Apakah merek dan promosisecara simultan berpengaruh terhadap keputusan pembelian produk abon ikan (MANISE).Populasi dalam penelitian ini adalah keseluruahan dari konsumen yang telah melakukan pembelianabon ikan “MANISE” pada UKM Setia Kawan Kota Kupang. Sedangkan responden berjumlah 196orang. Metode pengumpulan data menggunakan kuesioner. Data hasil penelitian dianalisis deganmenggunkan program SPSS versi 20 for windows.Hasil penelitian menunjukan 1). Secara parsial merek berpengaruh signifikan terhadap keputusanpembelian abon ikan “MANISE” pada UKM perikanan Setia Kawan Kota Kupang. 2) Secara parsialparsial promosi mempunyai pengaruh positif terhadap keputusan pembelian produk abon ikan“MANISE” pada UKM Perikanan Setia Kawan di Kota Kupang. 3) Secara simultan, variabel merekdan promosi memberikan pengaruh secara signifikan terhadap keputusan pembelian produk abon ikan“MANISE” pada UKM Perikanan Setia Kawan di Kota Kupang.Saran yang diajukan adalah untuk variabel merek dan promosi menjadi pusat perhatian bagi pihakpengurus UKM Perikanan Setia Kawan di Kota Kupang untuk menarik konsumen melakukankeputusan pembelian dengan cara meningkatkan asosiasi merek dan mempromosi jalur distribusidegan baik

    Analysis of Outpatient Satisfaction and the Quality of Health Services

    Quality is an important factor in health services. Bakunase Health Center is one of the primary accredited health centers in Kupang City. Based on data from the Health Department, there has been a decrease in the number of visits over the last three years. This happens due to external factors and internal factors. The external factor is the increasing degree of public health, while the internal factor is the decrease in the patients’ commitment to treatment. The purpose of this study is to describe the level of outpatient satisfaction with the quality of health services at Bakunase Health Center of Kupang City in 2020. This was a descriptive survey with a quantitative approach. The population in this study was all patients who visited and used outpatient services at Bakunase Health Center in Kupang City from January to October 2019. Accidental sampling was used in selecting the samples from each outpatient sub-group in 7 polyclinics at Bakunase Health Center using inclusion and exclusion criteria. The results indicated that the level of conformity between expectations and reality of the services are as follows:  98.78% tangible, 98.66% reliable, 98.33% responsive, 98.79% assurance 98.74% empathy. Based on the Cartesian Importance Performance Analysis (IPA) diagram, quadrant A was filled by one attribute from the responsiveness dimension, quadrant B was filled by most attributes from the reliability dimension, quadrant C was filled by most attributes from the empathy dimension and quadrant D was filled by one attribute from the responsiveness dimension

    MicroRNA regulation of type 2 innate lymphoid cell homeostasis and function in allergic inflammation.

    MicroRNAs (miRNAs) exert powerful effects on immunity through coordinate regulation of multiple target genes in a wide variety of cells. Type 2 innate lymphoid cells (ILC2s) are tissue sentinel mediators of allergic inflammation. We established the physiological requirements for miRNAs in ILC2 homeostasis and immune function and compared the global miRNA repertoire of resting and activated ILC2s and T helper type 2 (TH2) cells. After exposure to the natural allergen papain, mice selectively lacking the miR-17∼92 cluster in ILC2s displayed reduced lung inflammation. Moreover, miR-17∼92-deficient ILC2s exhibited defective growth and cytokine expression in response to IL-33 and thymic stromal lymphopoietin in vitro. The miR-17∼92 cluster member miR-19a promoted IL-13 and IL-5 production and inhibited expression of several targets, including SOCS1 and A20, signaling inhibitors that limit IL-13 and IL-5 production. These findings establish miRNAs as important regulators of ILC2 biology, reveal overlapping but nonidentical miRNA-regulated gene expression networks in ILC2s and TH2 cells, and reinforce the therapeutic potential of targeting miR-19 to alleviate pathogenic allergic responses

    ABSTRAK Steganografi adalah seni dan ilmu menulis pesan tersembunyi atau menyembunyikan pesan dengan suatu cara sehingga selain si pengirim dan si penerima, tidak ada seorangpun yang mengetahui atau menyadari bahwa ada suatu pesan rahasia. Metode yang digunakan untuk menyembunyikan pesan pada media digital tersebut berbedaâ€beda. Contohnya, pada berkas image pesan dapat disembunyikan dengan menggunakan cara menyisipkannya pada bit rendah atau bit yang paling kanan Least Significant Bit (LSB). Penelitian ini adalah untuk dapat memberikan keamanan pada pesan rahasia untuk menyembunyikan pesan tersebut pada gambar menggunakan Metode Least Significant Bit (LSB) dengan tools yang digunakan yaitu matlab.     Kelelahan kerja sering dikeluhkan sebagai salah satu permasalahan ditempat kerja, kelelahan sering atau dapat dirasakan oleh setiap manusia pada saat melakukan aktivitas, baik aktivitas ringan maupun aktivitas berat. Kelelahan kerja disebabkan oleh beberapa faktor yaitu faktor usia, jenis kelamin, penyakit dan beban kerja. Tujuan Penelitian ini adalah untuk mengetahui hubungan beban kerja fisik dengan kelelahan kerja pada pekerja bagian operator boiler dan turbin di PJBS Pembangkit Listrik Tenaga Uap Amurang.Metode penelitian menggunakan survei analitik dengan pendekatan cross sectional (potong lintang). Populasi dalam penelitian adalah semua pekerja operator boiler dan turbin yang berjumlah 41 responden dan seluruh populasi dijadikan subjek dalam penelitian ini. Instrumen yang digunakan dalam penelitian ini yaitu reaction timer dan Penilaian beban kerja berdasarkan, SNI 7269:200S. Analisis data menggunakan uji korelasi pearson. Penelitian ini menunjukkan terdapat hubungan antara beban kerja fisik dengan kelelahan kerja mendapatakan hasil p-value=0,000 (<0,05) dan nilai r= 0,578 atau korelasi sedang dengan arah hubungan yang positif. Kesimpulan dalam penelitian ini terdapat hubungan antara beban kerja fisik dengan kelelahan kerja pada pekerja Operator Boiler dan Turbin di PJBS Pembangkit Listrik Tenaga Uap Amurang. Kata Kunci: Beban Kerja, Kelelahan Kerja ABSTRACKWorking fatigue is often complained of as one of the problems in the workplace, frequent fatigue or can be felt by every human being during activity, both light activity and heavy activity. Working fatigue is caused by factors such as age, gender, illness and workload. The purpose of this research is to know the relationship of physical workload with working fatigue in the workers part of the boiler and turbine operators in PJBS Steam Power Plant Amurang. The research method uses an analytical survey with a cross sectional approach (cut latitude). The population in the study was all the workers of the boiler and turbine operators amounting to 41 respondents and the entire population was made a subject in this study. The instrument used in this research is the reaction timer and assessment of workload based, SNI 7269:200S. Data analysis using Pearson correlation test. This research shows that there is a relationship between physical workloads and work fatigue to be the result of P-value = 0,000 (< 0.05) and the value of R = 0.578 or medium correlation with the direction of a positive relationship. Conclusion in this study there is a link between physical workload with working fatigue in the workers of Boiler and turbine operators in PJBS Amurang Steam Power Plant. Keywoards : Workload, Working Fatigu

    Expression of Insoluble Influenza Neuraminidase Type 1 (NA1) Protein in Tobacco

    The avian influenza virus, particularly H5N1 strain, is highly virulent to poultry and mankind. Several expression systems, like yeast, baculovirus and mammalian cells, have been adopted to produce vaccine candidate for this lethal disease. The present research aimed at developing a recombinant vaccine candidate, neuraminidase type 1 (NA1), for the Malaysia isolate of H5N1 in Nicotiana benthamiana. The NA1 gene was fused directly in-frame in cowpea mosaic virus (CPMV)-based pEAQ-HT vector with C-terminal polyhistidine-tag incorporated to ease the subsequent purification step. The expression of the NA1 gene in tobacco was confirmed at RNA and protein levels at 6 days post-infiltration (Dpi). From the insoluble fraction of the protein, a recombinant glycosylated NA1 protein with a molecular weight of ~56 kDa was immunogenically detected by a specific anti-NA polyclonal antibody. We report for the first time the insolubility of the plant-made NA1 protein where a native sequence was used for its expression. This study signifies the necessity of the use of optimised sequences for expression work and provides great opportunity for the exploration of plant-manufactured NA1 protein as vaccine candidate

    Investigation of HIFU-induced anti-tumor immunity in a murine tumor model

    <p>Abstract</p> <p>Background</p> <p>High intensity focused ultrasound (HIFU) is an emerging non-invasive treatment modality for localized treatment of cancers. While current clinical strategies employ HIFU exclusively for thermal ablation of the target sites, biological responses associated with both thermal and mechanical damage from focused ultrasound have not been thoroughly investigated. In particular, endogenous danger signals from HIFU-damaged tumor cells may trigger the activation of dendritic cells. This response may play a critical role in a HIFU-elicited anti-tumor immune response which can be harnessed for more effective treatment.</p> <p>Methods</p> <p>Mice bearing MC-38 colon adenocarcinoma tumors were treated with thermal and mechanical HIFU exposure settings in order to independently observe HIFU-induced effects on the host's immunological response. <it>In vivo </it>dendritic cell activity was assessed along with the host's response to challenge tumor growth.</p> <p>Results</p> <p>Thermal and mechanical HIFU were found to increase CD11c+ cells 3.1-fold and 4-fold, respectively, as compared to 1.5-fold observed for DC injection alone. In addition, thermal and mechanical HIFU increased CFSE+ DC accumulation in draining lymph nodes 5-fold and 10-fold, respectively. Moreover, focused ultrasound treatments not only caused a reduction in the growth of primary tumors, with tumor volume decreasing by 85% for thermal HIFU and 43% for mechanical HIFU, but they also provided protection against subcutaneous tumor re-challenge. Further immunological assays confirmed an enhanced CTL activity and increased tumor-specific IFN-γ-secreting cells in the mice treated by focused ultrasound, with cytotoxicity induced by mechanical HIFU reaching as high as 27% at a 10:1 effector:target ratio.</p> <p>Conclusion</p> <p>These studies present initial encouraging results confirming that focused ultrasound treatment can elicit a systemic anti-tumor immune response, and they suggest that this immunity is closely related to dendritic cell activation. Because DC activation was more pronounced when tumor cells were mechanically lysed by focused ultrasound treatment, mechanical HIFU in particular may be employed as a potential strategy in combination with subsequent thermal ablations for increasing the efficacy of HIFU cancer treatment by enhancing the host's anti-tumor immunity.</p

    CD4 T lymphocyte autophagy is upregulated in the salivary glands of primary Sjögren’s syndrome patients and correlates with focus score and disease activity

    Background: Primary Sjögren’s syndrome (pSS) is a common chronic autoimmune disease characterized by lymphocytic infiltration of exocrine glands and peripheral lymphocyte perturbation. In the current study, we aimed to investigate the possible pathogenic implication of autophagy in T lymphocytes in patients with pSS. Methods: Thirty consecutive pSS patients were recruited together with 20 patients affected by sicca syndrome a nd/or chronic sialoadenitis and 30 healthy controls. Disease activity and damage were evaluated according to SS disease activity index, EULAR SS disease activity index, and SS disease damage index. T lymphocytes were analyzed for the expression of autophagy-specific markers by biochemical, molecular, and histological assays in peripheral blood and labial gland biopsies. Serum interleukin (IL)-23 and IL-21 levels were quantified by enzyme-linked immunosorbent assay. Results: Our study provides evidence for the first time that autophagy is upregulated in CD4+ T lymphocyte salivary glands from pSS patients. Furthermore, a statistically significant correlation was detected between lymphocyte autophagy levels, disease activity, and damage indexes. We also found a positive correlation between autophagy enhancement and the increased salivary gland expression of IL-21 and IL-23, providing a further link between innate and adaptive immune responses in pSS. Conclusions: These findings suggest that CD4+ T lymphocyte autophagy could play a key role in pSS pathogenesis. Additionally, our data highlight the potential exploitation of T cell autophagy as a biomarker of disease activity and provide new ground to verify the therapeutic implications of autophagy as an innovative drug target in pSS
