Thermodynamic analysis of inverted bifurcation
We present a thermodynamic analysis of inverted bifurcation
in binary mixtures heated from below. From this analysis it
follows that an inverted bifurcation is caused by the competition
between a stabilizing effect with a long relaxation time and a
destabilizing effect with a short relaxation time. These
conditions are precisely the same as those that give rise to
overstability. This might explain why overstability and inverted
bifurcation occur in the same systems
Applied Interventions in the Prevention and Treatment of Obesity Through the Research of Professor Jane Wardle
Purpose of Review
Obesity presents a challenge for practitioners, policy makers, researchers and for those with obesity themselves. This review focuses on psychological approaches to its management and prevention in children and adults.
Recent Findings
Through exploring the work of the late Professor Jane Wardle, we look at the earliest behavioural treatment approaches and how psychological theory has been used to develop more contemporary approaches, for example incorporating genetic feedback and habit formation theory into interventions. We also explore how Jane has challenged thinking about the causal pathways of obesity in relation to eating behaviour. Beyond academic work, Jane was an advocate of developing interventions which had real-world applications.

Competitive and Cooperative Interactions Mediate RNA Transfer from Herpesvirus Saimiri ORF57 to the Mammalian Export Adaptor ALYREF
The essential herpesvirus adaptor protein HVS ORF57, which has homologs in all other herpesviruses, promotes viral mRNA
export by utilizing the cellular mRNA export machinery. ORF57 protein specifically recognizes viral mRNA transcripts, and
binds to proteins of the cellular transcription-export (TREX) complex, in particular ALYREF. This interaction introduces viral
mRNA to the NXF1 pathway, subsequently directing it to the nuclear pore for export to the cytoplasm. Here we have used a
range of techniques to reveal the sites for direct contact between RNA and ORF57 in the absence and presence of ALYREF. A
binding site within ORF57 was characterized which recognizes specific viral mRNA motifs. When ALYREF is present, part of
this ORF57 RNA binding site, composed of an a-helix, binds preferentially to ALYREF. This competitively displaces viral RNA
from the a-helix, but contact with RNA is still maintained by a flanking region. At the same time, the flexible N-terminal
domain of ALYREF comes into contact with the viral RNA, which becomes engaged in an extensive network of synergistic
interactions with both ALYREF and ORF57. Transfer of RNA to ALYREF in the ternary complex, and involvement of individual
ORF57 residues in RNA recognition, were confirmed by UV cross-linking and mutagenesis. The atomic-resolution structure
of the ORF57-ALYREF interface was determined, which noticeably differed from the homologous ICP27-ALYREF structure.
Together, the data provides the first site-specific description of how viral mRNA is locked by a herpes viral adaptor protein in
complex with cellular ALYREF, giving herpesvirus access to the cellular mRNA export machinery. The NMR strategy used
may be more generally applicable to the study of fuzzy protein-protein-RNA complexes which involve flexible polypeptide
regions
Pigmentation plasticity enhances crypsis in larval newts: Associated metabolic cost and background choice behaviour
In heterogeneous environments, the capacity for colour change can be a valuable adaptation enhancing crypsis against predators. Alternatively, organisms might achieve concealment by evolving preferences for backgrounds that match their visual traits, thus avoiding the costs of plasticity. Here we examined the degree of plasticity in pigmentation of newt larvae (Lissotriton boscai) in relation to predation risk. Furthermore, we tested for associated metabolic costs and pigmentation-dependent background choice behaviour. Newt larvae expressed substantial changes in pigmentation so that light, high-reflecting environment induced depigmentation whereas dark, low-reflecting environment induced pigmentation in just three days of exposure. Induced pigmentation was completely reversible upon switching microhabitats. Predator cues, however, did not enhance cryptic phenotypes, suggesting that environmental albedo induces changes in pigmentation improving concealment regardless of the perceived predation risk. Metabolic rate was higher in heavily pigmented individuals from dark environments, indicating a high energetic requirement of pigmentation that could impose a constraint to larval camouflage in dim habitats. Finally, we found partial evidence for larvae selecting backgrounds matching their induced phenotypes. Physical comorbidities in men with mood and anxiety disorders: a population-based study
Background : The mind-body nexus has been a topic of growing interest. Further data are however required to understand the specific relationship between mood and anxiety disorders and individual physical health conditions, and to verify whether these psychiatric disorders are linked to overall medical burden. Methods : This study examined data collected from 942 men, 20 to 97 years old, participating in the Geelong Osteoporosis Study. A lifetime history of mood and anxiety disorders was identified using the Structured Clinical Interview for DSM-IV-TR Research Version, Non-patient edition (SCID-I/NP). The presence of medical conditions (lifetime) was self-reported and confirmed by medical records, medication use or clinical data. Anthropometric measurements and socioeconomic status (SES) were determined and information on medication use and lifestyle was obtained via questionnaire. Logistic regression models were used to test the associations. Results : After adjustment for age, socioeconomic status, and health risk factors (body mass index, physical activity and smoking), mood disorders were associated with gastro oesophageal reflux disease (GORD), recurrent headaches, blackouts and/or epilepsy, liver disorders and pulmonary disease in older people, whilst anxiety disorders were significantly associated with thyroid, GORD and other gastrointestinal disorders, and psoriasis. Increased odds of high medical burden were associated with both mood and anxiety disorders. Conclusions : Our study provides further population-based evidence supporting the link between mental and physical illness in men. Understanding these associations is not only necessary for individual management, but also to inform the delivery of health promotion messages and health care
Assessing the cost of global biodiversity and conservation knowledge
Knowledge products comprise assessments of authoritative information supported by
standards, governance, quality control, data, tools, and capacity building mechanisms.
Considerable resources are dedicated to developing and maintaining knowledge
products for biodiversity conservation, and they are widely used to inform policy and
advise decision makers and practitioners. However, the financial cost of delivering this
information is largely undocumented. We evaluated the costs and funding sources for
developing and maintaining four global biodiversity and conservation knowledge
products: The IUCN Red List of Threatened Species, the IUCN Red List of
Ecosystems, Protected Planet, and the World Database of Key Biodiversity Areas.
These are secondary data sets, built on primary data collected by extensive networks
of expert contributors worldwide. We estimate that US116-204
million), plus 293 person-years of volunteer time (range: 278-308 person-years) valued
at US12-16 million), were invested in these four knowledge
products between 1979 and 2013. More than half of this financing was provided
through philanthropy, and nearly three-quarters was spent on personnel costs. The
estimated annual cost of maintaining data and platforms for three of these knowledge
products (excluding the IUCN Red List of Ecosystems for which annual costs were not
possible to estimate for 2013 ) is US6.2-6.7 million).
We estimated that an additional US12 million. These costs
are much lower than those to maintain many other, similarly important, global
knowledge products. Ensuring that biodiversity and conservation knowledge products
are sufficiently up to date, comprehensive and accurate is fundamental to inform
decision-making for biodiversity conservation and sustainable development. Thus, the
development and implementation of plans for sustainable long-term financing for them
is critical
Microarray evidence of glutaminyl cyclase gene expression in melanoma: implications for tumor antigen specific immunotherapy
BACKGROUND: In recent years encouraging progress has been made in developing vaccine treatments for cancer, particularly with melanoma. However, the overall rate of clinically significant results has remained low. The present research used microarray datasets from previous investigations to examine gene expression patterns in cancer cell lines with the goal of better understanding the tumor microenvironment. METHODS: Principal Components Analyses with Promax rotational transformations were carried out with 90 cancer cell lines from 3 microarray datasets, which had been made available on the internet as supplementary information from prior publications. RESULTS: In each of the analyses a well defined melanoma component was identified that contained a gene coding for the enzyme, glutaminyl cyclase, which was as highly expressed as genes from a variety of well established biomarkers for melanoma, such as MAGE-3 and MART-1, which have frequently been used in clinical trials of melanoma vaccines. CONCLUSION: Since glutaminyl cyclase converts glutamine and glutamic acid into a pyroglutamic form, it may interfere with the tumor destructive process of vaccines using peptides having glutamine or glutamic acid at their N-terminals. Finding ways of inhibiting the activity of glutaminyl cyclase in the tumor microenvironment may help to increase the effectiveness of some melanoma vaccines

Targeting uracil-DNA glycosylases for therapeutic outcomes using insights from virus evolution
Ung-type uracil-DNA glycosylases are frontline defenders of DNA sequence fidelity in bacteria, plants, and animals; Ungs also directly assist both innate and humoral immunity. Critically important in viral pathogenesis, whether acting for or against viral DNA persistence, Ungs also have therapeutic relevance to cancer, microbial, and parasitic diseases. Ung catalytic specificity is uniquely conserved, yet selective antiviral drugging of the Ung catalytic pocket is tractable. However, more promising precision therapy approaches present themselves via insights from viral strategies, including sequestration or adaptation of Ung for non-canonical roles. A universal Ung inhibition mechanism, converged upon by unrelated viruses, could also inform design of compounds to inhibit specific distinct Ungs. Extrapolating current developments, the character of such novel chemical entities is proposed
