    Use of Aspergillus niger for improving the feeding value of rice offal

    Undegraded and degraded rice offals (RO) were used to compound rations for broiler birds for eight weeks. The undegraded was used at 7% inclusion level and the degraded was used at 3, 5 and 7% inclusion levels. A hundred and sixty five (165) day old chicks were randomly selected and allotted to five (5) treatments. Each treatment had three replicates and each replicate had eleven birds. Aspergillus niger was inoculated into RO through solid state fermentation for a period of seven (7) days and this was used as the degraded sample. There was improvement in the crude protein and ether extract of the degraded sample, while the crude fibre content reduced. The crude protein increased from 8.79 to 12.24% while the crude fibre reduced from 15.89 to 10.82% after biodegradation. The results from the feeding trials showed significant (P< 0.05) differences in the average feed intake and average body weight gain at the starter phase. The highest feed consumption was found in treatment 5 which contained 7% degraded RO (DRO) and it was 65.11 g/b/d. At the finisher phase, there were significant (P<0.05) differences in both the average feed intake and the average body weight gain of the birds. The highest feed intake was found in treatment 5 and it was 201.84 g/b/d. The highest body weight gain was also found in treatment 5 and it was 75.52 g/b/d. The relative cost benefit (RCB) showed that economically, the birds placed on degraded rice offal (DRO) gave better profit margin than the ones placed on the undegraded rice offal (URO).     Genetic variability and heritability studies of some reproductive traits in cowpea (Vigna unguiculate (L.) Walp.)

    The success of most crop improvement programs largely depends upon the genetic variability and the heritability of desirable traits. The magnitude and type of genetic variability help the breeder to determine the selection criteria and breeding schemes to be used for improvement purposes. A screen house experiment was carried out at Samaru, Nigeria in 1999 and 2000 dry seasons to estimate the genotypic variability of some reproductive traits and their heritability in some selected cowpea varieties. Results of the study showed that there was considerable variation among cultivars for duration of reproductive phase and rate of photosynthate partitioning. Genotypic coefficients of variation were also high for days to first flower, 100-seed weight, plant height, and harvest index. Broad-sense heritability estimate (h2) was 98.9% for 100-seed weight, 94% for duration of reproductive phase, 84.5% for days to first flower, 83.9% for days to maturity, and 77.3% for harvest index. This information showed that there is sufficient genetic variance to warrant selection for improvement in the cowpea genotypes studied. We concluded that considerable progress in cowpea breeding could be achieved by exploiting these traits

    Vitamin C inhibits platelet expression of CD40 ligand

    Upon stimulation with agonists, platelets express CD40 ligand (CD40L), a transmembrane protein implicated in the initiation and progression of atherosclerotic disease. We have recently discovered that oxidative stress plays a major role in platelet CD40L expression. In this study, we sought to determine whether vitamin C, a known antioxidant, is able to influence platelet CD40L expression. In vitro experiments were done by stimulating platelets with collagen in the presence or absence of vitamin C (50-100 mu M) or vehicle as control. An in vivo study was done in 10 healthy subjects who were randomized to intravenous infusion of placebo or 1 g vitamin C for 45 min in a crossover design. At the end of infusion platelet CD40L and O2- were measured. The in vitro study demonstrated that vitamin C dose dependently inhibited platelet CD40L expression without affecting agonist-induced platelet aggregation. In subjects treated with placebo no changes of platelet CD40L and O2- were observed; conversely, vitamin C infusion caused a significant and parallel decrease of platelet O2- (-70%, P < 0.001) and CD40L (-68%, P < 0.001). Platelet aggregation was not modified by either treatment. This study suggests that water-soluble antioxidants, which scavenge superoxide radicals, may reduce platelet CD40L expression.     Mindfulness-based interventions in epilepsy: a systematic review

    Mindfulness based interventions (MBIs) are increasingly used to help patients cope with physical and mental long-term conditions (LTCs). Epilepsy is associated with a range of mental and physical comorbidities that have a detrimental effect on quality of life (QOL), but it is not clear whether MBIs can help. We systematically reviewed the literature to determine the effectiveness of MBIs in people with epilepsy. Medline, Cochrane Central Register of Controlled Trials, EMBASE, CINAHL, Allied and Complimentary Medicine Database, and PsychInfo were searched in March 2016. These databases were searched using a combination of subject headings where available and keywords in the title and abstracts. We also searched the reference lists of related reviews. Study quality was assessed using the Cochrane Collaboration risk of bias tool. Three randomised controlled trials (RCTs) with a total of 231 participants were included. The interventions were tested in the USA (n = 171) and China (Hong Kong) (n = 60). Significant improvements were reported in depression symptoms, quality of life, anxiety, and depression knowledge and skills. Two of the included studies were assessed as being at unclear/high risk of bias - with randomisation and allocation procedures, as well as adverse events and reasons for drop-outs poorly reported. There was no reporting on intervention costs/benefits or how they affected health service utilisation. This systematic review found limited evidence for the effectiveness of MBIs in epilepsy, however preliminary evidence suggests it may lead to some improvement in anxiety, depression and quality of life. Further trials with larger sample sizes, active control groups and longer follow-ups are needed before the evidence for MBIs in epilepsy can be conclusively determined

    Bacterial membrane vesicles transport their DNA cargo into host cells

    © 2017 The Author(s). Bacterial outer membrane vesicles (OMVs) are extracellular sacs containing biologically active products, such as proteins, cell wall components and toxins. OMVs are reported to contain DNA, however, little is known about the nature of this DNA, nor whether it can be transported into host cells. Our work demonstrates that chromosomal DNA is packaged into OMVs shed by bacteria during exponential phase. Most of this DNA was present on the external surfaces of OMVs, with smaller amounts located internally. The DNA within the internal compartments of Pseudomonas aeruginosa OMVs were consistently enriched in specific regions of the bacterial chromosome, encoding proteins involved in virulence, stress response, antibiotic resistance and metabolism. Furthermore, we demonstrated that OMVs carry DNA into eukaryotic cells, and this DNA was detectable by PCR in the nuclear fraction of cells. These findings suggest a role for OMV-associated DNA in bacterial-host cell interactions and have implications for OMV-based vaccines

    CD98hc facilitates B cell proliferation and adaptive humoral immunity.

    The proliferation of antigen-specific lymphocytes and resulting clonal expansion are essential for adaptive immunity. We report here that B cell-specific deletion of the heavy chain of CD98 (CD98hc) resulted in lower antibody responses due to total suppression of B cell proliferation and subsequent plasma cell formation. Deletion of CD98hc did not impair early B cell activation but did inhibit later activation of the mitogen-activated protein kinase Erk1/2 and downregulation of the cell cycle inhibitor p27. Reconstitution of CD98hc-deficient B cells with CD98hc mutants showed that the integrin-binding domain of CD98hc was required for B cell proliferation but that the amino acid-transport function of CD98hc was dispensable for this. Thus, CD98hc supports integrin-dependent rapid proliferation of B cells. We propose that the advantage of adaptive immunity favored the appearance of CD98hc in vertebrates

    Optimal iterative methods for finding multiple roots of nonlinear equations using free parameters

    [EN] In this paper, we propose a family of optimal eighth order convergent iterative methods for multiple roots with known multiplicity with the introduction of two free parameters and three univariate weight functions.     Phospholipase C-eta enzymes as putative protein kinase C and Ca2+ signalling components in neuronal and neuroendocrine tissues

    Phosphoinositol-specific phospholipase C enzymes (PLCs) are central to inositol lipid signalling pathways, facilitating intracellular Ca2+ release and protein kinase C activation. A sixth class of phosphoinositol-specific PLC with a novel domain structure, PLC-eta (PLCeta) has recently been discovered in mammals. Recent research, reviewed here, shows that this class consists of two enzymes, PLCeta1 and PLCeta2. Both enzymes hydrolyze phosphatidylinositol 4,5-bisphosphate and are more sensitive to Ca2+ than other PLC isozymes and are likely to mediate G-protein-coupled receptor (GPCR) signalling pathways. Both enzymes are expressed in neuron-enriched regions, being abundant in the brain. We demonstrate that they are also expressed in neuroendocrine cell lines. PLCeta enzymes therefore represent novel proteins influencing intracellular Ca2+ dynamics and protein kinase C activation in the brain and neuroendocrine systems as putative mediation of GPCR regulation
