    A prática pedagógica: considerações críticas sobre as provas Saber Pro para licenciados

    The article analyzes the statements on teaching, training, and evaluation present in these guides and documents of the Framework of Reference for Educational Sciences for the Saber Pro test presented by teachers in training. As a result of the analysis, it is evident that these concepts are addressed in a limited way in these documents but, above all, that they displace pedagogical knowledge to a minor place. Hence, the text provides pedagogical conceptualizations for teaching, training, and evaluation, explicitly defending pedagogy as a conceptual field and, in this line, claiming the social recognition of the teacher. Consequently, the article argues that pedagogical practice allows the vindication of the teacher, but above all, it is a key epistemological tool for the training of graduates.Este artículo analiza los enunciados sobre enseñanza, formación y evaluación presentes en estas guías y documentos del Marco de Referencia para las Ciencias de la Educación para la prueba Saber Pro, que presentan los profesores en formación. Resultado del análisis, se evidencia que estos conceptos son abordados de manera limitada en estos documentos pero, sobre todo, que desplazan el saber pedagógico a un lugar menor. De allí que el texto brinde conceptualizaciones pedagógicas para la enseñanza, la formación y la evaluación, haciendo una defensa explícita por la pedagogía como campo conceptual y, en esa línea, reivindicando el reconocimiento social del profesor.     Symptoms of the viral annular spot disease associated with the African oil palm ring spot virus (AOPRV) on the interspecific hybrid palm OxG (Elaeis oleifera x Elaeis guineensis) in the early stages of commercial nursery and field planting agro-ecological conditions of Tumaco, Nariño State, Colombia, are described. The virus AOPRV associated with the disease was confirmed by RT-PCR molecular test.Se describen los síntomas que presentan las palmas del híbrido interespecífico OxG (Elaeis oleifera x Elaeis guineensis) al ser afectadas por la enfermedad viral de la mancha anular (MA) asociada al virus AOPRV (African Oil Palm Ring Spot Virus) y establecidas en las etapas de vivero y siembra comercial en campo en las condiciones agroecológicas de Tumaco, Departamento de Nariño, Colombia. La presencia del virus AOPRV, asociado con la enfermedad, se confirmó mediante pruebas moleculares RT-PCR. 

    Unraveling the effect of silent, intronic and missense mutations on VWF splicing : contribution of next generation sequencing in the study of mRNA

    Large studies in von Willebrand disease patients, including Spanish and Portuguese registries, led to the identification of >250 different mutations. It is a challenge to determine the pathogenic effect of potential splice site mutations on VWF mRNA. This study aimed to elucidate the true effects of 18 mutations on VWF mRNA processing, investigate the contribution of next-generation sequencing to in vivo mRNA study in von Willebrand disease, and compare the findings with in silico prediction. RNA extracted from patient platelets and leukocytes was amplified by RT-PCR and sequenced using Sanger and next generation sequencing techniques. Eight mutations affected VWF splicing: c.1533+1G>A, c.5664+2T>C and c.546G>A (p.=) prompted exon skipping; c.3223-7_3236dup and c.7082-2A>G resulted in activation of cryptic sites; c.3379+1G>A and c.7437G>A) demonstrated both molecular pathogenic mechanisms simultaneously; and the p.Cys370Tyr missense mutation generated two aberrant transcripts. Of note, the complete effect of three mutations was provided by next generation sequencing alone because of low expression of the aberrant transcripts. In the remaining 10 mutations, no effect was elucidated in the experiments. However, the differential findings obtained in platelets and leukocytes provided substantial evidence that four of these would have an effect on VWF levels. In this first report using next generation sequencing technology to unravel the effects of VWF mutations on splicing, the technique yielded valuable information. Our data bring to light the importance of studying the effect of synonymous and missense mutations on VWF splicing to improve the current knowledge of the molecular mechanisms behind von Willebrand disease. identifier:02869074

    Alternativas de desarrollo agropecuario con proyección sostenible para el distrito de riego del Zulia y su zona de influencia

    La asociación de usuarios del Distrito de Adecuación de Tierras de Gran Escala del Río Zulia (ASOZULIA) Norte de Santander, con un área de influencia de 45.536 hectáreas, está interesada en la planificación productiva de su territorio. Dentro de sus actividades agropecuarias se encuentran el arroz (12.000 a 17.000 ha), la palma de aceite (1.534 ha), cítricos (limón, naranja; 346 ha), caña de azúcar (100 ha) y la ganadería. Su principal sistema de producción durante más de 50 años es el cultivo de arroz, sistema que presenta reducción de la productividad (7 a 3 tha), degradación del suelo y problemas de plagas y enfermedades, debido entre otros al uso continuo del fangueo como sistema de preparación de suelos. Adicionalmente, a pesar de tener el distrito de riego del río Zulia una concesión de 13,5 m3.s1, en épocas de verano la oferta hidrica es mucho menor como, por ejemplo, la correspondiente a los meses de febrero a marzo de 2016, con un caudal en la bocatoma del distrito de 10,8 m3s'y una captación real del distrito de solo 6 m.s' Asimismo, la construcción del nuevo acueducto para el área metropolitana de la ciudad de Cúcuta tomará 2,95 m3s del caudal antes de la bocatoma que provee agua al distrito, lo que disminuirá aún más la disponibilidad de agua para riego en esa región. Por lo anterior, se requiere la recuperación de los suelos para el establecimiento de nuevos sistemas productivos que demanden un menor consumo de agua y sean una alternativa viable para los productores.Acelga-Remolacha de hoja, Beta vulgaris var.     Common variants in Alzheimer’s disease and risk stratification by polygenic risk scores

We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer's disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer's disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer's disease

    Molecular and clinical profile of von Willebrand disease in Spain (PCM-EVW-ES) : comprehensive genetic analysis by next-generation sequencing of 480 patients

    Molecular diagnosis of patients with von Willebrand disease is pending in most populations due to the complexity and high cost of conventional molecular analyses. The need for molecular and clinical characterization of von Willebrand disease in Spain prompted the creation of a multicenter project (PCM-EVW-ES) that resulted in the largest prospective cohort study of patients with all types of von Willebrand disease. Molecular analysis of relevant regions of the VWF, including intronic and promoter regions, was achieved in the 556 individuals recruited via the development of a simple, innovative, relatively low-cost protocol based on microfluidic technology and next-generation sequencing. A total of 704 variants (237 different) were identified along VWF, 155 of which had not been previously recorded in the international mutation database. The potential pathogenic effect of these variants was assessed by in silico analysis. Furthermore, four short tandem repeats were analyzed in order to evaluate the ancestral origin of recurrent mutations. The outcome of genetic analysis allowed for the reclassification of 110 patients, identification of 37 asymptomatic carriers (important for genetic counseling) and re-inclusion of 43 patients previously excluded by phenotyping results. In total, 480 patients were definitively diagnosed. Candidate mutations were identified in all patients except 13 type 1 von Willebrand disease, yielding a high genotype-phenotype correlation. Our data reinforce the capital importance and usefulness of genetics in von Willebrand disease diagnostics. The progressive implementation of molecular study as the first-line test for routine diagnosis of this condition will lead to increasingly more personalized and effective care for this patient population

    New insights into the genetic etiology of Alzheimer's disease and related dementias

    Characterization of the genetic landscape of Alzheimer's disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/'proxy' AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele

    Este título, sugestivo por los impactos durante la situación de la Covid 19 en el mundo, y que en Colombia lastimosamente han sido muy críticos, permiten asumir la obligada superación de tensiones sociales, políticas, y económicas; pero sobre todo científicas y tecnológicas. Inicialmente, esto supone la existencia de una capacidad de la sociedad colombiana por recuperar su estado inicial después de que haya cesado la perturbación a la que fue sometida por la catastrófica pandemia, y superar ese anterior estado de cosas ya que se encontraban -y aún se encuentran- muchos problemas locales mal resueltos, medianamente resueltos, y muchos sin resolver: es decir, habrá que rediseñar y fortalecer una probada resiliencia social existente - producto del prolongado conflicto social colombiano superado parcialmente por un proceso de paz exitoso - desde la tecnociencia local; como lo indicaba Markus Brunnermeier - economista alemán y catedrático de economía de la Universidad de Princeton- en su libro The Resilient Society…La cuestión no es preveerlo todo sino poder reaccionar…aprender a recuperarse rápido.This title, suggestive of the impacts during the Covid 19 situation in the world, and which have unfortunately been very critical in Colombia, allows us to assume the obligatory overcoming of social, political, and economic tensions; but above all scientific and technological.
