Renal lithiasis detection in uro-computed tomography using a non-parametric technique
Renal lithiasis is the pathology that causes nephritic colic, which is one of the most frequent reasons for consultation in emergency medical services. According to the size, location, hardness and number of stones present in the urinary system, usually in the human kidney, it is established to which form of treatment is suitable for the patient. These kidney stones can be analyzed by means of biopsy or imaging modalities such as computed tomography images. This type of images has challenging problems called noise, artifacts and low contrast. In this paper, in order to address these problems, a non-parametric semi-automatic computational technique is developed for detecting kidney stones, present in computed tomography images, using digital image processing techniques based on a smoothing filter and an edge detector. Finally, the size and position of the stones present in the images are calculated and a precision metric is considered to compare the manual segmentation, performed by an urologist, and the one generated by the NPCT, obtaining an excellent correlation. This technique can be useful in the renal lithiasis detection and if it is considering this kind of computational strategy, medical specialists can establish the clinic or surgical actions oriented to address this pathology
Update on and future perspectives for the diagnosis of alpha-1 antitrypsin deficiency in Brazil
Tècniques de genotipat; Deficiència d'alfa-1 antitripsinaTécnicas de genotipado; Deficiencia de alfa-1 antitripsinaGenotyping techniques; alpha-1 antitrypsin deficiencyAlpha-1 antitrypsin deficiency (AATD) is a rare genetic disorder caused by a mutation in the SERPINA1 gene, which encodes the protease inhibitor alpha-1 antitrypsin (AAT). Severe AATD predisposes individuals to COPD and liver disease. Early diagnosis is essential for implementing preventive measures and limiting the disease burden. Although national and international guidelines for the diagnosis and management of AATD have been available for 20 years, more than 85% of cases go undiagnosed and therefore untreated. In Brazil, reasons for the underdiagnosis of AATD include a lack of awareness of the condition among physicians, a racially diverse population, serum AAT levels being assessed in a limited number of individuals, and lack of convenient diagnostic tools. The diagnosis of AATD is based on laboratory test results. The standard diagnostic approach involves the assessment of serum AAT levels, followed by phenotyping, genotyping, gene sequencing, or combinations of those, to detect the specific mutation. Over the past 10 years, new techniques have been developed, offering a rapid, minimally invasive, reliable alternative to traditional testing methods. One such test available in Brazil is the A1AT Genotyping Test, which simultaneously analyzes the 14 most prevalent AATD mutations, using DNA extracted from a buccal swab or dried blood spot. Such advances may contribute to overcoming the problem of underdiagnosis in Brazil and elsewhere, as well as being likely to increase the rate detection of AATD and therefore mitigate the harmful effects of delayed diagnosis.This study received financial support from Grifols S.A., Barcelona, Spain

de imagenología médica como apoyo clínico tanto en
la generación de diagnósticos de un buen número de enfermedades
como en la planeación de tratamientos tendientes a
proporcionar una alternativa de solución a las personas que,
por una u otra razón, manifiestan algún desequilibrio en su
salud. Entre las modalidades imagenológicas que, frecuentemente,
se utilizan en el ámbito médico se pueden mencionar:
Ultrasonido (US), Resonancia Magnética (MRI), Tomografía
Computarizada sencilla (CT) y multicapa (MSCT), Tomografía
Computarizada por Emisión de Positrones (PET) y Tomografía
Computarizada por Emisión de Fotones simples (SPECT).
En este sentido, este artículo tiene como finalidad presentar
una descripción ordenada, coherente y sistemática de cada
una de las mencionadas modalidades y establecer la vinculación
de la MSCT con situaciones clínicas íntimamente relacionadas
con la anatomía cardiaca y procesos de hipertensión.
La razón por la cual se hace énfasis en la MSCT es debido a
que, por una parte, se cuenta con un número importante de
bases de datos tanto de sujetos fisiológicos como de sujetos
patológicos y, por la otra, que se tiene previsto presentar un
conjunto de técnicas computacionales que serán exploradas
en el desarrollo de futuras investigaciones en el contexto de
aspectos anatómico-cardiológicos que influyen directa o indirectamente
en la aparición, desarrollo y prevalencia de procesos
hipertensivos.Nowdays, several forms of medical imaging are usefulness in
clinical support for both diagnostics diseases generation and
treatment planning designed to provide an alternative solution
to people who, for one or another reason, manifest an
imbalance in your health. Among the imaging modalities that
frequently are used in the medical field may include: Ultrasound
(US), magnetic resonance imaging (MRI), simple computed
tomography (CT) and multilayer computed tomography
(MSCT), CT positron emission tomography (PET) Computed
tomography and single photon emission (SPECT). In this sense,
this article presents an orderly, coherent and systematic
description of these modalities and it establishes relationship
with the MSCT modality and clinical situations closely related
to cardiac anatomy and hypertension processes. The reason
emphasis on MSCT is done is because, on the one hand, we
have a large number of databases both physiological subjects
and pathological subjects and, on the other, we plan to
present a set of computational techniques that will be explored
in future research, in the context of cardiologic anatomical
aspects, with directly or indirectly influence in the emergence,

Therapeutic Effect of a Novel Oxazolidinone, DA-7867, in BALB/c Mice Infected with Nocardia brasiliensis
Actinomycetoma is an infectious disease of tropical and subtropical regions produced by actinobacteria of the genera Nocardia, Streptomyces, and Actinomadura. Therapeutic alternatives are scarce and include trimethoprim-sulfamethoxazole, diaminodiphenylsulfone, amoxicillin-clavulanate, imipenem, and amikacin. Oxazolidinones are a new class of antimicrobials with a completely different cellular target; the first compound in the market, linezolid, was introduced in the year 2000. It is active against many species of Nocardia and other aerobic actinomycetes; however, the long-term application in human subjects produces side effects including peripheral neuropathy and mielossupression. Therefore, it is important to screen other oxazolidinones with higher activity and less toxicity. In the present work, we tested DA-7867, a new oxazolidinone, in an experimental mouse model. The drug is active in vivo and decreases the production of lesions using only one dose a day in contrast to linezolid, which needs to be injected three times a day. Although it was tested on N. brasiliensis, it can possibly be active (once it is accepted for its use in humans) against Actinomadura spp and Streptomyces spp, which are frequently found in places of Africa and India where actinomycetoma is also an important consult in dermatology
A nature-inspired design yields a new class of steroids against trypanosomatids
This article belongs to the Special Issue Drug Discovery for Neglected DiseasesChagas disease and Leishmaniasis are neglected endemic protozoan diseases recognized as public health problems by the World Health Organization. These diseases affect millions of people around the world however, efficient and low-cost treatments are not available. Different steroid molecules with antimicrobial and antiparasitic activity were isolated from diverse organisms (ticks, plants, fungi). These molecules have complex structures that make de novo synthesis extremely difficult. In this work, we designed new and simpler compounds with antiparasitic potential inspired in natural steroids and synthesized a series of nineteen steroidal arylideneketones and thiazolidenehydrazines. We explored their biological activity against Leishmania infantum, Leishmania amazonensis, and Trypanosoma cruzi in vitro and in vivo. We also assayed their genotoxicity and acute toxicity in vitro and in mice. The best compound, a steroidal thiosemicarbazone compound 8 (ID_1260) was active in vitro (IC50 200 nM) and in vivo (60% infection reduction at 50 mg/kg) in Leishmania and T. cruzi. It also has low toxicity in vitro and in vivo (LD50 >2000 mg/kg) and no genotoxic effects, being a promising compound for anti-trypanosomatid drug development
Predictors and moderators of outcome of psychotherapeutic interventions for mental disorders in adolescents and young adults : protocol for systematic reviews
Background: Adolescence and young adulthood is a risk period for the emergence of mental disorders. There is strong evidence that psychotherapeutic interventions are effective for most mental disorders. However, very little is known about which of the different psychotherapeutic treatment modalities are effective for whom. This large systematic review aims to address this critical gap within the literature on non-specific predictors and moderators of the outcomes of psychotherapeutic interventions among adolescents and young adults with mental disorders. Methods: The protocol is being reported in accordance with the Preferred Reporting Items for Systematic Review and Meta-Analysis Protocols (PRISMA-P) Statement. PubMed and PsycINFO databases will be searched for randomized controlled and quasi-experimental/naturalistic clinical trials. Risk of bias of all included studies will be assessed by the Mixed Methods Appraisal Tool. The quality of predictor and moderator variables will be also assessed. A narrative synthesis will be conducted for all included studies. Discussion: This systematic review will strengthen the evidence base on effective mental health interventions for young people, being the first to explore predictors and moderators of outcome of psychotherapeutic interventions for a wide range of mental disorders in young people.Peer reviewe
