    Comparative transcriptomic analysis of plum fruit treated with 1-MCP

    Microarray technology has allowed the large scale transcriptomic analysis of fruit ripening. The μPEACH1.0 microarray containing 4,806 probes corresponding to genes expressed in peach fruit tissues has been used in a heterologous fashion in two studies of plums ripening behavior. Gene expression of different cultivars of plums treated with the ethylene antagonist, 1-methylcyclopropene (1-MCP) and stored for short periods at room temperature or for longer periods of cold storage was examined. In the first study, mature fruit of a suppressed ethylene climacteric cultivar 'Shiro' and a cultivar characterized by a typical increase of ethylene production during ripening ('Santa Rosa') were harvested and incubated for 24h in air (control) or 1-MCP and allowed to ripen at room temperature. Different levels of transcripts of genes implicated in cell wall metabolism, hormone (ethylene and auxin) regulation, stress and defense, and in the transcription/translation machinery, as well as others involved with ripening were identified. In the second study, the effects of 1-MCP on gene expression in relation to the development of chilling injury (CI) in the climacteric cultivars 'Ruby Red' (RR) and 'October Sun' (OS) and 'Zee Lady' peaches (ZP) were analyzed. The fruit were treated for 24h at room temperature with 1-MCP prior to storage at 0°C. For RR, there was no significant effect of 1-MCP on the level of CI symptoms, while 1-MCP significantly reduced CI symptoms in OS fruit and an increase of CI in treated ZP fruit. Microarray analysis showed that immediately following treatment, 186, 134 and 56 genes were differentially expressed between the control and 1-MCP-treated fruit of these cultivars, respectively: after 4 weeks cold storage, 311, 52 and 224 genes for RR, OS and ZP, respectively, were differentially expressed between control and treated fruit. Thus, for OS, the number of differentially expressed genes reduced during storage while the number increased in RR and ZP. Comparisons of the data suggest that the transcript profile is altered by 1-MCP more in plums than peaches.     Biosynthesis and Cellular Functions of Tartaric Acid in Grapevines

    Tartaric acid (TA) is an obscure end point to the catabolism of ascorbic acid (Asc). Here, it is proposed as a “specialized primary metabolite”, originating from carbohydrate metabolism but with restricted distribution within the plant kingdom and lack of known function in primary metabolic pathways. Grapes fall into the list of high TA-accumulators, with biosynthesis occurring in both leaf and berry. Very little is known of the TA biosynthetic pathway enzymes in any plant species, although recently some progress has been made in this space. New technologies in grapevine research such as the development of global co-expression network analysis tools and genome-wide association studies, should enable more rapid progress. There is also a lack of information regarding roles for this organic acid in plant metabolism. Therefore this review aims to briefly summarize current knowledge about the key intermediates and enzymes of TA biosynthesis in grapes and the regulation of its precursor, ascorbate, followed by speculative discussion around the potential roles of TA based on current knowledge of Asc metabolism, TA biosynthetic enzymes and other aspects of fruit metabolism

    Ethylene-induced differential gene expression during abscission of citrus leaves

    The main objective of this work was to identify and classify genes involved in the process of leaf abscission in Clementina de Nules (Citrus clementina Hort. Ex Tan.). A 7 K unigene citrus cDNA microarray containing 12 K spots was used to characterize the transcriptome of the ethylene-induced abscission process in laminar abscission zone-enriched tissues and the petiole of debladed leaf explants. In these conditions, ethylene induced 100% leaf explant abscission in 72 h while, in air-treated samples, the abscission period started later and took 240 h. Gene expression monitored during the first 36 h of ethylene treatment showed that out of the 12 672 cDNA microarray probes, ethylene differentially induced 725 probes distributed as follows: 216 (29.8%) probes in the laminar abscission zone and 509 (70.2%) in the petiole. Functional MIPS classification and manual annotation of differentially expressed genes highlighted key processes regulating the activation and progress of the cell separation that brings about abscission. These included cell-wall modification, lipid transport, protein biosynthesis and degradation, and differential activation of signal transduction and transcription control pathways. Expression data associated with the petiole indicated the occurrence of a double defensive strategy mediated by the activation of a biochemical programme including scavenging ROS, defence and PR genes, and a physical response mostly based on lignin biosynthesis and deposition. This work identifies new genes probably involved in the onset and development of the leaf abscission process and suggests a different but co-ordinated and complementary role for the laminar abscission zone and the petiole during the process of abscission

Il titolo del volume \ue8 spia indicati-va per descrivere l\u2019atteggiamento ermeneutico proprio di De Simone e di chi si dedichi allo studio delle lingue antiche, soprattutto di quel-le \uabrelitto\ubb: una fortunata con-giunzione di metodi (strutturali-smo, funzionalismo, sociolingui-stica) inquadrata in uno storici-smo ed in una attenzione al dato archeologico concreto influenzati anche dalla scuola di Massimo Pallottino ed aggiornati dalle pi\uf9 recenti tendenze della linguistica moderna. A questa impostazione, che ha fatto scuola, si devono i meriti di Carlo De Simone e le fortunate proposte d\u2019interpretazione testuale che caratterizzano tutta la sua produzione scientifica

    This chapter deals with the difficult process of constitutionalisation which characterised Italian Unification. Constitutionalisation is a long-term phenomenon which had the purpose of giving constitutional forms to the Nation. The promulgation of the Albertine Statute is more the start than the arrival of this phenomenon. The focus of this investigation is, therefore, to study the Constitution through its evolution paying particular attention to the process of legal integration within the structures of the Albertine Statute and to the amendment mechanisms of the constitutional text. The preamble of the Albertine Statute speaks of «perpetual and irrevocable fundamental law». The word «perpetual» meant the prohibition of revoking constitutional concession, while the word «irrevocable» was intended as a pact between the Sovereign and the Nation. Over the years, very few were the changes to the letter of the Albertine Statute. The interpretation and the practice represented the most important mechanisms of constitutional change (implicit constitutional changes). A primary role was acknowledged to non-written norms. In this perspective, it may well be said that the Italian Constitution consisted in something more than the written text and dwelt in the spirit and not in the letter of the Albertine Statute
