Pre-Study Walkthrough with a Commercial Pilot for a Preliminary Single Pilot Operations Experiment
The number of crew members in commercial flights has decreased to two members, down from the five-member crew required 50 years ago. One question of interest is whether the crew should be reduced to one pilot. In order to determine the critical factors involved in safely transitioning to a single pilot, research must examine whether any performance deficits arise with the loss of a crew member. With a concrete understanding of the cognitive and behavioral role of a co-pilot, aeronautical technologies and procedures can be developed that make up for the removal of the second aircrew member. The current project describes a pre-study walkthrough process that can be used to help in the development of scenarios for testing future concepts and technologies for single pilot operations. Qualitative information regarding the tasks performed by the pilots can be extracted with this technique and adapted for future investigations of single pilot operations
A review of the toxicology of oil in vertebrates : what we have learned following the Deepwater Horizon oil spill

This is National Marine Mammal Foundation Contribution #314 to peer-reviewed scientific literature.In the wake of the Deepwater Horizon (DWH) oil spill, a number of government agencies, academic institutions, consultants, and nonprofit organizations conducted lab- and field-based research to understand the toxic effects of the oil. Lab testing was performed with a variety of fish, birds, turtles, and vertebrate cell lines (as well as invertebrates); field biologists conducted observations on fish, birds, turtles, and marine mammals; and epidemiologists carried out observational studies in humans. Eight years after the spill, scientists and resource managers held a workshop to summarize the similarities and differences in the effects of DWH oil on vertebrate taxa and to identify remaining gaps in our understanding of oil toxicity in wildlife and humans, building upon the cross-taxonomic synthesis initiated during the Natural Resource Damage Assessment. Across the studies, consistency was found in the types of toxic response observed in the different organisms. Impairment of stress responses and adrenal gland function, cardiotoxicity, immune system dysfunction, disruption of blood cells and their function, effects on locomotion, and oxidative damage were observed across taxa. This consistency suggests conservation in the mechanisms of action and disease pathogenesis. From a toxicological perspective, a logical progression of impacts was noted: from molecular and cellular effects that manifest as organ dysfunction, to systemic effects that compromise fitness, growth, reproductive potential, and survival. From a clinical perspective, adverse health effects from DWH oil spill exposure formed a suite of signs/symptomatic responses that at the highest doses/concentrations resulted in multi-organ system failure.Publisher PDFPeer reviewe
Detection and characterization of subvisible aggregates of monoclonal lgG in serum
To detect and characterize the aggregation of therapeutic monoclonal antibodies in undiluted biological fluids.
Fluorescently labeled subvisible IgG aggregates formed by applying either heat stress or by pH-shift were investigated immediately after addition to human serum, and after 24 h. Unstressed and stressed IgG formulations were analyzed by fluorescence single particle tracking, confocal laser scanning microscopy and flow cytometry.
Unstressed formulations remained free from subvisible aggregates in serum, whereas heat-stressed and pH-shift stressed formulations showed dissimilar aggregation behaviors. The aggregation profile of the heat-stressed formulation diluted in serum remained practically the same as the one diluted in buffer, even after the 24 h incubation period. The pH-shift stressed formulation had strikingly smaller and more numerous subvisible aggregates immediately after dilution in serum compared to buffer. These aggregates became noticeably larger in both diluents after 24 h, but in serum they appeared to be formed by other types of constituents than the labeled protein itself.
These results show that subvisible therapeutic protein aggregates may undergo changes in number, type and size distribution upon contact with human serum. This emphasizes the importance of analytical strategies for monitoring aggregation in undiluted biological fluids
Facial trauma: a recurrent disease? The potential role of disease prevention.
PURPOSE: The objectives of this investigation were to determine a profile of facial trauma patients presenting to the emergency department of University Hospital, University of Medicine and Dentistry of New Jersey, Newark, NJ, and to assess patient interest in violence or stress reduction programs.
MATERIALS AND METHODS: A prospective study of the patients was conducted with the aid of a data collection form generated specifically for the purpose of this study. Data regarding patient age, race, gender, various aspects of social history, mechanism, and nature of injury were collected over a period of 1 year. In addition, all patients were asked to respond to 4 health promotion questions. All patients for whom the oral maxillofacial surgery service was consulted were included in the study. Descriptive statistical analysis of the data was used.
RESULTS: A total of 92 patients were enrolled, of whom 80% were males. The mean age of patients enrolled was 30.5 years with the peak incidence of injury occurring in the 20- to 30-year-old age group (n = 30). The most frequent etiology was assault (75%), followed by motor vehicle accidents (18.5%). The most frequently occurring injury was mandible fracture (46.7%), followed by lacerations (42.4%). Within the study sample, 42.4% had previous injuries. Patients with facial injuries were 1.5 times more likely to have experienced previous interpersonal violence. This patient group also expressed an overwhelming willingness to change their behavior patterns (91.3%).
CONCLUSIONS: The findings of this investigation indicated that most facial trauma patients are between the ages of 20 and 30 years and male. Assault is the most common etiologic agent resulting in facial trauma. Mandible fractures and lacerations are the most likely injuries in the facial trauma patient. Patients experiencing recurrent trauma due to assault are more responsive to violence reduction programs than those experiencing only 1 assault
Cognitive phenotypes predict response to restorative cognitive rehabilitation in multiple sclerosis
Background: Cognitive phenotyping may be useful for predicting rehabilitation response in multiple sclerosis. Objective: To evaluate the association between cognitive phenotype(s) and response to restorative cognitive rehabilitation (RRCR). Methods: In a post hoc retrospective analysis of the RRCR study including 51 multiple sclerosis patients, we evaluated both impairment within specific cognitive domains as well as overall global impairment severity to investigate their relationship to improvement following rehabilitation. Results: Greater improvement in executive function was predicted by impairment within this domain as well as by having fewer impaired cognitive domains overall. Similar results were observed for visuospatial memory. Conclusions: Patients most likely to benefit from restorative cognitive rehabilitation may exhibit impairment within the domain of interest yet lower cognitive burden overall
