    Advancing Stepped Wedge Cluster Randomized Trials Analysis: Bayesian Hierarchical Penalized Spline Models for Immediate and Time-Varying Intervention Effects

    Stepped wedge cluster randomized trials (SWCRTs) often face challenges with potential confounding by time trends. Traditional frequentist methods can fail to provide adequate coverage of the intervention's true effect using confidence intervals, whereas Bayesian approaches show potential for better coverage of intervention effects. However, Bayesian methods have seen limited development in SWCRTs. We propose two novel Bayesian hierarchical penalized spline models for SWCRTs. The first model is for SWCRTs involving many clusters and time periods, focusing on immediate intervention effects. To evaluate its efficacy, we compared this model to traditional frequentist methods. We further developed the model to estimate time-varying intervention effects. We conducted a comparative analysis of this Bayesian spline model against an existing Bayesian monotone effect curve model. The proposed models are applied in the Primary Palliative Care for Emergency Medicine stepped wedge trial to evaluate the effectiveness of primary palliative care intervention. Extensive simulations and a real-world application demonstrate the strengths of the proposed Bayesian models. The Bayesian immediate effect model consistently achieves near the frequentist nominal coverage probability for true intervention effect, providing more reliable interval estimations than traditional frequentist models, while maintaining high estimation accuracy. The proposed Bayesian time-varying effect model exhibits advancements over the existing Bayesian monotone effect curve model in terms of improved accuracy and reliability. To the best of our knowledge, this is the first development of Bayesian hierarchical spline modeling for SWCRTs. The proposed models offer an accurate and robust analysis of intervention effects. Their application could lead to effective adjustments in intervention strategies

    The glacial geomorphology of upper Godthåbsfjord (Nuup Kangerlua) in south-west Greenland

Information on outlet glacier dynamics, prior to this, are limited due to both a lack of observations and a paucity of mapped or mappable deglacial evidence which restricts our understanding of centennial to millennial timescale dynamics of the GrIS. Here we present glacial geomorphological mapping, for upper Godthåbsfjord, covering 5800 km 2 at a scale of 1:92,000, using a combination of ASTER GDEM V2, a medium-resolution DEM (error < 10 m horizontal and < 6 m vertical accuracy), panchromatic orthophotographs and ground truthing. This work provides a detailed geomorphological assessment for the area, compiled as a single map, comprising of moraines, meltwater channels, streamlined bedrock, sediment lineations, ice-dammed lakes, trimlines, terraces, gullied sediment and marine limits. Whilst some of the landforms have been previously identified, the new information presented here improves our understanding of ice margin behaviour and can be used for future numerical modelling and landform dating programmes. Data also form the basis for palaeoglaciological reconstructions and contribute towards understanding of the centennial to millennial timescale record of this sector of the GrIS.Peer reviewedFinal Published versio

    With the increasing enrollment of international students in North American universities, the need to support the adjustment of international students has also increased. One factor consistently identified as essential to successful student adjustment is having a strong support network, including familial support. Previous research investigating the needs of international students’ partners is limited and has suggested that partners may face greater barriers to adjustment than international students. The current study was conducted to better understand the needs of partners as well as potential barriers they face when attempting to access services. Although limited by sample size, the results suggested that partners are interested in increased support in helping their transition to local living and mainstream culture

    Oxidative stress and kidney dysfunction due to ischemia/reperfusion in rat: Attenuation by dehydroepiandrosterone.BackgroundThe pathogenesis of ischemia/reperfusion (I/R) involves generation of reactive oxygen and nitrogen species. This in vivo study investigates the effect of dehydroepiandrosterone (DHEA), a physiologic steroid with antioxidant properties, on oxidative balance and renal dysfunctions induced by monolateral I/R.MethodsNormal and DHEA-treated rats (4mg/day × 21 days, orally) were subjected to monolateral renal I/R (30 minutes/6 hours). The oxidative state was determined by measuring hydrogen peroxide level and activities of glutathione-peroxidase, catalase, and superoxide dismutase. Tumor necrosis factor-α (TNF-α) and nitric oxide production and inducible nitric oxide synthase (iNOS) levels were also measured. Hydroxynonenal content was used to probe lipid peroxidation. Functional parameters determined were creatinine levels and Na/K-ATPase activity. Immunohistochemical and morphologic studies were also performed.ResultsA markedly pro-oxidant state was evident in the kidney of rats subjected to I/R. Both hydrogen peroxide and reactive nitrogen species (nitric oxide and iNOS) increased, whereas antioxidants decreased. Oxidant species induce TNF-α increase, which, in turn, produces lipoperoxidative processes, as documented by the increased hydroxynonenal (HNE) level. As final result, impaired renal functionality, hydropic degeneration, and vacuolization of proximal convolute tubules were observed in kidneys of I/R rats. DHEA pretreatment improved the parameters considered.ConclusionI/R induces oxidative stress and consequently damages the proximal convolute renal tubules. Rats supplemented with DHEA and subjected to I/R had reduced pro-oxidant state, oxidative damage, and improved renal functionality, indicating an attenuation of oxidative injury and dysfunctions mediated by I/R

    La pseudomonas syringae pv. actinidiae es un patógeno responsable significativo de la afta bacteriana severa del kiwi (Actinidia sp.). Los bacteriófagos infectados de este fitopatógeno tienen potencial como agentes de control biológico como parte de un enfoque integrado de la gestión del cancro bacteriano, y para su uso como herramientas molecular para el estudio de esta bacteria. Una variedad de bacteriófagos fueron previamente aislados, antes de ser infectados con P. syringae pv. Actinidiae; y sus propiedades básicas fueron caracterizadas para proporcionar un marco para la formulación de estos fagos, como agentes de biocontrol. Aquí, hemos examinado con más detalle el φPsa17, un fago con la capacidad de infectar a una amplia gama de cepas P. syringae pv. Actinidiae, único miembro de la Podoviridae en esta colección. La morfología de partículas fue visualizada mediante criomicroscopía electrónica, el genoma fue secuenciado, y sus proteínas estructurales fueron analizados usando shotgun proteomics. Bacteriophages infecting this phytopathogen have potential as biocontrol agents as part of an integrated approach to the management of bacterial canker, and for use as molecular tools to study this bacterium. A variety of bacteriophages were previously isolated that infect P. syringae pv. actinidiae, and their basic properties were characterized to provide a framework for formulation of these phages as biocontrol agents. Here, we have examined in more detail φPsa17, a phage with the capacity to infect a broad range of P. syringae pv. actinidiae strains and the only member of the Podoviridae in this collection. Particle morphology was visualized using cryo-electron microscopy, the genome was sequenced, and its structural proteins were analysed using shotgun proteomics. These studies demonstrated that φPsa17 has a 40,525 bp genome, is a member of the T7likevirus genus and is closely related to the pseudomonad phages φPSA2 and gh-1. Eleven structural proteins (one scaffolding) were detected by proteomics and φPsa17 has a capsid of approximately 60 nm in diameter. No genes indicative of a lysogenic lifecycle were identified, suggesting the phage is obligately lytic.     Ultralow-light-level color image reconstruction using high-efficiency plasmonic metasurface mosaic filters

    As single-photon imaging becomes progressively more commonplace in sensing applications such as low-light-level imaging, three-dimensional profiling, and fluorescence imaging, there exist a number of fields where multispectral information can also be exploited, e.g., in environmental monitoring and target identification. We have fabricated a high-transmittance mosaic filter array, where each optical filter was composed of a plasmonic metasurface fabricated in a single lithographic step. This plasmonic metasurface design utilized an array of elliptical and circular nanoholes, which produced enhanced optical coupling between multiple plasmonic interactions. The resulting metasurfaces produced narrow bandpass filters for blue, green, and red light with peak transmission efficiencies of 79%, 75%, and 68%, respectively. After the three metasurface filter designs were arranged in a 64×64 format random mosaic pattern, this mosaic filter was directly integrated onto a CMOS single-photon avalanche diode detector array. Color images were then reconstructed at light levels as low as approximately 5 photons per pixel, on average, via the simultaneous acquisition of low-photon multispectral data using both three-color active laser illumination and a broadband white-light illumination source
