    ESPECTRA: Searching the Bipolar Spectrum in Eating Disorder patients

    <p>Abstract</p> <p>Background</p> <p>Bipolar Disorder (BD) is a chronic, recurrent and highly prevalent illness. Despite the need for correct diagnosis to allow proper treatment, studies have shown that reaching a diagnosis can take up to ten years due to the lack of recognition of the broader presentations of BD. Frequent comorbidities with other psychiatric disorders are a major cause of misdiagnosis and warrant thorough evaluation.</p> <p>Methods/Design</p> <p>ESPECTRA (<it>Occurrence of Bipolar Spectrum Disorders in Eating Disorder Patients</it>) is a single-site cross-sectional study involving a comparison group, designed to evaluate the prevalence of bipolar spectrum in an eating disorder sample. Women aged 18-45 years will be evaluated using the SCID-P and Zurich criteria for diagnosis and the HAM-D, YOUNG, SCI-MOODS, HCL-32, BIS-11, BSQ, WHOQoL and EAS instruments for rating symptoms and measuring clinical correlates.</p> <p>Discussion</p> <p>The classificatory systems in psychiatry are based on categorical models that have been criticized for simplifying the diagnosis and leading to an increase in comorbidities. Some dimensional approaches have been proposed aimed at improving the validity and reliability of psychiatric disorder assessments, especially in conditions with high rates of comorbidity such as BD and Eating Disorder (ED). The Bipolar Spectrum (BS) remains under-recognized in clinical practice and its definition is not well established in current diagnostic guidelines. Broader evaluation of psychiatric disorders combining categorical and dimensional views could contribute to a more realistic understanding of comorbidities and help toward establishing a prognosis.</p

    Analysis of a possible association between oral lichen planus and drug intake. A controlled study

    Objectives: To investigate whether daily systemic and/or topical medication contributes to the development of oral lichen planus (OLP) lesions. Study Design: The study involved 110 OLP patients and 76 control subjects, matched by age, race and sex. The analyzed data included medical records, drug intake and topical medication. Criteria for analysis of drug intake included: (1) ATC-code drug classification; (2) number of different drugs used daily in the categories of monopharmacy (1 drug), minor polypharmacy (2 4 drugs), and major polypharmacy (> 5 drugs); and (3) drugs implicated in lichenoid reactions (DILRs). Results: Sixty (54.5%) of the 110 OLP patients reported daily medication (prior to the appearance of the OLP lesion) compared to 52 (68.4%) of the 76 control subjects. No statistical difference was found between the two groups in terms of systemic diseases, number of medicated individuals in the categories of mono- and polypharmacy, nor use of DILRs (P > 0.05). Regarding the clinical forms and site of involvement, a statistically significant difference was only found for the clinical erosive form of OLP, seen more frequently in non-DILR (P = 0.04) and nonmedicated OLP patients (P = 0.02) than in DILR OLP patients. Daily use of topical oral medication was reported by 2 (1.8%) OLP patients and 1 (1.3%) control subject. Conclusions: It seems that the use of systemic medication does not lead to a significant increase in the incidence of OLP lesions. For their part, lichenoid drug reactions are likely to occur only in a very low percentage of patients. © Medicina Oral S. L

    Scaling of a standardized summary test (RESUMeV) for two primary school grades

    El objetivo del estudio fue el de mostrar evidencias de fiabilidad y validez para una prueba de resúmenes (RESUMev) que evalúa el grado de comprensión lectora en alumnos de cuarto y sexto de primaria. Participaron un total de 528 estudiantes de primaria, de los cuales 236 fueron estudiantes de 4º de primaria (9 y 10 años) y 292 a 6º de primaria (11 a 13 años). Todos estos alumnos procedían de 21 centros escolares. Para evaluar la consistencia interna, se calculó el alfa de Cronbach en los criterios de evaluación de resúmenes (contenido, coherencia y estilo) y su índice de homogeneidad (Hj). La validez se evaluó mediante la comparación entre niveles académicos. En cuanto a los resultados, se obtuvieron índices elevados significativos de fiabilidad y de validez. The sample of this study consisted of 528 children, 236 from fourth grade and 296 from sixth grade. They were drawn from 21 different primary schools. Several criteria were used. To evaluate the internal consistency, Cronbach’s alpha was calculated for all summary evaluation criteria (content, coherence, and style); a homogeneity index (Hj) was calculated too. The validity was evaluated by comparing academic levels. As for the results, both reliability and validity indices were high and significant.     CD44-high neural crest stem-like cells are associated with tumour aggressiveness and poor survival in neuroblastoma tumours

    BACKGROUND: Neuroblastoma is a paediatric tumour originated from sympathoadrenal precursors and characterized by its heterogeneity and poor outcome in advanced stages. Intra-tumoral cellular heterogeneity has emerged as an important feature in neuroblastoma, with a potential major impact on tumour aggressiveness and response to therapy. CD44 is an adhesion protein involved in tumour progression, metastasis and stemness in different cancers; however, there has been controversies about the significance of CD44 expression in neuroblastoma and its relationship with tumour progression. METHODS: We have performed transcriptomic analysis on patient tumour samples studying the outcome of patients with high CD44 expression. Adhesion, invasion and proliferation assays were performed in sorted CD44high neuroblastoma cells. Tumoursphere cultures have been used to enrich in undifferentiated stem-like cells and to asses self-renewal and differentiation potential. We have finally performed in vivo tumorigenic assays on cell line-derived or Patient-derived xenografts. FINDINGS: We show that high CD44 expression is associated with low survival in high-grade human neuroblastoma, independently of MYCN amplification. CD44 is expressed in a cell population with neural crest stem-like features, and with the capacity to generate multipotent, undifferentiated tumourspheres in culture. These cells are more invasive and proliferative in vitro. CD44 positive cells obtained from tumours are more tumorigenic and metastatic, giving rise to aggressive neuroblastic tumours at high frequency upon transplantation. INTERPRETATION: We describe an unexpected intra-tumoural heterogeneity within cellular entities expressing CD44 in neuroblastoma, and propose that CD44 has a role in neural crest stem-like undifferentiated cells, which can contribute to tumorigenesis and malignancy in this type of cancer.     BSA-PEG Hydrogel: A Novel Protein-Ligand Binding 3D Matrix

    Hydrogel materials have good biomimetic properties and high potential for biomedical and bioanalytical applications. In this work, a hydrogel of serum albumin crosslinked with poly-(ethylene glycol) was prepared and characterized for its water content, protein structure and stability. The ability of the hydrogel to bind small molecule ligands with different hydrophobicity was evaluated using a homologous series of amphiphiles (NBD-Cn, n = 4, 6 and 8) and the calculated binding affinities were similar to that of free protein in solution.     Quantum entanglement in exactly soluble atomic models: The Moshinsky model with three electrons, and with two electrons in a uniform magnetic field

    We investigate the entanglement-related features of the eigenstates of two exactly soluble atomic models: a one-dimensional three-electron Moshinsky model, and a three-dimensional two-electron Moshinsky system in an external uniform magnetic field. We analytically compute the amount of entanglement exhibited by the wavefunctions corresponding to the ground, first and second excited states of the three-electron model. We found that the amount of entanglement of the system tends to increase with energy, and in the case of excited states we found a finite amount of entanglement in the limit of vanishing interaction. We also analyze the entanglement properties of the ground and first few excited states of the two-electron Moshinsky model in the presence of a magnetic field. The dependence of the eigenstates' entanglement on the energy, as well as its behaviour in the regime of vanishing interaction, are similar to those observed in the three-electron system. On the other hand, the entanglement exhibits a monotonically decreasing behavior with the strength of the external magnetic field. For strong magnetic fields the entanglement approaches a finite asymptotic value that depends on the interaction strength. For both systems studied here we consider a perturbative approach in order to shed some light on the entanglement's dependence on energy and also to clarify the finite entanglement exhibited by excited states in the limit of weak interactions.     Comparative Assessment of Shrimp Hydrolyzates as Alternative Organic Fertilizers for Legumes

    The global annual production of shrimp is nearly 4 million metric tons generating almost half of this weight in waste. This study assessed the crop production of legumes fertilized with shrimp exoskeletons obtained by microwaves under greenhouse conditions. Plants were grown under the following fertilization regimes: (i) untreated shrimp waste, (ii) shrimp waste pellets, (iii) shrimp-based pellets having a hydrolysis degree of 42%, (iv) untreated cellulose pellets, (v) untreated soil, (vi) untreated cotton substrate, and (vii) two commercial fertilizers (CF1 and CF2). CF1 and CF2 showed the largest electric conductivity and ionic exchange capability, whereas the fertilizing pellets showed the lowest values. However, pH, densification and conductivity of soil were not affected by fertilization. Shrimp waste showed a high content of C, N, O, Ca and P mainly derived from chitin, proteins and minerals. All fertilizers showed typical type II isotherms, but the untreated soil and CF2 per se exhibited the largest water uptake. The soil microbiota increased during the growing cycle and then decreased as the reproductive phase started. Further, soil planted with Phaseolus vulgaris showed a larger microbial population than Pisum sativum. The best plant growth was achieved when treated with CF2, whereas the raw shrimp waste caused a beneficial plant growth and crop yield mainly in Phaseolus vulgaris

    Epigenetic and post-transcriptional regulation of somatostatin receptor subtype 5 (SST5) in pituitary and pancreatic neuroendocrine tumors

    Somatostatin receptor subtype 5 (SST5) is an emerging biomarker and actionable target in pituitary (PitNETs) and pancreatic (PanNETs) neuroendocrine tumors. Transcriptional and epigenetic regulation of SSTR5 gene expression and mRNA biogenesis is poorly understood. Recently, an overlapping natural antisense transcript, SSTR5-AS1, potentially regulating SSTR5 expression, was identified. We aimed to elucidate whether epigenetic processes contribute to the regulation of SSTR5 expression in PitNETs (somatotropinomas) and PanNETs. We analyzed the SSTR5/SSTR5-AS1 human locus in silico to identify CpG islands. SSTR5 and SSTR5-AS1 expression was assessed by quantitative real-time PCR (qPCR) in 27 somatotropinomas, 11 normal pituitaries (NPs), and 15 PanNETs/paired adjacent (control) samples. We evaluated methylation grade in four CpG islands in the SSTR5/SSTR5-AS1 genes. Results revealed that SSTR5 and SSTR5-AS1 were directly correlated in NP, somatotropinoma, and PanNET samples. Interestingly, selected CpG islands were differentially methylated in somatotropinomas compared with NPs. In PanNETs cell lines, SSTR5-AS1 silencing downregulated SSTR5 expression, altered aggressiveness features, and influenced pasireotide response. These results provide evidence that SSTR5 expression in PitNETs and PanNETs can be epigenetically regulated by the SSTR5-AS1 antisense transcript and, indirectly, by DNA methylation, which may thereby impact tumor behavior and treatment response.Junta de AndalucíaMinisterio de EconomíaMinisterio de Ciencia e Innovació

    Let's End HepC: modelling public health epidemiological policies applied to Hepatitis C in Spain

    Background: The WHO has defined international targets toward the elimination of hepatitis C by 2030. Most countries cannot be on track to achieve this goal unless many challenges are surpassed. The Let's End HepC (LEHC) tool aims to contribute to the control of hepatitis C. The innovation of this tool combines the modelling of public health policies (PHP) focused on hepatitis C with epidemiological modelling of the disease, obtaining a unique result that allows to forecast the impact of policy outcomes. The model was applied to several countries, including Spain. Methods: To address the stated objective, we applied the “Adaptive Conjoint Analysis” for PHP decision-making and Markov Chains in the LEHC modelling tool. The tool also aims to be used as an element of health literacy for patient advocacy through gamification mechanisms and country comparability. The LEHC project has been conducted in several countries, including Spain. The population segments comprised in the project are: People Who Inject Drugs (PWID), prisoners, blood products, remnant population. Results: A total of 24 PHP related to hepatitis C were included in the LEHC project. It was identified that Spain had fully implemented 14 of those policies to control hepatitis C. According to LEHC's model forecast, the WHO's Hepatitis C elimination goal on reducing the number of patients living with Hepatitis C to 10% can be achieved in Spain by 2026 if current policies are maintained. The model estimates that the total population in Spain, by 2026, is expected to comprise 26,367 individuals living with hepatitis C. Moreover, if the 24 PHP considered for this study are fully implemented in Spain, the elimination goal may be achieved in 2024, with 29,615 individuals living with hepatitis C by that year. Conclusion: The findings corroborate the view that Spain has set great efforts in directing PHP toward Hepatitis C Virus (HCV) elimination by 2030. However, there is still room for improvement, namely in further implementing 10 of the 24 PHP considered for the LEHC project. By maintaining the 14 PHP in force, the LEHC model estimates the HCV elimination in the country by 2026, and by 2024 if further measures are employed to control the disease.info:eu-repo/semantics/publishedVersio
