    <p>Abstract</p> <p>Background</p> <p>High throughput pyrosequencing (454 sequencing) is the major sequencing platform for producing long read high throughput data. While most other sequencing techniques produce reading errors mainly comparable with substitutions, pyrosequencing produce errors mainly comparable with gaps. These errors are less efficiently detected by most conventional alignment programs and may produce inaccurate alignments.</p> <p>Results</p> <p>We suggest a novel algorithm for calculating the optimal local alignment which utilises flowpeak information in order to improve alignment accuracy. Flowpeak information can be retained from a 454 sequencing run through interpretation of the binary SFF-file format. This novel algorithm has been implemented in a program named FAAST (Flow-space Assisted Alignment Search Tool).</p> <p>Conclusions</p> <p>We present and discuss the results of simulations that show that FAAST, through the use of the novel algorithm, can gain several percentage points of accuracy compared to Smith-Waterman-Gotoh alignments, depending on the 454 data quality.     Draft Genome Sequencing of Giardia intestinalis Assemblage B Isolate GS: Is Human Giardiasis Caused by Two Different Species?

    Giardia intestinalis is a major cause of diarrheal disease worldwide and two major Giardia genotypes, assemblages A and B, infect humans. The genome of assemblage A parasite WB was recently sequenced, and the structurally compact 11.7 Mbp genome contains simplified basic cellular machineries and metabolism. We here performed 454 sequencing to 16× coverage of the assemblage B isolate GS, the only Giardia isolate successfully used to experimentally infect animals and humans. The two genomes show 77% nucleotide and 78% amino-acid identity in protein coding regions. Comparative analysis identified 28 unique GS and 3 unique WB protein coding genes, and the variable surface protein (VSP) repertoires of the two isolates are completely different. The promoters of several enzymes involved in the synthesis of the cyst-wall lack binding sites for encystation-specific transcription factors in GS. Several synteny-breaks were detected and verified. The tetraploid GS genome shows higher levels of overall allelic sequence polymorphism (0.5 versus <0.01% in WB). The genomic differences between WB and GS may explain some of the observed biological and clinical differences between the two isolates, and it suggests that assemblage A and B Giardia can be two different species

    School Effects on the Wellbeing of Children and Adolescents

    Well-being is a multidimensional construct, with psychological, physical and social components. As theoretical basis to help understand this concept and how it relates to school, we propose the Self-Determination Theory, which contends that self-determined motivation and personality integration, growth and well-being are dependent on a healthy balance of three innate psychological needs of autonomy, relatedness and competence. Thus, current indicators involve school effects on children’s well-being, in many diverse modalities which have been explored. Some are described in this chapter, mainly: the importance of peer relationships; the benefits of friendship; the effects of schools in conjunction with some forms of family influence; the school climate in terms of safety and physical ecology; the relevance of the teacher input; the school goal structure and the implementation of cooperative learning. All these parameters have an influence in promoting optimal functioning among children and increasing their well-being by meeting the above mentioned needs. The empirical support for the importance of schools indicates significant small effects, which often translate into important real-life effects as it is admitted at present. The conclusion is that schools do make a difference in children’s peer relationships and well-being

    Coordinating knowledge creation: A systematic literature review on the interplay between operational excellence and industry 4.0 technologies

    In the process of creating new knowledge, literature has scarcely studied how bodies of knowledge arising from different sources should be coordinated to enhance performance. In particular, the present research focuses on two sources of newly created knowledge, i.e., operational excellence and Industry 4.0, to understand whether they should be implemented sequentially or simultaneously. Operational excellence refers to the implementation of practices such as just in time, total quality management, and Six Sigma that help a firm to create knowledge that facilitates waste reduction and customer value improvement. Industry 4.0 refers to the implementation of new technologies such as artificial intelligence, big data, robotics, Internet of Things, and laser cutting that help a firm to create knowledge to improve overall business performance. We identified and analyzed 30 papers published in 13 peer-reviewed journals and conference proceedings in the field of operations management. Our findings based on the systematic literature review suggest that the interplay between operational excellence and Industry 4.0 can be categorized into four groups: (1) Industry 4.0 supports operational excellence; (2) operational excellence supports Industry 4.0; (3) complementary; and (4) no interdependence. Majority of the papers under study are in the first category, suggesting Industry 4.0 technologies as enabler of operational excellence
