Novel mode of ISG15-mediated protection against influenza A virus and Sendai virus in mice
ISG15 is a diubiquitin-like modifier and one of the most rapidly induced genes upon type I interferon stimulation. Hundreds of host proteins and a number of viral proteins have been shown to be ISGylated, and understanding how these modifications affect the interferon response and virus replication has been of considerable interest. ISG15(−/−) mice exhibit increased susceptibility to viral infection, and in the case of influenza B virus and vaccinia virus, ISG15 conjugation has been shown to restrict virus replication in vivo. A number of studies have also found that ISG15 is capable of antagonizing replication of some viruses in tissue culture. However, recent findings have demonstrated that ISG15 can protect mice from Chikungunya virus infection without affecting the virus burden. In order to better understand the function of ISG15 in vivo, we characterized the pathogenesis of influenza A virus and Sendai virus in ISG15(−/−) mice. We found that ISG15 protects mice from virus induced lethality by a conjugation-dependent mechanism in both of these models. However, surprisingly, we found that ISG15 had minimal effect on virus replication and did not have an obvious role in the modulation of the acute immune response to infection. Instead, we observed an increase in the number of diseased small airways in mice lacking ISG15. This ability of ISG15 to protect mice in a conjugation-dependent, but nonantiviral, manner from respiratory virus infection represents a previously undescribed role for ISG15 and demonstrates the importance of further characterization of ISG15 in vivo. IMPORTANCE It has previously been demonstrated that ISG15(−/−) mice are more susceptible to a number of viral infections. Since ISG15 is one of the most strongly induced genes after type I interferon stimulation, analysis of ISG15 function has largely focused on its role as an antiviral molecule during acute infection. Although a number of studies have shown that ISG15 does have a small effect on virus replication in tissue culture, few studies have confirmed this mechanism of protection in vivo. In these studies we have found that while ISG15(−/−) mice are more susceptible to influenza A virus and Sendai virus infections, ISGylation does not appear to mediate this protection through the direct inhibition of virus replication or the modulation of the acute immune response. Thus, in addition to showing a novel mode of ISG15 mediated protection from virus infection, this study demonstrates the importance of studying the role of ISG15 in vivo

An overview of ISCAT 2000
The Investigation of Sulfur Chemistry in the Antarctic Troposphere (ISCAT) took place over the timer period of 15 November to 31 December in the year 2000. The study location was the Amundsen Scott Station in Antarctica. ISCAT 2000 defines the second phase of a program designed to explore tropospheric chemistry in Antarctica. As in 1998, the 2000 ISCAT study revealed a strong oxidizing environment at South Pole (SP). During the 2000 investigation, however, the suite of measurements was greatly expanded. These new measurements established the recycling of reactive nitrogen as a critical component of this unique environment. This paper first presents the historical background leading up to the ISCAT 2000 observations; then it focuses on providing a summary of the year 2000 results and contrasts these with those recorded during 1998. Important developments made during the 2000 study included the recording of SP data for several species being emitted from the snowpack. These included NO, H 2O2 and CH2O. In this context, eddy-diffusion flux measurements provided the first quantitative estimates of the SP NO and NOx snow-to-atmosphere fluxes. This study also revealed that HNO 3 and HO2NO2 were major sink species for HOx and NOx radicals. And, it identified the critical factors responsible for SP NO levels exceeding those at other polar sites by nearly an order of magnitude. Finally, it reports on the levels of gas phase sulfur species and provides evidence indicating that the absence of DMS at SP is most likely due to its greatly shorten chemical lifetime in the near vicinity of the plateau. It is proposed that this is due to the influence of NO on the distribution of OH in the lower free troposphere over a region that extends well beyond the plateau itself. The energy budget in Rayleigh-Benard convection
It is shown using three series of Rayleigh number simulations of varying
aspect ratio AR and Prandtl number Pr that the normalized dissipation at the
wall, while significantly greater than 1, approaches a constant dependent upon
AR and Pr. It is also found that the peak velocity, not the mean square
velocity, obeys the experimental scaling of Ra^{0.5}. The scaling of the mean
square velocity is closer to Ra^{0.46}, which is shown to be consistent with
experimental measurements and the numerical results for the scaling of Nu and
the temperature if there are strong correlations between the velocity and

Similarity theory and calculation of turbulent fluxes at the surface for the stably stratified atmospheric boundary layers
In this paper we revise the similarity theory for the stably stratified
atmospheric boundary layer (ABL), formulate analytical approximations for the
wind velocity and potential temperature profiles over the entire ABL, validate
them against large-eddy simulation and observational data, and develop an

Ship-based measurement of air-sea CO2exchange by eddy covariance
A system for the shipboard measurement of air-sea CO2 fluxes by eddy covariance was developed and tested. The system was designed to reduce two major sources of experimental uncertainty previously reported. First, the correction for in situ water vapor fluctuations (the “Webb” correction) was reduced by 97% by drying the air sample stream. Second, motion sensitivity of the gas analyzer was reduced by using an open-path type sensor that was converted to a closed-path configuration to facilitate drying of the air stream. High-quality CO2 fluxes were obtained during 429 14 min flux intervals during two cruises in the North Atlantic. The results suggest that the gas analyzer resolved atmospheric CO2 fluctuations well below its RMS noise level. This noise was uncorrelated with the vertical wind and therefore filtered out by the flux calculation. Using climatological data, we estimate that the techniques reported here could enable high-quality measurements of air-sea CO2 flux over much of the world oceans
A chimeric virus-mouse model system for evaluating the function and inhibition of papain-like proteases of emerging coronaviruses
To combat emerging coronaviruses, developing safe and efficient platforms to evaluate viral protease activities and the efficacy of protease inhibitors is a high priority. Here, we exploit a biosafety level 2 (BSL-2) chimeric Sindbis virus system to evaluate protease activities and the efficacy of inhibitors directed against the papain-like protease (PLpro) of severe acute respiratory syndrome coronavirus (SARS-CoV), a biosafety level 3 (BSL-3) pathogen. We engineered Sindbis virus to coexpress PLpro and a substrate, murine interferon-stimulated gene 15 (ISG15), and found that PLpro mediates removal of ISG15 (deISGylation) from cellular proteins. Mutation of the catalytic cysteine residue of PLpro or addition of a PLpro inhibitor blocked deISGylation in virus-infected cells. Thus, deISGylation is a marker of PLpro activity. Infection of alpha/beta interferon receptor knockout (IFNAR−/−) mice with these chimeric viruses revealed that PLpro deISGylation activity removed ISG15-mediated protection during viral infection. Importantly, administration of a PLpro inhibitor protected these mice from lethal infection, demonstrating the efficacy of a coronavirus protease inhibitor in a mouse model. However, this PLpro inhibitor was not sufficient to protect the mice from lethal infection with SARS-CoV MA15, suggesting that further optimization of the delivery and stability of PLpro inhibitors is needed. We extended the chimeric-virus platform to evaluate the papain-like protease/deISGylating activity of Middle East respiratory syndrome coronavirus (MERS-CoV) to provide a small-animal model to evaluate PLpro inhibitors of this recently emerged pathogen. This platform has the potential to be universally adaptable to other viral and cellular enzymes that have deISGylating activities
Hysteresis phenomenon in turbulent convection
Coherent large-scale circulations of turbulent thermal convection in air have
been studied experimentally in a rectangular box heated from below and cooled
from above using Particle Image Velocimetry. The hysteresis phenomenon in
turbulent convection was found by varying the temperature difference between
the bottom and the top walls of the chamber (the Rayleigh number was changed
within the range of ). The hysteresis loop comprises the one-cell
and two-cells flow patterns while the aspect ratio is kept constant (). We found that the change of the sign of the degree of the anisotropy of
turbulence was accompanied by the change of the flow pattern. The developed
theory of coherent structures in turbulent convection (Elperin et al. 2002;
2005) is in agreement with the experimental observations. The observed coherent
structures are superimposed on a small-scale turbulent convection. The
redistribution of the turbulent heat flux plays a crucial role in the formation

CD160-Associated CD8 T-Cell Functional Impairment Is Independent of PD-1 Expression.
Expression of co-inhibitory molecules is generally associated with T-cell dysfunction in chronic viral infections such as HIV or HCV. However, their relative contribution in the T-cell impairment remains unclear. In the present study, we have evaluated the impact of the expression of co-inhibitory molecules such as 2B4, PD-1 and CD160 on the functions of CD8 T-cells specific to influenza, EBV and CMV. We show that CD8 T-cell populations expressing CD160, but not PD-1, had reduced proliferation capacity and perforin expression, thus indicating that the functional impairment in CD160+ CD8 T cells may be independent of PD-1 expression. The blockade of CD160/CD160-ligand interaction restored CD8 T-cell proliferation capacity, and the extent of restoration directly correlated with the ex vivo proportion of CD160+ CD8 T cells suggesting that CD160 negatively regulates TCR-mediated signaling. Furthermore, CD160 expression was not up-regulated upon T-cell activation or proliferation as compared to PD-1. Taken together, these results provide evidence that CD160-associated CD8 T-cell functional impairment is independent of PD-1 expression

The challenge of policy coordination for sustainable sociotechnical transitions: the case of the zero-carbon homes agenda in England
Emerging in recent research on sociotechnical transitions towards a low-carbon economy is the question of the extent to which such transitions require centralised, intentional coordination by government. Drawing from Hayek's conceptualisation of coordination, I evaluate the effectiveness of policy for low-carbon and zero-carbon homes in England. A detailed analysis is presented of how policy makers address complex choices and trade-offs as well as significant uncertainty. Particular attention is given to those policy decisions which are widely agreed by stakeholders to cause distortive effects. The focus here on the impacts of policy definition and delivery in terms of multiple evaluative criteria can complement and enrich the more process-orientated cross-sector and multilevel analyses that predominate in existing research on policy coordination. Furthermore, the coordination problems identified yield further insights into the actual and potential effectiveness of policy processes in shaping complex sociotechnical transitions
