    Combination of GNSS and SLR observations using satellite co-locations

    Satellite Laser Ranging (SLR) observations to Global Navigation Satellite System (GNSS) satellites may be used for several purposes. On one hand, the range measurement may be used as an independent validation for satellite orbits derived solely from GNSS microwave observations. On the other hand, both observation types may be analyzed together to generate a combined orbit. The latter procedure implies that one common set of orbit parameters is estimated from GNSS and SLR data. We performed such a combined processing of GNSS and SLR using the data of the year 2008. During this period, two GPS and four GLONASS satellites could be used as satellite co-locations. We focus on the general procedure for this type of combined processing and the impact on the terrestrial reference frame (including scale and geocenter), the GNSS satellite antenna offsets (SAO) and the SLR range biases. We show that the combination using only satellite co-locations as connection between GNSS and SLR is possible and allows the estimation of SLR station coordinates at the level of 1-2cm. The SLR observations to GNSS satellites provide the scale allowing the estimation of GNSS SAO without relying on the scale of any a priori terrestrial reference frame. We show that the necessity to estimate SLR range biases does not prohibit the estimation of GNSS SAO. A good distribution of SLR observations allows a common estimation of the two parameter types. The estimated corrections for the GNSS SAO are 119mm and −13mm on average for the GPS and GLONASS satellites, respectively. The resulting SLR range biases suggest that it might be sufficient to estimate one parameter per station representing a range bias common to all GNSS satellites. The estimated biases are in the range of a few centimeters up to 5cm. Scale differences of 0.9ppb are seen between GNSS and SL

    Propylene oxide, a favorite target of experimental and theoretical studies of circular dichroism, was recently discovered in interstellar space, further amplifying the attention to its role in the current debate on protobiological homochirality. In the present work, a photoelectron-photoion-photoion coincidence technique, using an ion-imaging detector and tunable synchrotron radiation in the 18.0-37.0 eV energy range, permits us (i) to observe six double ionization fragmentation channels, their relative yields being accounted for about two-thirds by the couple (C2H4+, CH2O+) and one-fifth by (C2H3+, CH3O+); (ii) to measure thresholds for their openings as a function of photon energy; and (iii) to unravel a pronounced bimodality for a kinetic-energy-released distribution, fingerprint of competitive non-adiabatic mechanisms

    Ketamine administration in idiopathic epileptic and healthy control dogs: Can we detect differences in brain metabolite response with spectroscopy?

    IntroductionIn recent years ketamine has increasingly become the focus of multimodal emergency management for epileptic seizures. However, little is known about the effect of ketamine on brain metabolites in epileptic patients. Magnetic resonance spectroscopy (MRS) is a non-invasive technique to estimate brain metabolites in vivo. Our aim was to measure the effect of ketamine on thalamic metabolites in idiopathic epileptic (IE) dogs using 3 Tesla MRS. We hypothesized that ketamine would increase the glutamine—glutamate (GLX)/creatine ratio in epileptic dogs with and without antiseizure drug treatment, but not in control dogs. Furthermore, we hypothesized that no different responses after ketamine administration in other measured brain metabolite ratios between the different groups would be detected.MethodsIn this controlled prospective experimental trial IE dogs with or without antiseizure drug treatment and healthy client-owned relatives of the breeds Border Collie and Greater Swiss Mountain Dog, were included. After sedation with butorphanol, induction with propofol and maintenance with sevoflurane in oxygen and air, a single voxel MRS at the level of the thalamus was performed before and 2 min after intravenous administration of 1 mg/kg ketamine. An automated data processing spectral fitting linear combination model algorithm was used to estimate all commonly measured metabolite ratios. A mixed ANOVA with the independent variables ketamine administration and group allocation was performed for all measured metabolites. A p < 0.05 was considered statistically significant.ResultsTwelve healthy control dogs, 10 untreated IE and 12 treated IE dogs were included. No significant effects for GLX/creatine were found. However, increased glucose/creatine ratios were found (p < 0.001) with no effect of group allocation. Furthermore, increases in the GABA/creatine ratio were found in IEU dogs.DiscussionMRS was able to detect changes in metabolite/creatine ratios after intravenous administration of 1 mg/kg ketamine in dogs and no evidence was found that excitatory effects are induced in the thalamus. Although it is beyond the scope of this study to investigate the antiseizure potential of ketamine in dogs, results of this research suggest that the effect of ketamine on the brain metabolites could be dependent on the concentrations of brain metabolites before administration

    FAS-dependent cell death in α-synuclein transgenic oligodendrocyte models of multiple system atrophy

    Multiple system atrophy is a parkinsonian neurodegenerative disorder. It is cytopathologically characterized by accumulation of the protein p25α in cell bodies of oligodendrocytes followed by accumulation of aggregated α-synuclein in so-called glial cytoplasmic inclusions. p25α is a stimulator of α-synuclein aggregation, and coexpression of α-synuclein and p25α in the oligodendroglial OLN-t40-AS cell line causes α-synuclein aggregate-dependent toxicity. In this study, we investigated whether the FAS system is involved in α-synuclein aggregate dependent degeneration in oligodendrocytes and may play a role in multiple system atrophy. Using rat oligodendroglial OLN-t40-AS cells we demonstrate that the cytotoxicity caused by coexpressing α-synuclein and p25α relies on stimulation of the death domain receptor FAS and caspase-8 activation. Using primary oligodendrocytes derived from PLP-α-synuclein transgenic mice we demonstrate that they exist in a sensitized state expressing pro-apoptotic FAS receptor, which makes them sensitive to FAS ligand-mediated apoptosis. Immunoblot analysis shows an increase in FAS in brain extracts from multiple system atrophy cases. Immunohistochemical analysis demonstrated enhanced FAS expression in multiple system atrophy brains notably in oligodendrocytes harboring the earliest stages of glial cytoplasmic inclusion formation. Oligodendroglial FAS expression is an early hallmark of oligodendroglial pathology in multiple system atrophy that mechanistically may be coupled to α-synuclein dependent degeneration and thus represent a potential target for protective intervention

    Cluster temporal de metales en un testigo corto de sedimento del Cañón Submarino de Cascais (Margen de Portugal)

    The Cascais Canyon delivers contaminated sediments from the shelf to the deep marine environment. Multi-core PE252-32, located at 2100 m water depth in the canyon, records the latest 300 years of sedimentation. It was dated by 210Pb and analyzed texturally and geochemically for major elements and selected trace metals (Cu, Cr, Hg, Li, Ni, Pb and Zn). Cluster analysis performed on the down-core geochemical data identified four groups of variables related by grain-size, geochemical source and composition. Mercury, Pb and Zn were grouped in a cluster representing the anthropogenic component. Cluster analysis was applied again particularly to the latter cluster relatively to depth, in order to constrain the onset and temporal evolution of anthropogenic contamination. A second clustering, made on the basis of Hg, Pb and Zn, grouped samples by age and degree of anthropogenic contamination. One cluster contained relatively uncontaminated samples older than 1900 AD, and another cluster samples younger than 1900 AD with distinct metal enrichment. Maximum enrichments occurred during the early 1980s, followed by a slight recovery from the mid-1980s to the present. Mercury was the element with the highest enrichment factor (EFHg=5). Despite relatively low accumulation rates at this core location, our results show the importance of the Cascais Canyon as a transport route for contaminated sediments from the Tagus prodelta into the deep regions of the Portuguese Margin.El Cañón submarino de Cascais distribuye sedimentos contaminados de la plataforma continental al medio marino profundo. El testigo PE252-32, situado a 2100 metros de profundidad dentro del cañón, registra los últimos 300 años de su sedimentación. Se ha datado con 210Pb y ha sido analizada su granulometría, elementos mayoritarios y traza (cobre, cromo, mercurio, litio, níquel, plomo y zinc). Se ha llevado a cabo un análisis estadístico clúster, inicialmente aplicado a las series de datos geoquímicos en función de la profundidad, identificándose 4 grupos de variables, relacionados por la granulometría, procedencia y composición geoquímica. Los metales mercurio, plomo y zinc aparecen agrupados en un clúster que representa la componente antropogénica. A continuación el mismo análisis estadístico se aplicó específicamente a este grupo, en función de la profundidad, organizando las muestras según la edad de contaminación antropogénica. Como resultado las muestras quedan agrupadas en dos clústers principales, el de las muestras no contaminadas y más antiguas de 1900 AC, y el de las muestras más recientes de 1900 AC, marcando esta fecha el inicio de la contaminación por impacto de la actividad humana. Los máximos enriquecimientos ocurren durante el inicio de la década de los años ochenta, seguidos de un ligero descenso a partir de mediados de la misma década hasta la actualidad. De entre los demás, el mercurio es el elemento con el mayor grado de enriquecimiento (EFHg=5). Aunque la tasa de acumulación de sedimentos en este lugar es relativamente baja, los resultados obtenidos ponen en evidencia la importancia del Cañón submarino de Cascais como vía de transporte de sedimentos contaminados depositados en el pro-delta del Tajo a las regiones profundas del margen de Portugal

    The rearing environment persistently modulates mouse phenotypes from the molecular to the behavioural level.

    The phenotype of an organism results from its genotype and the influence of the environment throughout development. Even when using animals of the same genotype, independent studies may test animals of different phenotypes, resulting in poor replicability due to genotype-by-environment interactions. Thus, genetically defined strains of mice may respond differently to experimental treatments depending on their rearing environment. However, the extent of such phenotypic plasticity and its implications for the replicability of research findings have remained unknown. Here, we examined the extent to which common environmental differences between animal facilities modulate the phenotype of genetically homogeneous (inbred) mice. We conducted a comprehensive multicentre study, whereby inbred C57BL/6J mice from a single breeding cohort were allocated to and reared in 5 different animal facilities throughout early life and adolescence, before being transported to a single test laboratory. We found persistent effects of the rearing facility on the composition and heterogeneity of the gut microbial community. These effects were paralleled by persistent differences in body weight and in the behavioural phenotype of the mice. Furthermore, we show that environmental variation among animal facilities is strong enough to influence epigenetic patterns in neurons at the level of chromatin organisation. We detected changes in chromatin organisation in the regulatory regions of genes involved in nucleosome assembly, neuronal differentiation, synaptic plasticity, and regulation of behaviour. Our findings demonstrate that common environmental differences between animal facilities may produce facility-specific phenotypes, from the molecular to the behavioural level. Furthermore, they highlight an important limitation of inferences from single-laboratory studies and thus argue that study designs should take environmental background into account to increase the robustness and replicability of findings
