    Coherence in the age of statutes

    The High Court of Australia, in pursuing coherence between common law and statute law, has limited itself to ensuring that the rules of common law and statute law should be free of contradiction. The Court does not appear to have embraced the idea, which lies at the core of some major theories of private law, that a set of rules is coherent only if the set can be explained as the outworking of a single principle. Applying that idea to the relationship between common law and statute law is confronted by some serious challenges. In the past, coherence as non-contradiction (combined with the idea of parliamentary supremacy) has worked well as a means of reconciling common law with statute law, but the proliferation of legislation in recent years and the character of much modern legislation has drawn attention to the limitations of such an approach to the question. A more exacting approach to coherence of common law and statute law, on the other hand, would require the revision of some widely held assumptions about the nature of law, such as the core assumption of legal positivism that the ultimate criterion of the authority of the law is its pronouncement by an authoritative institution

    Double suicide genes selectively kill human umbilical vein endothelial cells

    <p>Abstract</p> <p>Background</p> <p>To construct a recombinant adenovirus containing CDglyTK double suicide genes and evaluate the killing effect of the double suicide genes driven by kinase domain insert containing receptor (KDR) promoter on human umbilical vein endothelial cells.</p> <p>Methods</p> <p>Human KDR promoter, <it>Escherichia coli </it>(<it>E. coli</it>) cytosine deaminase (CD) gene and the herpes simplex virus-thymidine kinase (TK) gene were cloned using polymerase chain reaction (PCR). Plasmid pKDR-CDglyTK was constructed with the KDR promoter and CDglyTK genes. A recombinant adenoviral plasmid AdKDR-CDglyTK was then constructed and transfected into 293 packaging cells to grow and harvest adenoviruses. KDR-expressing human umbilical vein endothelial cells (ECV304) and KDR-negative liver cancer cell line (HepG2) were infected with the recombinant adenoviruses at different multiplicity of infection (MOI). The infection rate was measured by green fluorescent protein (GFP) expression. The infected cells were cultured in culture media containing different concentrations of prodrugs ganciclovir (GCV) and/or 5-fluorocytosine (5-FC). The killing effects were measured using two different methods, i.e. annexin V-FITC staining and terminal transferase-mediated dUTP nick end-labeling (TUNEL) staining.</p> <p>Results</p> <p>Recombinant adenoviruses AdKDR-CDglyTK were successfully constructed and they infected ECV304 and HepG2 cells efficiently. The infection rate was dependent on MOI of recombinant adenoviruses. ECV304 cells infected with AdKDR-CDglyTK were highly sensitive to GCV and 5-FC. The cell survival rate was dependent on both the concentration of the prodrugs and the MOI of recombinant adenoviruses. In contrast, there were no killing effects in the HepG2 cells. The combination of two prodrugs was much more effective in killing ECV304 cells than GCV or 5-FC alone. The growth of transgenic ECV304 cells was suppressed in the presence of prodrugs.</p> <p>Conclusion</p> <p>AdKDR-CDglyTK/double prodrog system may be a useful method for suppressing tumor angiogenesis.</p

    Massively Parallel Sequencing and Analysis of the Necator americanus Transcriptome

    The blood-feeding hookworm Necator americanus infects hundreds of millions of people. To elucidate fundamental molecular biological aspects of this hookworm, the transcriptome of adult Necator americanus was studied using next-generation sequencing and in silico analyses. Contigs (n = 19,997) were assembled from the sequence data; 6,771 of them had known orthologues in the free-living nematode Caenorhabditis elegans, and most encoded proteins with WD40 repeats (10.6%), proteinase inhibitors (7.8%) or calcium-binding EF-hand proteins (6.7%). Bioinformatic analyses inferred that C. elegans homologues are involved mainly in biological pathways linked to ribosome biogenesis (70%), oxidative phosphorylation (63%) and/or proteases (60%). Comparative analyses of the transcriptomes of N. americanus and the canine hookworm, Ancylostoma caninum, revealed qualitative and quantitative differences. Essential molecules were predicted using a combination of orthology mapping and functional data available for C. elegans. Further analyses allowed the prioritization of 18 predicted drug targets which did not have human homologues. These candidate targets were inferred to be linked to mitochondrial metabolism or amino acid synthesis.     Diagnosing cross-shelf transport along an ocean front: an observational case study in the Gulf of Lion

    Exchanges between coastal regions and the open ocean are often associated with intermittent and localized processes such as eddies, fronts and filaments. Since these features are difficult to observe, their impact has been predominantly investigated using numerical models and remote sensing. In this study, satellite sea surface temperature maps, Lagrangian surface drifter trajectories, and ship-based surveys of currents and hydrography from the Latex10 campaign are used to quantify cross-shelf exchanges associated with a temperature front in the western Gulf of Lion. Satellite imagery and thermosalinograph sections provide the characterization of the various water masses associated with the front. Lagrangian drifter trajectories are used to identify the main transport structures and to quantify the velocity components associated with near-inertial oscillations. These are removed from the instantaneous ADCP observations with which the cross-shelf exchanges are then computed. The results indicate an average outflow of 0.074 ±0.013 Sv and an inflow of 0.021 ±0.006 Sv. Integrated over the two-week lifetime of the front, such outflow induced a total export of ∼ 90 ± 14 km3 of water, indicating that 3 to 4 of such events are sufficient to completely renew the surface waters of the Gulf of Lion. The total import was ∼ 25 ± 7 km3, suggesting larger inflows at depth or in the eastern part of the gulf to maintain its volume balance. These in-situ estimates represent a key term of comparison for the further development of numerical model- and satellite-based studies of cross-shelf exchanges associated with this type of processes

    Renal transport kinetics of furosemide in the isolated perfused rat kidney

    Direct quantitative data and corresponding theory are provided for the effect of protein binding on the renal transport of furosemide. Drug studies were performed with various combinations of bovine serum albumin and dextran. This resulted in a percent unbound ( fu ) of furosemide ranging from 0.785 to 85.8%. The corrected renal ( CLr/GFR ) and secretion ( CLs/GFR ) clearances of furosemide were observed to increase with percent free, but in a nonproportional manner. Plots of CLr/GFR or CLs/GFR vs. fu appeared to have a prominent y intercept as well as a convex ascending curve. In addition, the excretion ratio [ ER=CLr/ (fu · GFR) ] was reduced from 60.8 to 8.72 as fu increased. Overall, the data were best fitted to a model in which two Michaelis-Menten terms wre used to describe renal tubular transport, and secretion was dependent upon free drug concentrations in the perfusate. The results demonstrate that the renal mechanisms of furosemide excretion are more complex than previously reported and that active secretion may involve two different transport systems over the concentration range studied.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/45036/1/10928_2005_Article_BF01065259.pd
