    Obesity‐associated mutant melanocortin‐4 receptors with normal Gαs coupling frequently exhibit other discoverable pharmacological and biochemical defects

    Mutations in the melanocortin‐4 receptor (MC4R) are the most common cause of early syndromic obesity known. Most of these mutations result in a loss of protein expression, α‐melanocyte‐stimulating hormone binding, receptor trafficking or coupling to the stimulatory G‐protein, Gαs. However, approximately 26% of the obesity‐associated mutations characterised to date exhibit none of these pharmacological defects. In the present study, we investigated seven of these apparently normal mutant MC4R in more detail and found that the majority (five of the seven) exhibit marked defects including defective binding of another endogenous melanocortin ligand, defective glycosylation, and defective recruitment of β‐arrestin. These data provide support for two hypotheses: (i) that the majority of these rare, obesity‐associated mutations are likely defective and causative of obesity and (ii) that β‐arrestin recruitment is a valuable marker of normal MC4R function.     In vivo Spectroscopic and Imaging Studies of Photosensitizer in Photodynamic Therapy

    Photodynamic Therapy (PDT) has emerged as a useful cancer treatment modality which utilizes a tumor localizing dye and activating light to selectively destroy neoplastic tissue. In an effort to understand the newly synthesized photosensitizers, we are studying them in a mouse tumor model grown on the dorsal side of the foot by in vivo magnetic resonance techniques. We have synthesized several photosensitizers which are specifically labeled with fluorine. Several coils appropriate for the tumor study by 19F NMR were designed and constructed for this project. The solenoid coil tunable to both 1Hand 19F nuclei was used to monitor the 19F labeled photosensitizer in the mouse foot tumor. An in vivo 19F NMR technique was used to study the retention of the photosensitizer over time in the tumor. We have used 31P NMR to study the outcome of PDT after using the new photosensitizer

    Organic Centre Wales (OCW) was originally formed to act as an information source for producers enquiring into organic farming; in our second phase, in response to changes in the sector, we have an increased remit on policy issues. It has been a busy time, seeing the production of a second Organic Action Plan, the implementation of the CAP reform and development work on agri-environment reforms. The year also saw the start of organic maintenance payments for farmers. This Annual Report contains accounts of the progress that has been made on many fronts during the last 12 months, particularly in market knowledge due to the research for the recently published report, 'Organic Food: understanding the consumer and increasing sales'. This information, together with continuing product development work, will enable Welsh organic producers to be proactive in building a solid consumer base

    A Further Look at Potential Impact of Satlets on Design, Production, and Cost of Satellite Systems

    For the past 50 years, the morphology for satellites has remained fundamentally unchanged despite evolutions in manufacturing, communications, and software occurring in other industries. Primary spacecraft support systems—power, attitude control, and others—are designed in the same way, whether in space telescopes, large communications satellites, interplanetary spacecraft, or Cubesats. This paradigm has been the status quo in spacecraft design and construction and has precluded any industry-wide, large-scale cost savings while maintaining performance. To change this trend and ensure performance and utility at low cost, that can scale, DARPA postulated the concept of a cellularized satellite, or “satlet,” as a satellite architectural unit. In this new morphology, each satlet would provide some fraction of the overall functions that, when aggregated via hardware and software, provide spacecraft space system with its complete required capabilities. The DARPA Phoenix program has developed this satlet morphology in Phase I and plans to validate and demonstrate it in a series of steps that exercise various applications and levels of configuration flexibility enabled by a satlet architecture. The first system experiment is planned to be conducted on orbit in 2015. This paper aims to take a deeper look at the potential impact of space systems with cellular based designs, and using historical data showcases how design, production and ultimately cost can form the foundation for next generation spacecraft opportunities. A first order analysis conducted in a previous paper indicated that U.S.-launched satellites alone could create a market demand for 2,000-8,000 satlets flown per year, while the overall annual world satellite market could create demand for 10,000-40,000 satlets. This paper explores the instantiation of a cellular morphology to design, production and development to further quantify the impact of this revolutionary space system capability

    Increased signal complexity is associated with increased mating success

    The evolution of complex signals has often been explored by testing multiple functional hypotheses regarding how independent signal components provide selective benefits to offset the costs of their production. In the present study, we take a different approach by exploring the function of complexity per se. We test the hypothesis that increased vibratory signal complexity—based on both proportional and temporal patterning—provides selective benefits to courting male Schizocosa stridulans wolf spiders. In support of this hypothesis, all of our quantified metrics of vibratory signal complexity predicted the mating success of male S. stridulans. The rate of visual signalling, which is mechanistically tied to vibratory signal production, was also associated with mating success. We additionally found evidence that males can dynamically adjust the complexity of their vibratory signalling. Together, our results suggest that complexity per se may be a target of female choice
