    In vivo identification of brain structures functionally involved in spatial learning and strategy switch

    Spatial learning is a complex behavior which includes, among others, encoding of space, sensory and motivational processes, arousal and locomotor performance. Today, our view on spatial navigation is largely hippocampus-centrist. Less is known about the involvement of brain structures up- and downstream, or out of this circuit. Here, I provide the first in vivo assessment of the neural matrix underlying spatial learning, using functional manganese-enhanced MRI (MEMRI) and voxel-wise whole brain analysis. Mice underwent place-learning (PL) vs. response-learning (RL) in the water cross maze (WCM) and its readout was correlated to the Mn2+ contrasts. Thus, I identified structures involved in spatial learning largely overlooked in the past, due to methods focused on region of interest (ROI) analyses. These structures include several sensory-related structures and differ between place-learners and response-learners, with the former (PL) comprising mostly structures involved in different properties of visual processing, such as horizontal gaze (e.g. nucleus prepositus) and saccade (e.g. fastigial nucleus), or provide vision-input and eye movement information from parahippocampal (e.g. presubiculum, perirhinal, postrhinal and ectorhinal areas) and other regions (e.g. orbital area, superior colliculus and vestibular ocular-reflex from the vestibular nucleus) likely to head-direction, grid- and place-cells; and the latter (RL) presenting structures related to more basic rodent sensory computations, like odor (e.g main and accessory olfactory bulb, cortical amygdala, piriform, endopiriform and postpiriform areas) and acoustic stimuli representation (e.g. auditory area, nucleus of the lateral lemniscus and superior olivary complex), or sensory-motor properties, such as body representation (e.g. somatosensory area ? upper limbs) and head-direction signal. Add-on experiments pointed to preferential Mn2+ accumulation towards projection terminals, suggesting that our mapping was mostly formed by projection sites of the originally activated structures. This is corroborated by in-depth analysis of MEMRI data after WCM learning showing mostly downstream targets of the hippocampus. These differ between fornical afferences from vCA1 and direct innervation from dCA1/iCA1 (for PL), and structures along the longitudinal association bundle originating in vCA1 (for RL). To elucidate the pattern of Mn2+ accumulation seen on the scans, I performed c-fos expression analyses following learning in the WCM. This helped me identify the structures initially activated during spatial learning and its underlying connectivity to establish the matrix. Finally, to test the causal involvement of selected structures from our previous findings I inhibited them (through DREADDs) while mice performed the WCM task. I also focused on the causal involvement of the vHPC-mPFC circuit on strategy switch during WCM learning. I believe that this study might shed light into new brain structures involved in spatial learning and strategy switch and complement the current knowledge on these circuits? connectivity. Moreover, I elucidated some functional mechanisms of MEMRI, clarifying the interpretation of data obtained with this method and its possible future applications

    Bounds on topological Abelian string-vortex and string-cigar from information-entropic measure

    In this work we obtain bounds on the topological Abelian string-vortex and on the string-cigar, by using a new measure of configurational complexity, known as configurational entropy. In this way, the information-theoretical measure of six-dimensional braneworlds scenarios are capable to probe situations where the parameters responsible for the brane thickness are arbitrary. The so-called configurational entropy (CE) selects the best value of the parameter in the model. This is accomplished by minimizing the CE, namely, by selecting the most appropriate parameters in the model that correspond to the most organized system, based upon the Shannon information theory. This information-theoretical measure of complexity provides a complementary perspective to situations where strictly energy-based arguments are inconclusive. We show that the higher the energy the higher the CE, what shows an important correlation between the energy of the a localized field configuration and its associated entropic measure.Comment: 6 pages, 7 figures, final version to appear in Phys.     Toxocaríase humana: inquérito soroepidemiológico no município de Campinas (SP), Brasil

    The occurrence of human Toxocara infection was evaluated in three neighborhoods of the periphery of the Campinas municipality (Jardim Santa Mônica, Jardim São Marcos and Jardim Campineiro) in 1999. Forty residences and 138 residents were randomly selected by drawing lots and were submitted to a seroepidemiological survey, which included blood collection for the immunoenzymatic detection (ELISA) of anti-Toxocara antibodies and a blood count, and the application of a semi-structured questionnaire for the evaluation of epidemiological data. Significant levels of anti-Toxocara antibodies were detected in 23.9% of the 1999 samples. No significant difference in the frequency of infection according to age was observed. Environmental contamination with Toxocara eggs was observed in 12.3 and 14.0% of 57 soil samples collected in the same region in December 1998 and July 1999, respectively. Univariate analysis and multiple logistic regression of the data obtained from the questionnaires and of the results of the serological tests, suggest a significant influence of socioeconomic variables on the frequency of human infection with Toxocara under the conditions prevalent in the study area.Estudou-se a ocorrência de infecção humana por Toxocara em três bairros da periferia do município de Campinas (Jardim Santa Mônica, Jardim São Marcos e Jardim Campineiro). Em 1999 sortearam-se aleatoriamente 40 domicílios e 138 moradores foram submetidos a inquérito soroepidemiológico, que incluiu coleta de sangue para teste imunoenzimático (E.L.I.S.A.) visando pesquisa de anticorpos anti-Toxocara e hemograma, além de aplicação de questionário semi-estruturado para avaliação de dados de interesse epidemiológico. Observou-se presença de anticorpos anti-Toxocara em 23,9% das amostras, sem ocorrência de diferença significativa quando se considerou a idade ou sexo dos indivíduos examinados. Em 57 amostras de solo da mesma região, coletadas em dezembro de 1998 e novamente em julho de 1999, estudou-se a contaminação ambiental por ovos de Toxocara, verificando-se sua presença em, respectivamente, 12,3% e 14,0% das amostras. Análises univariada e multivariada por regressão logística dos dados obtidos a partir dos questionários e dos resultados da sorologia anti-Toxocara mostraram influência significativa de variáveis de ordem sócio-econômica na prevalência de infecção humana por Toxocara, nas condições presentes na área estudada

    Retinoblastoma in a pediatric oncology reference center in southern brazil

    Background: Retinoblastoma (Rb) is the most common intraocular tumor diagnosed in children in Brazil. However, detailed information is lacking regarding patient clinical demographics. This study aimed to determine the clinical profile of patients with Rb who were treated in a public university hospital in southern Brazil from 1983 to 2012. Methods: Patients’ medical records were reviewed to retrospectively identify patients with a principal diagnosis of Rb. Rb was classified as hereditary or non-hereditary. Clinical staging was reviewed by an ophthalmologist. Statistical analysis was performed using SPSS. Results: Of 165 patients with a diagnosis of Rb during this period, 140 were included in the study. Disease was unilateral in 65.0 % of patients, bilateral in 32.9 %, and trilateral in 2.1 %. The mean age at onset of the first sign/ symptom was 18.1 month, and 35.7 % of patients were diagnosed during the first year of life. The most common presenting signs were leukocoria (73.6 %) and strabismus (20.7 %). The mean age at diagnosis was 23.5 months, and time to diagnosis was 5.4 months. In patients with clinical features of hereditary Rb, both onset of the first sign/symptom and diagnosis were at an earlier age than in patients without these features (12.3 vs 21.6 months [P = 0.001] and 15.9 vs 28.0 months [P < 0.001], respectively). However, there was no significant difference in overall survival between the two groups. Ocular stage at diagnosis was advanced in 76.5 % (Reese V) and 78.1 % (International Classification D or E). Of patients with unilateral and bilateral disease, 35.2 % and 34.8 %, respectively, had extraocular disease at diagnosis; 10.7 % had metastatic disease at diagnosis. Enucleation was observed in 88.1 % and exenteration in 11.9 % of patients; 93.6 % patients were followed until 2012, and 22.9 % relapsed. Overall survival was 86.4 %. Conclusions: Most Rb diagnoses are still diagnosed in advanced stages of the disease, considerably reducing overall survival time and the rate of eye and vision preservation

    AGUIA: autonomous graphical user interface assembly for clinical trials semantic data services

    <p>Abstract</p> <p>Background</p> <p>AGUIA is a front-end web application originally developed to manage clinical, demographic and biomolecular patient data collected during clinical trials at MD Anderson Cancer Center. The diversity of methods involved in patient screening and sample processing generates a variety of data types that require a resource-oriented architecture to capture the associations between the heterogeneous data elements. AGUIA uses a semantic web formalism, resource description framework (RDF), and a bottom-up design of knowledge bases that employ the S3DB tool as the starting point for the client's interface assembly.</p> <p>Methods</p> <p>The data web service, S3DB, meets the necessary requirements of generating the RDF and of explicitly distinguishing the description of the domain from its instantiation, while allowing for continuous editing of both. Furthermore, it uses an HTTP-REST protocol, has a SPARQL endpoint, and has open source availability in the public domain, which facilitates the development and dissemination of this application. However, S3DB alone does not address the issue of representing content in a form that makes sense for domain experts.</p> <p>Results</p> <p>We identified an autonomous set of descriptors, the GBox, that provides user and domain specifications for the graphical user interface. This was achieved by identifying a formalism that makes use of an RDF schema to enable the automatic assembly of graphical user interfaces in a meaningful manner while using only resources native to the client web browser (JavaScript interpreter, document object model). We defined a generalized RDF model such that changes in the graphic descriptors are automatically and immediately (locally) reflected into the configuration of the client's interface application.</p> <p>Conclusions</p> <p>The design patterns identified for the GBox benefit from and reflect the specific requirements of interacting with data generated by clinical trials, and they contain clues for a general purpose solution to the challenge of having interfaces automatically assembled for multiple and volatile views of a domain. By coding AGUIA in JavaScript, for which all browsers include a native interpreter, a solution was found that assembles interfaces that are meaningful to the particular user, and which are also ubiquitous and lightweight, allowing the computational load to be carried by the client's machine.</p

    Transcriptome profiling of grapevine seedless segregants during berry development reveals candidate genes associated with berry weight

However, this is a quantitative and polygenic trait, and its genetic determination is still poorly understood. Considering its economic importance, it is relevant to determine its genetic architecture and elucidate the mechanisms involved in its expression. To approach this issue, an RNA-Seq experiment based on Illumina platform was performed (14 libraries), including seedless segregants with contrasting phenotypes for berry weight at fruit setting (FST) and 6–8 mm berries (B68) phenological stages. Results A group of 526 differentially expressed (DE) genes were identified, by comparing seedless segregants with contrasting phenotypes for berry weight: 101 genes from the FST stage and 463 from the B68 stage. Also, we integrated differential expression, principal components analysis (PCA), correlations and network co-expression analyses to characterize the transcriptome profiling observed in segregants with contrasting phenotypes for berry weight. After this, 68 DE genes were selected as candidate genes, and seven candidate genes were validated by real time-PCR, confirming their expression profiles. Conclusions We have carried out the first transcriptome analysis focused on table grape seedless segregants with contrasting phenotypes for berry weight. Our findings contributed to the understanding of the mechanisms involved in berry weight determination. Also, this comparative transcriptome profiling revealed candidate genes for berry weight which could be evaluated as selection tools in table grape breeding programs.http://bmcplantbiol.biomedcentral.com/articles/10.1186/s12870-016-0789-
