    Variational Trajectory Optimization Tool Set: Technical description and user's manual

    The algorithms that comprise the Variational Trajectory Optimization Tool Set (VTOTS) package are briefly described. The VTOTS is a software package for solving nonlinear constrained optimal control problems from a wide range of engineering and scientific disciplines. The VTOTS package was specifically designed to minimize the amount of user programming; in fact, for problems that may be expressed in terms of analytical functions, the user needs only to define the problem in terms of symbolic variables. This version of the VTOTS does not support tabular data; thus, problems must be expressed in terms of analytical functions. The VTOTS package consists of two methods for solving nonlinear optimal control problems: a time-domain finite-element algorithm and a multiple shooting algorithm. These two algorithms, under the VTOTS package, may be run independently or jointly. The finite-element algorithm generates approximate solutions, whereas the shooting algorithm provides a more accurate solution to the optimization problem. A user's manual, some examples with results, and a brief description of the individual subroutines are included

    Heterocyst placement strategies to maximize growth of cyanobacterial filaments

    Under conditions of limited fixed-nitrogen, some filamentous cyanobacteria develop a regular pattern of heterocyst cells that fix nitrogen for the remaining vegetative cells. We examine three different heterocyst placement strategies by quantitatively modelling filament growth while varying both external fixed-nitrogen and leakage from the filament. We find that there is an optimum heterocyst frequency which maximizes the growth rate of the filament; the optimum frequency decreases as the external fixed-nitrogen concentration increases but increases as the leakage increases. In the presence of leakage, filaments implementing a local heterocyst placement strategy grow significantly faster than filaments implementing random heterocyst placement strategies. With no extracellular fixed-nitrogen, consistent with recent experimental studies of Anabaena sp. PCC 7120, the modelled heterocyst spacing distribution using our local heterocyst placement strategy is qualitatively similar to experimentally observed patterns.     The Clustering of Massive Halos

    The clustering properties of dark matter halos are a firm prediction of modern theories of structure formation. We use two large volume, high-resolution N-body simulations to study how the correlation function of massive dark matter halos depends upon their mass and formation history. We find that halos with the lowest concentrations are presently more clustered than those of higher concentration, the size of the effect increasing with halo mass; this agrees with trends found in studies of lower mass halos. The clustering dependence on other characterizations of the full mass accretion history appears weaker than the effect with concentration. Using the integrated correlation function, marked correlation functions, and a power-law fit to the correlation function, we find evidence that halos which have recently undergone a major merger or a large mass gain have slightly enhanced clustering relative to a randomly chosen population with the same mass distribution.Comment: 10 pages, 8 figures; text improved, references and one figure added; accepted for publication in Ap

    Nitrogen and phosphorus loads to temperate seepage lakes associated with allochthonous dissolved organic carbon loads

    Terrestrial loads of dissolved organic matter (DOM) have increased in recent years in many north temperate lakes. While much of the focus on the “browning” phenomena has been on its consequences for carbon cycling, much less is known about how it influences nutrient loading to lakes. We characterize potential loads of nitrogen and phosphorus to seepage lakes in northern Wisconsin, USA, based on a laboratory soil leaching experiment and a model that includes landscape cover and watershed area. In these seepage lakes, nutrient concentrations are positively correlated with dissolved organic carbon concentrations (nitrogen: r = 0.68, phosphorus: r = 0.54). Using long‐term records of browning, we found that dissolved organic matter‐associated nutrient loadings may have resulted in substantial increases in nitrogen and phosphorus in seepage lakes and could account for currently observed nutrient concentrations in the lake. “Silent” nutrient loadings to brown‐water lakes may lead to future water‐quality concerns. PLAIN LANGUAGE SUMMARY: The color of many temperate lakes is changing; some lakes are becoming more darkly stained brown. The tea‐colored stain is due to dissolved organic matter from the surrounding landscape. Much of the research related to the causes and consequences of increased staining, or “brownification,” relate to its connection to the carbon cycle. However, by examining long‐term lake chemical records, analyzing the properties of the organic compounds, and modeling potential flows of the compounds, we find that carbon is not the only element that is influenced by browning.     Molecular basis for passive immunotherapy of Alzheimer's disease

    Amyloid aggregates of the amyloid-{beta} (A{beta}) peptide are implicated in the pathology of Alzheimer's disease. Anti-A{beta} monoclonal antibodies (mAbs) have been shown to reduce amyloid plaques in vitro and in animal studies. Consequently, passive immunization is being considered for treating Alzheimer's, and anti-A{beta} mAbs are now in phase II trials. We report the isolation of two mAbs (PFA1 and PFA2) that recognize A{beta} monomers, protofibrils, and fibrils and the structures of their antigen binding fragments (Fabs) in complex with the A{beta}(1–8) peptide DAEFRHDS. The immunodominant EFRHD sequence forms salt bridges, hydrogen bonds, and hydrophobic contacts, including interactions with a striking WWDDD motif of the antigen binding fragments. We also show that a similar sequence (AKFRHD) derived from the human protein GRIP1 is able to cross-react with both PFA1 and PFA2 and, when cocrystallized with PFA1, binds in an identical conformation to A{beta}(1–8). Because such cross-reactivity has implications for potential side effects of immunotherapy, our structures provide a template for designing derivative mAbs that target A{beta} with improved specificity and higher affinity

    SMASH 1 : A VERY FAINT GLOBULAR CLUSTER DISRUPTING in the OUTER REACHES of the LMC?

    We present the discovery of a very faint stellar system, SMASH 1, that is potentially a satellite of the Large Magellanic Cloud. Found within the Survey of the Magellanic Stellar History (SMASH), SMASH 1 is a compact (rh=9.1-3.4+5.9pc) and very low luminosity (Mv=-1.0±0.9,Lv=102.3±0.4L⊙ ) stellar system that is revealed by its sparsely populated main sequence and a handful of red giant branch candidate member stars. The photometric properties of these stars are compatible with a metal-poor ([Fe/H]=-2.2) and old (13 Gyr) isochrone located at a distance modulus of ∌18.8, i.e., a distance of . Situated at 11.°3 from the LMC in projection, its three-dimensional distance from the Cloud is 13 kpc, consistent with a connection to the LMC, whose tidal radius is at least . Although the nature of SMASH 1 remains uncertain, its compactness favors it being a stellar cluster and hence dark-matter free. If this is the case, its dynamical tidal radius is only â‰Č19 pc at this distance from the LMC, and smaller than the system's extent on the sky. Its low luminosity and apparent high ellipticity (Ï”=0.62-0.21+0.17) with its major axis pointing toward the LMC may well be the tell-tale sign of its imminent tidal demise.Peer reviewe

    Measurement invariance of three narcissism questionnaires across the United States, the United Kingdom, and Germany

    With a recent surge of research on narcissism, narcissism questionnaires are increasingly being translated and applied in various countries. The measurement invariance of an instrument across countries is a precondition for being able to compare scores across countries. We investigated the cross-cultural measurement invariance of three narcissism questionnaires (Brief Pathological Narcissism Inventory [B-PNI], Narcissistic Personality Inventory [NPI], and Narcissistic Admiration and Rivalry Questionnaire [NARQ]) and mean-level differences across samples from the United States (N = 2,464), the United Kingdom (N = 307), and Germany (N = 925). Overall, the B-PNI and NARQ functioned equivalently for the U.S. and U.K. participants. More violations of measurement invariance were found between Germany and the combined U.S. and U.K. samples, and for the NPI. In the B-PNI and NARQ, Americans scored higher than individuals from the United Kingdom regarding agentic aspects (self-sacrificing self-enhancement, admiration), while Germans scored lower than both Americans and U.K. individuals regarding antagonistic (entitlement rage, rivalry) and neurotic (hiding the self, contingent self-esteem) aspects. More inconsistent results were found for NPI facets. When noninvariance was present, observed means yielded biased results. Thus, the degree of measurement invariance across translated instrument versions should be considered in cross-cultural comparisons, even with culturally similar countries

    Sedimentation in an artificial lake -Lake Matahina, Bay of Plenty

    Lake Matahina, an 8 km long hydroelectric storage reservoir, is a small (2.5 km2), 50 m deep, warm monomictic, gorge-type lake whose internal circulation is controlled by the inflowing Rangitaiki River which drains a greywacke and acid volcanic catchment. Three major proximal to distal subenvironments are defined for the lake on the basis of surficial sediment character and dominant depositional process: (a) fluvial-glassy, quartzofeld-spathic, and lithic gravel-sand mixtures deposited from contact and saltation loads in less than 3 m depth; (b) (pro-)deltaic-quartzofeldspathic and glassy sand-silt mixtures deposited from graded and uniform suspension loads in 3-20 m depth; and (c) basinal-diatomaceous, argillaceous, and glassy silt-clay mixtures deposited from uniform and pelagic suspension loads in 20-50 m depth. The delta face has been prograding into the lake at a rate of 35-40 m/year and vertical accretion rates in pro-delta areas are 15-20 cm/year. Basinal deposits are fed mainly from river plume dispersion involving overflows, interflows, and underflows, and by pelagic settling, and sedimentation rates behind the dam have averaged about 2 cm/year. Occasional fine sand layers in muds of basinal cores attest to density currents or underflows generated during river flooding flowing the length of the lake along a sublacustrine channel marking the position of the now submerged channel of the Rangitaiki River
