    The report is one of a series of outputs from the national evaluation of the CBP, being undertaken by a team of researchers at the Policy Research Institute (PRI) at Leeds Metropolitan University and the Cities Research Unit at the University of West of England. The Capacity Building Programme for local government was launched in 2003 as a joint Department for Communities and Local Government (DCLG) / Local Government Association (LGA) initiative to support capacity building and improvement activities within local authorities in England. The evaluation of the Capacity Building Programme has been underway since late 2004. A scoping phase was conducted until May 2005, including a short evaluation of the Pilot Programmes. The main phase of the evaluation commenced in September 2005 and encompassed four main phases (see Section 1.3: p10)

    Using Unnatural Amino Acid Mutagenesis To Probe the Regulation of PRMT1

    Protein arginine methyltransferase 1 (PRMT1)-dependent methylation contributes to the onset and progression of numerous diseases (e.g., cancer, heart disease, ALS); however, the regulatory mechanisms that control PRMT1 activity are relatively unexplored. We therefore set out to decipher how phosphorylation regulates PRMT1 activity. Curated mass spectrometry data identified Tyr291, a residue adjacent to the conserved THW loop, as being phosphorylated. Natural and unnatural amino acid mutagenesis, including the incorporation of p-carboxymethyl-L-phenylalanine (pCmF) as a phosphotyrosine mimic, were used to show that Tyr291 phosphorylation alters the substrate specificity of PRMT1. Additionally, p-benzoyl-L-phenylalanine (pBpF) was incorporated at the Tyr291 position, and cross-linking experiments with K562 cell extracts identified several proteins (e.g., hnRNPA1 and hnRNP center dot center dot H3) that bind specifically to this site. Moreover, we also demonstrate that Tyr291 phosphorylation impairs PRMT1\u27s ability to bind and methylate both proteins. In total, these studies demonstrate that Tyr291 phosphorylation alters both PRMT1 substrate specificity and protein-protein interacions

    Revised Status of Rare and Endangered Unionaea (Mollusca: Margaritiferidae, Unionidae) in Arkansas

    Harris and Gordon (1987) reviewed the distribution and status of 18 rare and /or endangered unionacean bivalve species (commonly referred to as clams, mussels, freshwater mussels, naiads) that occur or have occurred in Arkansas. They discussed four species that were federally listed as endangered, four species that were considered endangered or extirpated within Arkansas, four species considered threatened within Arkansas, four species of special concern within Arkansas, and two species for which the conservation status was considered uncertain due to questions regarding taxonomic validity. Numerous unionacean field surveys have been performed during 1986 1996, and a substantial database of new distributional and relative abundance information has been accumulated. Two additional unionacean species have been listed as federally endangered, one additional species has been listed as federally threatened, and one endangered species has been newly discovered within Arkansas bringing the total number of federally protected unionacean species occurring within Arkansas to eight. The conservation status of 16 additional unionacean species occurring in Arkansas is discussed also

    MACiE: a database of enzyme reaction mechanisms.

    SUMMARY: MACiE (mechanism, annotation and classification in enzymes) is a publicly available web-based database, held in CMLReact (an XML application), that aims to help our understanding of the evolution of enzyme catalytic mechanisms and also to create a classification system which reflects the actual chemical mechanism (catalytic steps) of an enzyme reaction, not only the overall reaction.     The Eruption from a Sigmoidal Solar Active Region on 2005 May 13

    This paper presents a multiwavelength study of the M8.0 flare and its associated fast halo CME that originated from a bipolar active region NOAA 10759 on 2005 May 13. The source active region has a conspicuous sigmoid structure at TRACE 171 A channel as well as in the SXI soft X-ray images, and we mainly concern ourselves with the detailed process of the sigmoid eruption as evidenced by the multiwavelength data ranging from Halpha, WL, EUV/UV, radio, and hard X-rays (HXRs). The most important finding is that the flare brightening starts in the core of the active region earlier than that of the rising motion of the flux rope. This timing clearly addresses one of the main issues in the magnetic eruption onset of sigmoid, namely, whether the eruption is initiated by an internal tether-cutting to allow the flux rope to rise upward or a flux rope rises due to a loss of equilibrium to later induce tether cutting below it. Our high time cadence SXI and Halpha data shows that the first scenario is relevant to this eruption. As other major findings, we have the RHESSI HXR images showing a change of the HXR source from a confined footpoint structure to an elongated ribbon-like structure after the flare maximum, which we relate to the sigmoid-to-arcade evolution. Radio dynamic spectrum shows a type II precursor that occurred at the time of expansion of the sigmoid and a drifting pulsating structure in the flare rising phase in HXR. Finally type II and III bursts are seen at the time of maximum HXR emission, simultaneous with the maximum reconnection rate derived from the flare ribbon motion in UV.     Growth Curves of Four Species of Commercially Valuable Freshwater Mussels (Bivalva: Unionidae)

    North American freshwater mussels (Bivalvia: Unionidae) have been exploited commercially for over 100 years and have been regulated using shell size limits and/or harvest seasons. Presently, freshwater mussels are considered a threatened faunal group in North America due to the large numbers of endangered, threatened or special concern species. Therefore, management of this fauna should emphasize their long-term sustainability. The objectives of this study were 1) to construct von Bertalanffy growth curves for selected commercially-most-valuable species, Fusconaia ebena, Megalonaias nervosa, Amblema plicata and Quadrula quadrula, from five rivers and two reservoirs, 2) to compare species-specific von Bertalanffy growth curves from different rivers and reservoirs, and 3) to provide information on size at onset of sexual maturity in F. ebena and A. plicata. Von Bertalanffy growth curves of four commercially valuable Ambleminae species were used in this study to compare drainage-specific growth. Growth curves for all four species investigated were significantly different between pairs of drainages. Approximate size at onset of sexual maturity was determined for Arkansas F. ebena and A. plicata. Von Bertalanffy growth curves, coupled with life history and population dynamics information, could be useful in assessing and determining national/state harvest sizes and/or drainage specific harvest sizes once annual growth line formation is confirmed

    Two complementary approaches for intracellular delivery of exogenous enzymes.

    Intracellular delivery of biologically active proteins remains a formidable challenge in biomedical research. Here we show that biomedically relevant enzymes can be delivered into cells using a new DNA transfection reagent, lipofectamine 3000, allowing assessment of their intracellular functions. We also show that the J774.2 macrophage cell line exhibits unusual intracellular uptake of structurally and functionally distinct enzymes providing a convenient, reagent-free approach for evaluation of intracellular activities of enzymes

    Partially-erupting prominences: a comparison between observations and model-predicted observables

    <p><b>Aims:</b> We investigate several partially-erupting prominences to study their relationship with other CME-associated phenomena and compare these observations with observables predicted by a model of partially-expelled-flux-ropes (Gibson & Fan 2006a, ApJ, 637, L65; 2006b, J. Geophys. Res., 111, 12103).</p> <p><b>Methods:</b> We studied 6 selected events with partially-erupting prominences using multi-wavelength observations recorded by the Extreme-ultraviolet Imaging Telescope (EIT), Transition Region and Coronal Explorer (TRACE), Mauna Loa Solar Observatory (MLSO), Big Bear Solar Observatory (BBSO), and Soft X-ray Telescope (SXT). The observational features associated with partially-erupting prominences were then compared with the predicted observables from the model.</p> <p><b>Results:</b> The partially-expelled-flux-rope (PEFR) model can explain the partial eruption of these prominences, and in addition predicts a variety of other CME-related observables that provide evidence of internal reconnection during eruption. We find that all of the partially-erupting prominences studied in this paper exhibit indirect evidence of internal reconnection. Moreover, all cases showed evidence of at least one observable unique to the PEFR model, e.g., dimmings external to the source region and/or a soft X-ray cusp overlying a reformed sigmoid.</p> <p><b>Conclusions:</b> The PEFR model provides a plausible mechanism to explain the observed evolution of partially-erupting-prominence-associated CMEs in our study.</p&gt

    Not Pregnant Now in almost two-thirds of all municipalities in The Netherlands

    Het programma Nu Niet Zwanger is gericht op het voorkomen van onbedoelde zwangerschappen bij mensen in kwetsbare omstandigheden. Hulpverleners in het sociaal en medisch domein gaan daartoe tijdens reguliere contactmomenten met hun eigen cliënten (m/v) in gesprek over hun kinderwens. Ze ondersteunen hen bij het maken van een geïnformeerde en vrijwillige keuze, en bieden desgewenst begeleiding bij het kiezen en het realiseren van anticonceptie. Onderzoek laat zien dat de ervaringen van hulpverleners en cliënten met Nu Niet Zwanger positief zijn en leverde ook verbeterpunten op. Begin 2023 wordt het programma in bijna twee derde van de gemeenten in Nederland uitgevoerd en ligt de focus op structurele, landelijke borging.The program Not Pregnant Now aims to prevent unintended pregnancies in vulnerable populations. To this end, care providers start an openconversation during regular social and healthcare visits with their clients about their desire to have children. They support their clients in making an informed and voluntary decision about this and, ifwanted, offer support in choosing and realizing contraception. Research showed that the experiences of the care providers and clients with the program arepositive and also resulted in recommendations for improvement. At the beginning of 2023, the program runs in three quarters of the municipalities in TheNetherlands and the focus is on structural, national maintenance
