    Comparative study of the biological behaviour in hamster of two isolates of leishmania characterized respectively as L. major-like and L. donovani

    Hamster inoculated intraperitoneally with 1 x 10(7) parasites of L. donovani and L. major-like of the New World were studied in groups of 15, 30, 60 and 90 days of infection. The parasite load and density showed progressive increase with the evolution of the infection and was higher in the L. donovani groups than in the L. major-like groups. The L. major-like groups showed parasite density higher in the spleen than in the liver and was similar in both organs in L. donovani groups. The histopathology showed a diffuse marked hyperplasia and hypertrophy of the reticuloendothelial system with high parasitism in the L. donovani groups while there was focal involvement of these organs in L. major-like groups, forming nodules of macrophages that were scantly parasitised. The biological behaviour could be useful in the preliminary studies of Leishmania strain in regional laboratories and understanding the histopathology of lesions caused by different leishmania species.Experimentos utilizando-se hamsters inoculados intraperitonealmente com 1 x 10(7) parasitas de 2 cepas, L. donovani (MHOM/BR/72/LD 46) e L. major-like (MCAN/BR/73/LD 70) isoladas no Novo Mundo foram realizados e estudados em grupos de 15, 30, 60 e 90 dias de infecção. A carga e a densidade parasitária mostraram progressivo aumento com a evolução da infecção e foi maior nos grupos inoculados com L. donovani do que nos grupos inoculados com L. major-like. Os grupos inoculados com L. major-like mostraram densidade parasitária maior no baço que no fígado e foram semelhantes em ambos os órgãos nos grupos inoculados com L. donovani. A histopatologia mostrou intensa e difusa hyperplasia e hipertrofia do sistema reticuloendotelial com alto parasitismo nos grupos inoculados com L. donovani, enquanto foi encontrado envolvimento focai nestes órgãos nos grupos inoculados com L. major-like, formando nódulos de macrófagos discretamente parasitados. O comportamento biológico seria útil em estudos preliminares de identificação de cepas de Leishmania em laboratórios regionais e na compreensão da histopatologia das lesões causadas por diferentes espécimes de leishmanias

    Curvas de aprendizado aplicadas a análise da produtividade de soldadores

    Este artigo apresenta uma análise da produtividade de soldadores de estruturas navais utilizando o conceito de curvas de maturidade. O presente estudo caracteriza-se por ter uma abordagem quantitativa, o método escolhido é o estudo de caso e seu propósito exploratório. Foram coletados 82 amostras buscando relacionar a evolução da produtividade (hh/ton) e o aumento da tonelada produzida acumulada. O ajuste dos dados de desempenho foi realizado utilizando os modelos de curva de aprendizado: potencial; hiperbólico; exponencial e logarítmico. Os parâmetros foram obtidos através de regressão não- linear pelo método de Quasi-Newton por intermédio de software de análise estatística Statistica 7.0. Os resultados obtidos pelo ajuste da curva exponencial evidenciam b=0,198 próximo de zero, o que indica um moderado percentual de aprendizado e assimilação moderada de conhecimento, destreza e preceitos da tarefa. Como principal contribuição apresenta-se uma aplicação de modelos constituídos por funções matemáticas de complexidade diversas, que possibilitam melhor conhecer o processo de aprendizado de soldadores, e abre caminho para sua utilização em diversas atividades de planejamento de mão de obra e industrial

    Polymyalgia Rheumatica (PMR) Special Interest Group at OMERACT 11: outcomes of importance for patients with PMR

    We worked toward developing a core outcome set for clinical research studies in polymyalgia rheumatica (PMR) by conducting (1) patient consultations using modified nominal group technique; (2) a systematic literature review of outcome measures in PMR; (3) a pilot observational study of patients presenting with untreated PMR, and further discussion with patient research partners; and (4) a qualitative focus group study of patients with PMR on the meaning of stiffness, using thematic analysis. (1) Consultations included 104 patients at 4 centers. Symptoms of PMR included pain, stiffness, fatigue, and sleep disturbance. Function, anxiety, and depression were also often mentioned. Participants expressed concerns about diagnostic delay, adverse effects of glucocorticoids, and fear of relapse. (2) In the systematic review, outcome measures previously used for PMR include pain visual analog scores (VAS), morning stiffness, blood markers, function, and quality of life; standardized effect sizes posttreatment were large. (3) Findings from the observational study indicated that asking about symptom severity at 7 AM, or "on waking," appeared more relevant to disease activity than asking about symptom severity "now" (which depended on the time of assessment). (4) Preliminary results were presented from the focus group qualitative study, encompassing broad themes of stiffness, pain, and the effect of PMR on patients' lives. It was concluded that further validation work is required before a core outcome set in PMR can be recommended. Nevertheless, the large standardized effect sizes suggest that pain VAS is likely to be satisfactory as a primary outcome measure for assessing response to initial therapy of PMR. Dissection of between-patient heterogeneity in the subsequent treatment course may require attention to comorbidity as a potential confounding factor

    Size and frequency of natural forest disturbances and Amazon carbon balance

    Forest inventory studies in the Amazon indicate a large terrestrial carbon sink. However, field plots may fail to represent forest mortality processes at landscape-scales of tropical forests. Here we characterize the frequency distribution of disturbance events in natural forests from 0.01 ha to 2,651 ha size throughout Amazonia using a novel combination of forest inventory, airborne lidar and satellite remote sensing data. We find that small-scale mortality events are responsible for aboveground biomass losses of B1.28 Pg C y 1 over the entire Amazon region. We also find that intermediate-scale disturbances account for losses of B0.01 Pg C y 1 , and that the largest-scale disturbances as a result of blow-downs only account for losses of B0.003 Pg C y 1 . Simulation of growth and mortality indicates that even when all carbon losses from intermediate and large-scale disturbances are considered, these are outweighed by the net biomass accumulation by tree growth, supporting the inference of an Amazon carbon sink

    Conformational studies of peptides representing a segment of TM7 from H+-VO-ATPase in SDS micelles

    The conformation of a transmembrane peptide, sMTM7, encompassing the cytoplasmic hemi-channel domain of the seventh transmembrane section of subunit a from V-ATPase from Saccharomyces cerevisiae solubilized in SDS solutions was studied by circular dichroism (CD) spectroscopy and fluorescence spectroscopy of the single tryptophan residue of this peptide. The results show that the peptide adopts an α-helical conformation or aggregated β-sheet depending on the peptide-to-SDS ratio used. The results are compared with published data about a longer version of the peptide (i.e., MTM7). It is concluded that the bulky, positively charged arginine residue located in the center of both peptides has a destabilizing effect on the helical conformation of the SDS-solubilized peptides, leading to β-sheet formation and subsequent aggregation

    J-PLUS: analysis of the intracluster light in the Coma cluster

    The intracluster light (ICL) is a luminous component of galaxy clusters composed of stars that are gravitationally bound to the cluster potential but do not belong to the individual galaxies. Previous studies of the ICL have shown that its formation and evolution are intimately linked to the evolutionary stage of the cluster. Thus, the analysis of the ICL in the Coma cluster will give insights into the main processes driving the dynamics in this highly complex system. Using a recently developed technique, we measure the ICL fraction in Coma at several wavelengths, using the J-PLUS unique filter system. The combination of narrow- and broadband filters provides valuable information on the dynamical state of the cluster, the ICL stellar types, and the morphology of the diffuse light. We use the Chebyshev-Fourier Intracluster Light Estimator (CICLE) to disentangle the ICL from the light of the galaxies, and to robustly measure the ICL fraction in seven J-PLUS filters. We obtain the ICL fraction distribution of the Coma cluster at different optical wavelengths, which varies from ∼7%−21%\sim 7\%-21\%, showing the highest values in the narrowband filters J0395, J0410, and J0430. This ICL fraction excess is distinctive pattern recently observed in dynamically active clusters (mergers), indicating a higher amount of bluer stars in the ICL compared to the cluster galaxies. Both the high ICL fractions and the excess in the bluer filters are indicative of a merging state. The presence of younger/lower-metallicity stars the ICL suggests that the main mechanism of ICL formation for the Coma cluster is the stripping of the stars in the outskirts of infalling galaxies and, possibly, the disruption of dwarf galaxies during past/ongoing mergers.Comment: 10 pages, 3 figures, 1 table.     A Pilot Study on the Metabolic Impact of Mediterranean Diet in Type 2 Diabetes: Is Gut Microbiota the Key?

    The Mediterranean diet (MD) has been recommended for type 2 diabetes (T2D) treatment. The impact of diet in shaping the gut microbiota is well known, particularly for MD. However, the link between MD and diabetes outcome improvement is not completely clear. This study aims to evaluate the role of microbiota modulation by a nonpharmacological intervention in patients with T2D. In this 12-week single-arm pilot study, nine participants received individual nutritional counseling sessions promoting MD. Gut microbiota, biochemical parameters, body composition, and blood pressure were assessed at baseline, 4 weeks, and 12 weeks after the intervention. Adherence to MD [assessed by Mediterranean Diet Adherence Screener (MEDAS) score] increased after the intervention. Bacterial richness increased after 4 weeks of intervention and was negatively correlated with fasting glucose levels and Homeostatic Model Assessment for Insulin Resistance (HOMA-IR). Prevotella to Bacteroides ratio also increased after 4 weeks. In contrast, glycated haemoglobin (HbA1c) and HOMA-IR were only decreased at the end of study. Alkaline phosphatase activity was assessed in fecal samples and was negatively correlated with HbA1c and positively correlated with bacterial diversity. The results of this study reinforce that MD adherence results in a better glycemic control in subjects with T2D. Changes in gut bacterial richness caused by MD adherence may be relevant in mediating the metabolic impact of this dietary intervention.info:eu-repo/semantics/publishedVersio

    J-PLUS: A wide-field multi-band study of the M15 globular cluster. Evidence of multiple stellar populations in the RGB

    The Javalambre Photometric Local Universe Survey (J-PLUS) provides wide field-of-view images in 12 narrow, intermediate and broad-band filters optimized for stellar photometry. Here we have applied J-PLUS data for the first time for the study of Galactic GCs using science verification data obtained for the very metal-poor GC M\,15. Our J-PLUS data provide low-resolution spectral energy distributions covering the near-UV to the near-IR, allowing us to search for MPs based on pseudo-spectral fitting diagnostics. J-PLUS CMDs are found to be particularly useful to search for splits in the sequences formed by the upper red giant branch (RGB) and asymptotic giant branch (AGB) stars. We interpret these split sequences as evidence for the presence of MPs. This demonstrates that the J-PLUS survey will have sufficient spatial coverage and spectral resolution to perform a large statistical study of GCs through multi-band photometry in the coming years.Comment: 11 pages, 11 figures.
