    Diversity of Endophytic and Pathogenic Fungi of Saffron (Crocus sativus) Plants from Cultivation Sites in Italy

    Crocus sativus is an important crop for the production of saffron and bioactive compounds. Plant endophytic fungi are a source of secondary metabolites additional to those produced by the plant itself. We analysed the biodiversity of endophytic fungi present in corms, stems, leaves, tepals, and stigmas of C. sativus from ten Italian sites; furthermore, we isolated putative pathogenic fungi from rotten plants. We used an in vitro isolation approach followed by molecular analysis of the internal transcribed spacer (ITS rDNA) region. We obtained 165 strains belonging to 39 OTUs, spreading over 26 genera and 29 species. Dark septate endophytes of the genus Cadophora and the species Talaromyces pinophilus dominated in corms, while Alternaria alternata, Epicoccum spp., T. pinophilus, Mucor fragilis, and Stemphylium vesicarium dominated in other tissues. The most frequently isolated pathogens were Fusarium oxysporum and Rhizopus oryzae. Endophytic communities significantly differed among tissues and life stages, whereas differences among cultivation sites were not statistically supported. Several endophytes were hypothesized to have changing trophic modes and/or to be latent pathogens in C. sativus. All strains were conserved ex-situ for future bioactivity tests and production of metabolites

    A new advanced railgun system for debris impact study

    The growing quantity of debris in Earth orbit poses a danger to users of the orbital environment, such as spacecraft. It also increases the risk that humans or manmade structures could be impacted when objects reenter Earth's atmosphere. During the design of a spacecraft, a requirement may be specified for the surviv-ability of the spacecraft against Meteoroid / Orbital Debris (M/OD) impacts throughout the mission; further-more, the structure of a spacecraft is designed to insure its integrity during the launch and, if it is reusable, during descent, re-entry and landing. In addition, the structure has to provide required stiffness in order to allow for exact positioning of experiments and antennas, and it has to protect the payload against the space environment. In order to decrease the probability of spacecraft failure caused by M/OD, space maneuver is needed to avoid M/OD if the M/OD has dimensions larger than 10cm, but for M/OD with dimensions less than 1cm M/OD shields are needed for spacecrafts. It is therefore necessary to determine the impact-related failure mechanisms and associated ballistic limit equations (BLEs) for typical spacecraft components and subsys-tems. The methods that are used to obtain the ballistic limit equations are numerical simulations and la-borato-ry experiments. In order to perform an high energy ballistic characterization of layered structures, a new ad-vanced electromagnetic accelerator, called railgun, has been assembled and tuned. A railgun is an electrically powered electromagnetic projectile launcher. Such device is made up of a pair of parallel conducting rails, which a sliding metallic armature is accelerated along by the electromagnetic effect (Lorentz force) of a cur-rent that flows down one rail, into the armature and then back along the other rail, thanks to a high power pulse given by a bank of capacitors. A tunable power supplier is used to set the capacitors charging voltage at the desired level: in this way the Rail Gun energy can be tuned as a function of the desired bullet velocity. This facility is able to analyze both low and high velocity impacts. A numerical simulation is also performed by using the Ansys Autodyn code in order to analyze the damage. The experimental results and numerical simulations show that the railgun-device is a good candidate to perform impact testing of materials in the space debris energy range

    Clima de aula y estilos de enseñanza en una institución educativa privada

    La presente investigación tuvo como propósito analizar y describir los mejores climas de aula percibidos por los alumnos de cada sección del sexto de primaria. A la par, se buscó conocer el estilo de enseñanza del docente de cada curso elegido. En este estudio participaron 115 estudiantes y 12 maestros de una institución educativa básica privada de Lima Metropolitana. Para lograr los objetivos planteados se usó el instrumento Clima Social Escolar (CES) de Moos y Trickett validado por Dionicio (2016), y la Escala sobre Estilos de Enseñanza (ESEE) de González, López y Pino (2012), respectivamente para estudiantes y maestros. Se encontró que todos los docentes de los mejores climas de aula de cada sección poseían un estilo de enseñanza activo. Lo anterior podría ser tomado como parte de las características fundamentales que generan un clima de aula positivo, ya que permitiría el involucramiento activo de los estudiantes en la dinámica de la clase. Así mismo, otros rasgos encontrados fueron: reconocer las habilidades personales del alumnado, hallar nuevas formas para el aprendizaje a través de la creatividad, y la organización y el control de las clases. Esta última sería una característica necesaria para percibir un buen clima de aula, ya que propondría como gestor al maestro.The purpose of this research was to analyze and describe the best classroom climates perceived by students in each section of the sixth grade, as well as the teaching style of each teacher. The participants of this study were 115 students and 12 teachers from a private basic education institution in Lima Metropolitana. To achieve the proposed objectives, the Moos and Trickett instrument, validated by Dionicio (2016), and the Scale on Teaching Styles (ESEE) by González, López and Pino (2012) were used, respectively for students and teachers. According to the results, all teachers in the best classroom climates in each section had an active teaching style. This could be taken as part of the fundamental characteristics that generate a positive classroom climate, since it would allow the active involvement of students in the dynamics of the class. Likewise, other present features of positive classroom climate were: recognizing the personal skills of the students, finding new ways to learn through creativity, and the organization and control of classes. The last one mentioned would be a necessary characteristic to perceive a good classroom climate, since it would put the teacher in a management role

    Identification of new polymorphic regions and differentiation of cultivated olives (Olea europaea L.) through plastome sequence comparison

    <p>Abstract</p> <p>Background</p> <p>The cultivated olive (<it>Olea europaea </it>L.) is the most agriculturally important species of the Oleaceae family. Although many studies have been performed on plastid polymorphisms to evaluate taxonomy, phylogeny and phylogeography of <it>Olea </it>subspecies, only few polymorphic regions discriminating among the agronomically and economically important olive cultivars have been identified. The objective of this study was to sequence the entire plastome of olive and analyze many potential polymorphic regions to develop new inter-cultivar genetic markers.</p> <p>Results</p> <p>The complete plastid genome of the olive cultivar Frantoio was determined by direct sequence analysis using universal and novel PCR primers designed to amplify all overlapping regions. The chloroplast genome of the olive has an organisation and gene order that is conserved among numerous Angiosperm species and do not contain any of the inversions, gene duplications, insertions, inverted repeat expansions and gene/intron losses that have been found in the chloroplast genomes of the genera <it>Jasminum </it>and <it>Menodora</it>, from the same family as <it>Olea</it>.</p> <p>The annotated sequence was used to evaluate the content of coding genes, the extent, and distribution of repeated and long dispersed sequences and the nucleotide composition pattern. These analyses provided essential information for structural, functional and comparative genomic studies in olive plastids. Furthermore, the alignment of the olive plastome sequence to those of other varieties and species identified 30 new organellar polymorphisms within the cultivated olive.</p> <p>Conclusions</p> <p>In addition to identifying mutations that may play a functional role in modifying the metabolism and adaptation of olive cultivars, the new chloroplast markers represent a valuable tool to assess the level of olive intercultivar plastome variation for use in population genetic analysis, phylogenesis, cultivar characterisation and DNA food tracking.</p

    Substernal oxyphil parathyroid adenoma producing PTHrP with hypercalcemia and normal PTH level

    <p>Abstract</p> <p>Background</p> <p>Parathyroid adenoma is the most common cause of primary hyperparathyroidism. Preoperative serum calcium and intact-parathyroid hormone levels are the most useful diagnostic parameters that allow differentiating primary hyperparathyroidism from non-parathyroid-dependent hypercalcemia. Parathyroidectomy is the definitive treatment for primary hyperparathyroidism. Approximately 5% of patients who underwent parathyroidectomy present with persistent or recurrent hyperparathyroidism due to ectopic localization of the adenoma. Functioning oxyphil parathyroid adenoma is an uncommon histological form, seldom causing primary hyperparathyroidism. Parathyroid adenoma with hypercalcemia exhibiting normal parathyroid hormone level is rare. An incidence of 5% to 33% has been documented in the literature; no etiologic explanation has been given. In 1987, parathyroid-hormone-related peptide was isolated as a causative factor of humeral hypercalcemia of malignancy. The presence of parathyroid-hormone-related peptide in parathyroid tissue under normal and pathological conditions has been described in the literature; however, its role in causing hyperparathyroidism has not yet been defined.</p> <p>Case presentation</p> <p>We present a case of persistent hypercalcemia with a normal level of intact-parathyroid hormone due to a substernal parathyroid adenoma, treated with radioguided parathyroidectomy. The final histological diagnosis was oxyphil adenoma, positive for parathyroid-hormone-related peptide antigens.</p> <p>Conclusion</p> <p>In clinical practice, this atypical biochemical presentation of primary hyperparathyroidism should be considered in the differential diagnosis of hypercalcemia. The parathyroid-hormone-related peptide should be considered not only in the presence of malignancy.</p

    Phenotypic selection mosaic for flower length influenced by geographically varying hawkmoth pollinator proboscis length and abiotic environment

    Biotic and abiotic context may affect the intensity of interspecific interactions and subsequently drive locally particular phenotypic selection patterns on interacting traits. We evaluated the geographical variation of matching traits of the brush‐type flowers of Caesalpinia gilliesii and of the proboscis length of its guild of hawkmoth pollinators, as well as their relation with environmental variables. We assessed the geographical variation of interacting traits (style and filament vs. mean proboscis length of the guild of hawkmoths) across seven populations, and estimated phenotypic selection on the plant side. Interacting traits showed similar relationships with environmental variables. Phenotypic selection on the plant side was influenced by proboscis length and by environmental conditions. Mean proboscis length of the guild was shorter than previously recorded for the same study area thus probably shifting the selective optima of flower length. We observed two presumptive coevolutionary cold spots where one‐sided negative directional selection is acting on style length. The lack of selection on the pollinator‐side should be further confirmed. We provided joint evidence, mostly lacking, about the geographical variation of selective pressures on the plant side associated with both proboscis length and abiotic conditions.     Identification of Possible Pre-Slaughter Indicators to Predict Stress and Meat Quality: A Study on Heavy Pigs

    This study aimed at identifying possible pre-slaughter indicators and/or indexes to be used to predict pig stress response and meat quality variation. Data were collected on 44 shipments (loads) of Italian heavy pigs. For each shipment, several pre-slaughter parameters were recorded on farm, during transport, and at the slaughterhouse. Blood and meat samples were taken from 10 pigs from every of the 44 loads included in the study (N = 440). Blood samples were used to assess cortisol and creatine kinase levels, whereas meat samples were used to assess meat quality (pH, instrumental color, tenderness, water-holding capacity, and sensory analysis). Cluster analysis of blood parameters allowed the categorization of the shipments into two main clusters: Lower Stress (LS) and Higher Stress (HS). The variables/indexes statistically differing between the two clusters were: average vehicle speed during transport, welfare index at slaughter (i.e., \u201cslaughter score\u201d), overall transport and slaughter welfare index (TSWI), distance travelled, and behaviors (slips, falls, overlaps) during unloading, which appeared to be the best descriptors of the welfare conditions experienced by Italian heavy pigs during pre-slaughter handling. No consistent effects of the stress level experienced on meat quality was detected, which warrants the need for further studies conducted under more variable pre-slaughter conditions, with the aim of simplifying and improving the TSWI

    Addition of the tumour-stroma ratio to the 8th edition American Joint Committee on Cancer staging system improves survival prediction for patients with oral tongue squamous cell carcinoma

    Aims One of the objectives of current research is to customise the treatment of cancer patients. The achievement of this objective requires stratification of patients based on the most significant prognostic factors. The aims of this study were to evaluate the prognostic value of the tumour-stroma ratio (TSR), defined as the proportion of tumour cells relative to surrounding stroma, in patients with oral tongue squamous cell carcinoma (OTSCC), and to develop a prognostic nomogram based on the most significant clinicopathological features. Methods and results Clinicopathological data of 211 patients treated at 'Ospedali Riuniti' General Hospital (Ancona, Italy) for OTSCC were collected. One hundred and thirty-nine patients were restaged according to the 8th edition American Joint Committee on Cancer (AJCC) staging system. Evaluation of the TSR was performed on haematoxylin and eosin-stained slides, and correlation with survival outcomes was evaluated. In addition, with the aim of integrating the independent value of the TSR with the 8th edition AJCC staging system, a prognostic nomogram for OTSCC has been developed. OTSCC with a low TSR (i.e. a high proportion of stroma and a low proportion of tumour cells) was shown to have negative prognostic value in terms of disease-specific survival, with a hazard ratio (HR) of 1.883 and a 95% confidence interval (CI) of 1.033-3.432 (P = 0.039), and overall survival (HR = 1.747, 95% CI 0.967-3.154;P = 0.044), independently of other histological and clinical parameters. For the cohort of 139 patients restaged according to the 8th edition AJCC staging system, variables correlating with a poor prognosis were: the TSR, perineural invasion, and sex. The nomogram built on these parameters showed good predictive capacity, outperforming the 8th edition AJCC staging system in stratifying disease-specific survival in OTSCC patients. Conclusions Including the TSR in the predictive model could improve risk stratification of OTSCC patients and aid in making treatment decisions.Peer reviewe
