    The effect of smoking during pregnancy on severity and directionality of externalizing and internalizing symptoms: A genetically informed approach

    The objective was to examine the association between maternal smoking during pregnancy (SDP) and (I) severity and (II) directionality of externalizing and internalizing symptoms in a sample of sibling pairs while rigorously controlling for familial confounds. The Missouri Mothers and Their Children Study is a family study (N = 173 families) with sibling pairs (aged 7 to 16 years) who are discordant for exposure to SDP. This sibling comparison study is designed to disentangle the effects of SDP from familial confounds. An SDP severity score was created for each child using a combination of SDP indicators (timing, duration, and amount). Principal component analysis of externalizing and internalizing behavior, assessed with the Child Behavior Checklist and Teacher Report Form, was used to create symptom severity and directionality scores. The variance in severity and directionality scores was primarily a function of differences between siblings (71% and 85%, respectively) rather than differences across families (29% and 15%, respectively). The severity score that combines externalizing and internalizing symptom severity was not associated with SDP. However, a significant within-family effect of SDP on symptom directionality (b = 0.07

    Examining the Factor Structure of the Home Mathematics Environment to Delineate Its Role in Predicting Preschool Numeracy, Mathematical Language, and Spatial Skills

    A growing body of evidence suggests that the ways in which parents and preschool children interact in terms of home-based mathematics activities (i.e., the home mathematics environment; HME) is related to children’s mathematics development (e.g., primarily numeracy skills and spatial skills); however, this body of evidence is mixed with some research supporting the relation and others finding null effects. Importantly, few studies have explicitly examined the factor structure of the HME and contrasted multiple hypothesized models. To develop more precise models of how the HME supports children’s mathematics development, the structure of the HME needs to be examined and linked to mathematics performance. The purpose of this study was to extend prior work by replicating the factor structure of the HME (as one general HME factor and three specific factors of direct numeracy, indirect numeracy, and spatial) and using those factors to predict direct assessments of children’s numeracy, mathematical language, and spatial skills. It was hypothesized that the general HME factor would be related to each direct assessment, the direct numeracy factor would be related to both numeracy and mathematical language, and the spatial factor would be related to spatial skills. Using a sample of 129 preschool children (M age = 4.71 years, SD = 0.55; 46.5% female), a series of confirmatory factor analyses were conducted. Results diverged somewhat from prior work as the best fitting model was a bifactor model with a general HME factor and two specific factors (one that combined direct and indirect numeracy activities and another of spatial activities) rather than three specific factors as had previously been found. Further, structural equation modeling analyses suggested that, in contrast to expectations, only the direct + indirect numeracy factor was a significant predictor of direct child assessments when accounting for age, sex, and parental education. These findings provide evidence that a bifactor model is important in understanding the structure of the HME, but only one specific factor is related to children’s outcomes. Delineating the structure of the HME, and how specific facets of the HME relate to children’s mathematics skills, provides a strong foundation for understanding and enhancing the mechanisms that support mathematics development

    Drug Repurposing: A Systematic Approach to Evaluate Candidate Oral Neuroprotective Interventions for Secondary Progressive Multiple Sclerosis

    Objective: To develop and implement an evidence based framework to select, from drugs already licenced, candidate oral neuroprotective drugs to be tested in secondary progressive multiple sclerosis. Design: Systematic review of clinical studies of oral putative neuroprotective therapies in MS and four other neurodegenerative diseases with shared pathological features, followed by systematic review and meta-analyses of the in vivo experimental data for those interventions. We presented summary data to an international multi-disciplinary committee, which assessed each drug in turn using pre-specified criteria including consideration of mechanism of action. Results: We identified a short list of fifty-two candidate interventions. After review of all clinical and pre-clinical evidence we identified ibudilast, riluzole, amiloride, pirfenidone, fluoxetine, oxcarbazepine, and the polyunsaturated fatty-acid class (Linoleic Acid, Lipoic acid; Omega-3 fatty acid, Max EPA oil) as lead candidates for clinical evaluation. Conclusions: We demonstrate a standardised and systematic approach to candidate identification for drug rescue and repurposing trials that can be applied widely to neurodegenerative disorders

    Contextual Influences of Prenatal and Postnatal Environments on Executive Function Risk for Adolescent Substance Use

    Due to the great transitions and turmoil uniquely attributed to the period of adolescence, youth experience a greater risk for substance use and the multitude of concerns that coincide with the early onset of substance use. Many biological and environmental factors have been investigated as predictors of adolescent substance use. Executive function and disruptive behaviors are two important individual characteristics linked to adolescent substance use. Both smoking during pregnancy and sibling relationships are separate contexts that can mitigate or exacerbate the associations of executive function and adolescent substance use. The present study focuses on development of substance use through executive function deficits and disruptive behavior, while considering smoking during pregnancy and sibling relationships as unique moderators of these pathways. This work addresses a novel, interrelated set of questions with a series of three studies. The central hypothesis driving this program of research is that smoking during pregnancy and sibling relationships are under-studied contexts that can mitigate or exacerbate the associations of executive function, disruptive behavior, and adolescent substance use. This dissertation examines whether: (1) executive function mediates the smoking during pregnancy-disruptive behavior association and smoking during pregnancy exacerbates the executive functiondisruptive behavior association, (2) smoking during pregnancy exacerbates the association between executive function and disruptive behavior during adolescence using a sibling comparison design, and (3) sibling relationship quality moderates developmental trajectories of executive function on the transition from disruptive problems to adolescent substance use using a high-risk, longitudinal sample. Findings challenge the link between exposure to smoking during pregnancy and both executive function and disruptive behavior. Further, these findings reinforce the need to utilize genetically-informed designs when examining potential effects of smoking during pregnancy. Additionally, this dissertation found support for the link between executive function and disruptive behavior, but not executive function and substance use

    Peer Deviance, Adolescent Substance Use and The Moderating Role of Parental Monitoring

    Low parental monitoring (Barnes et al., 2006; Kiesner et al., 2010) and associating with deviant peers (Bahr et al., 2005; Barnes et al., 2006) has been linked to adolescents’ substance use. Studies examining parenting and peers together typically assess whether low monitoring has an effect on substance use through increasing youths’ ability to engage with deviant peers (Dishion et al., 2004; Pires & Jenkins, 2007). Fewer studies have investigated how these two factors interact to influence adolescent substance use, but suggest that the effect of peer deviance is stronger for youth experiencing less monitoring (Kiesner et al., 2010). The current study extends this work by examining mothers and fathers and older and younger siblings separately. We hypothesized that peer deviance would be more strongly associated with adolescent substance use when parental monitoring is low. Utilizing data from 395 families (father, mother, older sibling and younger sibling) from the US-based Nonshared Environmental in Adolescent Development study (NEAD; Neiderhiser et al., 2007), we ran a series of hierarchical regressions testing interactions of parental monitoring and peer deviance predicting adolescent substance use. For younger siblings, the effect of peer deviance was strongest when maternal monitoring was low (ÎČ = .07, p = .036). However, contrary to our hypothesis, for older siblings, the effect of peer deviance was strongest when paternal monitoring was high (ÎČ = -.13, p = .001). Results indicate that for younger siblings with highly deviant peers, maternal monitoring plays a protective role, but for older siblings monitoring may be too restrictive and exacerbate risk associated with deviant peers

    Reliability of prospective and retrospective maternal reports of prenatal experiences

    Abstract Background Extant perinatal research utilizes retrospective reports on the prenatal environment, but there are limited data on the validity of retrospective data compared with prospective data. The current study examined the reliability of birth mothers’ memory of prenatal stress and distress and perinatal risks at 6-months postpartum with maternal reports gathered across each trimester of pregnancy and explored whether recall varied with maternal socioeconomic status. Methods Surveys were collected from 34 pregnant women (M age = 29.14, SD = 5.06 years, 83% non-Hispanic White) on stress, distress, and pregnancy complications at 12(T1), 26(T2), and 38(T3) weeks of pregnancy, and at 6-month post-partum asking the same questions but specifically about the pregnancy. Cohen’s kappa and Pearson’s correlations were used to investigate maternal recall at post-partum with prospective reports at T1, T2, T3 and an average score of T1, T2, and T3. Correlations were also examined separately for those with high and relatively lower socioeconomic status. Results Birth mothers’ recall was generally reliable. Retrospective reports were most strongly related to prospective reports in T1 for perceived stress, T1 and T3 for anxiety symptoms and exposure to toxins, but T3 for depressive symptoms. Recall of pregnancy complications best reflected the average score across trimesters (rather than specific trimesters). Women with higher socioeconomic status better recalled prenatal (di)stress, but women with relatively lower socioeconomic status better recalled exposure to toxins. Conclusion This study provides support for utilizing retrospective reports of maternal prenatal experiences at 6-months post-partum, with implications for interpretation of specific recalled phenotypes

    Absence Makes the Heart Grow Fonder : A Qualitative Examination of Sibling Relationships During Emerging Adulthood

    The present study explored the unique experiences of siblings during emerging adulthood. Using a qualitative design, this study sought to better understand sibling relationship dynamics (e.g., closeness and negativity), communication strategies (e.g., frequency, quality, and form), and the implications of siblings\u27 proximity with one another. Importantly, the period of emerging adulthood provides a unique context in which these experiences might be especially salient. We posited the following research questions: What do sibling relationships look like during emerging adulthood? What does communication look like between siblings during emerging adulthood and how does it impact their relationship? and What implications does proximity have for the sibling relationship? To answer these questions, nine different qualitative focus groups (N = 45 participants) were conducted across two different university settings. Results suggested that siblings remained important in the lives of emerging adults, despite a lack of physical proximity and less frequent communication. Participants indicated that although communication was less frequent, the content of the discussions was often more meaningful. When sharing in-person conversations, participants described the relationships to have picked up where they had left off, with no time passing in between. Further, sibling relationship quality appeared to improve with participants, indicating they were happier with their sibling and felt more like equals and had a better understanding of one another. These findings help to fill a gap in knowledge about sibling relationships during emerging adulthood and elucidate our understanding of how these relationships are maintained and developed during this period of transition

    Parental Differential Treatment of Siblings and Adolescents\u27 Health-Related Behaviors: The Moderating Role of Personality

    Youth who receive comparatively poorer parental treatment than a sibling are at risk for maladaptive behaviors in a variety of domains, but research has yet to examine links with adolescents\u27 health-related behaviors nor consider how those links may vary based on adolescents\u27 personality traits, namely conscientiousness and agreeableness. Two siblings (n = 590 adolescents; 53% female; Mage = 15.86, SD = 1.73) from 295 families reported on their differential conflict and closeness with their fathers and mothers as well as on their personality, sleep habits, exercise habits, and general health habits. Multilevel modeling revealed that, generally, the less conscientious adolescents had better health habits when they had comparatively warmer relationships with their mothers. Less conscientiousness adolescents may be less distressed by inequality in the family, and thus may experience positive effects of relatively better treatment
