    High hypoxia status in pancreatic cancer is associated with multiple hallmarks of an immunosuppressive tumor microenvironment

    Introduction: Pancreatic ductal adenocarcinoma (PDAC), the most common form of pancreatic cancer, is a particularly lethal disease that is often diagnosed late and is refractory to most forms of treatment. Tumour hypoxia is a key hallmark of PDAC and is purported to contribute to multiple facets of disease progression such as treatment resistance, increased invasiveness, metabolic reprogramming, and immunosuppression.Methods: We used the Buffa gene signature as a hypoxia score to profile transcriptomics datasets from PDAC cases. We performed cell-type deconvolution and gene expression profiling approaches to compare the immunological phenotypes of cases with low and high hypoxia scores. We further supported our findings by qPCR analyses in PDAC cell lines cultured in hypoxic conditions.Results: First, we demonstrated that this hypoxia score is associated with increased tumour grade and reduced survival suggesting that this score is correlated to disease progression. Subsequently, we compared the immune phenotypes of cases with high versus low hypoxia score expression (HypoxiaHI vs. HypoxiaLOW) to show that high hypoxia is associated with reduced levels of T cells, NK cells and dendritic cells (DC), including the crucial cDC1 subset. Concomitantly, immune-related gene expression profiling revealed that compared to HypoxiaLOW tumours, mRNA levels for multiple immunosuppressive molecules were notably elevated in HypoxiaHI cases. Using a Random Forest machine learning approach for variable selection, we identified LGALS3 (Galectin-3) as the top gene associated with high hypoxia status and confirmed its expression in hypoxic PDAC cell lines.Discussion: In summary, we demonstrated novel associations between hypoxia and multiple immunosuppressive mediators in PDAC, highlighting avenues for improving PDAC immunotherapy by targeting these immune molecules in combination with hypoxia-targeted drugs.<br/

    Alternative splicing of CD200 is regulated by an exonic splicing enhancer and SF2/ASF

    CD200, a type I membrane glycoprotein, plays an important role in prevention of inflammatory disorders, graft rejection, autoimmune diseases and spontaneous fetal loss. It also regulates tumor immunity. A truncated CD200 (CD200tr) resulting from alternative splicing has been identified and characterized as a functional antagonist to full-length CD200. Thus, it is important to explore the mechanism(s) controlling alternative splicing of CD200. In this study, we identified an exonic splicing enhancer (ESE) located in exon 2, which is a putative binding site for a splicing regulatory protein SF2/ASF. Deletion or mutation of the ESE site decreased expression of the full-length CD200. Direct binding of SF2/ASF to the ESE site was confirmed by RNA electrophoretic mobility shift assay (EMSA). Knockdown of expression of SF2/ASF resulted in the same splicing pattern as seen after deletion or mutation of the ESE, whereas overexpression of SF2/ASF increased expression of the full-length CD200. In vivo studies showed that viral infection reversed the alternative splicing pattern of CD200 with increased expression of SF2/ASF and the full-length CD200. Taken together, our data suggest for the first time that SF2/ASF regulates the function of CD200 by controlling CD200 alternative splicing, through direct binding to an ESE located in exon 2 of CD200

    TRAIL Death Receptor-4, Decoy Receptor-1 and Decoy Receptor-2 Expression on CD8+ T Cells Correlate with the Disease Severity in Patients with Rheumatoid Arthritis

    BACKGROUND: Rheumatoid Arthritis (RA) is a chronic autoimmune inflammatory disorder. Although the pathogenesis of disease is unclear, it is well known that T cells play a major role in both development and perpetuation of RA through activating macrophages and B cells. Since the lack of TNF-Related Apoptosis Inducing Ligand (TRAIL) expression resulted in defective thymocyte apoptosis leading to an autoimmune disease, we explored evidence for alterations in TRAIL/TRAIL receptor expression on peripheral T lymphocytes in the molecular mechanism of RA development. METHODS: The expression of TRAIL/TRAIL receptors on T cells in 20 RA patients and 12 control individuals were analyzed using flow cytometry. The correlation of TRAIL and its receptor expression profile was compared with clinical RA parameters (RA activity scored as per DAS28) using Spearman Rho Analysis. RESULTS: While no change was detected in the ratio of CD4+ to CD8+ T cells between controls and RA patient groups, upregulation of TRAIL and its receptors (both death and decoy) was detected on both CD4+ and CD8+ T cells in RA patients compared to control individuals. Death Receptor-4 (DR4) and the decoy receptors DcR1 and DcR2 on CD8+ T cells, but not on CD4+ T cells, were positively correlated with patients' DAS scores. CONCLUSIONS: Our data suggest that TRAIL/TRAIL receptor expression profiles on T cells might be important in revelation of RA pathogenesis

    Cause of Cambrian Explosion - Terrestrial or Cosmic?

    We review the salient evidence consistent with or predicted by the Hoyle-Wickramasinghe (H-W) thesis of Cometary (Cosmic) Biology. Much of this physical and biological evidence is multifactorial. One particular focus are the recent studies which date the emergence of the complex retroviruses of vertebrate lines at or just before the Cambrian Explosion of ∼500 Ma. Such viruses are known to be plausibly associated with major evolutionary genomic processes. We believe this coincidence is not fortuitous but is consistent with a key prediction of H-W theory whereby major extinction-diversification evolutionary boundaries coincide with virus-bearing cometary-bolide bombardment events. A second focus is the remarkable evolution of intelligent complexity (Cephalopods) culminating in the emergence of the Octopus. A third focus concerns the micro-organism fossil evidence contained within meteorites as well as the detection in the upper atmosphere of apparent incoming life-bearing particles from space. In our view the totality of the multifactorial data and critical analyses assembled by Fred Hoyle, Chandra Wickramasinghe and their many colleagues since the 1960s leads to a very plausible conclusion - life may have been seeded here on Earth by life-bearing comets as soon as conditions on Earth allowed it to flourish (about or just before 4.1 Billion years ago); and living organisms such as space-resistant and space-hardy bacteria, viruses, more complex eukaryotic cells, fertilised ova and seeds have been continuously delivered ever since to Earth so being one important driver of further terrestrial evolution which has resulted in considerable genetic diversity and which has led to the emergence of mankind

    Regulatory T cells in oral and self-tolerance

    Several mechanisms of immunologic tolerance have been proposed, including deletion, anergy and active suppression. Deletion and anergy have been reported based on data from both in vivo and in vitro experiments, but evidence to support the involvement of active suppression in tolerance has been elusive, principally because of the difficulty in defining regulatory T (Tr) cells with immunosuppressive activity. However, data characterizing the function of Tr cells has recently begun to emerge both from experiments in the field of oral tolerance and from others utilizing transgenic (Tg) mice. One such model which we described uses Tg mice expressing a foreign physiological soluble antigen, beef insulin (BI), while another uses T cell receptor (α/β) Tg mice, whose T cell receptors (TCR) are specific for ovalbumin (OVA). Using such models, adoptive transfer of the Tr clones specific for myelin basic protein (MBP) from orally tolerant mice into naive mice was shown to protect the animals from experimental autoimmune encephalomyelitis (EAE). Transforming growth factor-beta (TGF-β) has been shown to be responsible for the immuno-suppression. Tr clones functioning in self-tolerance have also been obtained from mice expressing TCR or BI transgenes. T cell cultures from OVA specific TCR expressing Tg mice were stimulated in the presence of interleukin-10 (IL-10) and OVA peptide to produce Tr clones. The Tr clones secreted IL-10 and TGF-β as immunosuppressive factors. Tr clones with a different characteristic have been obtained from BI Tg mice. Adoptive transfer of the Tr clones into normal mice suppressed the BI specific antibody response. These Tr clones have type II Th cytokine profile and produced TGF-β as an inhibitory cytokine. TGF-β production also led to functional bystander suppression in the BI Tg mice. These three different types of Tr clones show considerable heterogeneity in the cytokine profiles. Further investigation of Tr cells will be important for our understanding of autoimmunity and the development of tactics for the therapy of autoimmune disease

    Bakalaura darba "Rīgas iedzīvotāju apmierinātība ar sabiedrisko transportu Ziepniekkalna apkaimē" galvenais mērķis bija izpētīt to, cik lielā mērā ar dažādiem sabiedriskā transporta aspektiem Ziepniekkalna apkaimē ir apmierināti Rīgas iedzīvotāji. Tēma tika izvēlēta, ņemot vērā to, ka sabiedriskais transports ir svarīgs elements pilsētas dzīvē, bet Ziepniekkalna apkaime robežojas ar Rīgas pilsētas robežu un tajā izbūvēts lielizmēra dzīvojamais rajons. Pētījumā tika izmantotas gan kvantitatīvās, gan kvalitatīvās metodes, kā teorētisko pamatu izmantojot mobilitātes un patērētāju apmierinātības teorijas. Ar aptaujas anketas palīdzību, kurā indivīdiem, kas izmanto sabiedrisko transportu Ziepniekkalnā, tika uzdoti jautājumi par sabiedriskā transporta izmantošanas paradumiem, apmierinātību ar dažādiem faktoriem, kas saistīti ar sabiedrisko transportu, tāpat bija jānovērtē piekrišana dažādiem ar sabiedrisko transportu saistītiem faktoriem. Kvalitatīvās metodes tika izmantotas, lai analizētu "Rīgas Satiksmes" attīstības stratēģiju. Galvenie bakalaura darba secinājumi vēsta, ka kopumā respondenti izrādīja apmierinātību ar sabiedrisko transportu Ziepniekkalna apkaimē.The main purpose of bachelor thesis "Riga's citizens satisfaction with public transport in Ziepniekkalns neighbourhood" was to define satisfaction level of Riga's citizens with different factors related to public transport in Ziepniekkalns neighbourhood. Both quantitative and qualitative methods were used in this research. Main part of research was conducted with analysis of internet survey where participants were asked questions about their satisfaction with public transporti in Ziepniekkalns, about their habits of using the public transport and about how much they agree with different statements related to public transport in Ziepniekkalns. Qualitative methods were used to analyse a development strategy of "Rīgas Satiksme". The main conclusions of this research tells that participants of conducted survey are mainly satisfied with public transportation in Ziepniekkalns, although there were factors where there were mainly dissatisfacted participants
