    Effects of an iron-based fuel-borne catalyst and a diesel particle filter on exhaust toxicity in lung cells in vitro

    Metal-containing fuel additives catalyzing soot combustion in diesel particle filters are used in a widespread manner, and with the growing popularity of diesel vehicles, their application is expected to increase in the near future. Detailed investigation into how such additives affect exhaust toxicity is therefore necessary and has to be performed before epidemiological evidence points towards adverse effects of their application. The present study investigates how the addition of an iron-based fuel additive (Satacen®3, 40 ppm Fe) to low-sulfur diesel affects the in vitro cytotoxic, oxidative, (pro-)inflammatory, and mutagenic activity of the exhaust of a passenger car operated under constant, low-load conditions by exposing a three-dimensional model of the human airway epithelium to complete exhaust at the air–liquid interface. We could show that the use of the iron catalyst without and with filter technology has positive as well as negative effects on exhaust toxicity compared to exhaust with no additives: it decreases the oxidative and, compared to a non-catalyzed diesel particle filter, the mutagenic potential of diesel exhaust, but increases (pro-)inflammatory effects. The presence of a diesel particle filter also influences the impact of Satacen®3 on exhaust toxicity, and the proper choice of the filter type to be used is of importance with regards to exhaust toxicity

    Biological response of an in vitro human 3D lung cell model exposed to brake wear debris varies based on brake pad formulation

    Wear particles from automotive friction brake pads of various sizes, morphology, and chemical composition are significant contributors towards particulate matter. Knowledge concerning the potential adverse effects following inhalation exposure to brake wear debris is limited. Our aim was, therefore, to generate brake wear particles released from commercial low-metallic and non-asbestos organic automotive brake pads used in mid-size passenger cars by a full-scale brake dynamometer with an environmental chamber simulating urban driving and to deduce their potential hazard in vitro. The collected fractions were analysed using scanning electron microscopy via energy-dispersive X-ray spectroscopy (SEM-EDS) and Raman microspectroscopy. The biological impact of the samples was investigated using a human 3D multicellular model consisting of human epithelial cells (A549) and human primary immune cells (macrophages and dendritic cells) mimicking the human epithelial tissue barrier. The viability, morphology, oxidative stress, and (pro-)inflammatory response of the cells were assessed following 24 h exposure to similar to 12, similar to 24, and similar to 48 A mu g/cm(2) of non-airborne samples and to similar to 3.7 A mu g/cm(2) of different brake wear size fractions (2-4, 1-2, and 0.25-1 A mu m) applying a pseudo-air-liquid interface approach. Brake wear debris with low-metallic formula does not induce any adverse biological effects to the in vitro lung multicellular model. Brake wear particles from non-asbestos organic formulated pads, however, induced increased (pro-)inflammatory mediator release from the same in vitro system.     Lock-in thermography as an analytical tool for magnetic nanoparticles: measuring heating power and magnetic fields

    Magnetic nanoparticles and their ability to convert electromagnetic energy into heat are of explicit interest for various applications. However, precise quantification of their heating efficiency is not always upfront, and several parameters render comparative studies challenging. This paper describes the theory behind lock-in thermography, a new technique for quantifying the heating properties of magnetic nanoparticles. This technique allows the investigation of some of the potential sources of variability: key factors such as magnetic field inhomogeneity and its effects on the heating power are explored in detail. The presented results, obtained from various nanoparticle batches of different origins, highlight the importance of pursuing a standardized and systematic approach when quantifying the heating efficiency of magnetic nanoparticles
