Crucial cross-talk of interleukin-1β and progesterone in human choriocarcinoma

This article can be accessed from the links below.This article has been made available through the Brunel Open Access Publishing Fund.Choriocarcinoma is a highly malignant epithelial tumour that is most often associated with hydatidiform mole and presents the most common emergency medical problem in the management of trophoblast disease. We hypothesise that the hormones/cytokines present within the tumour microenvironment play key roles in the development of choriocarcinoma. In this study we assessed the effects of interleukin-1β (IL-1β) on cell death in the presence or absence of the sex hormone progesterone using two choriocarcinoma cell lines (BeWo and JEG-3) as in vitro experimental models. Although IL-1β induced cell death in both cell lines, the effect was more pronounced in JEG-3 cells, where cell death reached 40% compared to 15% in BeWo cells. Cell death of JEG-3 cells in response to IL-1β was significantly decreased by co-treatment with 100 nM and 1000 nM progesterone and completely abolished at a progesterone concentration of 1000 nM. Progesterone was also able to induce phosphorylation of ERK1/2 in these cells. Pretreatment of JEG-3 cells with a specific MAPK inhibitor (UO126) inhibited progesterone's inhibitory effect on cell death. Collectively, these data provide evidence of cross-talk between progesterone and IL-1β in this aggressive and poorly understood tumour that involves activation of a MAPK pathway and involvement of numerous progesterone receptors.This research was funded by a National Institutes of Health Grant ESO12961. Novel sputtering-technology for grain-size control
In this paper, we present a description of a novel high-rate plasma sputtering system that allows the control of grain size in sputtered films. Additionally, the system has the advantage of a better utilization of the target material (around 80% to 90%) by eliminating the race track at the target as in conventional plasma magnetron sputtering systems. The potential and capabilities of this novel plasma sputtering device are demonstrated in this paper by the deposition of a number of different Cr thin films suitable for underlayers in thin-film media and for which we have performed a systematic X-ray and TEM analysis to determine the grain-size histograms, mean grain diameters, and their relationship to the sputtering processes
Evolutionary game dynamics of controlled and automatic decision-making
We integrate dual-process theories of human cognition with evolutionary game
theory to study the evolution of automatic and controlled decision-making
processes. We introduce a model where agents who make decisions using either
automatic or controlled processing compete with each other for survival. Agents
using automatic processing act quickly and so are more likely to acquire
resources, but agents using controlled processing are better planners and so
make more effective use of the resources they have. Using the replicator
equation, we characterize the conditions under which automatic or controlled
agents dominate, when coexistence is possible, and when bistability occurs. We
then extend the replicator equation to consider feedback between the state of
the population and the environment. Under conditions where having a greater
proportion of controlled agents either enriches the environment or enhances the
competitive advantage of automatic agents, we find that limit cycles can occur,
leading to persistent oscillations in the population dynamics. Critically,
however, these limit cycles only emerge when feedback occurs on a sufficiently
long time scale. Our results shed light on the connection between evolution and
human cognition, and demonstrate necessary conditions for the rise and fall of

Kinematics of diffuse ionized gas in the disk halo interface of NGC 891 from Fabry-P\'erot observations
The properties of the gas in halos of galaxies constrain global models of the
interstellar medium. Kinematical information is of particular interest since it
is a clue to the origin of the gas. Here we report observations of the
kinematics of the thick layer of the diffuse ionized gas in NGC 891 in order to
determine the rotation curve of the halo gas. We have obtained a Fabry-P\'erot
data cube in Halpha to measure the kinematics of the halo gas with angular
resolution much higher than obtained from HI 21 cm observations. The data cube
was obtained with the TAURUS II spectrograph at the WHT on La Palma. The
velocity information of the diffuse ionized gas extracted from the data cube is
compared to model distributions to constrain the distribution of the gas and in
particular the halo rotation curve. The best fit model has a central
attenuation tau_H-alpha=6, a dust scale length of 8.1 kpc, an ionized gas scale
length of 5.0 kpc. Above the plane the rotation curve lags with a vertical
gradient of -18.8 km/s/kpc. We find that the scale length of the H-alpha must
be between 2.5 and 6.5 kpc. Furthermore we find evidence that the rotation
curve above the plane rises less steeply than in the plane. This is all in
agreement with the velocities measured in the HI.Comment: A&A, in press. 13 pages, 19 figure

Expression of membrane and nuclear progesterone receptors in two human placental choriocarcinoma cell lines (JEG-3 and BeWo): Effects of syncytialization

Copyright @ 2011 Spandidos Publications Ltd.A vital function of the human placenta is to produce steroid hormones such as progesterone, which are essential for the maintenance of pregnancy and the onset of parturition. Although choriocarcinoma cell lines are valuable placental models for investigations of steroid hormone actions, little is known about the expression of progesterone receptors (PRs) in these cell lines. Therefore, in this study, the expression of membrane and nuclear PRs was investigated in cultures of fusigenic (BeWo) and non-fusigenic (JEG-3) human choriocarcinoma cell lines. In addition, the effects of an inducer of syncytialization (forskolin) on the PR expression in BeWo cells were assessed. Quantitative RT-PCR revealed that in fully syncytialized BeWo cells (treated with 50 mu M forskolin for 72 h) there was a significant down-regulation of mPR alpha and up-regulation of mPR beta and of the progesterone membrane component-1 (PGRMC1) when compared with non-syncytialized BeWo cells. Expression of all the mPR and PGRMC1 mRNAs was significantly lower in JEG-3 cells compared to non-syncytialized BeWo cells. Interestingly, expression of PR-B was unaltered between the two BeWo states but was significantly higher in JEG-3 cells. Immunofluorescence analysis revealed that mPR proteins are differentially expressed in these choriocarcinoma cell lines as well as in the human placenta. The data demonstrate that human choriocarcinoma cell lines have a complex system of progesterone signalling involving multiple classes of PRs. The finding that syncytialization is accompanied by changes in the expression of these receptors may suggest that this process influences progesterone signalling
Time-resolved imaging of non-diffusive carrier transport in long-lifetime halide perovskite thin films
Owing to their exceptional semiconducting properties, hybrid
inorganic-organic perovskites show great promise as photovoltaic absorbers. In
these materials, long-range diffusion of charge carriers allows for most of the
photogenerated carriers to contribute to the photovoltaic efficiency. Here,
time-resolved photoluminescence (PL) microscopy is used to directly probe
ambipolar carrier diffusion and recombination kinetics in hybrid perovskites.
This technique is applied to thin films of methylammonium lead tri-iodide
MAPbI obtained with two different fabrication routes, methylammonium lead
tribromide (MAPbBr), and an alloy of formamidinium lead tri-iodide
(FAPbI) and methylammonium lead bromide
FAMAPb(IBr_). Average diffusion
coefficients in the films leading to the highest device efficiencies and
longest lifetimes, i.e., in FAMAPb(IBr)
and acetonitrile-processed MAPbI, are found to be several orders of
magnitude lower than in the other films. Further examination of the
time-dependence shows strong evidence for non-diffusive transport. In
particular, acetonitrile-processed MAPbI shows distinct diffusion regimes
on short and long timescales with an effective diffusion constant varying over
2 orders of magnitude. Our results also highlight the fact that increases in
carrier lifetime in this class of materials are not necessarily concomitant
with increased diffusion lengths and that the PL quantum efficiency under solar
cell operating conditions is a greater indication of material, and ultimately
device, quality
Memento Mori: The development and validation of the Death Reflection Scale
Despite its potential for advancing organizational behavior (OB) research, the topic of death awareness has been vastly understudied. Moreover, research on death awareness has predominantly focused on the anxiety‐provoking aspect of death‐related cognitions, thus overlooking the positive aspect of death awareness, death reflection. This gap is exacerbated by the lack of a valid research instrument to measure death reflection. To address this issue, we offer a systematic conceptualization of death reflection, develop the Death Reflection Scale, and assess its psychometric properties across four studies. Further, using a sample of 268 firefighters, we examine whether death reflection buffers the detrimental impact of mortality cues at work on employee well‐being and safety performance. Results provide strong support for the psychometric properties of the Death Reflection Scale. Further, moderation analysis indicates death reflection weakens the negative effect of mortality cues on firefighters' safety performance. Overall, these findings suggest the newly developed Death Reflection Scale will prove useful in future research on death‐related cognitions

A continuously updated, geospatially rectified database of utility-scale wind turbines in the United States.
Over 60,000 utility-scale wind turbines are installed in the United States as of October, 2019, representing over 97 gigawatts of electric power capacity; US wind turbine installations continue to grow at a rapid pace. Yet, until April 2018, no publicly-available, regularly updated data source existed to describe those turbines and their locations. Under a cooperative research and development agreement, analysts from three organizations collaborated to develop and release the United States Wind Turbine Database (USWTDB) - a publicly available, continuously updated, spatially rectified data source of locations and attributes of utility-scale wind turbines in the United States. Technical specifications and wind facility data, incorporated from five sources, undergo rigorous quality control. The location of each turbine is visually verified using high-resolution aerial imagery. The quarterly-updated data are available in a variety of formats, including an interactive web application, comma-separated values (CSV), shapefile, and application programming interface (API). The data are used widely by academic researchers, engineers and developers from wind energy companies, government agencies, planners, educators, and the general public
Properties of synchronization in the systems of non-identical coupled van der Pol and van der Pol - Duffing oscillators. Broadband synchronization
The particular properties of dynamics are discussed for the dissipatively
coupled van der Pol oscillators, non-identical in values of parameters
controlling the Hopf bifurcation. Possibility of a special synchronization
regime in an infinitively long band between oscillation death and quasiperiodic
areas is shown for such system. Features of the bifurcation picture are
discussed for different values of the control parameters and for the case of
additional Duffing-type nonlinearity. Analysis of the abridged equations is
