    Home-Based Therapy for Young Children in Low-Income Families: A Student Training Program

    The purpose of this project was to develop an internship training program that offered in-home therapy for young children with significant emotional and behavior problems. The children lived in single-parent, low-income homes in unsafe neighborhoods of a large, urban area. A year-long, training and supervision program was implemented with 10 second-year, graduate students enrolled in 5 different university programs that prepared mental health professionals. Students received specialized instruction in working with diverse families living in poverty and in an evidence-based treatment program. They initially observed veteran counselors implementing the treatment program in homes and gradually assumed responsibility for conducting sessions on their own. Students’ scores on a measure of counseling self-efficacy improved significantly from pre-to post-internship. Students reported high levels of satisfaction with the training program and significantly improved confidence levels in their counseling skills at the conclusion of their internship experience. The limitations of these preliminary outcomes for this pilot program are discussed along with the need for more research in this understudied area

    Intravital Multiphoton Microscopy with Fluorescent Bile Salts in Rats as an In Vivo Biomarker for Hepatobiliary Transport Inhibition

    The bile salt export pump (BSEP) is expressed at the canalicular domain of hepatocytes, where it mediates the elimination of monovalent bile salts into the bile. Inhibition of BSEP is considered a susceptibility factor for drug-induced liver injury that often goes undetected during nonclinical testing. Although in vitro assays exist for screening BSEP inhibition, a reliable and specific method for confirming Bsep inhibition in vivo would be a valuable follow up to a BSEP screening strategy, helping to put a translatable context around in vitro inhibition data, incorporating processes such as metabolism, protein binding, and other exposure properties that are lacking in most in vitro BSEP models. Here, we describe studies in which methods of quantitative intravital microscopy were used to identify dose-dependent effects of two known BSEP/Bsep inhibitors, 2-[4-[4-(butylcarbamoyl)-2-[(2,4-dichlorophenyl)sulfonylamino]phenoxy]-3-methoxyphenyl]acetic acid (AMG-009) and bosentan, on hepatocellular transport of the fluorescent bile salts cholylglycyl amidofluorescein and cholyl-lysyl-fluorescein in rats. Results of these studies demonstrate that the intravital microscopy approach is capable of detecting Bsep inhibition at drug doses well below those found to increase serum bile acid levels, and also indicate that basolateral efflux transporters play a significant role in preventing cytosolic accumulation of bile acids under conditions of Bsep inhibition in rats. Studies of this kind can both improve our understanding of exposures needed to inhibit Bsep in vivo and provide unique insights into drug effects in ways that can improve our ability interpret animal studies for the prediction of human drug hepatotoxicity

    Delivering Bio-Mems & Microfluidic Education Around Accessible Technologies

    Electronic Systems are now being deployed in al-most all aspects of daily life as opposed to being confined to consumer electronics, computing, communication and control applications as was the case in the 90’s. One of the more significant growth areas is medical instrumentation, health care, bio-chemical analysis and environmental monitoring. Most of these applications will in the future require the integration of fluidics and biology within complex electronic systems. We are now seeing technologies emerging together with access services such as the FP6 “INTEGRAMplus” and “MicroBuilder” programs that offer competitive solutions for companies wishing to de-sign and prototype microfluidic systems. For successful deployment of these systems, a new breed of electronic engineers are needed that understand how to deliver bio-chemistry and living cells to transducers and integrate the required technologies reliably into robust systems.     Shifts in Gamma Phase–Amplitude Coupling Frequency from Theta to Alpha Over Posterior Cortex During Visual Tasks

    The phase of ongoing theta (4–8 Hz) and alpha (8–12 Hz) electrophysiological oscillations is coupled to high gamma (80–150 Hz) amplitude, which suggests that low-frequency oscillations modulate local cortical activity. While this phase–amplitude coupling (PAC) has been demonstrated in a variety of tasks and cortical regions, it has not been shown whether task demands differentially affect the regional distribution of the preferred low-frequency coupling to high gamma. To address this issue we investigated multiple-rhythm theta/alpha to high gamma PAC in two subjects with implanted subdural electrocorticographic grids. We show that high gamma amplitude couples to the theta and alpha troughs and demonstrate that, during visual tasks, alpha/high gamma coupling preferentially increases in visual cortical regions. These results suggest that low-frequency phase to high-frequency amplitude coupling is modulated by behavioral task and may reflect a mechanism for selection between communicating neuronal networks

    Concrete Box Beam Risk Assessment and Mitigation: Volume 2—Evaluation and Structural Behavior

    Adjacent box beam bridges have a history of poor long-term performance including premature deterioration and failures. Leaking joints between box beams allow chloride-laden water to migrate through the superstructure and initiate corrosion. The nature of this deterioration leads to uncertainty of the extent and effect of deterioration on structural behavior. Due to limitations in previous research and understanding of the strength of deteriorated box beam bridges, conservative assumptions are made for the assessment and load rating of these bridges. Furthermore, the design of new box beam bridges, which can offer an efficient and economical solution, is often discouraged due to poor past performance. The objective of this research is to develop recommendations for inspection, load-rating, and design of adjacent box beam bridges. The research is presented in two volumes. Volume 1 focuses on the evolution of box beam design in Indiana to understand the lack of performance and durability. The Indiana Department of Transportation (INDOT) standards and bridge design manuals were reviewed to track the historical development of box beam bridges in the State. Two timelines were produced tracking important updates to box beam design. Adjacent box beam bridges within INDOT’s bridge database were also analyzed. Superstructure ratings were compared with bridge age as well as bridge characteristics to highlight possible causes for deterioration. Analyzing the INDOT inventory, data shows that the condition of adjacent box beam bridges may be affected by location, type of wearing surface, and the use of deck membranes. Six bridges were then inspected to identify common deficiencies and specific problems. Exterior beams and beams within the wheel load path tend to have higher levels of deterioration. Furthermore, leaking joints between beams leads to corrosion of reinforcement, ultimately resulting in spalling, fracture of prestressing strands, and loss of structural capacity. Volume 2 focuses on evaluating the capacity of deteriorated adjacent box beams, the development of improved load rating procedures, and new box beam design. Through a series of bridge inspections, deteriorated box beams were identified and acquired for experimental testing. The extent of corrosion was determined through visual inspection, non-destructive evaluation, and destructive evaluation. Non-destructive tests (NDT) included the use of connectionless electrical pulse response analysis (CEPRA), ground penetrating radar (GPR), and half-cell potentials. Deteriorated capacity was determined through structural testing, and an analysis procedure was developed to estimate deteriorated behavior. A rehabilitation procedure was also developed to restore load transfer of adjacent beams in cases where shear key failures are suspected. Based on the understanding of deterioration developed through study of deteriorated adjacent box beam bridges, improved inspection and load rating procedures are provided along with design recommendations for the next generation of box beam bridges

    Bait the hook to suit the phish, not the phisherman: A field experiment on security networks of teams to withstand spear phishing attacks on online social networks

    In this paper, we present our research in progress of a field experiment conducted to observe the impact of collective security behavior of teams when being targeted with a spear phishing attack on online social networks. To observe the shaping of security networks in teams, fifteen different honeypot profiles were created to send spear phishing messages after an initial bonding of eight weeks to the target group of 76 people. The experiment simulated a regular communication on online social networks of three teams of an international organization. The team members were entangled in personal and individual chats on an online social network to later react to an unexpected and unforeseen spear phishing message. As previous research has shown, various aspects influence the spear phishing susceptibility, but the collective security behavior has currently been neglected. This work plans to evaluate how security networks are being formed, the factors relevant to shape those networks and efforts to protect against spear phishing attacks

    Spring 2014 Internship Diffuser Data Analysis

    J-2X engine testing on the A-2 test stand at the NASA John C. Stennis Space Center (SSC) has recently concluded. As part of that test campaign, the engine was operated at lower power levels in support of expanding the use of J-2X to other missions. However, the A-2 diffuser was not designed for engine testing at the proposed low power levels. To evaluate the risk of damage to the diffuser, computer simulations were created of the rocket engine exhaust plume inside the 50ft long, water-cooled, altitude-simulating diffuser. The simulations predicted that low power level testing would cause the plume to oscillate in the lower sections of the diffuser. This can possibly cause excessive vibrations, stress, and heat transfer from the plume to the diffuser walls. To understand and assess the performance of the diffuser during low power level engine testing, nine accelerometers and four strain gages were installed around the outer surface of the diffuser. The added instrumentation also allowed for the verification of the rocket exhaust plume computational model. Prior to engine hot-fire testing, a diffuser water-flow test was conducted to verify the proper operation of the newly installed instrumentation. Subsequently, two J-2X engine hot-fire tests were completed. Hot-Fire Test 1 was 11.5 seconds in duration, and accelerometer and strain data verified that the rocket engine plume oscillated in the lower sections of the diffuser. The accelerometers showed very different results dependent upon location. The diffuser consists of four sections, with Section 1 being closest to the engine nozzle and Section 4 being farthest from the engine nozzle. Section 1 accelerometers showed increased amplitudes at startup and shutdown, but low amplitudes while the diffuser was started. Section 3 accelerometers showed the opposite results with near zero G amplitudes prior to and after diffuser start and peak amplitudes to +/- 100G while the diffuser was started. Hot-Fire Test 1 strain gages showed different data dependent on section. Section 1 strains were small, and were in the range of 50 to 150 microstrain, which would result in stresses from 1.45 to 4.35 ksi. The yield stress of the material, A-285 Grade C Steel, is 29.7 ksi. Section 4 strain gages showed much higher values with strains peaking at 1600 microstrain. This strain corresponds to a stress of 46.41 ksi, which is in excess of the yield stress, but below the ultimate stress of 55 to 75 ksi. The decreased accelerations and strain in Section 1, and the increased accelerations and strain in Sections 3 and 4 verified the computer simulation prediction of increased plume oscillations in the lower sections of the diffuser. Hot-Fire Test 2 ran for a duration of 125 seconds. The engine operated at a slightly higher power level than Hot-Fire Test 1 for the initial 35 seconds of the test. After 35 seconds the power level was lowered to Hot-Fire Test 1 levels. The acceleration and strain data for Hot-Fire Test 2 was similar during the initial part of the test. However, just prior to the engine being lowered to the Hot-Fire Test 1 power level, the strain gage data in Section 4 showed a large decrease to strains near zero microstrain from their peak at 1500 microstrain. Future work includes further strain and acceleration data analysis and evaluation

    Elongin B-Mediated Epigenetic Alteration of Viral Chromatin Correlates with Efficient Human Cytomegalovirus Gene Expression and Replication

    Elongins B and C are members of complexes that increase the efficiency of transcriptional elongation by RNA polymerase II (RNAPII) and enhance the monoubiquitination of histone H2B, an epigenetic mark of actively transcribed genes. Here we show that, in addition to its role in facilitating transcription of the cellular genome, elongin B also enhances gene expression from the double-stranded DNA genome of human cytomegalovirus (HCMV), a pathogenic herpesvirus. Reducing the level of elongin B by small interfering RNA- or short hairpin RNA-mediated knockdown decreased viral mRNA expression, viral protein accumulation, viral DNA replication, and infectious virion production. Chromatin immunoprecipitation analysis indicated viral genome occupancy of the elongating form of RNAPII, and monoubiquitinated histone H2B was reduced in elongin B-deficient cells. These data suggest that, in addition to the previously documented epigenetic regulation of transcriptional initiation, HCMV also subverts cellular elongin B-mediated epigenetic mechanisms for enhancing transcriptional elongation to enhance viral gene expression and virus replication
