    Spiers Memorial Lecture: Molecular mechanics and molecular electronics

    We describe our research into building integrated molecular electronics circuitry for a diverse set of functions, and with a focus on the fundamental scientific issues that surround this project. In particular, we discuss experiments aimed at understanding the function of bistable [2]rotaxane molecular electronic switches by correlating the switching kinetics and ground state thermodynamic properties of those switches in various environments, ranging from the solution phase to a Langmuir monolayer of the switching molecules sandwiched between two electrodes. We discuss various devices, low bit-density memory circuits, and ultra-high density memory circuits that utilize the electrochemical switching characteristics of these molecules in conjunction with novel patterning methods. We also discuss interconnect schemes that are capable of bridging the micrometre to submicrometre length scales of conventional patterning approaches to the near-molecular length scales of the ultra-dense memory circuits. Finally, we discuss some of the challenges associated with fabricated ultra-dense molecular electronic integrated circuits

    Metabolomics should be deployed in the identification and characterization of gene-edited crops

    Abstract Gene editing techniques are currently revolutionizing biology allowing far greater precision than previous mutagenic and transgenic approaches. They are becoming applicable to a wide range of plant species and biological processes. Gene editing can rapidly improve a range of crop traits including disease resistance, abiotic stress tolerance, yield, nutritional quality and additional consumer traits. Unlike transgenic approaches, however, it is not facile to forensically detect gene-editing events at the molecular level, as no foreign DNA exists in the elite line. These limitations in molecular detection approaches are likely to focus more attention on the products generated from the technology, than the process per se. Rapid advances in sequencing and genome assembly increasingly facilitate genome sequencing as a means of characterizing new varieties generated by gene editing techniques. Nevertheless, subtle edits such as single base changes or small deletions may be difficult to distinguish from normal variation within a genotype. Given these emerging scenarios, downstream ā€˜omicsā€™ technologies reflective of edited affects, such as metabolomics, need to be utilized in a more prominent manner to fully assess compositional changes in novel foodstuffs. To achieve this goal, metabolomics or ā€œnon-targeted metabolite analysisā€ needs to make significant advances to deliver greater representation across the metabolome. With the emergence of new edited crop varieties we advocate; (i) concerted efforts in the advancement of ā€˜omicsā€™ technologies such as metabolomics and (ii) redress the use of the technology in the regulatory assessment for metabolically-engineered biotech crops

    Low-Dose Sodium Nitrite Attenuates Myocardial Ischemia and Vascular Ischemia-Reperfusion Injury in Human Models

    ObjectivesThe aim of this study was to assess the potential benefits of inorganic nitrite in 2 clinical models: stress-induced myocardial ischemia and whole-arm ischemia-reperfusion.BackgroundInorganic nitrite, traditionally considered a relatively inert metabolite of nitric oxide, may exert vasomodulatory and vasoprotective effects. Despite promising results from animal models, few have shown effectiveness in human model systems, and none have fully translated to the clinical setting.MethodsIn 10 patients with inducible myocardial ischemia, saline and low-dose sodium nitrite (NaNO2) (1.5 Ī¼mol/min for 20Ā min) were administered in a double-blind fashion during dobutamine stress echocardiography, at separate visits and in a random order; long-axis myocardial function was quantified by peak systolic velocity (Vs) and strain rate (SR) responses. In 19 healthy subjects, flow-mediated dilation was assessed before and after whole-arm ischemia-reperfusion; nitrite was given before ischemia or during reperfusion.ResultsComparing saline and nitrite infusions, Vs and SR at peak dobutamine increased in regions exhibiting ischemia (VsĀ from 9.5 Ā± 0.5 cm/s to 12.4 Ā± 0.6 cm/s, SR fromĀ āˆ’2.0 Ā± 0.2 sāˆ’1 toĀ āˆ’2.8 Ā± 0.3 sāˆ’1), whereas they did not change inĀ normally functioning regions (Vs from 12.6 Ā± 0.4 cm/s to 12.6 Ā± 0.6 cm/s, SR fromĀ āˆ’2.6 Ā± 0.3 sāˆ’1 toĀ āˆ’2.3 Ā± 0.1Ā sāˆ’1) (pĀ < 0.001, analysis of variance). With NaNO2, the increment of Vs (normalized for increase in heart rate) increased only in poorly functioning myocardial regions (+122%, pĀ < 0.001). Peak flow-mediated dilation decreased by 43% after ischemia-reperfusion when subjects received only saline (6.8 Ā± 0.7% vs. 3.9 Ā± 0.7%, pĀ < 0.01); administration of NaNO2 before ischemia prevented this decrease in flow-mediated dilation (5.9 Ā± 0.7% vs. 5.2 Ā± 0.5%, pĀ = NS), whereas administration during reperfusion did not.ConclusionsLow-dose NaNO2 improves functional responses in ischemic myocardium but has no effect on normal regions. Low-dose NaNO2 protects against vascular ischemia-reperfusion injury only when it is given before the onset of ischemia

    This document provides brief reports, or case studies, on a number of investigations and data set development activities sponsored by the Calibration and Validation Team (CVT) within the Sea-viewing Wide Field-of-view Sensor (SeaWiFS) Project. Chapter 1 is a comparison with the atmospheric correction of Coastal Zone Color Scanner (CZCS) data using two independent radiative transfer formulations. Chapter 2 is a study on lunar reflectance at the SeaWiFS wavelengths which was useful in establishing the SeaWiFS lunar gain. Chapter 3 reports the results of the first ground-based solar calibration of the SeaWiFS instrument. The experiment was repeated in the fall of 1993 after the instrument was modified to reduce stray light; the results from the second experiment will be provided in the next case studies volume. Chapter 4 is a laboratory experiment using trap detectors which may be useful tools in the calibration round-robin program. Chapter 5 is the original data format evaluation study conducted in 1992 which outlines the technical criteria used in considering three candidate formats, the hierarchical data format (HDF), the common data format (CDF), and the network CDF (netCDF). Chapter 6 summarizes the meteorological data sets accumulated during the first three years of CZCS operation which are being used for initial testing of the operational SeaWiFS algorithms and systems and would be used during a second global processing of the CZCS data set. Chapter 7 describes how near-real time surface meteorological and total ozone data required for the atmospheric correction algorithm will be retrieved and processed. Finally, Chapter 8 is a comparison of surface wind products from various operational meteorological centers and field observations. Surface winds are used in the atmospheric correction scheme to estimate glint and foam radiances

    Reporting randomised trials of social and psychological interventions: the CONSORT-SPI 2018 Extension

    Background: Randomised controlled trials (RCTs) are used to evaluate social and psychological interventions and inform policy decisions about them. Accurate, complete, and transparent reports of social and psychological intervention RCTs are essential for understanding their design, conduct, results, and the implications of the findings. However, the reporting of RCTs of social and psychological interventions remains suboptimal. The CONSORT Statement has improved the reporting of RCTs in biomedicine. A similar high-quality guideline is needed for the behavioural and social sciences. Our objective was to develop an official extension of the Consolidated Standards of Reporting Trials 2010 Statement (CONSORT 2010) for reporting RCTs of social and psychological interventions: CONSORT-SPI 2018. Methods: We followed best practices in developing the reporting guideline extension. First, we conducted a systematic review of existing reporting guidelines. We then conducted an online Delphi process including 384 international participants. In March 2014, we held a 3-day consensus meeting of 31 experts to determine the content of a checklist specifically targeting social and psychological intervention RCTs. Experts discussed previous research and methodological issues of particular relevance to social and psychological intervention RCTs. They then voted on proposed modifications or extensions of items from CONSORT 2010. Results: The CONSORT-SPI 2018 checklist extends 9 of the 25 items from CONSORT 2010: background and objectives, trial design, participants, interventions, statistical methods, participant flow, baseline data, outcomes and estimation, and funding. In addition, participants added a new item related to stakeholder involvement, and they modified aspects of the flow diagram related to participant recruitment and retention. Conclusions: Authors should use CONSORT-SPI 2018 to improve reporting of their social and psychological intervention RCTs. Journals should revise editorial policies and procedures to require use of reporting guidelines by authors and peer reviewers to produce manuscripts that allow readers to appraise study quality, evaluate the applicability of findings to their contexts, and replicate effective interventions

    In situ correlative observation of humping-induced cracking in directed energy deposition of nickel-based superalloys

    Directed energy deposition (DED) is a promising additive manufacturing technique for repair; however, DED is prone to surface waviness (humping) in thin-walled sections, which increases residual stresses and crack susceptibility, and lowers fatigue performance. Currently, the crack formation mechanism in DED is not well understood due to a lack of operando monitoring methods with high spatiotemporal resolution. Here, we use inline coherent imaging (ICI) to optically monitor surface topology and detect cracking in situ, coupled with synchrotron X-ray imaging for observing sub-surface crack healing and growth. For the first time, ICI was aligned off-axis (24Ā° relative to laser), enabling integration into a DED machine with no alterations to the laser delivery optics. We achieved accurate registration laterally (0.93), directly tracking surface roughness and waviness. We intentionally seed humping into thin-wall builds of nickel super-alloy CM247LC, locally inducing cracking in surface valleys. Crack openings as small as 7 Āµm were observed in situ using ICI, including sub-surface signal. By quantifying both humping and cracking, we demonstrate that ICI is a viable tool for in situ crack detection

    Tissue-specific transcriptional programming of macrophages controls the microRNA transcriptome targeting multiple functional pathways

    Recent interest in the biology and function of peritoneal tissue resident macrophages (pMĪ¦) has led to a better understanding of their cellular origin, programming and renewal. The programming of pMĪ¦ is dependent on microenvironmental cues and tissue specific transcription factors, including GATA6. However, the contribution of microRNAs remains poorly defined. We conducted a detailed analysis of the impact of GATA6-deficiency on microRNA expression in mouse pMĪ¦. Our data suggest that for many of the pMĪ¦, microRNA composition may be established during tissue specialization, and that the effect of GATA6 knockout is largely unable to be rescued in the adult by exogenous GATA6. The data are consistent with GATA6 modulating the expression pattern of specific microRNAs, directly or indirectly, and including miR-146a, -223, and -203 established by the lineage-determining transcription factor PU.1, to achieve a differentiated pMĪ¦ phenotype. Lastly, we showed a significant dysregulation of miR-708 in pMĪ¦ in the absence of GATA6 during homeostasis and in response to LPS/IFN-Ī³ stimulation. Overexpression of miR-708 in mouse pMĪ¦ in vivo altered 167 mRNA species demonstrating functional downregulation of predicted targets, including cell immune responses and cell cycle regulation. In conclusion, we demonstrate dependence of the microRNA transcriptome on tissue-specific programming of tissue macrophages as exemplified by the role of GATA6 in pMĪ¦ specialization
