    'Word from the street' : when non-electoral representative claims meet electoral representation in the United Kingdom

    Taking the specific case of street protests in the UK – the ‘word from the street’– this article examines recent (re)conceptualizations of political representation, most particularly Saward’s notion of ‘representative claim’. The specific example of nonelectoral claims articulated by protestors and demonstrators in the UK is used to illustrate: the processes of making, constituting, evaluating and accepting claims for and by constituencies and audiences; and the continuing distinctiveness of claims based upon electoral representation. Two basic questions structure the analysis: first, why would the political representative claims of elected representatives trump the nonelectoral claims of mass demonstrators and, second, in what ways does the ‘perceived legitimacy’ of the former differ from the latter

    Dual Requirement for Yeast hnRNP Nab2p in mRNA poly(A) Tail Length Control and Nuclear Export

    Recent studies of mRNA export factors have provided additional evidence for a mechanistic link between mRNA 3′‐end formation and nuclear export. Here, we identify Nab2p as a nuclear poly(A)‐binding protein required for both poly(A) tail length control and nuclear export of mRNA. Loss of NAB2 expression leads to hyperadenylation and nuclear accumulation of poly(A)+ RNA but, in contrast to mRNA export mutants, these defects can be uncoupled in a nab2 mutant strain. Previous studies have implicated the cytoplasmic poly(A) tail‐binding protein Pab1p in poly(A) tail length control during polyadenylation. Although cells are viable in the absence of NAB2 expression when PAB1 is overexpressed, Pab1p fails to resolve the nab2Δ hyperadenylation defect even when Pab1p is tagged with a nuclear localization sequence and targeted to the nucleus. These results indicate that Nab2p is essential for poly(A) tail length control in vivo, and we demonstrate that Nab2p activates polyadenylation, while inhibiting hyperadenylation, in the absence of Pab1p in vitro.     The growing demand of ecosystem services provided by forests increased the need for fast and accurate field survey. The recent technological innovations fostered the application of geomatic tools and processes to different fields of the forestry sector. In this study we compared the efficiency and the accuracy of Mobile Laser Scanner (MLS), combined with Simultaneous Localization and Mapping (SLAM) technology, and traditional field survey for the mensuration of main forest dendrometric variables like stem diameter at breast height (DBH), individual tree height (H), crown base height (CBH) and branch-free stem volume (VOL). With ground truth measurements taken from 50 felled trees, we tested the applicability of MLS technology for individual tree parameters esti-mation in a conifer plantation in central Italy. Our results showed no bias of DBH estimates and the corre-sponding RMSE was equal to 10.8% (2.7 cm). H and CBH measured with MLS were underestimated compared to the ground truth (bias of-8.6% for H and-13.3% for CBH). VOL values showed a bias and a RMSE of-4.1% (-0.01 m(3)) and 12.4% (0.04 m3) respectively. Tree height is not perfectly estimated due to laser obstruction by crowns layer, but the acquisition speed of this survey, joined with a suitable accuracy of parameters extraction, suggests sufficient suitability of the method for operational applications in simple forest structures (e.g. one-layered stands)

    Cryopreservation of hazelnut (Corylus avellana l.) axillary buds from in vitro shoots using the droplet vitrification method

    Cryopreservation by droplet vitrification was applied to hazelnut (Corylus avellana L.). axillary buds of the Italian cultivated variety Tonda Gentile Romana, which were collected from in vitro growing shoots, immersed in ice cooled PVS2 or PVS3 for 60 or 90 min, then transferred to a droplet of vitrification solution, placed on a strip of aluminium foil, and plunged into liquid nitrogen (LN). Additionally, the effect on the recovery of the mother plant after cryopreservation was evaluated, following a cold pre-treatment at 4◦C for 3 months. The highest regrowth percentage (56.7%) was obtained after applying PVS3 for 60 min, while the application of PVS2 for the same amount of time reduced regrowth to 41.5%. Increasing the exposure to vitrification solutions to 90 min reduced regrowth to 43.3% when PVS3 was applied, and 35.6% if PVS2 was used. The cold pre-treatment on the mother plant did not significantly improve overall regrowth. The cryopreservation process did not decline the rooting ability of the recovered shoot

    The 3′ Region of the Chicken Hypersensitive Site-4 Insulator Has Properties Similar to Its Core and Is Required for Full Insulator Activity

    Chromatin insulators separate active transcriptional domains and block the spread of heterochromatin in the genome. Studies on the chicken hypersensitive site-4 (cHS4) element, a prototypic insulator, have identified CTCF and USF-1/2 motifs in the proximal 250 bp of cHS4, termed the “core”, which provide enhancer blocking activity and reduce position effects. However, the core alone does not insulate viral vectors effectively. The full-length cHS4 has excellent insulating properties, but its large size severely compromises vector titers. We performed a structure-function analysis of cHS4 flanking lentivirus-vectors and analyzed transgene expression in the clonal progeny of hematopoietic stem cells and epigenetic changes in cHS4 and the transgene promoter. We found that the core only reduced the clonal variegation in expression. Unique insulator activity resided in the distal 400 bp cHS4 sequences, which when combined with the core, restored full insulator activity and open chromatin marks over the transgene promoter and the insulator. These data consolidate the known insulating activity of the canonical 5′ core with a novel 3′ 400 bp element with properties similar to the core. Together, they have excellent insulating properties and viral titers. Our data have important implications in understanding the molecular basis of insulator function and design of gene therapy vectors

    Penelitian ini dilatarbelakangi oleh kurangnya pemahaman, penerimaan, dan sikap orang tua terhadap hambatan yang dihadapi anak. Hal ini berdampak pada kemampuan keluarga dalam memberikan layanan intervensi dini di rumah. Tujuan dari penelitian ini adalah untuk merumuskan program intervensi dini bersumber daya keluarga yang akan menjadi pedoman orang tua dalam melaksanakan intervensi. Penelitian ini menggunakan pendekatan kualitatif dengan metode deskriptif. Subjek dalam penelitian ini adalah satu keluarga yang memiliki anak Cerebral Palsy dengan hambatan komunikasi di Ciamis. Teknik pengumpulan data yang digunakan adalah observasi, wawancara, dan studi dokumentasi. Hasil penelitian menunjukkan bahwa: 1) anak memiliki hambatan komunikasi yang pencapaian perkembangan komunikasi verbalnya setara dengan anak usia 2-3 tahun dan usia kalender saat ini 5 tahun. 2) rendahnya tingkat pemahaman orangtua terhadap hambatan anak. 3). Rumusan program intervensi dini bersumber daya keluarga ini berdasarkan kondisi objektif serta kebutuhan keluarga dan anak. 4). tingkat pemahaman orang tua mengenai hambatan anak meningkat serta terjadi perubahan sikap dan perilaku orang tua kepada anak;--- The study was motivated by the lack of understanding, acceptance as well as the attitude of the parents toward their child’s obstacles. It somehow has an impact on family’s capacity to give early intervention support at home. This study was aimed at formulating an early intervention program to the child. The study used descriptive qualitative method involving a single family having a child suffering from Cerebral Palsy causing communication barrier at Ciamis. The techniques used in collecting the data were observations, interviews, and document analysis. The study revealed several findings as follows: 1) the child had communication barrier which made her communication verbal developmental stage was equal to 2-3 years old children. 2) The lack of understanding, acceptance and attitude of the parents toward their child’s communication barrier as well as the support in delivering early intervention at home. 3) The early intervention family-based program’s formulation was based on the objective condition as well as the needs’ of both the child and the parents. 4) The level understanding parents of the barriers to child increase and a change in attitudes and behavior of parents to child

    Cutting edge: extracellular high mobility group box-1 protein is a proangiogenic cytokine.

    The chromosomal high mobility group box-1 (HMGB1) protein acts as a proinflammatory cytokine when released in the extracellular environment by necrotic and inflammatory cells. In the present study, we show that HMGB1 exerts proangiogenic effects by inducing MAPK ERK1/2 activation, cell proliferation, and chemotaxis in endothelial cells of different origin. Accordingly, HMGB1 stimulates membrane ruffling and repair of a mechanically wounded endothelial cell monolayer and causes endothelial cell sprouting in a three-dimensional fibrin gel. In keeping with its in vitro properties, HMGB1 stimulates neovascularization when applied in vivo on the top of the chicken embryo chorioallantoic membrane whose blood vessels express the HMGB1 receptor for advanced glycation end products (RAGE). Accordingly, RAGE blockade by neutralizing Abs inhibits HMGB1-induced neovascularization in vivo and endothelial cell proliferation and membrane ruffling in vitro. Taken together, the data identify HMGB1/RAGE interaction as a potent proangiogenic stimulus
