Evaluation Of Biochemical And Serological Methods To Identify And Clustering Yeast Cells Of Oral Candida Species By Chromagar Test, Sds-page And Elisa.
The purpose of this work was to evaluate biochemical and serological methods to characterize and identify Candida species from the oral cavity. The strains used were five Candida species previously identified: C. albicans, C. guilliermondii, C. parapsilosis, C. krusei, C. tropicalis, and Kluyveromyces marxianus, as a negative control. The analyses were conducted through the SDS-PAGE associated with statistical analysis using software, chromogenic medium, and CHROMagar Candida (CA), as a differential medium for the isolation and presumptive identification of clinically important yeasts and an enzyme-linked immunoabsorbent assay (ELISA), using antisera produced against antigens from two C. albicans strains. This method enabled the screening of the three Candida species: C. albicans, C. tropicalis, and C. krusei, with 100% of specificity. The ELISA using purified immunoglobulin G showed a high level of cross-reaction against protein extracts of Candida species. The SDS-PAGE method allowed the clustering of species-specific isolates using the Simple Matching coefficient, S(SM) = 1.0. The protein profile analysis by SDS-PAGE increases what is known about the taxonomic relationships among oral yeasts. This methodology showed good reproducibility and allows collection of useful information for numerical analysis on information relevant to clinical application, and epidemiological and systematical studies.64231732
VIP enhances TRH-stimulated prolactin secretion of pituitary tumours Studies with 31P NMR
AbstractIntravenous thyrotrophin releasing hormone (TRH) caused a 6.5-fold increase in plasma prolactin (PRL) in rats carrying implanted pituitary tumours. Vasoactive intestinal polypeptide (VIP) had no effect, but TRH given after VIP raised TRH stimulated secretion 13-fold above basal. 31P NMR spectroscopy showed that VIP caused a decrease in high energy metabolites (depleted phosphocreatine, elevated inorganic phosphate and lowered intracellular pH). TRH alone caused a similar but smaller effect; given after VIP, it caused no detectable depletion. We suggest that the changes in high energy metabolite cencentrations reflect increased cellular energy consumption consistent with a priming process (stage 1) in PRL secretion, followed by hormone release (stage 2). Membrane-associated proteins of ejaculated sperm from Morada Nova rams
AbstractThe objective was to describe the profile of membrane proteins from sperm of tropically adapted Morada Nova rams (N = 5). Samples from protein-enriched fractions of ejaculated sperm (containing 400 μg of protein) were separated by two-dimensional electrophoresis and respective maps analyzed using PDQuest software (version 7.3.0; Bio-Rad). Proteins were identified using tandem mass spectrometry. Also, membrane proteins were incubated with antibodies against binder of sperm protein (BSP) 1 and bodhesin 2 (Bdh-2), components of vesicular gland secretion. For membrane proteins of ejaculated sperm, an average of 133 ± 4.6 spots were detected per gel, of which, 107 spots were consistently present on all gels. Sixty-eight spots and 37 proteins were identified using mass spectrometry, corresponding to 71.6% of the intensity of all spots detected. Three major spots identified as ram seminal vesicle protein (RSVP) 14 represented approximately 30% of the intensity of all spots. Two of the most intense spots in the gel reacted against anti-BSP1, at 14 kDa. In addition, four low molecular weight spots reacted with anti-Bdh-2 antibodies. Proteins RSVP and Bdh-2 belong to the BSP and spermadhesin families, respectively, and were previously reported as major components of ram seminal proteins. Additional proteins identified in the sperm membrane two-dimensional maps included alpha-2-heparan sulfate-glycoprotein, plasma glutamate carboxypeptidase, arylsulfatase A, cathelicidin, heat shock protein 70 kDa, angiotensin-converting enzyme, leucine aminopeptidase, and clusterin. Some proteins were present as multiple isoforms, such as tubulin (12), alpha-2-heparan sulfate-glycoprotein (5), ATP synthase (5), Bdh-2 (4) and RSVP14 (3). Based on gene ontology analysis, the most common biological processes associated with the membrane proteins were cellular processes (34%), response to stimulus (14%), and metabolic processes (11%). Binding (37%) and catalytic activity (32%) corresponded to the most frequent molecular functions for those proteins. In conclusion, we identified a diverse cohort of components of membrane proteins in ram sperm. Major proteins previously reported in seminal plasma, such as RSVP14 and Bdh-2, were also extracted from sperm membranes. Knowledge of sperm proteins is crucial for elucidating mechanisms underlying their association with sperm function

Occurrence and exposure of 3-monochloropropanediol diesters in edible oils and oil-based foodstuffs from the Spanish market
During the industrial refining process of edible oils and the manufacture of oil-based foodstuff, contaminants such as 3-monochloropropanediol (3-MCPD) fatty acid diesters can be produced. One hundred samples of different edible oils and related fatty food purchased from local Spanish markets were analyzed to evaluate the occurrence of these contaminants. Data of seven 3-MCPD diesters together with corresponding total 3-MCPD equivalents are presented. The procedure is based on a modified QuEChERS protocol followed by LC-MS/MS analysis. Extra virgin olive oil (EVOO) and unrefined oils did not contain detectable levels of the target analytes. The highest levels of 3-MCPD diesters were found in palm oils, for 1,2-Dilinoleoyl-3-chloropropanediol (LILI) and 1–2-Bispalmitoyl-3-chloropropanediol (PAPA) with concentrations close to 10 mg kg−1 and in the lipid fraction of margarines (8.09, 3.77 and 3.72 mg kg−1 for LILI, PAPA and 1-Oleoyl-2-linoleoyl-3-chloropropanediol (OLLI), respectively).Spanish Ministry of Science and Innovation (Project AGL-2014-53647-R) and FEDER fund
In Vitro Activity Of Artemisia Annua L (asteraceae) Extracts Against Rhipicephalus (boophilus) Microplus [atividade In Vitro De Extratos De Artemisia Annua L (asteraceae) Sobre Rhipicephalus (boophilus) Microplus]
The activity of plant extracts on parasites may indicate groups of substances that are potentially useful for controlling Rhipicephalus (Boophilus) microplus. The aim of the present study was to investigate the in vitro action of Artemisia annua extracts on this tick. The concentrations of the sesquiterpene lactones artemisinin and deoxyartemisinin present in plant extracts were quantified via high-performance liquid chromatography. Four extracts produced from the concentrated crude extract (CCE) were evaluated on larvae using the impregnated paper method, with readings after 24 hours of incubation. The engorged females were immersed in the CCE and in its four derived extracts for five minutes, with incubation for subsequent analysis of biological parameters. The extracts were not effective on the larvae at the concentrations tested (3.1 to 50 mg.mL-1). The CCE showed greater efficacy on engorged females (EC50 of 130.6 mg.mL-1 and EC90 of 302.9 mg.mL-1) than did the derived extracts. These results tend to confirm that the action of artemisinin on engorged females of R. (B.) microplus is conditional to their blood intake. In this case, in vitro methods would be inadequate for effective evaluation of the action of A. annua on R. Influence of Insulin-like Growth Factor I (IGF-I) on the survival and the in vitro development of caprine preantral follicles
The aim of this study was to investigate the effects of the insulin-like growth factor -I (IGF-I) on survival, activation (transition from primordial to primary follicles) and growth of caprine preantral follicles cultured in vitro. Fragments of ovarian cortex were cultured for one and seven days in the absence or presence of IGF-I (0, 50 and 100ng/ml). The non-cultured and cultured tissues were processed and analyzed by histology and transmission electron microscopy. The culture for one day in a medium with 100ng/ml of IGF-I showed 86.7% of morphologically normal follicles. These results were similar (P>0.05) to the percentage of normal follicles found in the control (96.7%). It was also found that this medium increased the percentage of follicular activation (developing follicles) with one day of culture. The oocyte and follicular diameters remained similar to the control by culturing for one day in a medium containing 100ng/ml of IGF-I. The ultrastructural analysis did not confirm the integrity of the follicular fragments in a medium containing IGF-I (100ng/ml) after one and seven days of culture. In conclusion, this study demonstrated that the addition of 100 ng/ml of IGF-I in the culture medium enables the development of preantral follicles of goats with one day of culture. However, it is not sufficient to maintain the follicular integrity and the follicular survival rate after seven days of culture.O objetivo do presente estudo foi investigar os efeitos do fator de crescimento semelhante a insulina-I (IGF-I) na sobrevivência, ativação (transição de folículos primordiais para primários) e crescimento de folículos pré-antrais caprinos cultivados in vitro. Fragmentos de córtex ovariano foram cultivados por um e sete dias na ausência ou presença de IGF-I (0, 50 e 100ng/mL). Os tecidos não cultivados e cultivados foram processados e analisados por histologia e microscopia eletrônica de transmissão. O cultivo por um dia em meio com 100ng/mL de IGF-I apresentou 86,7% de folículos morfologicamente normais. Estes resultados foram semelhantes (P>0,05) ao percentual de folículos normais encontrados no controle (96,7%). Verificou-se ainda que este meio aumentou o percentual de ativação folicular (folículos em desenvolvimento) com um dia de cultivo. Os diâmetros ovocitário e folicular mantiveram-se semelhantes ao controle ao cultivar por um dia em meio contendo 100ng/mL de IGF-I. As análises ultraestruturais não confirmaram a integridade folicular dos fragmentos em meio contendo IGF-I (100 ng/mL) após um e sete dias de cultivo. Em conclusão, esse estudo demonstrou que a adição de 100 ng/mL de IGF-I no meio de cultivo ativa o desenvolvimento de folículos pré-antrais de caprinos com um dia de cultivo. Entretanto, não é suficiente para manter a integridade folicular e a taxa de sobrevivência folicular após sete dias de cultivo
Seasonal approach to forecast the suitability of spawning habitats of a temperate small pelagic fish under a high-emission climate change scenario
ABSTRACT: Spawning habitats of cold-water, European small pelagic fishes have shifted
poleward in the last three decades coincident with gradual ocean warming. We
predicted present-day, season-specific habitat suitability for spawning by
European sardine Sardina pichardus in the Atlantic Ocean and Mediterranean
and Black Seas, and projected climate-driven changes in suitable areas from
2050-2099 under the IPCC – RCP 8.5 scenario. Sea surface temperature and
distance to the coast had the greater influences in spawning habitats, reflecting
the temperature- and coastal-dependent spawning of sardines. Chlorophyll-a
was the third most important explanatory variable for spawning in winter to
summer. Winds were predominantly important during autumn, whilst sea
surface salinity was an important driver during spring and summer. Presentday, “hotspots” for spawning were identified in regions of highly productive,
salty waters, where SST was between 6 and 18°C from autumn to spring or 16
and 25°C during summer and favourable winds occurred that would retain eggs
and larvae closer to the coast (< 250 km). For future scenarios, forecasts
indicate that environmental optima for spawning is projected to be in regions
where SST varies between 11°C and 18°C from autumn to spring; and between
18°C and 24°C during summer. However, a negative relationship between
phytoplankton productivity and habitat suitability induced by warming is likely
to occur in the future. Projections suggest that suitable spawning habitats in all
seasons will shift to higher latitudes, with a prominent range expansion along
the coast of Norway during winter and autumn (> 83%). The total spawning
area, however, was projected to contract in the future during spring (-10.5%)
and autumn (-4.1%) due to losses of currently suitable areas along the Atlantic
African Coast and Mediterranean Sea. Such regions currently support the greatest sardine stocks but climate-driven warming and decreased plankton
productivity are projected to make these areas unsuitable for spawning and

Technical and economic pre-feasibility study for the construction of septic tank-filter-sinkhole with alternative material

facilitate and spread the use of this sewage treatment system in places that are not assisted by
municipal sewage systems and in the rural area, which despite having a smaller number of
inhabitants compared to the urban area This study aims to carry out a technical and economic
evaluation of the concrete and tires using in the construction of septic tanks-filter-sinkhole. The
wastewater treatment systems were built according to the recommendations in NBR 7229/93 and
13969/97. To evaluate the efficiency of each system built, the following parameters were
analyzed: chemical oxygen demand (COD), the potential of hydrogen (pH), alkalinity, acidity,
and temperature. In the economic evaluation, the materials and labor required to install the
systems were considered using the Brazilian cost database (SINAPI), and an economic and
financial feasibility study was carried out. According to the technical and economic analysis of
construction, both systems showed the same technical performance, however, the concrete design
proved to be more advantageous than the tire design, considering the difficulty in acquiring the
tires and the high cost if it is necessary to buy them, in addition to the greater difficulty in handling
and installing the tire system compared to the concrete one
PROFIT: a new alternative for emission-line PROfile FITting
I briefly describe a simple routine for emission-line profiles fitting by
Gaussian curves or Gauss-Hermite series. The PROFIT (line-PROfile FITting)
routine represent a new alternative for use in fits data cubes, as those from
Integral Field Spectroscopy or Fabry-Perot Interferometry, and may be useful to
better study the emission-line flux distributions and gas kinematics in
distinct astrophysical objects, such as the central regions of galaxies and
star forming regions. The PROFIT routine is written in IDL language and is
available at http://www.ufsm.br/rogemar/software.html.
The PROFIT routine was used to fit the [Fe II]1.257um emission-line profiles
for about 1800 spectra of the inner 350 pc of the Seyfert galaxy Mrk1066
obtained with Gemini NIFS and shows that the line profiles are better
reproduced by Gauss-Hermite series than by the commonly used Gaussian curves.
The two-dimensional map of the h_3 Gauss-Hermite moment shows its highest
absolute values in regions close to the edge of the radio structure. These high
values may be originated in an biconical outflowing gas associated with the
radio jet - previously observed in the optical [O III] emission. The analysis
of this kinematic component indicates that the radio jet leaves the center of
the galaxy with the north-west side slightly oriented towards us and the
