    Probabilistic risk assessment of the environmental impacts of pesticides in the Crocodile (west) Marico catchment, North-West Province

    External agricultural inputs, such as pesticides, may pose risks to aquatic ecosystems and affect aquatic populations, communities and ecosystems. To predict these risks, a tiered approach was followed, incorporating both the PRIMET and PERPEST models. The first-tier PRIMET model is designed to yield a relatively worst-case risk assessment requiring a minimum of input data, after which the effects of the risks can be refined using a higher tier PERPEST model. The risk assessment initially depends on data supplied from local landowners, pesticide characteristic, application scheme and physical scenario of the environment under question. Preliminary results are presented, together with ecotoxicological data on several frequently-used pesticides in a section of the Crocodile (west) Marico Water Management Area (WMA) in South Africa. This area is historically known to have a high pesticide usage, with deltamethrin, aldicarb, parathion, cypermethrin and dichlorvos being the main pesticides used. Deltamethrin was indicated as having the highest probability of risks to aquatic organisms occurring in the study area. Cypermethrin, parathion, dichlorvos, carbaryl, romoxynil, linuron, methomyl and aldicarb were all indicated as having possible risks (ETR 1-100) to the aquatic environment. The pesticides posing a possible risk to the aquatic ecosystem were evaluated further to determine their effects on 8 grouped endpoints using the PERPEST effect model. Deltamethrin and cypermethrin were again noted as posing the greatest risk and clear effects were eminent for aquatic insects and macro-crustaceans, followed by micro-crustaceans and rotifers. High percentages of clear effects on insects were also observed for carbaryl, parathion and dichlorvos. Linuron was indicated as having minimal clear effects on community metabolism, macrophytes and phytoplankton classes, while lesser clear effects of bromoxynil occurred on periphyton communities. Application of both the lower-tier PRIMET and higher-tier PERPEST models showed similar trends in that they both ranked the top 5 pesticides in the same order of risk. This approach offers a significant improvement over the presently-used simulation models or use of safety factors. It is therefore especially useful in developing countries such as South Africa, where pesticide environmental risk information is scarce.     Deep-water antipatharians: Proxies of environmental change

    Deep-water (307–697 m) antipatharian (black coral) specimens were collected from the southeastern continental slope of the United States and the north-central Gulf of Mexico. The sclerochronology of the specimens indicates that skeletal growth takes place by formation of concentric coeval layers. We used 210Pb to estimate radial growth rate of two specimens, and to establish that they were several centuries old. Bands were delaminated in KOH and analyzed for carbon and nitrogen stable isotopes. Carbon values ranged from _16.4‰ to _15.7‰; the oldest specimen displayed the largest range in values. Nitrogen values ranged from 7.7‰ to 8.6‰. Two specimens from the same location and depth had similar 15N signatures, indicating good reproducibility between specimens

    There is a wide range of sensory therapies using sound, music and visual stimuli. Some focus on soothing or distracting stimuli such as natural sounds or classical music as analgesic, while other approaches emphasize the active performance of producing music as therapy. This paper proposes an immersive multi-sensory Exposure Therapy for people suffering from anxiety disorders, based on a rich, detailed surround-soundscape. This soundscape is composed to include the users’ own idiosyncratic anxiety triggers as a form of habituation, and to provoke psychological catharsis, as a non-verbal, visceral and enveloping exposure. To accurately pinpoint the most effective sounds and to optimally compose the soundscape we will monitor the participants’ physiological responses such as electroencephalography, respiration, electromyography, and heart rate during exposure. We hypothesize that such physiologically optimized sensory landscapes will aid the development of future immersive therapies for various psychological conditions, Sound is a major trigger of anxiety, and auditory hypersensitivity is an extremely problematic symptom. Exposure to stress-inducing sounds can free anxiety sufferers from entrenched avoidance behaviors, teaching physiological coping strategies and encouraging resolution of the psychological issues agitated by the sound

    Developing oral chronotherapy for cortisol replacement in congenital adrenal hyperplasia

    The sun imposes a 24-h periodicity to life and circadian rhythms have evolved to maintain homoeostasis through the day/night cycle. In humans, there is a central clock that controls the sleep/wake cycle which is paralleled metabolically by a fast/feed cycle. The clock maintains homoeostasis by synchronising metabolism to the time of feeding. Loss of synchrony between the clock and hormonal rhythms results in loss of homoeostasis as evidenced by obesity, depression, and diabetes in people undertaking shift work. Cortisol has a distinct circadian rhythm; peaking on waking and low at sleep onset. Loss of this rhythm in adrenal insufficiency is associated with a poor quality of life and increased mortality. To replace the cortisol rhythm requires chronotherapy and for this you need to define the key parameters of the target rhythm, create a formulation to replicate that rhythm, and then prove clinical benefit. The physiology of hormones is more complex than that of nonnative drugs. Hormones are secreted with varied rhythms, bound to multiple cognate binding proteins, and actively transported and cleared through enzymatic pathways in multiple organs. We have examined the diurnal rhythm of cortisol in healthy volunteers, created physiologically-based pharmacokinetic models, and tested various oral delayed and sustained formulations of hydrocortisone (development name, Chronocort) in clinical trials. The outcome from this work was the manufacture of modified-release hydrocortisone hard capsules (tradename Efmody, Diurnal Ltd), that replicate the cortisol diurnal rhythm and improve the disease control of congenital adrenal hyperplasia the commonest hereditary form of adrenal insufficiency

    In this inspirational note, we describe the development of an endocrine chronotherapy to restore the physiological rhythm of the essential adrenal stress hormone, cortisol. The challenges included demonstrating the circadian rhythm of the drug target, creating a drug formulation that replicated that rhythm and then proving benefit in clinical trials. The physiological cortisol circadian rhythm is well defined with cortisol levels high on waking and low on going to sleep. We experimented with different formulation technologies including modified-release tablets and multi-particulates to replicate the cortisol rhythm where absent through disease. We describe the development of Efmody®, a modified-release formulation of hydrocortisone, which replicates the cortisol diurnal rhythm and improves the disease control of congenital adrenal hyperplasia, the commonest hereditary form of adrenal insufficiency. This program shows it is possible, through modified-release technology, to treat chronic endocrine diseases with physiological replacement to preserve health for life

    Subglacial lakes and hydrology across the Ellsworth Subglacial Highlands, West Antarctica

    Subglacial water plays an important role in ice sheet dynamics and stability. Subglacial lakes are often located at the onset of ice streams and have been hypothesised to enhance ice flow downstream by lubricating the ice– bed interface. The most recent subglacial-lake inventory of Antarctica mapped nearly 400 lakes, of which ∼ 14 % are found in West Antarctica. Despite the potential importance of subglacial water for ice dynamics, there is a lack of detailed subglacial-water characterisation in West Antarctica. Using radio-echo sounding data, we analyse the ice–bed interface to detect subglacial lakes. We report 33 previously uncharted subglacial lakes and present a systematic analysis of their physical properties. This represents a ∼ 40 % increase in subglacial lakes in West Antarctica. Additionally, a new digital elevation model of basal topography of the Ellsworth Subglacial Highlands was built and used to create a hydropotential model to simulate the subglacial hydrological network. This allows us to characterise basal hydrology, determine subglacial water catchments and assess their connectivity. We show that the simulated subglacial hydrological catchments of the Rutford Ice Stream, Pine Island Glacier and Thwaites Glacier do not correspond to their ice surface catchments

    Duality between Electric and Magnetic Black Holes

    A number of attempts have recently been made to extend the conjectured SS duality of Yang Mills theory to gravity. Central to these speculations has been the belief that electrically and magnetically charged black holes, the solitons of quantum gravity, have identical quantum properties. This is not obvious, because although duality is a symmetry of the classical equations of motion, it changes the sign of the Maxwell action. Nevertheless, we show that the chemical potential and charge projection that one has to introduce for electric but not magnetic black holes exactly compensate for the difference in action in the semi-classical approximation. In particular, we show that the pair production of electric black holes is not a runaway process, as one might think if one just went by the action of the relevant instanton. We also comment on the definition of the entropy in cosmological situations, and show that we need to be more careful when defining the entropy than we are in an asymptotically-flat case.Comment: 23 pages, revtex, no figures.     Threshold criterion for wetting at the triple point

    Grand canonical simulations are used to calculate adsorption isotherms of various classical gases on alkali metal and Mg surfaces. Ab initio adsorption potentials and Lennard-Jones gas-gas interactions are used. Depending on the system, the resulting behavior can be nonwetting for all temperatures studied, complete wetting, or (in the intermediate case) exhibit a wetting transition. An unusual variety of wetting transitions at the triple point is found in the case of a specific adsorption potential of intermediate strength. The general threshold for wetting near the triple point is found to be close to that predicted with a heuristic model of Cheng et al. This same conclusion was drawn in a recent experimental and simulation study of Ar on CO_2 by Mistura et al. These results imply that a dimensionless wetting parameter w is useful for predicting whether wetting behavior is present at and above the triple temperature.
