    GAMBUT field experiment of peatland wildfires in Sumatra: from ignition to spread and suppression

    Peat wildfires can burn over large areas of peatland, releasing ancient carbon and toxic gases into the atmosphere over prolonged periods. These emissions cause haze episodes of pollution and accelerate climate change. Peat wildfires are characterised by smouldering - the flameless, most persistent type of combustion. Mitigation strategies are needed in arctic, boreal, and tropical areas but are hindered by incomplete scientific understanding of smouldering. Here, we present GAMBUT, the largest and longest to-date field experiment of peat wildfires, conducted in a degraded peatland of Sumatra. Temperature, emission and spread of peat fire were continuously measured over 4-10 days and nights, and three major rainfalls. Measurements of temperature in the soil provide field experimental evidence of lethal fire severity to the biological system of the peat up to 30 cm depth. We report that the temperature of the deep smouldering is 13% hotter than shallow layer during daytime. During night-time, both deep and shallow smouldering had the same level of temperature. The experiment was terminated by suppression with water. Comparison of rainfall with suppression confirms the existence of a critical water column height below which extinction is not possible. GAMBUT provides a unique understanding of peat wildfires at field conditions that can contribute to mitigation strategies

    First Observation of Coherent π0\pi^0 Production in Neutrino Nucleus Interactions with Eν<E_{\nu}< 2 GeV

    The MiniBooNE experiment at Fermilab has amassed the largest sample to date of π0\pi^0s produced in neutral current (NC) neutrino-nucleus interactions at low energy. This paper reports a measurement of the momentum distribution of π0\pi^0s produced in mineral oil (CH2_2) and the first observation of coherent π0\pi^0 production below 2 GeV. In the forward direction, the yield of events observed above the expectation for resonant production is attributed primarily to coherent production off carbon, but may also include a small contribution from diffractive production on hydrogen. Integrated over the MiniBooNE neutrino flux, the sum of the NC coherent and diffractive modes is found to be (19.5 ±\pm1.1 (stat) ±\pm2.5 (sys))% of all exclusive NC π0\pi^0 production at MiniBooNE. These measurements are of immediate utility because they quantify an important background to MiniBooNE's search for νμνe\nu_{\mu} \to \nu_e oscillations.Comment: Submitted to Phys. Lett.

    Validity of the Patient Experiences and Satisfaction with Medications (PESaM) Questionnaire

    Background: This study assessed the validity and reliability of the generic module of the recently developed Patient Experiences and Satisfaction with Medications (PESaM) questionnaire in a sample of patients in the Netherlands. Methods: The generic module of the PESaM questionnaire consists of 18 items related to the domains effectiveness, side effects and ease of use of medications. It assesses patients’ experiences regarding the impact of the medication on daily life, health and satisfaction. In 2017, the PESaM questionnaire was sent out to idiopathic pulmonary fibrosis patients using pirfenidone or nintedanib, atypical haemolytic uraemic syndrome patients receiving eculizumab and patients using tacrolimus after kidney transplantation. Mean scores for each domain were calculated applying a scoring algorithm. Construct validity and reliability were assessed using recommended methods. Results: 188 participants completed the generic module, of whom 48% used pirfenidone, 36% nintedanib, 11% tacrolimus and 5% eculizumab. The generic module has good structural properties. Internal consistency values of the domains were satisfactory (i.e. Cronbach’s coefficient alpha above 0.7). Confirmatory factor analysis provided further evidence for construct validity, with good convergent and discriminant validity. The PESaM questionnaire also showed different scores for patients using different medications, in line with expectations, and was therefore able to differentiate between patient groups. Test–retest reliability of the items and domains were rated as moderate to fair (i.e. intraclass coefficients ranged between 0.18 and 0.76). Conclusions: The PESaM questionnaire is a unique patient-reported outcome measure evaluating patient experiences and satisfaction with medications. It has been developed in conjunction with patients, ensuring coverage of domains and issues relevant from the patient’s perspective. This study has shown promising validity of the generic module of the PESaM questionnaire. Further research is recommended to assess reliability in greater detail as well as the responsiveness of the measure. Trial registration: The study

    Change and continuity in Japanese compensation practices: the case of occupational pensions since the early 2000s

    This article analyses changes in the provision of Japanese occupational pensions since the early 2000s. It shows how Japanese companies have followed strategies of cost and risk reduction by creating multi-layered benefit systems that offer a combination of defined benefit (DB) and defined contribution (DC) plans whose benefits are becoming increasingly performance-oriented. Analysing the reasons behind the resilience of DB schemes in Japan, the article concludes that enterprise union behaviour has had less influence than regulatory issues and continued corporate commitment to long-standing employment practices for regular workers. These findings highlight the embeddedness of Japanese employment practices in their institutional context

    Cofactor Tpr2 combines two TPR domains and a J domain to regulate the Hsp70/Hsp90 chaperone system

    In the eukaryotic cytosol, Hsp70 and Hsp90 cooperate with various co-chaperone proteins in the folding of a growing set of substrates, including the glucocorticoid receptor (GR). Here, we analyse the function of the co-chaperone Tpr2, which contains two chaperone-binding TPR domains and a DnaJ homologous J domain. In vivo, an increase or decrease in Tpr2 expression reduces GR activation, suggesting that Tpr2 is required at a narrowly defined expression level. As shown in vitro, Tpr2 recognizes both Hsp70 and Hsp90 through its TPR domains, and its J domain stimulates ATP hydrolysis and polypeptide binding by Hsp70. Furthermore, unlike other co-chaperones, Tpr2 induces ATP-independent dissociation of Hsp90 but not of Hsp70 from chaperone–substrate complexes. Excess Tpr2 inhibits the Hsp90-dependent folding of GR in cell lysates. We propose a novel mechanism in which Tpr2 mediates the retrograde transfer of substrates from Hsp90 onto Hsp70.
