    Oral cancer secretome: Identification of cancer-associated proteins

    This study aims to identify cancer-associated proteins in the secretome of oral cancer cell lines. We have successfully established four primary cell cultures of normal cells with a limited lifespan without human telomerase reverse transcriptase (hTERT) immortalization. The secretome of these primary cell cultures were compared with that of oral cancer cell lines using 2DE. Thirty five protein spots were found to have changed in abundance. Unambiguous identification of these proteins was achieved by MALDI TOF/TOF. In silico analysis predicted that 24 of these proteins were secreted via classical or nonclassical mechanisms. The mRNA expression of six genes was found to correlate with the corresponding protein abundance. Ingenuity Pathway Analysis (IPA) core analysis revealed that the identified proteins were relevant in, and related to, cancer development with likely involvements in tumor growth, metastasis, hyperproliferation, tumorigenesis, neoplasia, hyperplasia, and cell transformation. In conclusion, we have demonstrated that a comparative study of the secretome of cancer versus normal cell lines can be used to identify cancer-associated proteins.Article Link: http://onlinelibrary.wiley.com/doi/10.1002/elps.201300126/abstrac

    Geometric and process design of ultra-thin junctionless double gate vertical MOSFETs

    The junctionless MOSFET architectures appear to be attractive in realizing the Moore’s law prediction. In this paper, a comprehensive 2-D simulation on junctionless vertical double-gate MOSFET (JLDGVM) under geometric and process consideration was introduced in order to obtain excellent electrical characteristics. Geometrical designs such as channel length (Lch) and pillar thickness (Tp) were considered and the impact on the electrical performance was analyzed. The influence of doping concentration and metal gate work function (WF) were further investigated for achieving better performance. The results show that the shorter Lch can boost the drain current (ID) of n-JLDGVM and p-JLDGVM by approximately 68% and 70% respectively. The ID of the n-JLVDGM and p-JLVDGM could possibly boost up to 42% and 78% respectively as the Tp is scaled down from 11nm to 8nm. The channel doping (Nch) is also a critical parameter, affecting the electrical performance of both n-JLDGVM and p-JLDGVM in which 15% and 39% improvements are observed in their respective ID as the concentration level is increased from 1E18 to 9E18 atom/cm3.     Performance analysis of ultrathin junctionless double gate vertical MOSFETs

    The main challenge in MOSFET minituarization is to form an ultra-shallow source/drain (S/D) junction with high doping concentration gradient, which requires an intricate S/D and channel engineering. Junctionless MOSFET configuration is an alternative solution for this issue as the junction and doping gradients is totally eliminated. A process simulation has been developed to investigate the impact of junctionless configuration on the double-gate vertical MOSFET. The result proves that the performance of junctionless double-gate vertical MOSFETs (JLDGVM) are superior to the conventional junctioned double-gate vertical MOSFETs (JDGVM). The results reveal that the drain current (ID) of the n-JLVDGM and p-JLVDGM could be tremendously enhanced by 57% and 60% respectively as the junctionless configuration was applied to the double-gate vertical MOSFET. In addition, junctionless devices also exhibit larger ION/IOFF ratio and smaller subthreshold slope compared to the junction devices, implying that the junctionless devices have better power consumption and faster switching capability

    Determination of Leptospira borgpetersenii serovar Javanica and Leptospira interrogans serovar Bataviae as the persistent Leptospira serovars circulating in the urban rat populations in peninsular Malaysia

    Background: Leptospirosis is an emerging infectious disease of global significance, and is endemic in tropical countries, including Malaysia. Over the last decade, a dramatic increase of human cases was reported; however, information on the primary vector, the rat, and the Leptospira serovars circulating among the rat population is limited. Therefore, the present study was undertaken to isolate Leptospira and characterise the serovars circulating in the urban rat populations from selected main cities in Peninsular Malaysia. Methods: Rat trappings were carried out between October 2011 to February 2014 in five urban cities which were chosen as study sites to represent different geographical locations in Peninsular Malaysia. Microscopic agglutination test (MAT) and PCR were carried out to identify the Leptospiral serogroup and determine the pathogenic status of the isolates, respectively while pulsed-field gel electrophoresis (PFGE) and random amplified polymorphic DNA (RAPD)-PCR were used to characterize the isolates. Results: Three rat species were identified from the three hundred and fifty seven rats captured with Rattus rattus, being the dominant rat species (285, 80 %) followed by Rattus norgevicus (53, 15 %) and Rattus exulans (19, 5 %). Only 39 samples (11.0 %) were positive by culture and further confirmed as pathogenic Leptospira by PCR. Significant associations were shown between host infection with locality, season, host-age and species. Based on MAT, two serogroups were identified in the population namely; L. borgpetersenii serogroup Javanica (n = 16) and L. interrogans serogroup Bataviae (n = 23). Pulsed-field gel electrophoresis (PFGE) distinguished the two serovars in the urban rat populations: L. borgpetersenii serovar Javanica (41 %), and L. interrogans serovar Bataviae (59 %). RAPD-PCR yielded 14 distinct patterns and was found to be more discriminative than PFGE. Conclusions: This study confirms two Leptospira serovars circulating among the urban rats population in Peninsular Malaysia namely; L. borgpetersenii serovar Javanica and L. interrogans serovars Bataviae. Despite the low number of isolates obtained from the rat population, this study suggests that rodent control programs and disease surveillance may help to reduce the possible risk of disease transmission

    Susceptibility Status of The Malaria Vector Anopheles Arabiensis To Insecticides in Khartoum State, Sudan

    Background: Increasing insecticide resistancein the major anopheline vectors remain the main obstacle for malaria control programmes in African countries including Sudan.Objectives: To assess the susceptibility status of Anopheles arabiensis the malaria vector to different classes of insecticides in Khartoum State.Materials and Methods: Using WHO procedure, susceptibility tests were conducted on adults of An.arabiensis from nine sentinel sites in Khartoum State. Mortality rates and knockdown times of insecticides for An. arabiensis were calculated.Results: A total of 8345 females of An.arabiensis were tested againsteight insecticides, these wereDDT 4%, fenitrothion 1%, malathion 5%, propoxur 0.1%, permethrin 0.75%, deltamethrin 0.05% and lambdacyhalothrin 0.05%. Of these insecticides tested, An. arabiensis from Khartoum State wassusceptible to only fenitrothion 1% and lambdacyhalothrin 0.05% with overall mean percentage mortalities 99±0.12 and 100±0.45 respectively.In addition, the overall results, revealed a low mean mortality rates in An. arabiensis indicating resistance to the other remaining insecticides.Furthermore, all populations of An. arabiensis from different sentinel sites showed variation in mortality rates for the tested insecticides. The exception were for fenitrothion 1% and lambdacyhalothrin 0.05% which were resulted in 100% mean mortality in An. Arabiensis in each sentinel sites. Similarly, the knock down time (KDT) of all insecticide tested for 50% and 95% of An. arabiensis varied between populations of sentinel sites.Conclusion: In conclusion, An. arabiensis the main malariavector in Khartoum State is mainly susceptible to fenitrothion and lambdacyhalothrin.     Ultrasensitive Detection of Cancer Biomarkers in the Clinic by Use of a Nanostructured Microfluidic Array

    Multiplexed biomarker protein detection holds unrealized promise for clinical cancer diagnostics due to lack of suitable measurement devices and lack of rigorously validated protein panels. Here we report an ultrasensitive electrochemical microfluidic array optimized to measure a four-protein panel of biomarker proteins, and we validate the protein panel for accurate oral cancer diagnostics. Unprecedented ultralow detection into the 5-50 fg.mL(-1) range was achieved for simultaneous measurement of proteins interleukin 6 (IL-6), IL-8, vascular endothelial growth factor (VEGF), and VEGF-C in diluted serum. The immunoarray achieves high sensitivity in 50 min assays by using off-line protein capture by magnetic beads carrying 400 000 enzyme labels and similar to 100 000 antibodies. After capture of the proteins and washing to inhibit nonspecific binding, the beads are magnetically separated and injected into the array for selective capture by antibodies on eight nanostructured sensors. Good correlations with enzyme-linked immunosorbent assays (ELISA) for protein determinations in conditioned cancer cell media confirmed the accuracy of this approach. Normalized means of the four protein levels in 78 oral cancer patient serum samples and 49 controls gave clinical sensitivity of 89 and specificity of 98 for oral cancer detection, demonstrating high diagnostic utility. The low-cost, easily fabricated immunoarray provides a rapid Serum test for diagnosis and personalized therapy of oral cancer.     Stabilization of Tropical Peat Soil from Sarawak with Different Stabilizing Agents

    This paper describes a study on tropical peat soil stabilization to improve its physical properties by using different stabilizing agents. The samples were collected from six different locations of Sarawak, Malaysia, to evaluate their physical or index properties. Out of them, sample having the highest percentage of organic content has been selected for stabilization purposes. In this study, ordinary portland cement (OPC), quick lime (QL), and class F fly ash (FA) were used as stabilizer. The amount of OPC, QL, and FA added to the peat soil sample, as percentage of dry soil mass, were in the range of 5–20%; 5–20% and 2–8%, respectively for the curing periods of 7, 14, and 28 days. The Unconfined Compressive Strength (UCS) test was carried out on treated/stabilized samples with the above mentioned percentages of the stabilizer and the result shows that the UCS value increases significantly with the increase of all stabilizing agent used and also with curing periods. However, in case of FA and QL, the UCS value increases up to 15 and 6%, respectively with a curing period of 28 days but decreases rather steady beyond this percentage. Some UCS tests have been conducted with a mixture of FA and QL to study the combined effect of the stabilizer. In addition, Scanning Electron Microscope (SEM) study was carried out on original peat soil and FA, as well as some treated samples in order to study their microstructures

    Correlation Between Different Physical and Engineering Properties of Tropical Peat Soils from Sarawak

    The present paper describes the physical and engineering properties of tropical peat soils from Sarawak, Malaysia. Different physical properties such as organic content (OC), liquid limit (LL), fibre content (FC), specific gravity (Gs) and engineering properties (mainly the standard Proctor test) have been conducted on remoulded peat soil samples. The results show that, the value of LL, FC, Optimum Moisture Content (OMC) increases with an increase in OC. Also the results show that, Gs and MDD decrease with increasing values of OC. Furthermore, the plot of OC against Gs from the current study is compared with other researchers; whereby the correlation shows that R2 values ranges from 0.71 to 0.95. This demonstrates that findings on local peat soils from Sarawak are in good agreement with other researchers as similar trends are observed. Consequently, geotechnical engineers can refer these correlations to comprehend the preliminary behavior of peat soil, where the geotechnical data are not readily available

    Study of artificial intelligence into checkers game using HTML and Javascript

    Artificial Intelligence (AI) in the game industry matures and continues to expand due to its capabilities to explore and exploit in bringing gaming to life. In this paper, we described the game design for the game Checkers, which is developed by using an AI heuristic approach using HTML and JavaScript. A group of people chosen in this study to test the game, which is a validation process to confirm that the game goes as per requirement. The testing activity ends after the final bug fix and the result shows that the game executes properly as expected. The result indicates that the game meets the objective of the system, which is the game, may learn and explore by itself as a heuristic element

    Measurement of the quasi-elastic axial vector mass in neutrino-oxygen interactions

    The weak nucleon axial-vector form factor for quasi-elastic interactions is determined using neutrino interaction data from the K2K Scintillating Fiber detector in the neutrino beam at KEK. More than 12,000 events are analyzed, of which half are charged-current quasi-elastic interactions nu-mu n to mu- p occurring primarily in oxygen nuclei. We use a relativistic Fermi gas model for oxygen and assume the form factor is approximately a dipole with one parameter, the axial vector mass M_A, and fit to the shape of the distribution of the square of the momentum transfer from the nucleon to the nucleus. Our best fit result for M_A = 1.20 \pm 0.12 GeV. Furthermore, this analysis includes updated vector form factors from recent electron scattering experiments and a discussion of the effects of the nucleon momentum on the shape of the fitted distributions.Comment: 14 pages, 10 figures, 6 table
