    Analysis of virulence genes in Escherichia coli isolated from grated cheese

    This research aimed to verify the presence of virulence genes in strains of Escherichia coli isolated from grated cheese sold in farmers' markets of Cuiaba-MT, Brazil. Forty samples of this food were submitted for microbiological analysis and 22 (55%) tested positive for E. coli. Next, 64 strains of E. coli were isolated from the positive samples and screened by the polymerase chain reaction (PCR) for the presence of the genes encoding the following virulence factors: stx1 and stx2 (verotoxin types 1 and 2), eae (intimin), lt1 (heat-labile toxin type 1), st1 (heat-stable toxin type 1), cnf1 and cnf2 (cytotoxic necrozing factor types 1 and 2), and cdtB (cytolethal distending toxin). All the isolates were negative for the genes stx1, stx2, eae, lt1, st1, cnf1, and cdtB, and five strains (7.81%) were positive for cnf2. A low prevalence of E. coli positive for virulence factors associated with the pathogenesis of diarrhoea was observed in this study. However, the presence of CNF-2 producing strains and the possibility of occurrence and scattering of other virulence factors that were not surveyed in the work indicate the risk related to the consumption of grated cheese from farmers' markets.31110610

    Prevalence of enterotoxigenic and Shiga toxin-producing Escherichia coli in pigs slaughtered in Mato Grosso, Brazil

    Introduction: This study aimed to estimate the prevalence of enterotoxigenic Escherichia coli (ETEC) and Shiga toxin-producing Escherichia coli (STEC) strains in pigs slaughtered in abattoirs located in the state of Mato Grosso, Brazil. Methodology: Intestinal samples from 74 animals were aseptically dissected and lumen content was plated on MacConkey agar. Confluent colonies from each plate were screened for the presence of ETEC and STEC strains by PCR assays. Results: It was verified that the prevalence of STEC and ETEC carriers was 1.35% and 9.46% respectively. One (1.35%) of the 74 samples tested was positive for the stx2 gene, and seven (9.46%) for st1, of which two (2.70%) were also positive for lt1. Conclusion: The results provided represent a benchmark for future research on pathogenic E. coli of porcine origin in Mato Grosso.5212312

    The fourth wave of Portuguese emigration: Austerity policies, European peripheries and postcolonial continuities

    Little is known about emigration in European countries. Migratory pressure and the recent refugee crisis have helped keep academic attention over the last few decades focused on immigration, asylum and integration in Europe. However, these dynamics promoting entries into European countries coexist with other fair-ly significant dynamics promoting departures from these countries. The sovereign debt crisis coupled with austerity policies that asymmetrically affected Europe’s peripheral countries have increased emigration in various European countries. Our book aims to counter the invisibility of emigration from European countries in the literature by examining the particularities of the Portuguese case. In methodological terms, the book compiles the work of authors from different academic backgrounds who have conducted empirical research using a wide vari-ety of extensive and intensive methods. It is argued that when analysing recent Portuguese emigration it is important to examine in further detail: i) the impact of the 2008 economic and financial crisis and the austerity policies that followed in its wake; ii) south-north emigration in Europe; iii) north-south emigration outside Europe and post-colonial continuities; iv) the importance of reassessing the exist-ing model of Southern European migration; v) highly skilled and less skilled mi-gration; and finally, vi) emigrants’ and their descendants’ identities.info:eu-repo/semantics/publishedVersio

    Evolutionary and Experimental Assessment of Novel Markers for Detection of Xanthomonas euvesicatoria in Plant Samples

    BACKGROUND: Bacterial spot-causing xanthomonads (BSX) are quarantine phytopathogenic bacteria responsible for heavy losses in tomato and pepper production. Despite the research on improved plant spraying methods and resistant cultivars, the use of healthy plant material is still considered as the most effective bacterial spot control measure. Therefore, rapid and efficient detection methods are crucial for an early detection of these phytopathogens. METHODOLOGY: In this work, we selected and validated novel DNA markers for reliable detection of the BSX Xanthomonas euvesicatoria (Xeu). Xeu-specific DNA regions were selected using two online applications, CUPID and Insignia. Furthermore, to facilitate the selection of putative DNA markers, a customized C program was designed to retrieve the regions outputted by both databases. The in silico validation was further extended in order to provide an insight on the origin of these Xeu-specific regions by assessing chromosomal location, GC content, codon usage and synteny analyses. Primer-pairs were designed for amplification of those regions and the PCR validation assays showed that most primers allowed for positive amplification with different Xeu strains. The obtained amplicons were labeled and used as probes in dot blot assays, which allowed testing the probes against a collection of 12 non-BSX Xanthomonas and 23 other phytopathogenic bacteria. These assays confirmed the specificity of the selected DNA markers. Finally, we designed and tested a duplex PCR assay and an inverted dot blot platform for culture-independent detection of Xeu in infected plants. SIGNIFICANCE: This study details a selection strategy able to provide a large number of Xeu-specific DNA markers. As demonstrated, the selected markers can detect Xeu in infected plants both by PCR and by hybridization-based assays coupled with automatic data analysis.     Apparent and content validation of maternal self-efficiency scale for prevention of childhood diarrhea

    AIM: The aim of this paper is to describe the apparent and content validation for the Maternal Self-Efficiency Scale for the Prevention of Childhood Diarrhoea. METHOD: Methodological study with the execution of apparent and content validation by seven judges; semantic analysis, by 30 mothers of children under 5 years old and also a pre-test involving 31 mothers who have been selected through convenience. It has been considered necessary to have the agreement of at least 70% of the judges for apparent validation and a minimum of 80% for pertinence and Index of Content Validation. RESULTS: This paper shows that most items have been considered clear, comprehensive and relevant by the judges. The final Content Validity Index of the scale was 0.96. The suggestions of the mothers were accepted. CONCLUSION: The scale ended up having 25 items and two domains (family hygiene and general/eating practices) which assess the maternal self-efficiency for the prevention of diarrhea in their children, thereby contributing to the planning of nursing interventions

    The HepHIV 2023 Madrid conference: A call to action for political leadership in reaching the sustainable development goals on earlier testing and linkage to care for HIV, viral hepatitis, and sexually transmitted infections

    Introduction and Objectives: The HepHIV 2023 Conference, held in Madrid in November 2023, highlighted how Europe is not on track to meet the United Nations (UN) sustainable development goals and Joint UN Programme on HIV/AIDS (UNAIDS) targets. This article presents the outcomes of the conference, which focus on ways to improve testing and linkage to care for HIV, viral hepatitis, and other sexually transmitted infections. HIV-related stigma and discrimination, a major barrier to progress, was a key concept of the conference and on the agenda of the Spanish Presidency of the European Union. Methods: The HepHIV 2023 organizing committee, alongside the Spanish Ministry of Health, oversaw the conference organization and prepared the scientific programme based on abstract rankings. Key outcomes are derived from conference presentations and discussions. Results: Conference presentations covered the obstacles that HIV-related stigma and discrimination continue to pose to access to services, models for data collection to better monitor progress in the future, and examples of legislative action that can be taken at national levels. Diversification of testing approaches was also highlighted, to reach key populations, (e.g. migrant populations), to increase testing offered in healthcare settings (e.g. emergency departments), and to account for different stages of epidemics across the region. Conclusion: With a strong call for intensified action to address the impact of HIV-related stigma and discrimination on testing uptake, the conference concluded that strengthened collaboration is required between governments and implementers around testing and linkage to care. There is also an ongoing need to ensure sustainable political commitment and appropriate resource allocation to address gaps and inequalities in access for key populations and to focus on the implementation of integrated responses to HIV, viral hepatitis, and sexually transmitted infections. © 2024 The Author(s).
