    At an important tephrostratigraphic crossroads: cryptotephra in Late Glacial to Early Holocene lake sediments from the Carpathian Mountains, Romania

    Understanding the temporal and spatial environmental response to past climate change during the Last Glacial-Interglacial Transition (LGIT, 16-8 ka) across Europe relies on precise chronologies for palaeoenvironmental records. Tephra layers (volcanic ash) are a powerful chronological tool to synchronise disparate records across the continent. Yet, some regions remain overlooked in terms of cryptotephra investigations. Building on earlier work at the same sites, we present the first complete LGIT high-resolution cryptotephra investigation of two lake records in the Carpathian Mountains in Romania, Lake Brazi and Lake Lia. Numerous volcanic glass shards have been recognised as originating from various volcanic regions, including: Iceland (Katla, Askja, and Torfajokull), Italy (Campi Flegrei, Ischia, Lipari, and Pantelleria), and central Anatolia (Acigol and Ericyes). In total, four distinct tephra horizons have now been identified in these records: 1) an LGIT Lipari tephra (11,515–12,885 cal BP, 95.4% range); 2) Askja-S (11,070–10,720 cal BP, 95.4% range); 3) an Early Holocene Lipari tephra,(12,590–10,845 cal BP, 95.4% range) and; 4) an Early Holocene Ischia tephra (11,120–10,740 cal BP, 95.4% range). The use of trace element analysis on selected cryptotephra layers provided additional important information in identifying volcanic source and facilitating correlations. These tephra layers, along with numerous other discrete cryptotephra layers, offer promise as significant future isochrons for comprehending the spatial and temporal fluctuations in past climate change throughout Europe and the Mediterranean area.     Soothing the Threatened Brain: Leveraging Contact Comfort with Emotionally Focused Therapy

    Social relationships are tightly linked to health and well-being. Recent work suggests that social relationships can even serve vital emotion regulation functions by minimizing threat-related neural activity. But relationship distress remains a significant public health problem in North America and elsewhere. A promising approach to helping couples both resolve relationship distress and nurture effective interpersonal functioning is Emotionally Focused Therapy for couples (EFT), a manualized, empirically supported therapy that is strongly focused on repairing adult attachment bonds. We sought to examine a neural index of social emotion regulation as a potential mediator of the effects of EFT. Specifically, we examined the effectiveness of EFT for modifying the social regulation of neural threat responding using an fMRI-based handholding procedure. Results suggest that EFT altered the brain\u27s representation of threat cues in the presence of a romantic partner. EFT-related changes during stranger handholding were also observed, but stranger effects were dependent upon self-reported relationship quality. EFT also appeared to increase threat-related brain activity in regions associated with self-regulation during the no-handholding condition. These findings provide a critical window into the regulatory mechanisms of close relationships in general and EFT in particular

    HLA Class I and Genetic Susceptibility to Type 1 Diabetes: Results From the Type 1 Diabetes Genetics Consortium

    OBJECTIVE-We report here genotyping data and type 1 diabetes association analyses for HLA class I loci (A, B, and C) on 1,753 multiplex pedigrees from the Type 1 Diabetes Genetics Consortium (T1DGC), a large international collaborative study. RESEARCH DESIGN AND METHODS-Complete eight-locus HLA genotyping data were generated. Expected patient class I (HLA-A, -B, and -C) allele frequencies were calculated, based on linkage disequilibrium (LD) patterns with observed HLA class II DRB1-DQA1-DQB1 haplotype frequencies. Expected frequencies were compared to observed allele frequencies in patients. RESULTS-Significant type 1 diabetes associations were observed at all class I HLA loci. After accounting for LD with HLA class II, the most significantly type 1 diabetes-associated alleles were B*5701 (odds ratio 0.19; P = 4 x 10(-11)) and B*3906 (10.31; P = 4 X 10(-10)). Other significantly type 1 diabetes-associated alleles included A*2402, A*0201, B*1801, and C*0501 (predisposing) and A*1101, A*3201, A*6601, B*0702, B*4403, B*3502, C*1601, and C*0401 (protective). Some alleles, notably B*3906, appear to modulate the risk of all DRB1-DQA1-DQB1 haplotypes on which they reside, suggesting a class I effect that is independent of class H. Other class I type 1 diabetes associations appear to be specific to individual class H haplotypes. Some apparent associations (e.g., C*1601) could be attributed to strong LD to another class I susceptibility locus (B*4403). CONCLUSIONS-These data indicate that HLA class I alleles, in addition to and independently from HLA class H alleles, are associated with type 1 diabetes. Diabetes 59:2972-2979, 201

    Differential Post-Exercise Blood Pressure Responses between Blacks and Caucasians

    Post-exercise hypotension (PEH) is widely observed in Caucasians (CA) and is associated with histamine receptors 1- and 2- (H1R and H2R) mediated post-exercise vasodilation. However, it appears that blacks (BL) may not exhibit PEH following aerobic exercise. Hence, this study sought to determine the extent to which BL develop PEH, and the contri- bution of histamine receptors to PEH (or lack thereof) in this population. Forty-nine (22 BL, 27 CA) young and healthy subjects completed the study. Subjects were randomly assigned to take either a combined H1R and H2R antagonist (fexofenadine and ranitidine) or a con- trol placebo. Supine blood pressure (BP), cardiac output and peripheral vascular resistance measurements were obtained at baseline, as well as at 30 min, 60 min and 90 min after 45 min of treadmill exercise at 70% heart rate reserve. Exercise increased diastolic BP in young BL but not in CA. Post-exercise diastolic BP was also elevated in BL after exercise with histamine receptor blockade. Moreover, H1R and H2R blockade elicited differential responses in stroke volume between BL and CA at rest, and the difference remained follow- ing exercise. Our findings show differential BP responses following exercise in BL and CA, and a potential role of histamine receptors in mediating basal and post-exercise stroke vol- ume in BL. The heightened BP and vascular responses to exercise stimulus is consistent with the greater CVD risk in BL
