    Cryptic female choice favours sperm from major histocompatibility complex-dissimilar males

    Cryptic female choice may enable polyandrous females to avoid inbreeding or bias offspring variability at key loci after mating. However, the role of these genetic benefits in cryptic female choice remains poorly understood. Female red junglefowl, Gallus gallus, bias sperm use in favour of unrelated males. Here, we experimentally investigate whether this bias is driven by relatedness per se, or by similarity at the major histocompatibility complex (MHC), genes central to vertebrate acquired immunity, where polymorphism is critical to an individual's ability to combat pathogens. Through experimentally controlled natural matings, we confirm that selection against related males' sperm occurs within the female reproductive tract but demonstrate that this is more accurately predicted by MHC similarity: controlling for relatedness per se, more sperm reached the eggs when partners were MHC-dissimilar. Importantly, this effect appeared largely owing to similarity at a single MHC locus (class I minor). Further, the effect of MHC similarity was lost following artificial insemination, suggesting that male phenotypic cues might be required for females to select sperm differentially. These results indicate that postmating mechanisms that reduce inbreeding may do so as a consequence of more specific strategies of cryptic female choice promoting MHC diversity in offspring

    Challenges for funders in monitoring compliance with policies on clinical trials registration and reporting: analysis of funding and registry data in the UK

    Objectives: To evaluate compliance by researchers with funder requirements on clinical trial transparency, including identifying key areas for improvement; to assess the completeness, accuracy and suitability for annual compliance monitoring of the data routinely collected by a research funding body. / Design: Descriptive analysis of clinical trials funded between February 2011 and January 2017 against funder policy requirements. / Setting: Public medical research funding body in the UK. / Data sources: Relevant clinical trials were identified from grant application details, post-award grant monitoring systems and the International Standard Randomised Controlled Trial Number (ISRCTN) registry. / Main outcome measure: The proportion of all Medical Research Council (MRC)-funded clinical trials that were (a) registered in a clinical trial registry and (b) publicly reported summary results within 2 years of completion. / Results: There were 175 grants awarded that included a clinical trial and all trials were registered in a public trials registry. Of 62 trials completed for over 24 months, 42 (68%) had publicly reported the main findings by 24 months after trial completion; 18 of these achieved this within 12 months of completion. 11 (18%) trials took >24 months to report and 9 (15%) completed trials had not yet reported findings. Five datasets were shared with other researchers. / Conclusions: Compliance with the funder policy requirements on trial registration was excellent. Reporting of the main findings was achieved for most trials within 24 months of completion; however, the number of unreported trials remains a concern and should be a focus for future funder policy initiatives. Identifying trials from grant management and grant monitoring systems was challenging therefore funders should ensure investigators reliably provide trial registries with information and regularly update entries with details of trial publications and protocols

    CERN@school: bringing CERN into the classroom

    CERN@school brings technology from CERN into the classroom to aid with the teaching of particle physics. It also aims to inspire the next generation of physicists and engineers by giving participants the opportunity to be part of a national collaboration of students, teachers and academics, analysing data obtained from detectors based on the ground and in space to make new, curiosity-driven discoveries at school. CERN@school is based around the Timepix hybrid silicon pixel detector developed by the Medipix 2 Collaboration, which features a 300 μm thick silicon sensor bump-bonded to a Timepix readout ASIC. This defines a 256-by-256 grid of pixels with a pitch of 55 μm, the data from which can be used to visualise ionising radiation in a very accessible way. Broadly speaking, CERN@school consists of a web portal that allows access to data collected by the Langton Ultimate Cosmic ray Intensity Detector (LUCID) experiment in space and the student-operated Timepix detectors on the ground; a number of Timepix detector kits for ground-based experiments, to be made available to schools for both teaching and research purposes; and educational resources for teachers to use with LUCID data and detector kits in the classroom. By providing access to cutting-edge research equipment, raw data from ground and space-based experiments, CERN@school hopes to provide the foundation for a programme that meets the many of the aims and objectives of CERN and the project's supporting academic and industrial partners. The work presented here provides an update on the status of the programme as supported by the UK Science and Technology Facilities Council (STFC) and the Royal Commission for the Exhibition of 1851. This includes recent results from work with the GridPP Collaboration on using grid resources with schools to run GEANT4 simulations of CERN@school experiments

    Male-female interactions drive the (un)repeatability of copula duration in an insect.

    Across the animal kingdom the duration of copulation varies enormously from a few seconds to several days. Functional explanations for this variation are largely embedded within sperm competition theory in which males modulate the duration of copula in order to optimise their fitness. However, copulation is the union of two protagonists which are likely to have separate and often conflicting reproductive interests, yet few experimental designs specifically assess the effect of male-female interactions on the duration of copulation. This can result in inexact assertions over which sex controls copulatory behaviour. Here we analyse the repeatability of copulatory behaviour in the seed beetle Callosobruchus maculatus to determine which sex exerts primary influence over copulation duration. In C. maculatus, copulation follows two distinct phases: an initial quiescent phase followed by a period of vigorous female kicking behaviour that culminates in the termination of copulation. When males or females copulated with several novel mates, copulatory behaviour was not significantly repeatable. By contrast, when males or females mated repeatedly with the same mate, copula duration was repeatable. These data suggest copulatory behaviour in C. maculatus to be largely the product of male-female interactions rather than the consistent, sex-specific modulation of copula duration of one protagonist in response to the phenotypic variation presented by the other protagonist

    High-throughput amplicon sequencing reveals distinct communities within a corroding concrete sewer system

    This study investigated the variation in microbially induced concrete corrosion communities at different circumferential locations of a real sewer pipe and the effects of a wastewater flooding event on the community. Three distinct microbial community groups were found in different corrosion samples. The physico-chemical properties of the corrosion layers and the microbial communities were distinct for the cross-sectional positions within the pipe, ie ceiling, wall and tidal zones. The microbial communities detected from the same positions in the pipe were consistent over the length of the pipe, as well as being consistent between the replicate pipes. The dominating ceiling communities were members of the bacterial orders Rhodospirillales, Acidithiobacillales, Actinomycetales, Xanthomonadales and Acidobacteriales. The wall communities were composed of members of the Xanthomonadales, Hydrogenophilales, Chromatiales and Sphingobacteriales. The tidal zones were dominated by eight bacterial and one archaeal order, with the common physiological trait of anaerobic metabolism. Sewage flooding within the sewer system did not change the tidal and wall communities, although the corrosion communities in ceiling samples were notably different, becoming more similar to the wall and tidal samples. This suggests that sewage flooding has a significant impact on the corrosion community in sewers

    Omega-3 polyunsaturated fatty acids status and cognitive function in young women

    © 2019 The Author(s). Background: Research indicates that low omega-3 polyunsaturated fatty acid (n-3 PUFA) may be associated with decreased cognitive function. This study examined the association between n-3 PUFA status and cognitive function in young Australian women. Methods: This was a secondary outcome analysis of a cross-sectional study that recruited 300 healthy women (18-35 y) of normal weight (NW: BMI 18.5-24.9 kg/m2) or obese weight (OB: BMI ≥30.0 kg/m2). Participants completed a computer-based cognition testing battery (IntegNeuro™) evaluating the domains of impulsivity, attention, information processing, memory and executive function. The Omega-3 Index (O3I) was used to determine n-3 PUFA status (percentage of EPA (20:5n-3) plus DHA (22:6n3) in the red cell membrane) and the participants were divided into O3I tertile groups: T1 6.75%. Potential confounding factors of BMI, inflammatory status (C-reactive Protein), physical activity (total MET-min/wk), alpha1-acid glycoprotein, serum ferritin and hemoglobin, were assessed. Data reported as z-scores (mean ± SD), analyses via ANOVA and ANCOVA. Results: Two hundred ninety-nine women (26.9 ± 5.4 y) completed the study (O3I data, n = 288). The ANOVA showed no overall group differences but a significant group × cognition domain interaction (p < 0.01). Post hoc tests showed that participants in the low O3I tertile group scored significantly lower on attention than the middle group (p = 0.01; ES = 0.45 [0.15-0.74]), while the difference with the high group was borderline significant (p = 0.052; ES = 0.38 [0.09-0.68]). After confounder adjustments, the low group had lower attention scores than both the middle (p = 0.01) and high (p = 0.048) groups. These findings were supported by univariate analyses which found significant group differences for the attention domain only (p = 0.004). Conclusions: Cognitive function in the attention domain was lower in women with lower O3I, but still within normal range. This reduced but normal level of cognition potentially provides a lower baseline from which cognition would decline with age. Further investigation of individuals with low n-3 PUFA status is warranted

    Observation of An Evolving Magnetic Flux Rope Prior To and During A Solar Eruption

    Explosive energy release is a common phenomenon occurring in magnetized plasma systems ranging from laboratories, Earth's magnetosphere, the solar corona and astrophysical environments. Its physical explanation is usually attributed to magnetic reconnection in a thin current sheet. Here we report the important role of magnetic flux rope structure, a volumetric current channel, in producing explosive events. The flux rope is observed as a hot channel prior to and during a solar eruption from the Atmospheric Imaging Assembly (AIA) telescope on board the Solar Dynamic Observatory (SDO). It initially appears as a twisted and writhed sigmoidal structure with a temperature as high as 10 MK and then transforms toward a semi-circular shape during a slow rise phase, which is followed by fast acceleration and onset of a flare. The observations suggest that the instability of the magnetic flux rope trigger the eruption, thus making a major addition to the traditional magnetic-reconnection paradigm.Comment: 13 pages, 3 figure

    Noncontacting laser Ultrasonic Generation and Detection at the Surface of Molten Metal

    The use of pulsed lasers for noncontacting generation of ultrasound in solid materials is expanding rapidly [1], as is optical detection of ultrasound [2]. The noncontacting nature of laser ultrasonics is opening new areas of research where physical contact of transducers to the material under study is impossible or inadvisable. One example is in the titanium melting industry. Currently, vacuum arc remelting (VAR) is used to produce much of the nation’s titanium from Kroll process sponge. However, the process provides only limited means of removing oxynitride and carbide inclusions from the melt, which can become stress intensifiers in the ingot. VAR of titanium can be replaced with plasma or electron beam hearth melting, both of which have the potential to eliminate these stress-intensifying inclusions by increasing the residence time of the molten titanium in the hearth so that the oxynitrides dissolve and the carbides settle out of the melt. This process is so important that industry is starting to replace VAR with hearth melting for titanium to be used in critical applications such as rotating turbine parts. The new process has other advantages as well. Processing steps will be eliminated because sponge will no longer need to be consolidated into electrodes and fewer melting steps will be required. The improved quality of the melted product will result in less scrap, and the ability to recycle scrap into high value products will also be a major improvement. The most important aspect, though, is the capability to produce superior ingots with the potential of allowing turbine engines to be lighter and more efficient. However, industry has identified a critical requirement for these hearth melting processes: measurement of the volume of molten metal to ensure sufficient residence time in the melt. Ultrasonic sensing is one possible way for locating the interface between molten and solid metal so that the depth of the molten metal, the volume, and thus the residence time may be determined. Because the titanium hearth operates at high temperatures (1650°C), contacting transducers with buffer rods are not practical; it is also a potential source of melt contamination. Therefore, a totally noncontacting sensor system is needed. This sensing technology would also be widely applicable to other metals, including other reactive and refractory metals, superalloys, and steel.</p

    Integrating personality research and animal contest theory: aggressiveness in the green swordtail <i>Xiphophorus helleri</i>

    &lt;p&gt;Aggression occurs when individuals compete over limiting resources. While theoretical studies have long placed a strong emphasis on context-specificity of aggression, there is increasing recognition that consistent behavioural differences exist among individuals, and that aggressiveness may be an important component of individual personality. Though empirical studies tend to focus on one aspect or the other, we suggest there is merit in modelling both within-and among-individual variation in agonistic behaviour simultaneously. Here, we demonstrate how this can be achieved using multivariate linear mixed effect models. Using data from repeated mirror trials and dyadic interactions of male green swordtails, &lt;i&gt;Xiphophorus helleri&lt;/i&gt;, we show repeatable components of (co)variation in a suite of agonistic behaviour that is broadly consistent with a major axis of variation in aggressiveness. We also show that observed focal behaviour is dependent on opponent effects, which can themselves be repeatable but were more generally found to be context specific. In particular, our models show that within-individual variation in agonistic behaviour is explained, at least in part, by the relative size of a live opponent as predicted by contest theory. Finally, we suggest several additional applications of the multivariate models demonstrated here. These include testing the recently queried functional equivalence of alternative experimental approaches, (e. g., mirror trials, dyadic interaction tests) for assaying individual aggressiveness.&lt;/p&gt
