    Consumer understanding of nutrition and health claims: sources of evidence

    Provided that they are scientifically substantiated, nutrition and health (NH) claims linked to food products can help consumers make well-informed food choices. The new European legislation on NH claims made on foods entered into force on 19 January 2007. The law sets out conditions for their use, establishes a system for their scientific evaluation, and will create European lists of authorised claims. An important aspect of this proposed legislation is that it states, in article 5.2, ‘the use of nutrition and health claims shall only be permitted if the average consumer can be expected to understand the beneficial effects expressed in the claim'. The present review examines consumer understanding of NH claims from a consumer science perspective. It focuses on the type of data and information that could be needed to provide evidence that the average consumer adequately understands a particular NH claim. After exploring several different methodologies, it proposes a case-specific approach using a stepwise procedure for assessing consumer understanding of a NH clai

    Fisher choice may increase prevalence of green turtle fibropapillomatosis disease

But when healthy individuals are removed and diseased individuals are left in the population, it is anticipated that prevalence of disease increases. Although this scenario is presumably common in exploited populations where infected individuals are less marketable, it is not widely reported in the literature. We describe this scenario in a marine turtle fishery in the Turks and Caicos Islands (TCI), where green turtles are harvested for local consumption. During a 2-year period, we recorded the occurrence of fibropapillomatosis (FP) disease in green turtles (Chelonia mydas) captured during in-water surveys and compared it with that of turtles landed in the fishery. 13.4% (n = 32) of turtles captured during in-water surveys showed externally visible signs of FP. FP occurred at specific geographic locations where fishing also occurred. Despite the disease being prevalent in the size classes selected by fishers, FP was not present in any animals landed by the fishery (n = 162). The majority (61%) of fishers interviewed expressed that they had caught turtles with FP. Yet, 82% of those that had caught turtles with the disease chose to return their catch to the sea, thereby selectively harvesting healthy turtles and leaving those with the disease in the population.     Assessing recent warming using instrumentally homogeneous sea surface temperature records

    Sea surface temperature (SST) records are subject to potential biases due to changing instrumentation and measurement practices. Significant differences exist between commonly used composite SST reconstructions from the National Oceanic and Atmospheric Administration’s Extended Reconstruction Sea Surface Temperature (ERSST), the Hadley Centre SST data set (HadSST3), and the Japanese Meteorological Agency’s Centennial Observation-Based Estimates of SSTs (COBE-SST) from 2003 to the present. The update from ERSST version 3b to version 4 resulted in an increase in the operational SST trend estimate during the last 19 years from 0.07° to 0.12°C per decade, indicating a higher rate of warming in recent years. We show that ERSST version 4 trends generally agree with largely independent, near-global, and instrumentally homogeneous SST measurements from floating buoys, Argo floats, and radiometer-based satellite measurements that have been developed and deployed during the past two decades. We find a large cooling bias in ERSST version 3b and smaller but significant cooling biases in HadSST3 and COBE-SST from 2003 to the present, with respect to most series examined. These results suggest that reported rates of SST warming in recent years have been underestimated in these three data sets

    Predicting seabed burial of cylinders by wave-induced scour : application to the sandy inner shelf off Florida and Massachusetts

The definitive version was published in IEEE Journal of Oceanic Engineering 32 (2007): 167-183, doi:10.1109/JOE.2007.890958.A simple parameterized model for wave-induced burial of mine-like cylinders as a function of grain-size, time-varying, wave orbital velocity and mine diameter was implemented and assessed against results from inert instrumented mines placed off the Indian Rocks Beach (IRB, FL), and off the Martha’s Vineyard Coastal Observatory (MVCO, Edgartown, MA). The steady flow scour parameters provided by Whitehouse (1998) for self-settling cylinders worked well for predicting burial by depth below the ambient seabed for Ο (0.5 m) diameter mines in fine sand at both sites. By including or excluding scour pit infilling, a range of percent burial by surface area was predicted that was also consistent with observations. Rapid scour pit infilling was often seen at MVCO but never at IRB, suggesting that the environmental presence of fine sediment plays a key role in promoting infilling. Overprediction of mine scour in coarse sand was corrected by assuming a mine within a field of large ripples buries only until it generates no more turbulence than that produced by surrounding bedforms. The feasibility of using a regional wave model to predict mine burial in both hindcast and real-time forecast mode was tested using the National Oceanic and Atmospheric Administration (NOAA, Washington, DC) WaveWatch 3 (WW3) model. Hindcast waves were adequate for useful operational forcing of mine burial predictions, but five-day wave forecasts introduced large errors.     High-resolution mapping of mines and ripples at the Martha's Vineyard Coastal Observatory

The definitive version was published in IEEE Journal of Oceanic Engineering 32 (2007): 133-149, doi:10.1109/JOE.2007.890953.High-resolution multibeam sonar and state-of-the- art data processing and visualization techniques have been used to quantify the evolution of seafloor morphology and the degree of burial of instrumented mines and mine-shapes as part of the U.S. Office of Naval Research (ONR, Arlington, VA) mine burial experiment at the Martha's Vineyard Coastal Observatory (MVCO, Edgartown, MA). Four surveys were conducted over two years at the experiment site with a 455-kHz, Reson 8125 dynamically focused multibeam sonar. The region is characterized by shore-perpendicular alternating zones of coarse-grained sand with 5?25-cm-high, wave orbital-scale ripples, and zones of finer grained sands with smaller (2?5-cm-high) anorbital ripples and, on occasion, medium scale 10?20-cm-high, chaotic or hummocky bedforms. The boundaries between the zones appear to respond over periods of days to months to the predominant wave direction and energy. Smoothing and small shifts of the boundaries to the northeast take place during fair-weather wave conditions while erosion (scalloping of the boundary) and shifts to the north-northwest occur during storm conditions. The multibeam sonar was also able to resolve changes in the orientation and height of fields of ripples that were directly related to the differences in the prevailing wave direction and energy. The alignment of the small scale bedforms with the prevailing wave conditions appears to occur rapidly (on the order of hours or days) when the wave conditions exceed the threshold of sediment motion (most of the time for the fine sands) and particularly during moderate storm conditions. During storm events, erosional ?windows? to the coarse layer below appear in the fine-grained sands. These ?window? features are oriented parallel to the prevailing wave direction and reveal orbital-scale ripples that are oriented perpendicular to the prevailing wave direction. The resolution of the multibeam sonar combined with 3-D visualization techniques provided realistic looking images of both both instrumented and noninstrumented mines and mine-like objects (including bomb, Manta, and Rockan shapes) that were dimensionally correct and enabled unambiguous identification of the mine type. In two of the surveys (October and December 2004), the mines in the fine-grained sands scoured into local pits but were still perfectly visible and identifiable with the multibeam sonar. In the April 2004 survey, the mines were not visible and apparently were completely buried. In the coarse-grained sand zone, the mines were extremely difficult to detect after initial scour burial as the mines bury until they present the same hydrodynamic roughness as the orbital-scale bedforms and thus blend into the ambient ripple field. Given the relatively large, 3-D, spatial coverage of the multibeam sonar along with its ability to measure the depth of the seafloor and the depth and dimensions of the mine, it is possible to measure directly, the burial by depth and burial by surface area of the mines.     Photorespiration: metabolic pathways and their role in stress protection

    Photorespiration results from the oxygenase reaction catalysed by ribulose-1,5-bisphosphate carboxylase/ oxygenase. In this reaction glycollate-2-phosphate is produced and subsequently metabolized in the photorespiratory pathway to form the Calvin cycle intermediate glycerate-3-phosphate. During this metabolic process, CO2 and NH3 are produced and ATP and reducing equivalents are consumed, thus making photorespiration a wasteful process. However, precisely because of this ine¤ciency, photorespiration could serve as an energy sink preventing the overreduction of the photosynthetic electron transport chain and photoinhibition, especially under stress conditions that lead to reduced rates of photosynthetic CO2 assimilation. Furthermore, photorespiration provides metabolites for other metabolic processes, e.g. glycine for the synthesis of glutathione, which is also involved in stress protection. In this review, we describe the use of photorespiratory mutants to study the control and regulation of photorespiratory pathways. In addition, we discuss the possible role of photorespiration under stress conditions, such as drought, high salt concentrations and high light intensities encountered by alpine plants

    The Rice Miniature Inverted Repeat Transposable Element mPing Is an Effective Insertional Mutagen in Soybean

    Insertional mutagenesis of legume genomes such as soybean (Glycine max) should aid in identifying genes responsible for key traits such as nitrogen fixation and seed quality. The relatively low throughput of soybean transformation necessitates the use of a transposon-tagging strategy where a single transformation event will produce many mutations over a number of generations. However, existing transposon-tagging tools being used in legumes are of limited utility because of restricted transposition (Ac/Ds: soybean) or the requirement for tissue culture activation (Tnt1: Medicago truncatula). A recently discovered transposable element from rice (Oryza sativa), mPing, and the genes required for its mobilization, were transferred to soybean to determine if it will be an improvement over the other available transposon-tagging tools. Stable transformation events in soybean were tested for mPing transposition. Analysis of mPing excision at early and late embryo developmental stages revealed increased excision during late development in most transgenic lines, suggesting that transposition is developmentally regulated. Transgenic lines that produced heritable mPing insertions were identified, with the plants from the highest activity line producing at least one new insertion per generation. Analysis of the mPing insertion sites in the soybean genome revealed that features displayed in rice were retained including transposition to unlinked sites and a preference for insertion within 2.5 kb of a gene. Taken together these findings indicate that mPing has the characteristics necessary for an effective transposon-tagging resource

    Adenosine receptor expression and function in rat striatal cholinergic interneurons

Cholinergic neurons were identified in rat striatal slices by their size, membrane properties, sensitivity to the NK(1) receptor agonist (Sar(9), Met(O(2))(11)) Substance P, and expression of choline acetyltransferase mRNA. A(1) receptor mRNA was detected in 60% of the neurons analysed, and A(2A) receptor mRNA in 67% (n=15). 2. The A(1) receptor agonist R-N(6)-(2-phenylisopropyl)adenosine (R-PIA) hyperpolarized cholinergic neurons in a concentration dependent manner sensitive to the A(1) antagonist 8-cyclopentyl-1,3-dipropylxanthine (DPCPX, 100 nM). 3. In dual stimulus experiments, the A(2A) receptor antagonist 8-(3-chlorostyryl)caffeine (CSC, 500 nM) decreased release of [(3)H]-acetylcholine from striatal slices (S2/S1 0.78±0.07 versus 0.95±0.05 in control), as did adenosine deaminase (S2/S1 ratio 0.69±0.05), whereas the A(1) receptor antagonist DPCPX (100 nM) had no effect (S2/S1 1.05±0.14). 4. In the presence of adenosine deaminase the adenosine A(2A) receptor agonist 2-p-((carboxyethyl)phenylethylamino)-5′-N-ethylcarboxamidoadenosine (CGS21680, 10 nM) increased release (S2/S1 ratio 1.03±0.05 versus 0.88±0.05 in control), an effect blocked by the antagonist CSC (500 nM, S2/S1 0.68±0.05, versus 0.73±0.08 with CSC alone). The combined superfusion of bicuculline (10 μM), saclofen (1 μM) and naloxone (10 μM) had no effect on the stimulation by CGS21680 (S2/S1 ratio 0.99±0.04). 5. The A(1) receptor agonist R-PIA (100 nM) inhibited the release of [(3)H]-acetylcholine (S2/S1 ratio 0.70±0.03), an effect blocked by DPCPX (S2/S1 ratio 1.06±0.07). 6. It is concluded that both A(1) and A(2A) receptors are expressed on striatal cholinergic neurons where they are functionally active
