome in twins at risk of necrotising enterocolitis and sepsis

    The preterm gut microbiome is a complex dynamic community influenced by genetic and environmental factors and is implicated in the pathogenesis of necrotising enterocolitis (NEC) and sepsis. We aimed to explore the longitudinal development of the gut microbiome in preterm twins to determine how shared environmental and genetic factors may influence temporal changes and compared this to the expressed breast milk (EBM) microbiome. Stool samples (n = 173) from 27 infants (12 twin pairs and 1 triplet set) and EBM (n = 18) from 4 mothers were collected longitudinally. All samples underwent PCR-DGGE (denaturing gradient gel electrophoresis) analysis and a selected subset underwent 454 pyrosequencing. Stool and EBM shared a core microbiome dominated by Enterobacteriaceae, Enterococcaceae, and Staphylococcaceae. The gut microbiome showed greater similarity between siblings compared to unrelated individuals. Pyrosequencing revealed a reduction in diversity and increasing dominance of Escherichia sp. preceding NEC that was not observed in the healthy twin. Antibiotic treatment had a substantial effect on the gut microbiome, reducing Escherichia sp. and increasing other Enterobacteriaceae. This study demonstrates related preterm twins share similar gut microbiome development, even within the complex environment of neonatal intensive care. This is likely a result of shared genetic and immunomodulatory factors as well as exposure to the same maternal microbiome during birth, skin contact and exposure to EBM. Environmental factors including antibiotic exposure and feeding are additional significant determinants of community structure, regardless of host genetics

    HIV-1-Transmitted Drug Resistance and Transmission Clusters in Newly Diagnosed Patients in Portugal Between 2014 and 2019

    Objective: To describe and analyze transmitted drug resistance (TDR) between 2014 and 2019 in newly infected patients with HIV-1 in Portugal and to characterize its transmission networks. Methods: Clinical, socioepidemiological, and risk behavior data were collected from 820 newly diagnosed patients in Portugal between September 2014 and December 2019. The sequences obtained from drug resistance testing were used for subtyping, TDR determination, and transmission cluster (TC) analyses. Results: In Portugal, the overall prevalence of TDR between 2014 and 2019 was 11.0%. TDR presented a decreasing trend from 16.7% in 2014 to 9.2% in 2016 (p for-trend = 0.114). Multivariate analysis indicated that TDR was significantly associated with transmission route (MSM presented a lower probability of presenting TDR when compared to heterosexual contact) and with subtype (subtype C presented significantly more TDR when compared to subtype B). TC analysis corroborated that the heterosexual risk group presented a higher proportion of TDR in TCs when compared to MSMs. Among subtype A1, TDR reached 16.6% in heterosexuals, followed by 14.2% in patients infected with subtype B and 9.4% in patients infected with subtype G. Conclusion: Our molecular epidemiology approach indicates that the HIV-1 epidemic in Portugal is changing among risk group populations, with heterosexuals showing increasing levels of HIV-1 transmission and TDR.     A large outbreak of Legionnaires’ Disease in an industrial town in Portugal

    Background We describe the investigation and control of an outbreak of Legionnaires’ disease in Portugal in October, November and December 2014. Methods Confirmed cases were individuals with pneumonia, laboratory evidence of Legionella pneumophila serogroup 1 and exposure, by residence, occupational or leisure to the affected municipalities. 49 possible sources were reduced to four potential sources, all industries with wet cooling system, following risk assessment. We geo-referenced cases’ residences and the location of cooling towers defining four study areas 10 km buffer centered on each cooling tower system. We compared the number of cases with expected numbers, calculated from the outbreak's attack rates applied to 2011 census population. Using Stones’ Test, we tested observed to expected ratios for decline in risk, with distance up to 10 km four directions. Isolates of Legionella pneumophila were compared using molecular methods. Results We identified 403 cases, 377 of which were confirmed, 14 patients died. Patients became ill between 14 October and 2 December. A NE wind and thermal inversion were recorded during the estimated period of exposure. Disease risk was highest in people living south west from all of the industries identified and decreased with distance (p < 0.001). 71 clinical isolates demonstrated an identical SBT profile to an isolate from a cooling tower. Whole genome sequencing identified an unusual L. pneumophila subsp. fraseri serogroup 1 as the outbreak causative strain, and confirmed isolates’ relatedness. Conclusions Industrial wet cooling systems, bacteria with enhanced survival characteristics and a combination of climatic conditions contributed to the second largest outbreak of Legionnaires’ disease recorded internationally.info:eu-repo/semantics/publishedVersio

    Teacher training in the field of health promotion: a proposal for International collaboration and preparation of a symposium for the 20th IUHPE World Conference

    Schools are considered to be settings for both health education and health promotion. But the core business of schools is actually focussed on educational outcomes, not reducing health problems. In most countries, schools give low priority to health promotion and school staffs, mainly teachers, are not aware of their role in health promotion. Studies show that teachers who have received health promotion training tend to be involved more frequently in health promotion projects and have a more comprehensive approach to health education. Pre-service and In-service staff training is then a main challenge. That’s the reason why we have launched an initiative to join international forces to strengthen and advocate for teacher training in health promotion. The main goals are: develop research, affirm and reinforce the work done in teacher training in health promotion, support the institutes/colleges/universities in the provision of initial and in-service teacher training and stimulate international partnership work.LIBEC/CIFPEC - unidade de investigação 16/644 da FCT
