Do Avanafil and Zaprinast Change Some Selected Cytokine Levels In Ovariectomized Rat’s Liver?
Studies reported that phosphodiesterase-5 inhibitors (PDE-5Is) positively contributed to bone-mineral-density and thickness in rats with ovariectomy, which have the same condition with postmenapozal period. To explain the positive contribution mechanism on bone mineral density of PDE-5Is, we investigated the effect of zaprinast and avanafil on levels of some pro -or anti-resorptive cytokines in ovariectomized-rats. Albino female rats (8 months and 250-350 g) were used and four groups of equal-number were randomly assigned (n=6). Groups; was the sahm operated, positive control (OVX), Zaprinast and OVX, Avanafil and OVX groups, respectively. The levels of Estrogen, IL-1β, IL-6, IL-8, IL-10 and TNF-α were measured by ELISA kits, in liver of rats. IL-1β, IL-6, IL-8 and TNF-α levels were high in groups with OVX compared to sham group, while IL-10 levels were low. Also, IL-1β, IL-6, IL-8 and TNF-α levels were low in zaprinast and especially avanafil-treated groups with OVX and were similar to the sahm group values (p=0.001 for IL-1β, p=0.045 for IL-6, p=0.008 for IL-8, p=0.006 for IL-10, p=0.026 for TNF-α). Zaprinast and especially avanafil inhibited IL-1β, 8 and TNF-α and increased the IL-10 levels compared to the OVX group. This may support opinion that PDE-5Is enhance bone mineralization by inhibiting proresorptive cytokines
Tip 2 Diyabetli Bir Hastada Linagliptine Bağlı Lokalize Ağrılı Döküntü
The number of patients with type 2 diabetes using dipeptidylpeptidase-4 (DPP-4) inhibitors is increasing across the world.Although this class of antidiabetic medications is generallysafe and associated with less side effects compared to otheroral antidiabetic medications, they could also cause some sideeffects such as skin rashes. Herein we report a case of type 2diabetes patient who developed a painful maculopapular rashinduced by linagliptin, a widely used DPP-4 inhibitor.A localized painful maculopapular rash developed on thepalmar faces of the patient’s hands almost 1 day after theinitiation of the drug. The patient was using intensive insulintherapy before linagliptin was started. There was no eruptionon the other body parts except the palmar faces of the hands.Following the discontinuation of the drug, the rash disappearedin about four days. The patient had no history of urticaria anddid not use an ACE inhibitor. As far as we know, this is the firstcase report of a skin rash induced by linagliptin in the Turkishliterature. This case report highlights a rare and lesser knownside effect of linagliptin, a new member of DPP-4 inhibitors.Dipeptidil peptidaz-4 (DPP-4) inhibitörleri kullanan tip 2 diyabet hastalarının sayısı dünya genelinde artmaktadır. Bu antidiyabetik ilaç sınıfı genelde güvenli olmakla birlikte diğer oral antidiyabetik ilaçlarla karşılaştırıldığında daha az yan etki ile ilişkili olsa da, deri döküntüleri gibi bazı yan etkilerle ilişkili olabilirler. Burada yaygın olarak kullanılan bir DPP-4 inhibitörü olan linagliptin ile indüklenen ağrılı makülopapüler döküntü gelişen tip 2 diyabetli bir hastayı sunuyoruz. Linagliptin başlandıktan 1 gün sonra hastamızın ellerinin palmar yüzünde lokalize ağrılı makülopapüler bir döküntü gelişti. Linagliptin başlanmadan önce hasta intensif insülin tedavisi altındaydı. Vücudun diğer bölgelerinde herhangi bir döküntü yoktu. Linagliptin kesildikten sonra 4 gün içerisinde döküntü kayboldu. Hastanın ürtiker öyküsü yoktu ve Anjiyotensin dönüştürücü enzim inhibitörü kullanmıyordu. Bildiğimiz kadarıyla bu, Türk literatüründe Linagliptine bağlı deri döküntüsü geliştirdiği rapor edilen ilk olgudur. Bu olgu sunumu, DPP-4 inhibitörlerinin nadir görülen ve daha az bilinen bir yan etkisini vurgulamaktadır
The Effects of Some Phosphodiesterase 5 Inhibitors on Oxidative Stress, VEGF, BMP 2 and 9 in the Liver Tissue of Ovariectomized Rats
INTRODUCTION: Osteoporosis is an important health problem and there is no effective treatment yet. phosphodiesterase 5 inhibitors are promising agents for the treatment of osteoporosis. In this study, we aimed to determine the effects of phosphodiesterase 5 inhibitors (vardenafil, tadalafil, and udenafil) on bone morphogenic protein-2 and 9 (BMP-2 and 9), vascular endothelial growth factor (VEGF), and oxidative stress markers (malondialdehyde and CoQ10) in liver tissue of rats with ovariectomy-induced osteoporosis. METHODS: 50 Albino wistar female rats were randomly divided into 5 groups of 10 rats per group. Groups were the sham-operated, ovariectomise (OVEX), OVEX + Tadalafil, OVEX + udenafil and OVEX + vardenafil, respectively. VEGF, BMP-2 and 9 levels were measured by ELISA kits. To detect levels of MDA and CoQ10, we used high pressure liquid chromatography method. RESULTS: The levels of VEGF, BMP-2 and 9 levels in the groups that applied PDE-5 inhibitors were significantly higher than in the sham and OVEX groups. There was no significant difference between the OVEX+vardenafil and OVEX+udenafil groups in terms of VEGF, BMP-2 and 9 levels. The levels of MDA and CoQ10 were significantly lower in the groups that applied PDE5 inhibitors than in the OVEX group. When the histological and immunohistochemical results were examined, it was seen that angiogenesis was significantly higher in PDE-5 inhibitor groups. DISCUSSION AND CONCLUSION: In conclusion, we can say that these inhibitors may have positi ve effects on bone mineralization and remodelling by inducing the expression of VEGF, BMP-2 and 9 in liver tissue
Association of Insulin Receptor Substrate-2 Gene Polymorphism with Ovarian Cancer
Objective: The insulin receptor substrate (IRS) proteins have been implicated in contributing to all stages of tumorigenesis in many cancers, from initiating events to metastatic progression. In this study, we aimed to evaluate the association of IRS-2 gene polymorphism with ovarian cancer. Material and Methods: The study group consisted of 185 women: 45 women with ovarian cancer and 140 control subjects. All the patients with ovarian cancer were primarily treated by surgical intervention. Genetic polymorphism of IRS-2 G1057D was detected by using polymerase chain reaction (PCR)-based restriction fragment-length polymorphism (RFLP).Results: For IRS-2 G1057D polymorphism, the frequencies of GG, GD and DD genotypes were 60%, 33.3%, and 6.7%, respectively in the ovarian cancer cases and 53.6%, 38.6% and 7.9 % in controls. The risk for ovarian cancer was not significantlydifferent in the individuals with the IRS-2 DD genotype compared to the GG genotype (95% CI: 0.203-3.04, P: 0.787). Also, there was no association between carriage of the D allele and ovarian cancer risk. We found no significant difference between the genotypes in the ovarian cancer group and control group (P:0.545).Conclusion: These results do not support an association between carriage of the G1057D variant of IRS-2 gene and ovarian cancer

Knowledge sharing behaviors in e-learning community: Exploring the role of academic self-efficacy and sense of community
The purpose of this study was to explore the structural relationships
between knowledge sharing behaviors (KSB), academic self-efficacy (ASE)
and sense of community (SoC) of university students in e-learning
community. The study was carried out with students who joined Facebook
learning community that was created for the Computing I course which was
taught with blended learning method. Data were collected from 316
university students by utilizing three self-report instruments: KSB
scale, ASE scale (sub-scales: `social status', `cognitive applications'
and `technical skills') and classroom community (CC) scale (sub-scales:
`connectedness' and `learning'). The path analyses with structural
equation modeling (SEM) further verified that students' KSB were related
to their ASE and SoC in e-learning community. The results of the study
revealed that the ASE and SoC of the students positively affect their
KSB. And in terms of sub-scales, the connectedness to the community,
learning perception in the community, the self efficacy of the students
on the cognitive applications in the courses and their social status in
the community positively affect KSB. However, students' self-efficacy
perceptions on their technical skills affect KSB positively but its
affect size was smaller compared to other sub-scales: Further research
studies and implications are presented and discussed. (C) 2016 Elsevier
