    Sun safety in construction: a UK intervention study

    Background: Interventions to promote sun safety in the UK construction sector are warranted given the high incidence of skin cancer attributable to sun exposure relative to other occupational groups. Aims: To evaluate change in sun safety knowledge and practices among construction workers in response to an educational intervention. Methods: A baseline questionnaire was administered, followed by a bespoke sector-specific DVD-based intervention. At 12-month follow-up participants completed a further questionnaire. Results: Analyses were conducted on a sample of 120 workers (intervention group, n = 70; comparison group, n = 50). At follow-up the proportion of intervention group participants that reported correct sun safety knowledge was not significantly greater than at baseline. However, the intervention group demonstrated significant positive change on nine out of ten behavioural measures, the greatest change being use of a shade/cover when working in the sun followed by regularly checking skin for moles or unusual changes. Conclusions: Exposure to this intervention was linked to some specific positive changes in construction workers’ self-reported sun safety practices. These findings highlight the potential for educational interventions to contribute to tackling skin cancer in the UK construction sector. The findings support the development of bespoke educational interventions for other high-risk outdoor worker groups

    Aneuploidy in oocytes is prevented by sustained CDK1 activity through degron masking in cyclin B1

    Successful mitosis requires that cyclin B1:CDK1 kinase activity remains high until chromosomes are correctly aligned on the mitotic spindle. It has therefore been unclear why, in mammalian oocyte meiosis, cyclin B1 destruction begins before chromosome alignment is complete. Here, we resolve this paradox and show that mouse oocytes exploit an imbalance in the ratio of cyclin B1 to CDK1 to control CDK1 activity; early cyclin B1 destruction reflects the loss of an excess of non-CDK1-bound cyclin B1 in late prometaphase, while CDK1-bound cyclin B1 is destroyed only during metaphase. The ordered destruction of the two forms of cyclin B1 is brought about by a previously unidentified motif that is accessible in free cyclin B1 but masked when cyclin B1 is in complex with CDK1. This protects the CDK1-bound fraction from destruction in prometaphase, ensuring a period of prolonged CDK1 activity sufficient to achieve optimal chromosome alignment and prevent aneuploidy

    Alasdair Whittle has had a career-long interest in the Neolithic of the Avebury area (Fig. 17.1). In the late 1980s and early 1990s he undertook a major research project in the region to investigate the Neolithic sequence and its environment (Whittle 1993). This included a series of excavations of early Neolithic sites including the causewayed enclosure at Windmill Hill (Whittle et al. 1999), the chambered tomb at Millbarrow (Whittle 1994), and an earthen long barrow at Easton Down (Whittle et al. 1993). A series of trenches were also cut through two palisade enclosures at West Kennet (Whittle 1997). This campaign of new excavation was accompanied by research into the archives of previous investigations, particularly the publication and subsequent dating of Richard Atkinson’s excavation on and within Silbury Hill in 1968–70 (Bayliss et al. 2007a; Whittle 1997) and a reassessment of the date and development of Avebury itself (Pitts and Whittle 1992). The subsequent decades have seen continued work in the Avebury region, given focus by the Archaeological Research Agenda for the Avebury World Heritage site (AAHRG 2001). Alasdair himself has been instrumental in producing refined chronologies for the West Kennet long barrow (Bayliss et al. 2007b) and the causewayed enclosures at Windmill Hill and Knap Hill (Whittle et al. 2011, chapter 3), and in producing synthetic narratives of early Neolithic sites in the region and beyond (Whittle et al. 2007; 2011, chapters 14 and 15). Further understanding of the late Neolithic landscape has been gained through research excavations at the Beckhampton Avenue and Longstones Cove and ditched enclosure (Gillings et al. 2008), and through rescue excavations undertaken in advance of consolidation works at Silbury Hill (Leary et al. 2013a). The latter in particular has done much to improve our understanding of the development and date of Silbury Hill (Marshall et al. 2013; in prep. a; in prep. b), which has been put into context by a recent synthesis of the available scientific dating evidence for the Avebury area by Frances Healy (2016)

    A veritable confusion: use and abuse of isotope analysis in archaeology

    The expansion of isotope analyses has transformed the study of past migration and mobility, sometimes providing unexpected and intriguing results. This has, in turn, led to media attention (and concomitant misrepresentation) and scepticism from some archaeologists. Such scepticism is healthy and not always without foundation. Isotope analysis is yet to reach full maturity and challenging issues remain, concerning diagenesis, biosphere mapping resolution and knowledge of the drivers of variation. Bold and over-simplistic interpretations have been presented, especially when relying on single isotope proxies, and researchers have at times been accused of following specific agendas. It is therefore vital to integrate archaeological and environmental evidence to support interpretation. Most importantly, the use of multiple isotope proxies is key: isotope analysis is an exclusive approach and therefore single analyses provide only limited resolution. The growth in isotope research has led to a growth in rebuttals and counter-narratives. Such rebuttals warrant the same critical appraisal that is applied to original research, both of evidence for their assertions and the potential for underlying agendas.     The dietary impact of the Norman Conquest: A multiproxy archaeological investigation of Oxford, UK

    Archaeology has yet to capitalise on the opportunities offered by bioarchaeological approaches to examine the impact of the 11th-century AD Norman Conquest of England. This study utilises an integrated multiproxy analytical approach to identify and explain changes and continuities in diet and foodways between the 10th and 13th centuries in the city of Oxford, UK. The integration of organic residue analysis of ceramics, carbon (δ13C) and nitrogen (δ15N) isotope analysis of human and animal bones, incremental analysis of δ13C and δ15N from human tooth dentine and palaeopathological analysis of human skeletal remains has revealed a broad pattern of increasing intensification and marketisation across various areas of economic practice, with a much lesser and more short-term impact of the Conquest on everyday lifestyles than is suggested by documentary sources. Nonetheless, isotope data indicate short-term periods of instability, particularly food insecurity, did impact individuals. Evidence of preferences for certain foodstuffs and cooking techniques documented among the elite classes were also observed among lower-status townspeople, suggesting that Anglo-Norman fashions could be adopted across the social spectrum. This study demonstrates the potential for future archaeological research to generate more nuanced understanding of the cultural impact of the Norman Conquest of England, while showcasing a method which can be used to elucidate the undocumented, everyday implications of other large-scale political events on non-elites

    2MASS Galaxies in the Fornax Cluster Spectroscopic Survey

    The Fornax Cluster Spectroscopic Survey (FCSS) is an all-object survey of a region around the Fornax Cluster of galaxies undertaken using the 2dF multi-object spectrograph on the Anglo-Australian Telescope. Its aim was to obtain spectra for a complete sample of all objects with 16.5 < b_j < 19.7 irrespective of their morphology (i.e. including `stars', `galaxies' and `merged' images). We explore the extent to which (nearby) cluster galaxies are present in 2MASS. We consider the reasons for the omission of 2MASS galaxies from the FCSS and vice versa. We consider the intersection (2.9 square degrees on the sky) of our data set with the infra-red 2 Micron All-Sky Survey (2MASS), using both the 2MASS Extended Source Catalogue (XSC) and the Point Source Catalogue (PSC). We match all the XSC objects to FCSS counterparts by position and also extract a sample of galaxies, selected by their FCSS redshifts, from the PSC. We confirm that all 114 XSC objects in the overlap sample are galaxies, on the basis of their FCSS velocities. A total of 23 Fornax Cluster galaxies appear in the matched data, while, as expected, the remainder of the sample lie at redshifts out to z = 0.2 (the spectra show that 61% are early type galaxies, 18% are intermediate types and 21% are strongly star forming).The PSC sample turns out to contain twice as many galaxies as does the XSC. However, only one of these 225 galaxies is a (dwarf) cluster member. On the other hand, galaxies which are unresolved in the 2MASS data (though almost all are resolved in the optical) amount to 71% of the non-cluster galaxies with 2MASS detections and have redshifts out to z=0.32.Comment: 5 pages, accepted by A&A, resubmitted due to missing reference

    Extracellular vesicles produced by the human commensal gut bacterium Bacteroides thetaiotaomicron affect host immune pathways in a cell-type specific manner that are altered in inflammatory bowel disease

    The gastrointestinal (GI) tract harbours a complex microbial community, which contributes to its homeostasis. A disrupted microbiome can cause GI-related diseases, including inflammatory bowel disease (IBD), therefore identifying host-microbe interactions is crucial for better understanding gut health. Bacterial extracellular vesicles (BEVs), released into the gut lumen, can cross the mucus layer and access underlying immune cells. To study BEV-host interactions, we examined the influence of BEVs generated by the gut commensal bacterium, Bacteroides thetaiotaomicron, on host immune cells. Single-cell RNA sequencing data and host-microbe protein-protein interaction networks were used to predict the effect of BEVs on dendritic cells, macrophages and monocytes focusing on the Toll-like receptor (TLR) pathway. We identified biological processes affected in each immune cell type and cell-type specific processes including myeloid cell differentiation. TLR pathway analysis highlighted that BEV targets differ among cells and between the same cells in healthy versus disease (ulcerative colitis) conditions. The in silico findings were validated in BEV-monocyte co-cultures demonstrating the requirement for TLR4 and Toll-interleukin-1 receptor domain-containing adaptor protein (TIRAP) in BEV-elicited NF-kB activation. This study demonstrates that both cell-type and health status influence BEV-host communication. The results and the pipeline could facilitate BEV-based therapies for the treatment of IBD
