    Modafinil induces rapid-onset behavioral sensitization and cross-sensitization with cocaine in mice: implications for the addictive potential of modafinil

The rapid-onset type of behavioral sensitization (i.e., a type of sensitization that develops within a few hours from the drug priming administration) has been emerged as a valuable tool to study binge-like patterns of drug abuse and the neuroplastic changes that occur quickly after drug administration that ultimately lead to drug abuse. Our aim was to investigate the possible development of rapid-onset behavioral sensitization to modafinil and bidirectional rapid-onset cross sensitization with cocaine in male Swiss mice. A priming injection of a high dose of modafinil (64 mg/kg) induced rapid-onset behavioral sensitization to challenge injections of modafinil at the doses of 16, 32, and 64 mg/kg, administered 4 h later. Furthermore, rapid-onset cross-sensitization was developed between modafinil and cocaine (64 mg/kg modafinil and 20 mg/kg cocaine), in a bidirectional way. These results were not due to residual levels of modafinil as the behavioral effects of the priming injection of modafinil were no longer present and modafinil plasma concentration was reduced at 4 h post-administration.     The Combined Use of Melatonin and an Indoleamine 2,3-Dioxygenase-1 Inhibitor Enhances Vaccine-Induced Protective Cellular Immunity to HPV16-Associated Tumors

    Immunotherapy has become an important ally in the fight against distinct types of cancer. However, the metabolic plasticity of the tumor environment frequently influences the efficacy of therapeutic procedures, including those based on immunological tools. In this scenario, immunometabolic adjuvants arise as an alternative toward the development of more efficient cancer therapies. Here we demonstrated that the combination of melatonin, a neuroimmunomodulator molecule, and an indoleamine 2,3-dioxygenase (IDO) inhibitor (1-methyl-DL-tryptophan, DL-1MT) improves the efficacy of an immunotherapy (gDE7) targeting human papillomavirus (HPV)-associated tumors. Melatonin or IDO inhibitors (D-1MT and DL-1MT) directly reduced proliferation, migration, adhesion and viability of a tumor cell line (TC-1), capable to express the HPV-16 E6 and E7 oncoproteins, but could not confer in vivo antitumor protection effects. Nonetheless, combination of gDE7 with melatonin or D-1MT or DL-1MT enhanced the antitumor protective immunity of gDE7-based vaccine in mice. Notably, expression of IDO1 in stromal cells and/or immune cells, but not in tumor cells, inhibited the antitumor effects of the gDE7, as demonstrated in IDO1-deficient mice. Finally, co-administration of gDE7, melatonin and DL-1MT further improved the protective antitumor effects and the numbers of circulating E7-specific CD8+ T cells in mice previously transplanted with TC-1 cells. The unprecedented combination of melatonin and IDO inhibitors, as immunometabolic adjuvants, thus, represents a new and promising alternative for improving the efficacy of immunotherapeutic treatments of HPV-associated tumors

    Electrocardiographic evaluation of horses after polo exercise

    A eletrocardiografia pode ser usada para quantificar o treinamento e o desempenho atlético. Pode ainda avaliar a função cardíaca sob efeito do exercício identificando a influência de anomalias cardíacas, assim como os efeitos deletérios do esforço frente à função cardíaca. Considerando a complexidade do esforço físico inerente a cavalos em competições de polo, juntamente com a carência de relatos na literatura sobre a demanda cardíaca resultante. O objetivo deste estudo foi o de identificar e avaliar eletrocardiograficamente os efeitos cardíacos do exercício de polo em equinos de forma a subsidiar dados para a compreensão da demanda fisiológica cardíaca desta modalidade. Foram avaliados 27 equinos praticante de polo em repouso e entre cinco e dez minutos após o exercício. Os resultados obtidos demonstraram que as alterações observadas na duração e amplitude da onda P e duração do intervalo PR e QT foram consideradas fisiológicas em resposta ao aumento da frequência cardíaca. A onda P difásica encontrada em repouso representou hipertrofia atrial e quando bífida representou diferentes pontos de ativação do nó sinoatrial. Como esperado o complexo QRS não sofreu alterações consideráveis entre os dois momentos. O supradesnível do segmento ST e as variações da onda T observadas após o exercício poderiam representar efeito adverso ao miocárdio, entretanto estudos analisando múltiplos fatores são necessários para confirmar esta associação e definir sua real causa. O aumento do QTc sugeriu fadiga miocárdica leve representando alta demanda cardíaca para esta modalidade. O escore cardíaco demonstrou que estes animais estavam dentro do padrão de adaptação cardíaca para um cavalo atleta. A rápida recuperação cardíaca demonstrou bom condicionamento atlético. O marca-passo atrial mutável foi um achado normal encontrado nos equinos de polo. Foi observada baixa incidência de alterações no ritmo cardíaco.Electrocardiography can be used to quantify the training and athletic performance as well as evaluating the cardiac function under the effect of exercise identifying the influence of cardiac anomalies, and deleterious effects of stress ahead of heart function. Considering the complexity of the physical efforts inherent in polo ponies in competitions, together with the lack of reports in the literature, on the demand resulting for heart. The aim of this study was to identify and evaluate the cardiac effects of electrocardiographic exercise in polo horses in order to support data for understanding the physiological cardiac demand of this sport. We evaluated 27 equine practitioner polo at rest and between five and ten minutes after exercise. The results showed that the observed changes in the duration and P wave amplitude and duration of PR and QT interval were considered in physiological response to increased heart rate. The diphasic P wave found at rest and when atrial hypertrophy represented bifida represented different points of activation of the sinoatrial node. As expected, the QRS complex has not undergone major changes. The ST-segment elevation and T wave changes observed after exercise could pose adverse effects to the myocardium, but studies examining multiple factors are needed to confirm this association and define your actual question. The increase in QTc suggested fatigue mild myocardial represented high heart demand for this type of exercise. The heart score showed that these animals were within the pattern of cardiac adaptation to a horse athlete. The rapid recovery of HR showed good conditioning of these animals. The pacemaker migration was observed in most animals proven to be a normal finding horse athlete. We observed a low incidence of changes in cardiac rhythm

    Measurement of the cosmic ray spectrum above 4×10184{\times}10^{18} eV using inclined events detected with the Pierre Auger Observatory

    A measurement of the cosmic-ray spectrum for energies exceeding 4×10184{\times}10^{18} eV is presented, which is based on the analysis of showers with zenith angles greater than 6060^{\circ} detected with the Pierre Auger Observatory between 1 January 2004 and 31 December 2013. The measured spectrum confirms a flux suppression at the highest energies. Above 5.3×10185.3{\times}10^{18} eV, the "ankle", the flux can be described by a power law EγE^{-\gamma} with index γ=2.70±0.02(stat)±0.1(sys)\gamma=2.70 \pm 0.02 \,\text{(stat)} \pm 0.1\,\text{(sys)} followed by a smooth suppression region. For the energy (EsE_\text{s}) at which the spectral flux has fallen to one-half of its extrapolated value in the absence of suppression, we find Es=(5.12±0.25(stat)1.2+1.0(sys))×1019E_\text{s}=(5.12\pm0.25\,\text{(stat)}^{+1.0}_{-1.2}\,\text{(sys)}){\times}10^{19} eV.Comment: Replaced with published version.     Sintomas Cardiopulmonares Pós-COVID-19: Preditores e Características de Imagem de Pacientes após a Alta Hospitalar

    Resumo Fundamento A maioria da evidência sobre o impacto da síndrome COVID pós-aguda (PACS, do inglês, post-acute COVID-19 syndrome) descreve sintomas individuais sem correlacioná-los com exames de imagens. Objetivos Avaliar sintomas cardiopulmonares, seus preditores e imagens relacionadas em pacientes com COVID-19 após alta hospitalar. Métodos Pacientes consecutivos, que sobreviveram à COVID-19, foram contatados 90 dias após a alta hospitalar. A equipe de desfechos clínicos (cega quanto aos dados durante a internação) elaborou um questionário estruturado avaliando sintomas e estado clínico. Uma análise multivariada foi realizada abordando a evolução da COVID-19, comorbidades, ansiedade, depressão, e estresse pós-traumático durante a internação, e reabilitação cardíaca após a alta. O nível de significância usado nas análises foi de 5%. Resultados Foram incluídos 480 pacientes (idade 59±14 anos, 67,5% do sexo masculino) que receberam alta hospitalar por COVID-19; 22,3% necessitaram de ventilação mecânica. A prevalência de pacientes com sintomas cardiopulmonares relacionados à PACS (dispneia, cansaço/fadiga, tosse e desconforto no peito) foi de 16,3%. Vários parâmetros de tomografia computadorizada do tórax e de ecocardiograma foram similares entre os pacientes com e sem sintomas cardiopulmonares. A análise multivariada mostrou que sintomas cardiopulmonares foram relacionados de maneira independente com sexo feminino (OR 3,023; IC95% 1,319-6,929), trombose venosa profunda durante a internação (OR 13,689; IC95% 1,069-175,304), nível elevado de troponina (OR 1,355; IC95% 1,048-1,751) e de proteína C reativa durante a internação (OR 1,060; IC95% 1,023-1,097) e depressão (OR 6,110; IC95% 2,254-16,558). Conclusão Os sintomas cardiopulmonares relacionados à PACS 90 dias após a alta hospitalar são comuns e multifatoriais. Além dos marcadores trombóticos, inflamatórios e de lesão miocárdica durante a internação, sexo feminino e depressão foram associados independentemente com sintomas cardiopulmonares relacionados à PACS.
