    Matrix-assisted laser desorption/ionization time of flight mass spectrometry identification of Vibrio (Listonella) anguillarum isolated from sea bass and sea bream

    Vibrio (Listonella) anguillarum is a pathogenic bacterium causing septicaemia in a wide range of marine organisms and inducing severe mortalities, thus it is crucial to conduct its accurate and rapid identification. The aim of this study was to assess MALDI-TOF MS as a method of choice for identification of clinical V. anguillarum isolates from affected marine fish. Since the method accuracy might be influenced by the type of the medium used, as well as by the incubation conditions, we tested V. anguillarum isolates grown on standard media with and without the addition of NaCl, cultured at three incubation temperatures, and at three incubation periods. The best scores were retrieved for V. anguillarum strains grown on NaCl-supplemented tryptone soy agar (TSA) at 22°C and incubated for 48h (100% identification to species level; overall score 2.232), followed by incubation at 37°C and 48h (100% to species level; score 2.192). The strains grown on non-supplemented TSA gave the best readings when incubated at 22°C for 72h (100% identification to species level; overall score 2.182), followed by incubation at 15°C for 72h (100% to species level; score 2.160). Unreliable identifications and no-identifications were growing with the incubation duration at 37°C, on both media, amounting to 88.89% for 7d incubation on supplemented TSA, and 92.60% for 7d incubation on non-supplemented TSA. The age of the cultured strains and use of media significantly impacted the mass spectra, demonstrating that for reliable identification, MALDI-TOF MS protein fingerprinting with the on-target extraction should be performed on strains grown on a NaCl-supplemented medium at temperatures between 15 and 22°C, incubated for 48-72 hours

Journal of Consumer Behaviour, 19(3), 277-290. https://doi.org/10.1002/cb.1816By analyzing three experimental studies, this research tests how and when sophisticated service environment designs (compared to modest service designs) can minimize consumers' negative emotions and increase repurchase intentions after a failure. Drawing on part-list cueing literature, this research proposes that when a service failure occurs in a sophisticated (vs. modest) environment, consumers will rely on the sophisticated style of design as cues for service quality. We argue that sophisticated (vs. modest) service designs work as strong cues for quality that restrict the retrieval of negative information by consumers and can minimize the negative impacts of service failure, reducing consumers' negative emotions and increasing repurchase intentions. We further advance our theorizing by showing how choice failure consequences (i.e., the risk or consequence related to the service choice) moderate the effects via associative pathways of retrieval. The findings contribute to theory and practice by revealing how service designs can serve as cues to mitigate adverse consequences of service failure.authorsversionpublishe

    As demandas atuais na prestação de serviços de educação exigem que as Instituições de Ensino Superior apresentem critérios de gestão bem definidos. Neste contexto, o presente estudo tem por objetivo desenvolver um modelo de gestão de egressos aplicável às Faculdades Integradas Promove de Brasília. Foi realizado um levantamento de dados por meio de pesquisa bibliográfica gerando um Modelo de Gestão com instrumentos de execução para cada uma de suas etapas. Em seguida, um Grupo de Trabalho (GT) formado por alunos egressos avaliou o modelo elaborado com base em sua realidade acadêmica. Os resultados demonstraram que a implantação de um modelo de gestão de egressos é de suma importância para a realidade da IES, e que as proposições da teoria e modelos de gestão já aplicados em outras instituições pesquisadas refletem a realidade do ambiente deste estudo. Por fim, observou-se que a implantação de um modelo de gestão de egressos proporciona a IES uma interação contínua com seu alunado. Palavras-chaves: Gestão de Pessoas. Egressos.     Strength and thermal stability of Cu-Al2O3 composite obtained by internal oxidation

    The objective of the work is to study the effects of the high-energy milling on strengthening, thermal stability and electrical conductivity of Cu-Al2O3 composite. The prealloyed copper powders, atomized in inert gas and containing 3 wt. % Al, were milled up to 20 h in the planetary ball mill to oxidize in situ aluminium with oxygen from the air. Composite compacts were obtained by hot-pressing in an argon atmosphere at 800 degrees C for 3 h under the pressure of 35 MPa. The microstructural characterization was performed by the optical microscope, scanning electron microscope (SEM), transmission electron microscope (TEM) and X-ray diffraction analysis (XRD). The microhardness, electrical conductivity and density measurements were also carried out. The effect of internal oxidation and high-energy milling on strengthening of Cu-Al2O3 composite was significant, The increase of the microhardness of composite compacts (292 HV) is almost threefold comparing to compacts processed from the as-received Cu-3 wt. % Al powder (102 HV). The grain size of Cu-Al2O3 compacts processed from 5 and 20 h-milled powders was 75 and 45 nm, respectively. The small increase in the grain size and the small microhardness drop indicate the high thermal stability of Cu-Al2O3 composite during high-temperature exposure at 800 degrees C

    Using Soil and Water Conservation Contests for Extension: Experiences from the Bolivian Mountain Valleys

    Soil and water conservation (SWC) contests among farmer groups were organized in five rural villages in the Bolivian mountain valleys. The contests were aimed at quickly achieving widespread sustainable results. This article analyzes the effectiveness of these contests as an extension tool. Mixed results were obtained. In three villages, participation rates in the SWC activities introduced in the contests were still high even 2 years after project withdrawal. These were all villages where a solid foundation for sustainable development had been laid before the contests were held. Two years later, most families were still involved in maintenance of the SWC practices introduced in the contests, and many farmers had started to experiment with different soil management practices. However, replications of these SWC practices were not widespread, Conservation Leaders did not continue with their training activities, and the quality of maintenance of the practices was often not satisfactory. In order to become a more effective extension tool and achieve widespread impact, SWC contests must receive continued support by a catalyst agency. Moreover, other SWC contests should also be organized in which practices are not predefined. Given that SWC contests are a low-budget extension tool, local municipalities could become more actively involved

    Enhanced mercury reduction in the South Atlantic Ocean during carbon remineralization

    Highlights • Dissolved gaseous mercury can be calculated from modeled dissolved inorganic carbon. • Modeled dissolved gaseous mercury agrees well with worldwide observations. • Dissolved gaseous mercury is related to depth and macronutrients concentrations. Mercury (Hg) in seawater is subject to interconversions via (photo)chemical and (micro)biological processes that determine the extent of dissolved gaseous mercury (DGM) (re)emission and the production of monomethylmercury. We investigated Hg speciation in the South Atlantic Ocean on a GEOTRACES cruise along a 40°S section between December 2011 and January 2012 (354 samples collected at 24 stations from surface to 5250 m maximum depth). Using statistical analysis, concentrations of methylated mercury (MeHg, geometric mean 35.4 fmol L−1) were related to seawater temperature, salinity, and fluorescence. DGM concentrations (geometric mean 0.17 pmol L−1) were related to water column depth, concentrations of macronutrients and dissolved inorganic carbon (DIC). The first-ever observed linear correlation between DGM and DIC obtained from high-resolution data indicates possible DGM production by organic matter remineralization via biological or dark abiotic reactions. DGM concentrations projected from literature DIC data using the newly discovered DGM–DIC relationship agreed with published DGM observations

    Financial market development, global financial crisis and economic growth : evidence from developing nations

    Emerging and frontier markets in Africa have witnessed various economic and financial reforms aimed at integrating the domestic markets into the global financial market to attract investment. Whether these reforms promote high economic growth remains inconclusive. The paper applies the pooled mean group estimation technique to empirically re-investigate the link between financial market development, global financial crisis, and economic growth in selected African economies. The results strongly support our hypotheses that stock market and banking sector development promotes economic growth in the selected countries. Moreover, financial crisis reduce the positive effects of both the stock market and banking sector developments on economic growth. The study suggests that both the banking sector and stock market are important to deliver the long-run economic growth that the African region desired.     Spatial Distribution of the Pathways of Cholesterol Homeostasis in Human Retina

    The retina is a light-sensitive tissue lining the inner surface of the eye and one of the few human organs whose cholesterol maintenance is still poorly understood. Challenges in studies of the retina include its complex multicellular and multilayered structure; unique cell types and functions; and specific physico-chemical environment.We isolated specimens of the neural retina (NR) and underlying retinal pigment epithelium (RPE)/choroid from six deceased human donors and evaluated them for expression of genes and proteins representing the major pathways of cholesterol input, output and regulation. Eighty-four genes were studied by PCR array, 16 genes were assessed by quantitative real time PCR, and 13 proteins were characterized by immunohistochemistry. Cholesterol distribution among different retinal layers was analyzed as well by histochemical staining with filipin. Our major findings pertain to two adjacent retinal layers: the photoreceptor outer segments of NR and the RPE. We demonstrate that in the photoreceptor outer segments, cholesterol biosynthesis, catabolism and regulation via LXR and SREBP are weak or absent and cholesterol content is the lowest of all retinal layers. Cholesterol maintenance in the RPE is different, yet the gene expression also does not appear to be regulated by the SREBPs and varies significantly among different individuals.This comprehensive investigation provides important insights into the relationship and spatial distribution of different pathways of cholesterol input, output and regulation in the NR-RPE region. The data obtained are important for deciphering the putative link between cholesterol and age-related macular degeneration, a major cause of irreversible vision loss in the elderly
