    Seeing the body distorts tactile size perception

    Vision of the body modulates somatosensation, even when entirely non-informative about stimulation. For example, seeing the body increases tactile spatial acuity, but reduces acute pain. While previous results demonstrate that vision of the body modulates somatosensory sensitivity, it is unknown whether vision also affects metric properties of touch, and if so how. This study investigated how non-informative vision of the body modulates tactile size perception. We used the mirror box illusion to induce the illusion that participants were directly seeing their stimulated left hand, though they actually saw their reflected right hand. We manipulated whether participants: (a) had the illusion of directly seeing their stimulated left hand, (b) had the illusion of seeing a non-body object at the same location, or (c) looked directly at their non-stimulated right-hand. Participants made verbal estimates of the perceived distance between two tactile stimuli presented simultaneously to the dorsum of the left hand, either 20, 30, or 40 mm apart. Vision of the body significantly reduced the perceived size of touch, compared to vision of the object or of the contralateral hand. In contrast, no apparent changes of perceived hand size were found. These results show that seeing the body distorts tactile size perception

    More than skin-deep: integration of skin-based and musculo-skeletal reference frames in localisation of touch

    The skin of the forearm is, in one sense, a flat 2D sheet, but in another sense approximately cylindrical, mirroring the 3D volumetric shape of the arm. The role of frames of reference based on the skin as a 2D sheet versus based on the musculo-skeletal structure of the arm remains unclear. When we rotate the forearm from a pronated to a supinated posture, the skin on its surface is displaced. Thus, a marked location will slide with the skin across the underlying flesh, and the touch perceived at this location should follow this displacement if it is localised within a skin-based reference frame. We investigated, however, if the perceived tactile locations were also affected by the rearrangement in underlying musculo-skeletal structure, i.e. displaced medially and laterally on a pronated and supinated forearm, respectively. Participants pointed to perceived touches (Experiment 1), or marked them on a three-dimensional size-matched forearm on a computer screen (Experiment 2). The perceived locations were indeed displaced medially after forearm pronation in both response modalities. This misperception was reduced (Experiment 1), or absent altogether (Experiment 2) in the supinated posture when the actual stimulus grid moved laterally with the displaced skin. The grid was perceptually stretched at medial-lateral axis, and it was displaced distally, which suggest the influence of skin-based factors. Our study extends the tactile localisation literature focused on the skin-based reference frame and on the effects of spatial positions of body parts by implicating the musculo-skeletal factors in localisation of touch on the body

    A simplified edge illumination set-up for quantitative phase contrast mammography with synchrotron radiation at clinical doses

    This work presents the first study of x-ray phase contrast imaging based on a simple implementation of the edge illumination method (EIXPCi) in the field of mammography with synchrotron radiation. A simplified EIXPCi set-up was utilized to study a possible application in mammography at clinical doses. Moreover, through a novel algorithm capable of separating and quantifying absorption and phase perturbations of images acquired in EIXPCi modality, it is possible to extract quantitative information on breast images, allowing an accurate tissue identification. The study was carried out at the SYRMEP beamline of Elettra synchrotron radiation facility (Trieste, Italy), where a mastectomy specimen was investigated with the EIXPCi technique. The sample was exposed at three different energies suitable for mammography with synchrotron radiation in order to test the validity of the novel algorithm in extracting values of linear attenuation coefficients integrated over the sample thickness. It is demonstrated that the quantitative data are in good agreement with the theoretical values of linear attenuation coefficients calculated on the hypothesis of the breast with a given composition. The results are promising and encourage the current efforts to apply the method in mammography with synchrotron radiation

    Reconstructing neural representations of tactile space

    Psychophysical experiments have demonstrated large and highly systematic perceptual distortions of tactile space. Such a space can be referred to our experience of the spatial organisation of objects, at representational level, through touch, in analogy with the familiar concept of visual space. We investigated the neural basis of tactile space by analysing activity patterns induced by tactile stimulation of nine points on a 3 × 3 square grid on the hand dorsum using functional magnetic resonance imaging. We used a searchlight approach within pre-defined regions of interests to compute the pairwise Euclidean distances between the activity patterns elicited by tactile stimulation. Then, we used multidimensional scaling to reconstruct tactile space at the neural level and compare it with skin space at the perceptual level. Our reconstructions of the shape of skin space in contralateral primary somatosensory and motor cortices reveal that it is distorted in a way that matches the perceptual shape of skin space. This suggests that early sensorimotor areas critically contribute to the distorted internal representation of tactile space on the hand dorsum

    Pathways of L-glutamic acid transport in cultured human fibroblasts.

    The transport of L-glutamic acid has been studied in skin-derived diploid human fibroblasts. Competition analysis in the presence and absence of Na+ and mathematical discrimination by nonlinear regression indicated that L-glutamic acid enters the cell by at least three transport systems: 1) a high affinity Na+-dependent system which has been found to be identical to the previously described system for anionic amino acids (Gazzola, G. C., Dall'Asta, V., Bussolati, O., Makowske, M., and Christensen, H. N. (1981) J. Biol. Chem. 256, 6054-6059) and which is provisionally designated as System X-AG; this route was shared by L-aspartic acid; 2) a low affinity Na+-dependent system resembling the ASC System for neutral amino acids (Franchi-Gazzola, R., Gazzola, G. C., Dall'Asta, V., and Guidotti, G. G. (1982) J. Biol. Chem. 257, 9582-9587); its reactivity toward L-glutamic acid was strongly inhibited by L-serine, but not by 2-(methyl-amino)isobutyric acid; and 3) a Na+-independent system similar to System XC- described in fetal human lung fibroblasts (Bannai, S., and Kitamura, E. (1980) J. Biol. Chem. 255, 2372-2376). The XC- system served for L-glutamic acid and L-cystine, the latter amino acid behaving as a potent inhibitor of L-glutamic acid uptake. Amino acid starvation did not change the uptake of L-glutamic acid by the two Na+-dependent systems, but enhanced the activity of System XC- by increasing its Vmax. L-Glutamic acid transport was also affected by the density of the culture. An increased cell density lowered the uptake of the amino acid by Systems ASC and XC- and promoted the uptake by System X-AG. All these variations were dependent upon changes in Vmax

    Characterization of breast tissues in density and effective atomic number basis via spectral X-ray computed tomography

    Differentiation of breast tissues is challenging in X-ray imaging because tissues might share similar or even the same linear attenuation coefficients μ\mu. Spectral computed tomography (CT) allows for more quantitative characterization in terms of tissue density and effective atomic number by exploiting the energy dependence of μ\mu. In this work, 5 mastectomy samples and a phantom with inserts mimicking breast soft tissues were evaluated in a retrospective study. The samples were imaged at three monochromatic energy levels in the range of 24 - 38 keV at 5 mGy per scan using a propagation-based phase-contrast setup at SYRMEP beamline at the Italian national synchrotron Elettra. A custom-made algorithm incorporating CT reconstructions of an arbitrary number of spectral energy channels was developed to extract the density and effective atomic number of adipose, fibro-glandular, pure glandular, tumor, and skin from regions selected by a radiologist. Preliminary results suggest that, via spectral CT, it is possible to enhance tissue differentiation. It was found that adipose, fibro-glandular and tumorous tissues have average effective atomic numbers (5.94 ±\pm 0.09, 7.03 ±\pm 0.012, and 7.40 ±\pm 0.10) and densities (0.90 ±\pm 0.02, 0.96 ±\pm 0.02, and 1.07 ±\pm 0.03 g/cm3^{3}) and can be better distinguished if both quantitative values are observed together.Comment: 26 pages, 7 figures, submitted to Physics in Medicine and Biolog

    Staphylococcus saprophyticus Recovered from Humans, Food, and Recreational Waters in Rio de Janeiro, Brazil.

    Staphylococcus saprophyticus is an important agent of urinary tract infection (UTI) in young women, but information about this pathogen in human microbiota and in common environment is lacking. The aim of this study was to characterize S. saprophyticus isolates from genitoanal microbiota of 621 pregnant women, 10 minas cheese packs, and five beaches in Rio de Janeiro city and compare PFGE profiles of these isolates with five UTI PFGE clusters described in this city. We investigated 65 S. saprophyticus isolates from microbiota, 13 from minas cheese, and 30 from beaches and 32 UTI isolates. Antimicrobial resistance was determined by disk diffusion, MIC by agar dilution, and PCR. Erythromycin-resistance genes erm(C), msr(A), msr(B), mph(C), and lin(A) were found in 93% of isolates. Trimethoprim-sulfamethoxazole resistance correlated with dfrG or dfrA genes. Three cefoxitin-resistant isolates carried the mecA gene. All isolates obtained from cheese were susceptible to all antimicrobial agents. Six of 10 pregnant women with >1 isolate had monoclonal colonization. Isolates from pregnant women shared 100% similarity with UTI PFGE cluster types A and E obtained almost 10 years previously, suggesting temporal persistence of S. saprophyticus. Antimicrobial resistance of beach isolates reflected the profiles of human isolates. Taken together, results indicate a shared source for human and environmental isolates

    Mind the gap: the effects of temporal and spatial separation in localization of dual touches on the hand

    In this study, we aimed to relate the findings from two predominantly separate streams of literature, one reporting on the localisation of single touches on the skin, and the other on the distance perception of dual touches. Participants were touched with two points, delivered either simultaneously or separated by a short delay to various locations on their left hand dorsum. They then indicated on a size-matched hand silhouette the perceived locations of tactile stimuli. We quantified the deviations between the actual stimulus grid and the corresponding perceptual map which was constructed from the perceived tactile locations, and we calculated the precision of tactile localisation (i.e. the variability across localisation attempts). The evidence showed that the dual touches, akin to single touch stimulations, were mislocalised distally and that their variable localisation error was reduced near joints, particularly near knuckles. However, contrary to single-touch localisation literature, we observed for the dual touches to be mislocalised towards the ulnar side of the hand, particularly when they were presented sequentially. Further, the touches presented in a sequential order were slightly ‘repelled’ from each other and their perceived distance increased, while the simultaneous tactile pairs were localised closer to each other and their distance was compressed. Whereas the sequential touches may have been localised with reference to the body, the compression of tactile perceptual space for simultaneous touches was related in the previous literature to signal summation and inhibition and the low-level factors, including the innervation density and properties of receptive fields of somatosensory neurons
