    Understanding the role of E2F7 in the regulation of cellular proliferation and DNA damage responses.

    188 p.E2F transcription factors control diverse biological processes through regulation of target gene expression. The identification of a large set of genes regulated by each individual E2F, including those coding for microRNAs, has led to a better understanding of the functions performed by the different members of the family. Many studies have detailed the role of classical E2Fs in cell cycle control and DNA damage response. By contrast, the contribution of the atypical members of the family, E2F7 and E2F8, to these processes has not been clearly defined. A recent study from our group identified a set of novel microRNAs and protein-coding genes regulated by E2F7. These genes are involved in processes such as cell cycle regulation or DNA damage response. In this work, we have examined the role that E2F7 plays in the regulation of these processes through the transcriptional regulation of its target genes. We have identified E2F7 as a transcription factor required for the repression of a set of microRNAs that promote cellular proliferation. We show that miR-25, miR-92 and miR-7 expression is controlled at the transcriptional level by the antagonistic activity of E2F7 and E2F1-3. Interestingly, we find that several E2F7-repressed microRNAs downregulate the expression of cell cycle progression inhibitors and promote cellular proliferation, suggesting that E2F7 restrains cell cycle progression through repression of proliferation-promoting microRNAs.Importantly, we show that E2F7 plays a key role in the maintenance of genomic stability. We present evidence of E2F7-dependent transcriptional and non-transcriptional mechanisms for modulating cellular responses to genotoxic exposure. We identify an E2F7-dependent transcriptional regulation program that restricts homologous recombination-mediated DNA repair and cellular recovery upon induction of DNA lesions that interfere with replication fork progression (DNA interstrand cross-links and PARP1 inhibition).Additionally, we present evidence of a non-transcriptional mechanism by which E2F7 modulates cellular responses to alkylating DNA damage, possibly involving interaction with the repair protein XRRC1. Loss of E2F7 confers an increased resistance to chemotherapy in homologous recombination-deficient cells, a potentially harmful outcome for cancer treatment. Altogether, results in this work reveal a key role for E2F7 in limiting cellular proliferation and promoting genomic stability by ensuring the timely expression of protein-coding and microRNA genes that are required for cell cycle progression and DNA damage repair

    OFAR-CM: Efficient Dragonfly networks with simple congestion management

    Dragonfly networks are appealing topologies for large-scale Data center and HPC networks, that provide high throughput with low diameter and moderate cost. However, they are prone to congestion under certain frequent traffic patterns that saturate specific network links. Adaptive non-minimal routing can be used to avoid such congestion. That kind of routing employs longer paths to circumvent local or global congested links. However, if a distance-based deadlock avoidance mechanism is employed, more Virtual Channels (VCs) are required, what increases design complexity and cost. OFAR (On-the-Fly Adaptive Routing) is a previously proposed routing that decouples VCs from deadlock avoidance, making local and global misrouting affordable. However, the severity of congestion with OFAR is higher, as it relies on an escape sub network with low bisection bandwidth. Additionally, OFAR allows for unlimited misroutings on the escape sub network, leading to unbounded paths in the network and long latencies. In this paper we propose and evaluate OFAR-CM, a variant of OFAR combined with a simple congestion management (CM) mechanism which only relies on local information, specifically the credit count of the output ports in the local router. With simple escape sub networks such as a Hamiltonian ring or a tree, OFAR outperforms former proposals with distance-based deadlock avoidance. Additionally, although long paths are allowed in theory, in practice packets arrive at their destination in a small number of hops. Altogether, OFAR-CM constitutes the first practicable mechanism to the date that supports both local and global misrouting in Dragonfly networks.The research leading to these results has received funding from the European Research Council under the European Union’s Seventh Framework Programme (FP/2007-2013) / ERC Grant Agreement n.     Este artículo forma parte de un proyecto de investigación más extenso, encaminado a analizar y traducir los textos freudianos inéditos en español. El objetivo de este artículo es ofrecer la traducción y el comentario del informe que resume el contenido de la exposición sobre histeria que Sigmund Freud dictara, en tres partes, en octubre de 1895 ante los integrantes del Colegio Médico de Viena. Se pone el énfasis aquí en el modo en que ese material esclarece un momento esencial de su pensamiento.     La transformación experimentada en el mercado de trabajo en los últimos años ha supuesto que los jóvenes se encuentren en una situación de desventaja. Las cifras revelan un panorama laboral bastante eclipsado para este colectivo, pues la tasa de paro se ha triplicado desde 2008 (INE, 2017). Tomando como referencia la Teoría de la Autoeficacia de Bandura (1977), la percepción de empleabilidad puede constituir un elemento fundamental que determine cómo los jóvenes se enfrentan a la inserción del mundo laboral. Esta hace referencia a las posibilidades de un individuo para obtener o conservar un trabajo en un contexto sociolaboral determinado (Fugate, Kinicki y Ashforth, 2004; Thijssen, Van der Heijden y Rocco, 2008). En el contexto de globalización neoliberal se aprecia una redirección y decantación de las mujeres al mercado del trabajo en los espacios rurales, poniendo de manifiesto: a) la necesidad de incorporación de más miembros del grupo doméstico en la búsqueda de ingresos, b) la multiplicación de sus jornadas de trabajo, c) prácticas poco justas y equitativas dentro y fuera del hogar con respecto al trabajo, y d) mayor tensión entre los miembros del hogar en el momento de la toma de decisiones respecto al trabajo fuera del hogar por parte de ésta

    Contrasting Response to Drought and Climate of Planted and Natural Pinus pinaster Aiton Forests in Southern Spain

    Extreme drought events and increasing aridity are leading to forest decline and tree mortality, particularly in populations near the limits of the species distribution. Therefore, a better understanding of the growth response to drought and climate change could show the vulnerability of forests and enable predictions of future dieback. In this study, we used a dendrochronological approach to assess the response to drought in natural and planted forests of the maritime pine (Pinus pinaster Aiton) located in its southernmost distribution (south of Spain). In addition, we investigated how environmental variables (climatic and site conditions) and structural factors drive radial growth along the biogeographic and ecological gradients. Our results showed contrasting growth responses to drought of natural and planted stands, but these differences were not significant after repeated drought periods. Additionally, we found differences in the climate–growth relationships when comparing more inland sites (wet previous winter and late spring precipitation) and sites located closer to the coast (early spring precipitation). Response functions emphasized the negative effect of defoliation and drought, expressed as the June standard precipitation-evapotranspiration index calculated for the 12-month temporal scale and the mean temperature in the current February, on growth. The strong relationship between climatic variables and growth enabled acceptable results to be obtained in a modeling approach. The study and characterization of this tree species’ response to drought will help to improve the adaptive management of forests under climate change

    Los países Hispanoamericanos aspiran dentro del marco de sus políticas públicas a un crecimiento que incluya a todos sus ciudadanos. Con esta investigación de corte hermenéutico nos hemos propuesto contribuir al desarrollo de un sólido sistema de protección social. Este sistema es un mecanismo que permite generar condiciones que brinden seguridad a las personas durante toda su vida, garantizándoles derechos sociales para reducir los riesgos en empleo, salud, educación y previsión, creando condiciones de mayor igualdad y de oportunidades de progreso. Motivados por el requerimiento continuo de proveer información adecuada para la toma de decisiones, como resultado de esta investigación hemos formulado un modelo de gestión y/o política pública para Colombia que permita la generación de estilos de vida saludables en los lugares de trabajo bajo el marco conceptual de la Protección Social Hispano – Americana. Reconociendo que la salud presenta múltiples dimensiones y la necesidad de incorporar diversas miradas, ponemos a disposición de ustedes nuestra propuesta de modelo. Esperamos que este documento, y los que se entregarán a futuro, incentiven la participación de todos ustedes en el desarrollo e implementación de intervenciones que vayan en beneficio de mejorar la salud y calidad de vida de la población Hispano- Americana, en especial la Colombiana.The Hispanic countries inspire within their frame of their public politics a development where all the citizens are included. With this investigation of hermeneutic court we are proposing to contribute to a development of a solid system of Social Protection. This system is a mechanism that permits to generate conditions to give security to the people during all their lives, guarantying social rights to reduce the risks of jobs, health, education and forecast, creating conditions of mayor equality and opportunities to progress. Motivated by the continued requirement to provide adequate information to take decisions, like result of this investigation we formulated a model of gestation and or public politic for Colombia that permits the generation of healthy styles of life in places of work under the conceptual frame of The Social Protection Hispanic- American.
