    Who can help me? Reconstructing users' psychological journeys in depression-related social media interactions

    Social media are increasingly being used as self-help boards, where individuals can disclose personal experiences and feelings and look for support from peers or experts. Here we investigate several popular mental health-related Reddit boards about depression while proposing a novel psycho-social framework. We reconstruct users' psychological/linguistic profiles together with their social interactions. We cover a total of 303,016 users, engaging in 378,483 posts and 1,475,044 comments from 01/05/2018 to 01/05/2020. After identifying a network of users' interactions, e.g., who replied to whom, we open an unprecedented window over psycholinguistic, cognitive, and affective digital traces with relevance for mental health research. Through user-generated content, we identify four categories or archetypes of users in agreement with the Patient Health Engagement model: the emotionally turbulent/under blackout, the aroused, the adherent-yet-conflicted, and the eudaimonically hopeful. Analyzing users' transitions over time through conditional Markov processes, we show how these four archetypes are not consecutive stages. We do not find a linear progression or sequential patient journey, where users evolve from struggling to serenity through feelings of conflict. Instead, we find online users to follow spirals towards both negative and positive archetypal stages. Through psychological/linguistic and social network modelling, we can provide compelling quantitative pieces of evidence on how such a complex path unfolds through positive, negative, and conflicting online contexts.     Aflibercept and Ranibizumab modulate retinal pigment epithelial cells function by acting on their cross talk with vascular endothelial cells

    Background/Aims: We performed co-culture experiments between human RPE cells (ARPE-19) and human umbilical vascular endothelial cells (HUVEC) in order to evaluate how anti-VEGF drugs could affect NO release, mitochondrial function, the oxidative status, proliferation and migration of RPE cells through modulation of their cross talk with vascular endothelial cells. Materials: The co-culture HUVEC/RPE, was exposed to Ranibizumab/Aflibercept in the absence/presence of the NO synthase (NOS) inhibitor, the phosphatidylinositol 3\u2032-kinase (PI3K), the extracellular-signal-regulated kinases 1/2 (ERK1/2) and the p38 mitogen-activated protein kinase (p38 MAPK) blockers. Specific kits were used for cell viability, mitochondrial membrane potential, NO, ROS and GSH production. Western blot was performed for apoptosis markers, NOS isoforms, and others kinases detection. Cell migration was analyzed by scratch assay, whereas cell proliferation and cell cycle through xCELLigence and flow cytometry. Results: In RPE cells co-cultured with HUVEC in physiological conditions, Aflibercept/Ranibizumab increased NO release in a dose and time-dependent way. Opposite results were obtained in peroxidative conditions. Both anti-VEGF agents were able to prevent the fall of cell viability and mitochondrial membrane potential, an effect which was reduced by various inhibitors, and increased cell migration. Aflibercept/Ranibizumab counteracted the changes of apoptosis markers, NOS expression/activation, PI3K and ERK1/2 activation caused by peroxidation. These results were confirmed by cell cycle analysis. Conclusion: This study has shown new mechanisms at the basis of protective effects elicited by Aflibercept/Ranibizumab in RPE cells. HUVEC stimulated with Aflibercept/Ranibizumab, could release some paracrine factors that can modulate the RPE cells response in both physiologic and peroxidative conditions

    Association of Microvesicles With Graft Patency in Patients Undergoing CABG Surgery

    Abstract Background Graft patency is one of the major determinants of long-term outcome following coronary artery bypass graft surgery (CABG). Biomarkers, if indicative of the underlying pathophysiological mechanisms, would suggest strategies to limit graft failure. The prognostic value of microvesicles (MVs) for midterm graft patency has never been tested. Objectives The aim of this study was to evaluate whether MV pre-operative signature (number, cellular origin, procoagulant phenotype) could predict midterm graft failure and to investigate potential functional role of MVs in graft occlusion. Methods This was a nested case-control substudy of the CAGE (CoronAry bypass grafting: factors related to late events and Graft patency) study that enrolled 330 patients undergoing elective CABG. Of these, 179 underwent coronary computed tomography angiography 18 months post-surgery showing 24% graft occlusion. Flow cytometry MV analysis was performed in 60 patients (30 per group with occluded [cases] and patent [control subjects] grafts) on plasma samples collected the day before surgery and at follow-up. Results Before surgery, cases had 2- and 4-fold more activated platelet-derived and tissue-factor positive MVs respectively than control subjects. The MV procoagulant capacity was also significantly greater. Altogether this MV signature properly classified graft occlusion (area under the curve 0.897 [95% confidence interval: 0.81 to 0.98]; p Conclusions The pre-operative signature of MVs is independently associated with midterm graft occlusion in CABG patients and a cumulative MV score stratifies patients' risk. Because the MV signature mirrors platelet activation, patients with a high MV score could benefit from a personalized antiplatelet therapy

    Liquid vs. Gas Phase CO2 Photoreduction Process: Which Is the Effect of the Reaction Medium?

    The use of carbon dioxide, the most concerning environmental issue of the 21st century, as a feedstock for fuels productions still represents an innovative, yet challenging, task for the scientific community. CO2 photoreduction processes have the potential to transform this hazardous pollutant into important products for the energy industry (e.g., methane and methanol) employing a photocatalyst and light as the only energy input. In order to design an effective process, the high sustainability of this reaction should be matched with the perfect reaction conditions to allow the reactant, photocatalyst, and light source to come together: therefore, the choice of reaction conditions, and in particular its medium, is a crucial issue that needs to be investigated. Throughout this paper, a careful study of carbon dioxide photoreduction in liquid and vapour phases are reported, focusing on their effect on catalyst performances in terms of light harvesting, productivity, and selectivity. Different from most papers in the literature, catalytic tests were performed under extremely low light irradiance, in order to minimise the primary energy input, highlighting that this experimental variable has a great effect on the reaction pathway and, thus, product distribution

    Factors Controlling Hydrothermal Nickel and Cobalt Mineralization—Some Suggestions from Historical Ore Deposits in Italy

    We compare three poorly known, historical Ni–Co-bearing hydrothermal deposits in dierent geological settings in Italy: The Ni–Co–As–Sb–Au-bearing Arburese vein system (SW Sardinia), the Co–Ni–As-rich Usseglio vein system (Piedmont), and the small Cu–Ag–Co–Ni–Pb–Te–Se stockwork at Piazza (Liguria). These deposits share various (mineralogical, chemical, thermal, and stable isotopic) similarities to the Five Element Vein-type ores but only the first two were economic for Co–Ni. The Sardinian Ni-rich veins occur in Paleozoic basement near two Variscan plutons. Like the Co-rich Usseglio vein system, the uneconomic Piazza deposit is hosted in an ophiolite setting anomalous for Co. The Sardinian and Usseglio deposits share a polyphasic assemblage with Ni–Co–As–Sb–Bi followed by Ag-base metal sulfides, in siderite-rich gangue, whereas Piazza shows As-free, Ag–Pb–Te–Se-bearing Co–Ni–Cu sulfides, in prehnite–chlorite gangue. Fluid inclusions indicated Co–Ni arsenide precipitation at 170 C for Usseglio, whereas for the Sardinian system late sulfide deposition occurred within the 52–126 C range. Ore fluids in both systems are NaCl-CaCl2-bearing basinal brines. The chlorite geothermometer at Piazza provides the range of 200–280 C for ore deposition from CO2-poor fluids. Enrichments in Se and negative 13C in carbonates suggest interaction with carbonaceous shales. These deposits involve issues about source rocks, controls on Co/Ni and possible role of arsenic and carbonate components towards economic mineralization

    Anti-Vascular Endothelial Growth Factors Protect Retinal Pigment Epithelium Cells Against Oxidation by Modulating Nitric Oxide Release and Autophagy

    Background/Aims: the anti-vascular endothelial growth factors (VEGF), Aflibercept and Ranibizumab, are used for the treatment of macular degeneration. Here we examined the involvement of nitric oxide (NO), mitochondria function and of apoptosis/autophagy in their antioxidant effects in human retinal pigment epithelium cells (RPE). Methods: RPE were exposed to Ranibizumab/Aflibercept in the absence or presence of NO synthase (NOS) inhibitor and of autophagy activator/blocker, rapamicyn/3-methyladenine. Specific kits were used for cell viability, NO and reactive oxygen species detection and mitochondrial membrane potential measurement, whereas Western Blot was performed for apoptosis/ autophagy markers and other kinases detection. Results: In RPE cultured in physiological conditions, Aflibercept/Ranibizumab increased NO release in a dose and time-dependent way. Opposite results were obtained in RPE pretreated with hydrogen peroxide. Moreover, both the anti-VEGF agents were able to prevent the fall of cell viability and of mitochondrial membrane potential. Those effects were reduced by the NOS inhibitor and 3-methyladenine and were potentiated by rapamycin. Finally, Aflibercept and Ranibizumab counteracted the changes of apoptosis/autophagy markers, NOS, Phosphatidylinositol-3-Kinase/Protein Kinase B and Extracellular signal–regulated kinases 1/2 caused by peroxidation. Conclusion: Aflibercept and Ranibizumab protect RPE against peroxidation through the modulation of NO release, apoptosis and autophagy

    The Relation between the Theory of Mind and Socio-Emotional Functioning in a Sample of Older Adults

    We explored the relationship between the “Theory of Mind” (ToM) and socio-emotional functioning among primary aging individuals. Our sample was comprised of 266 older adults ranging from 65 years old to 94 years old. On all participants, we measured ToM and calculated indexes of performance such as causal connection and psychological lexicon. In addition, we assessed emotional (depression, alexithymia, distress, life satisfaction, empathy and hostility) and social variables (perception of the future, loneliness, social avoidance and perception of social support) with a battery of tests. The results of our cognitive screening indicated that 189 older adults presented normal or primary aging patterns. The results obtained with the sample of normal older adults showed that: a) social variables are independent from ToM; and b) emotional functioning predicted the making of causal connection and the use of psychological lexicon among participants who responded to ToM tasks. We discuss our work within a socio-emotional conceptual framework
