    Ultraestructura de la red alveolar y su relación con el recubrimiento de las paredes vasculares en olmos infectados con Ophiostoma novo-ulmi y en otras plantas infectadas con enfermedades similares de marchitamiento

    In elms infected with Dutch elm disease, alveolar networks, demarcated by filamentous-like bands and confluent with similar matter (the coating) accumulating on vessel walls, occurred regularly in vessel elements. Similar material lined vessel walls in inoculated, sterilized, thin elm wood sections fixed by high pressure freezing. The coating was observed to connect with fungal cells and occasionally contained small opaque particles, the size of ribosomes, membranous and vesicular structures, and, following incubation of wood chips taken from diseased samples incubated on an agar medium, it still displayed similar matter. Coating and alveolar bands increased in thickness by confluence of other bands or membranous structures. Similar matter and structures also occurred in other plants affected by similar fungal wilt diseases. In all systems, the compact coating did not label for chitin, cellulose and pectin. In staghorn sumac, the probe for DNA attached to the coating. Altogether, in the light of these data, it appears that the coating and alveolar networks are not inert components, a fact which indicates their primordial probable pathogen origin. It is proposed that these elements might be important not only in the initial infection stages but also in older or recurrent infections at a time when host resistance mechanisms are ineffective.En olmos afectados por la grafiosis, la red alveolar, demarcada por bandas filamentosas, y confluente con acumulaciones de la misma sustancia (cubrición) presentes en las paredes de los vasos, aparece regularmente en los elementos conductores. Sustancias similares tapizan las paredes de los vasos en secciones de madera delgada de olmo inoculada y esterilizada, y posteriormente criofijadas a altas presiones. Se observó que la cubrición se conecta con las células del micelio y que ocasionalmente contenía pequeñas partículas opacas del tamaño de los ribosomas, estructuras membranosas y vesiculares, así como que, tras la incubación de astillas leñosas cogidas de muestras enfermas incubadas en agar, aún se presentaba una sustancia similar. La cubrición y las bandas alveolares aumentaron su espesor en la confluencia con otras bandas o estructuras membranosas. Estructuras y sustancias similares aparecieron también en otras plantas afectadas por enfermedades similares originadas por hongos que producen marchitamiento. En todos los sistemas, la cubrición compacta no pudo ser marcada como quitina, celulosa ni pectina. En zumaque (Rhus typhina), la sonda de ADN se pegó a la cubrición. En resumen, a la vista de estos datos, parece ser que la cubrición y la red alveolar no están formados por componentes inertes, un hecho que indica su probable origen patogénico. Se sugiere que esos elementos podrían ser importantes no sólo en las fases iniciales de la infección, sino también, en infecciones más desarrolladas o recurrentes, en el momento en que los mecanismos de resistencia del hospedante no son efectivos

    First Astronomical Use of Multiplexed Transition Edge Bolometers

    We present performance results based on the first astronomical use of multiplexed superconducting bolometers. The Fabry-Perot Interferometer Bolometer Research Experiment (FIBRE) is a broadband submillimeter spectrometer that achieved first light in June 2001 at the Caltech Submillimeter Observatory (CSO). FIBRE'S detectors are superconducting transition edge sensor (TES) bolometers read out by a SQUID multiplexer. The Fabry-Perot uses a low resolution grating to order sort the incoming light. A linear bolometer array consisting of 16 elements detects this dispersed light, capturing 5 orders simultaneously from one position on the sky. With tuning of the Fabry-Perot over one free spectral range, a spectrum covering Δλ/λ= 1/7 at a resolution of δλ/λ ≈ 1/1200 can be acquired. This spectral resolution is sufficient to resolve Doppler-broadened line emission from external galaxies. FIBRE operates in the 350 µm and 450 µm bands. These bands cover line emission from the important star formation tracers neutral carbon [Cl] and carbon monoxide (CO). We have verified that the multiplexed bolometers are photon noise limited even with the low power present in moderate resolution spectrometry

    The Complex and Important Cellular and Metabolic Functions of Saturated Fatty Acids

    This review summarizes recent findings on the metabolism and biological functions of saturated fatty acids (SFA). Some of these findings show that SFA may have important and specific roles in the cells. Elucidated biochemical mechanisms like protein acylation (N-myristoylation, S-palmitoylation) and regulation of gene transcription are presented. In terms of physiology, SFA are involved for instance in lipogenesis, fat deposition, polyunsaturated fatty acids bioavailability and apoptosis. The variety of their functions demonstrates that SFA should no longer be considered as a single group

    Virus-Induced Cancers of the Skin and Mucosa:Are We Dealing with "Smoking Guns" or "Smoke and Mirrors" in the Operating Theatre?

    Introduction: Human papillomavirus (HPV) alone is thought to cause ~610,000 cases of cancer per year, and is the dominant aetiological agent for ano-genital (esp. cervical) and head and neck cancers (esp. oropharyngeal). Merkel cell polyomavirus (MCV) is a more recently discovered virus which causes Merkel cell carcinoma, a rare but highly aggressive skin malignancy. Methods: We explored the available published evidence to see if transmission of live HPV or MCV virus in smoke generated by laser or diathermy was feasible, and would pose an infection risk. Long-term infection with such carcinogenic viruses would then pose an increased risk for the development of virus-induced cancers in medical personnel. Results: The morphological structures of both HPV and MCV are very similar, and the size, external capsids and genomic structures show striking similarity. Both viruses have a non-enveloped external protein capsid consisting of 72 capsomeres, and a double-stranded DNA core. Sizes of both viruses range from 50 to 60 nm. There are now recent data demonstrating live and infectious HPV in smoke, and that these viruses can be used to infect cells in vitro. Further, anecdotal reports of virus transmission leading to disease causation in the production of respiratory airway viral warts (benign disease), and, finally, reports of HPV-induced oropharyngeal carcinoma (malignant disease) in two gynaecological surgeons as an occupational health hazard have been published recently. Conclusion: There is now sufficient evidence to support the hypotheses that live infectious carcinogenic viruses can be transmitted via smoke generated from surgical procedures, and, in rare instances, actually cause significant disease. Protective measures such as smoke extraction and airway protection should be instituted for all healthcare personnel, particularly those with multiple repeated exposures such as gynaecological surgeons

    Promising role of preoperative neutrophil-to-lymphocyte ratio in patients treated with radical nephroureterectomy.

    Several retrospective studies with small cohorts reported neutrophil-to-lymphocyte ratio (NLR) as a prognostic marker in upper tract urothelial carcinoma (UTUC) following radical nephroureterectomy (RNU). We aimed at validating the predictive and prognostic role of NLR in a large multi-institutional cohort. Preoperative NLR was assessed in a multi-institutional cohort of 2477 patients with UTUC treated with RNU. Altered NLR was defined by a ratio >2.7. Logistic regression analyses were performed to assess the association between NLR and lymph node metastasis, muscle-invasive and non-organ-confined disease. The association of altered NLR with recurrence-free survival (RFS) and cancer-specific survival (CSS) was evaluated using Cox proportional hazards regression models. Altered NLR was observed in 1428 (62.8 %) patients and associated with more advanced pathological tumor stage, lymph node metastasis, lymphovascular invasion, tumor necrosis and sessile tumor architecture. In a preoperative model that included age, gender, tumor location and architecture, NLR was an independent predictive factor for the presence of lymph node metastasis, muscle-invasive and non-organ-confined disease (p < 0.001). Within a median follow-up of 40 months (IQR 20-76 months), 548 (24.1 %) patients experienced disease recurrence and 453 patients (19.9 %) died from their cancer. Compared to patients with normal NLR, those with altered NLR had worse RFS (0.003) and CSS (p = 0.002). In multivariable analyses that adjusted for the effects of standard clinicopathologic features, altered NLR did not retain an independent value. In the subgroup of patients treated with lymphadenectomy in addition to RNU, NLR was independently associated with CSS (p = 0.03). In UTUC, preoperative NLR is associated with adverse clinicopathologic features and independently predicts features of biologically and clinically aggressive UTUC such as lymph node metastasis, muscle-invasive or non-organ-confined status. NLR may help better risk stratify patients with regard to lymphadenectomy and conservative therapy
