    Evidence of population-level impacts and resiliency for Gulf of Mexico shelf taxa following the Deepwater Horizon oil spill

    The goal of this paper was to review the evidence of population-level impacts of the Deepwater Horizon Oil Spill (DWH) on Gulf of Mexico (GOM) continental shelf taxa, as well as evidence of resiliency following the DWH. There is considerable environmental and biological evidence that GOM shelf taxa were exposed to and suffered direct and indirect impacts of the DWH. Numerous assessments, from mesocosm studies to analysis of biopsied tissue or tissue samples from necropsied animals, revealed a constellation of physiological effects related to DWH impacts on GOM biota, some of which clearly or likely resulted in mortality. While the estimated concentrations of hydrocarbons in shelf waters and sediments were orders of magnitude lower than measured in inshore or deep GOM environments, the level of mortality observed or predicted was substantial for many shelf taxa. In some cases, such as for zooplankton, community shifts following the spill were ephemeral, likely reflecting high rates of population turnover and productivity. In other taxa, such as GOM reef fishes, impacts of the spill are confounded with other stressors, such as fishing mortality or the appearance and rapid population growth of invasive lionfish (Pterois spp.). In yet others, such as cetaceans, modeling efforts to predict population-level effects of the DWH made conservative assumptions given the species’ protected status, which post-DWH population assessments either failed to detect or population increases were estimated. A persistent theme that emerged was the lack of precise population-level data or assessments prior to the DWH for many taxa, but even when data or assessments did exist, examining evidence of population resiliency was confounded by other stressors impacting GOM biota. Unless efforts are made to increase the resolution of the data or precision of population assessments, difficulties will likely remain in estimating the scale of population-level effects or resiliency in the case of future large-scale environmental catastrophes

    Analysis of expressed sequence tags generated from full-length enriched cDNA libraries of melon

    Abstract Background Melon (Cucumis melo), an economically important vegetable crop, belongs to the Cucurbitaceae family which includes several other important crops such as watermelon, cucumber, and pumpkin. It has served as a model system for sex determination and vascular biology studies. However, genomic resources currently available for melon are limited. Result We constructed eleven full-length enriched and four standard cDNA libraries from fruits, flowers, leaves, roots, cotyledons, and calluses of four different melon genotypes, and generated 71,577 and 22,179 ESTs from full-length enriched and standard cDNA libraries, respectively. These ESTs, together with ~35,000 ESTs available in public domains, were assembled into 24,444 unigenes, which were extensively annotated by comparing their sequences to different protein and functional domain databases, assigning them Gene Ontology (GO) terms, and mapping them onto metabolic pathways. Comparative analysis of melon unigenes and other plant genomes revealed that 75% to 85% of melon unigenes had homologs in other dicot plants, while approximately 70% had homologs in monocot plants. The analysis also identified 6,972 gene families that were conserved across dicot and monocot plants, and 181, 1,192, and 220 gene families specific to fleshy fruit-bearing plants, the Cucurbitaceae family, and melon, respectively. Digital expression analysis identified a total of 175 tissue-specific genes, which provides a valuable gene sequence resource for future genomics and functional studies. Furthermore, we identified 4,068 simple sequence repeats (SSRs) and 3,073 single nucleotide polymorphisms (SNPs) in the melon EST collection. Finally, we obtained a total of 1,382 melon full-length transcripts through the analysis of full-length enriched cDNA clones that were sequenced from both ends. Analysis of these full-length transcripts indicated that sizes of melon 5' and 3' UTRs were similar to those of tomato, but longer than many other dicot plants. Codon usages of melon full-length transcripts were largely similar to those of Arabidopsis coding sequences. Conclusion The collection of melon ESTs generated from full-length enriched and standard cDNA libraries is expected to play significant roles in annotating the melon genome.     Safety and Usage of C1-Inhibitor in Hereditary Angioedema: Berinert Registry Data

    BackgroundThe plasma-derived, highly purified, nanofiltered C1-inhibitor concentrate (Berinert; “pnfC1-INH”) is approved in the United States for treating hereditary angioedema (HAE) attacks and in many European countries for attack treatment and short-term prophylaxis.ObjectiveThe objective of this study was to describe safety and usage patterns of pnfC1-INH.MethodsA multicenter, observational, registry was conducted between 2010 and 2014 at 30 United States and 7 European sites to obtain both prospective (occurring after enrollment) and retrospective (occurring before enrollment) safety and usage data on subjects receiving pnfC1-INH for any reason.ResultsOf 343 enrolled patients, 318 received 1 or more doses of pnfC1-INH for HAE attacks (11,848 infusions) or for prophylaxis (3142 infusions), comprising the safety population. Median dosages per infusion were 10.8 IU/kg (attack treatment) and 16.6 IU/kg (prophylaxis). Approximately 95% of infusions were administered outside of a health care setting. No adverse events (AEs) were reported in retrospective data. Among prospective data (n = 296 subjects; 9148 infusions), 252 AEs were reported in 85 (28.7%) subjects (rate of 0.03 events/infusion); 9 events were considered related to pnfC1-INH. Two thromboembolic events were reported in subjects with thrombotic risk factors. No patient was noted to have undergone viral testing for suspected blood-borne infection during registry participation.ConclusionsThe findings from this large, international patient registry documented widespread implementation of pnfC1-INH self-administration outside of a health care setting consistent with current HAE guidelines. These real-world data revealed pnfC1-INH usage for a variety of reasons in patients with HAE and showed a high level of safety regardless of administration setting or reason for use

    Retention Time Variability as a Mechanism for Animal Mediated Long-Distance Dispersal

    Long-distance dispersal (LDD) events, although rare for most plant species, can strongly influence population and community dynamics. Animals function as a key biotic vector of seeds and thus, a mechanistic and quantitative understanding of how individual animal behaviors scale to dispersal patterns at different spatial scales is a question of critical importance from both basic and applied perspectives. Using a diffusion-theory based analytical approach for a wide range of animal movement and seed transportation patterns, we show that the scale (a measure of local dispersal) of the seed dispersal kernel increases with the organisms' rate of movement and mean seed retention time. We reveal that variations in seed retention time is a key determinant of various measures of LDD such as kurtosis (or shape) of the kernel, thinkness of tails and the absolute number of seeds falling beyond a threshold distance. Using empirical data sets of frugivores, we illustrate the importance of variability in retention times for predicting the key disperser species that influence LDD. Our study makes testable predictions linking animal movement behaviors and gut retention times to dispersal patterns and, more generally, highlights the potential importance of animal behavioral variability for the LDD of seeds

    The Canadian Healthy Infant Longitudinal Development (CHILD) birth cohort study: Assessment of environmental exposures

    The Canadian Healthy Infant Longitudinal Development birth cohort was designed to elucidate interactions between environment and genetics underlying development of asthma and allergy. Over 3600 pregnant mothers were recruited from the general population in four provinces with diverse environments. The child is followed to age 5 years, with prospective characterization of diverse exposures during this critical period. Key exposure domains include indoor and outdoor air pollutants, inhalation, ingestion and dermal uptake of chemicals, mold, dampness, biological allergens, pets and pests, housing structure, and living behavior, together with infections, nutrition, psychosocial environment, and medications. Assessments of early life exposures are focused on those linked to inflammatory responses driven by the acquired and innate immune systems. Mothers complete extensive environmental questionnaires including time-activity behavior at recruitment and when the child is 3, 6, 12, 24, 30, 36, 48, and 60 months old. House dust collected during a thorough home assessment at 3–4 months, and biological specimens obtained for multiple exposure-related measurements, are archived for analyses. Geo-locations of homes and daycares and land-use regression for estimating traffic-related air pollution complement time-activity-behavior data to provide comprehensive individual exposure profiles. Several analytical frameworks are proposed to address the many interacting exposure variables and potential issues of co-linearity in this complex data set

    Since Jerry Herron begins his forum essay, “Notes toward an Excellent Marxist-Elitist Honors Admissions Policy,” with his anecdotal True Genealogical Confessions, I feel obligated to begin in a similar mode. One side of my family was in the real estate business in St. Louis, and the other operated on the production side of industry—garment manufacturing, in the schmatta business so to speak. Like Herron, I have benefitted from a familial confluence of disparate skill sets in my position as Director of the Georgia Perimeter College Honors Program, which during the recruiting and registration season I would liken to that of the Buddhist monk selling water from a haphazardly constructed lemonadesque stand situated on the bank of a river. Of course, what unwary wayfaring students to GPC’s educational waters do not know is that my suitemate, who has for too many years endured overhearing my recruiting spiels, calls me a silver-tongued devil.     In past pages of JNCHC, I have argued the inappropriateness of believing that educational institutions should adhere to a business model or think of students as customers (“Business”). For supporting evidence, one need not peer far beyond the Savings & Loan Crisis, the housing bubble, the insider manipulations and profiteering on Wall Street, or the relationship between college tuition and the real costs of educating a student. I worked on Wall Street and at Rockefeller Center in the mid-1970s as a junior businessman. I was involved with the silver bullion trade when the Hunts manipulated the market, sending silver prices skyrocketing to unprecedented heights before the inevitable collapse. The playing field in the world of finance is not level: the brokers on Wall Street have access to information that the public does not
